A Study on the Protective Effects of Polygalae Radix on Neurotoxicity Induced by N-methyl-D-aspartic acid(NMDA) (원지(遠志)가 NMDA로 유발된 선경세포 손상에 미치는 효과)

  • Objectives : Polygalae Radix (PR) from Polygalae tenuifolia (Polygalaceae) has been clinically used as a sedative, anti-inflammatory, and anti-bacterial agent. To extend pharmacological effects of PR in the central nervous system (CNS) on the basis of its CNS protective effect, the present study was conducted to identify the effect of PR, whether it shows the neuroprotective action against excitatory neurotoxicity. Methods : To identify the protective effect of PR to excitatory neuro-toxic agent, the present study was focused on the PR effect on cell death, that was caused by applying NMDA to nerve cell, elevation of $(Ca^{2+})_i$, releasement of glutamate, and ROS generation. Result : 1. PR methanol extract, at the concentration range of 0.05 to 5 g/ml, significantly inhibited NMDA (1 mM)-induced neuronal cell death as well as MK-801 (non competitive NMDA antagonist). 2. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited NMDA (1 mM)-induced elevation of cytosolic calcium concentration $[Ca^{2+}]_i$. NMDA application in the presence of MK-801 $(10\;{\mu}M)$ failed to produce the increase of $[Ca^{2+}]_i$ through all the measurement time. 3. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of glutamate release. Also, MK-801 showed similar protective effects. 4. PR methanol extract $(0.5\;{\mu}g/ml)$ inhibited the NMDA-induced elevation of ROS generation. Also, MK-801 showed similar protective effects. Conclusion : The present study provides the availability of PR to exert its protective effect on the neuronal cell death in various neurodegenerative pathophysiological conditions.

Electrochemical Properties of Cu(I)hexafluoroacetylacetonate (Cu(I)hexafluoroacetylacetonate 착화합물들의 전기화학적 성질)

  • Complexes of $Cu(I)(hfac)PR_3$(P: phosphine and R: Me, Et and Bu) as Cu(I)(${\beta}$-diketonate) compounds have been synthesized and their electrochemical properties have been investigated using glassy carbon and carbon microelectrode in aprotic solvent. Reduction process of $Cu(I)(hfac)PR_3$ compounds carried out one electron pathway to Cu(0) by cyclic voltammetry in acetonitrile solution. Chronoamperometric curve using carbon microelectrode shows that these complexes are one electron process and diffusion coefficients are $4.5{\sim}6.7{\times}10^{-6}cm^2$/sec.

Effects of Polygonati Rhizoma Extracts on the Collagenase Activity and Procollagen Synthesis in Hs68 Human Fibroblasts and Tyrosinase Activity

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.

Application of a New Method to Reproduce the Enthalpies of Transfer of NaI from Water to Aqueous Methanol, Ethanol and iPrOH Solvent Systems at 298 K

  • The enthalpies of transfer, ${\Delta}H_t{\Theta}$, of NaI from water to aqueous methanol, ethanol and isopropanol, iPrOH, systems are reported. These data have been analysed in terms of the new solvation theory. These data are considered in terms of the new developed solvation theory including variable ($\alpha$n + $\beta$N), the net effect of the solute on the solvent-solvent bonding, is positive if there is a net breaking or weakening of solvent-solvent bonds. The solvation parameters recovered from the analyses indicate that the net affect of NaI on solvent structure is a breaking of solvent-solvent bonds and that NaI is preferentially solvated by water in all aqueous alcohol systems considered. ($\alpha$n + $\beta$N) values increase with increasing in the size of the alcohol alkyl residue from methanol to iPrOH.

Effects on the Initiation of Simian Virus 40 DNA Replication by Antisense RNA

  • When DNA replication of simian virus 40 (SV40) is initiated on the replication origin, the regions containing the initiation sites of DNA primase, which participates in the transient RNA primer synthesis for formation of Okazaki fragments in the lagging strand, were chosen as the target sites of antisense RNA for studies of the inhibition of SV40 DNA replication. Four recombinant transcription vectors, pUC-PrI, pUC-PrII, pGEM-PrBS, and pGEM-PrSN, coding antisense RNA, were constructed. Four antisense RNAs (named as I, II, BS, and SN) having the size of 18, 19,58, and