This study was focused on finding out the potential problems associated with organic farming system. The effect of composted animal manures subsequent inoculation of microbes on growth and yield of tomatoes (Lycopersicon esculentum Mill. 'Minicarol') were examined to develop a proper organic farming practice. Plant heights were greater in composted manure treatment than in conventional practice, whereas widths of leaves were higher in conventional field. Chlorophyll contents between various amount of composted manure application were gradually decreased and showed no significant differences after 45 days of planting. The yield in the treatment applied 12 ton of composted animal manure per 10a as pre-planting fertilizer and following microbial inoculation were only 50-60% compared to that of conventional farming. However, yield increased up to 80% when additional composts were applied to the treatment received 6 ton of composted animal manure per 10a in the middle of cultivation. Microbial inoculation followed by composted manure application induced rapid decrease of nitrogen content in soil. However, the density of microorganisms was significantly increased. Tomatoes produced through organic farming were clear in color, Further, soluble solid and acid content were increased. The highest level of acid and solids were observed in the treatment applied 12 ton of digested swine manure per 10a. Although nitrogen content including ammonium and nitrate rapidly increased after application of composts, these were significantly reduced approximately 4-5 weeks after planting. The level of phosphorus, potassium, magnesium and calcium showed gradual decrease compared to nitrogen.

Purpose: The purpose of this study was to study the effects of the utilization of ethanol solution in infection control of dental implant hand drivers, a common practice in dental prosthodontic clinics. Materials and Methods: Infection control methods were divided into two groups. One swabbed with 83% ethanol gauze and the other immersed in 83% ethanol solution for 30, 60, 90, 120, 150, 180 and 300 second intervals after inoculation of the dental implant hand drivers with Staphylococcus aureus. After measuring the number of colony forming units and analyzing the optical density, the effects of infection control in the experimental group were compared with the positive control group without infection control after inoculation with bacteria and the negative control group without inoculation with bacteria after sterilization. Results: The number of colony forming units and optical density analysis showed a statistically significant difference compared to the positive control. On the other hand, there was no statistically significant difference between the negative control and the group immersed in the 83% ethanol solution for more than 150 seconds. Conclusion: It is recommended to use the ethanol solution as a pre-cleaning process before sterilization, since the intermediate-level disinfection method using ethanol solution alone for the infection control of the dental implant hand driver cannot clinically secure the sterility.
Objective: To investigate the therapeutic potential of human embryonic stem cells (hESCs) as a vaccine to induce an immune response and provide antitumor protection in a rat model. Methods: Cross-reactivity of antigens between hESCs and tumour cells was screened by immunohistochemistry. Fischer 344 rats were divided into 7 groups, with 6 rats in each, immunized with: Group 1, hESC; Group 2, pre-inactivated mitotic NuTu-19; Group 3 PBS; Group 4, hESC; Group 5, pre-inactivated mitotic NuTu-19; Group 6, PBS; Group 7, hESC only. At 1 (Groups 1-3) or 4 weeks (Groups 4-6) after the last vaccination, each rat was challenged intraperitoneally with NuTu-19. Tumor growth and animal survival were closely monitored. Rats immunized with H9 and NuTu-19 were tested by Western blot analysis of rat orbital venous blood for cytokines produced by Th1 and Th2 cells. Results: hESCs presented tumour antigens, markers, and genes related to tumour growth, metastasis, and signal pathway interactions. The vaccine administered to rats in Group 1 led to significant antitumor responses and enhanced tumor rejection in rats with intraperitoneal inoculation of NuTu-19 cells compared to control groups. In contrast, rats in Group 4 did not display any elevation of antitumour responses. Western blot analysis found cross-reactivity among antibodies generated between H9 and NuTu-19. However, the cytokines did not show significant differences, and no side effects were detected. Conclusion: hESC-based vaccination is a promising modality for immunotherapy of ovarian cancer.

In this study, we analyzed for the changes of low organic compounds during 4 weeks incubation though inoculation of harmful microorganisms on commercial feed. Two percent of overnight cultures of Exiguobacterium acetylicum, Acinetobacter calcoaceticus, Aspergillus flavus and Fusarium graminearum were inoculated in feed, respectively. After adjusting moisture level to 50% for the promotion of feed spoilage, pH was decreased to 4.58~5.03 and microorganism was ranged to $6{\sim}10log_{10}CFU/g$. The compounds were compare between aflatoxin G1 producing feeds and aflatoxin G1 non-producing feeds. Aflatoxins G1 were detected by the immunoaffinity column clean-up method with HPLC-FLD, and were confirmed in samples incoulated by Aspergillus flavus and Acinetobacter calcoaceticus. Koiganal II, cyclohexanol and butadien-one were detected from samples (the non-sterilized inoculated feed) by Aspergillus flavus and Acinetobacter calcoaceticus. Respectively as aflatoxin G1 pre-detected substance, Koiganal II, cyclohexanol and butadien-one may be useful substance for the pre-detection of aflatoxin G1.
Anthers of hybrids and inbred lines of Chinese cabbage (Brassica campestris ssp. pekinensis) were inoculated on the modified solid or liquid B$_{5}$ medium after several pre-treatments. Anthers were then maintained at 25 $^{\circ}C$ after being subjected to various post-treatment. Somatic embryos began to appear 9 days after inoculation and ended at 13th days. Low temperature pre-treatment did not increase embryoid production whereas high post-treatment temperature of 4$0^{\circ}C$ or 45$^{\circ}C$ enhance the production. There were considerable levels differences in embryogenesis between lines used, but not between the culture methods. Somatic embryo yield was also increased by subjecting anthers to one day at 35$^{\circ}C$ and then another one day at 3$0^{\circ}C$ after plating. Histological observations showed several stages in haploid development ranging from a few celled to large multiple-celled embryoids.s.
Chlorella is one of the microorganisms which can live autotrophically by their own photosynthesis. It was previously revealed that pre-treatment of Chlorella fusca caused a suppression of appressorium formation on the cucumber leaves after inoculation with Colletothrichum orbiculare. In this study, the ultrastructures of C. orbiculare on the cucumber leaves pretreated with C. fusca were observed using both scanning electron microscope (SEM) and transmission electron microscope (TEM). The SEM images revealed that most fungal conidia and hyphae were attached with lots of C. fusca cells. Also, the conidia could germinate but not form appressorium, which is necessary to penetrate into host tissue. These observations suggested that C. fusca adjoined to the fungus may play a role in suppression of the appressorium formation. On the other hand, the observations of TEM showed no remarkable cytological differences on the ultrastructures of the intracellular hyphae between in the pre-treated and untreated leaves. It seemed that the fungus could grow in the pre-treated plant tissues as in the untreated one. Based on these observations, it is suggested that the suppression of appressorium on the leaf surfaces by the C. fusca cells may be a main cause of the reduction of the anthracnose disease.

Pseudomonas syringae pv. tomato (Pst) causes a bacterial speck disease in tomato and Arabidopsis. In Chinese cabbage, in which host-pathogen interactions are not well understood, Pst does not cause disease but rather elicits a hypersensitive response. Pst induces localized cell death and $H_2O_2$ accumulation, a typical hypersensitive response, in infiltrated cabbage leaves. Pre-inoculation with Pst was found to induce resistance to Erwinia carotovora subsp. carotovora, a pathogen that causes soft rot disease in Chinese cabbage. An examination of the expression profiles of 12 previously identified Pst-inducible genes revealed that the majority of these genes were activated by salicylic acid or BTH; however, expressions of the genes encoding PR4 and a class IV chitinase were induced by ethephon, an ethylene-releasing compound, but not by salicylic acid, BTH, or methyl jasmonate. This implies that Pst activates both salicylate-dependent and salicylate-independent defense responses in Chinese cabbage.
An experiment was carried out to investigate the effects of pre-inoculation with the mycorrhizal fungus Glomus clarum and foliar application of kinetin on the growth of mungbean plant irrigated wht different dilution of seawater. Arbuscular-mycorrhizal(AM) infection significantly increased dry weight, height, chlorophyll, sugar and protein content, nitrogen and phosphorus-use efficiencies, leaf conductivity, transpiration rate, nitrogenase, acid and alkaline phosphates activities of all salinized mungbean plants in comparison with control and non-mycorrhizal plants irrespective of the presence or absence of kinetin. Mycorrrhizal plants showed higher concentrations of N, P, K, Ca and Mg and lower Na/N, Na/P, Na/K, Na/Ca and Na/Mg ratios than non-mycorrhizal plants when irrigated with certain dilution of seawater. Mungbean plants showed 597% and 770% dependency on AM fungus G. clarum in absence and presence of kinetin, respectively, for biomass production under a level of 30% of seawater. The average value of tolerance index for mycorrhizal plants accounted 267% and 364% in absence and presence kinetin respectively. This study provides evidence for the benefits of kinetin which are actually known for mycorrhizal than non-mycorrhzal plants. AM fungus and kinetin protected the host plants against the detrimental effects of salt. However, mycorrhizal infection was much more effective than kinetin applications. Thus management applications of this arbuscular mycorrhizal endophyte(G. clarum) with kinetin could be of importance in using seawater in certain dilution for irrigation in agriculture.
The objective of the study is to determine differences between cucurbits in the pathogenicity of Didymella bryoniae isolated from the naturally infected seeds of cucumber and pumpkin. Primary seedling infection of cucumber(Cucumis sativus L.), oriental melon(Cucumis melo var. makuwa Makino), pumpkin(Cucurbita pepo L.) and watermelon (Citrullus vulgaris Shrad.) occurred on the radicle, hypocotyl and cotyledons and symptoms on each crop were very similar. Infection of the radicle generally caused pre-emergence rot, while infection on the hypocotyl and cotyledons provided further inoculum for infection of the first true leaves and the stem. In cross inoculation tests, all isolates of D. bryoniae could infect cucumber, oriental melon, pumpkin and watermelon at different growth stages and there were not much differences in pathogenicity or susceptibility between isolates of the pathogen and crops tested. The susceptibility of cucumber and pumpkin was markedly influenced by prevailing humid conditions.
A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.
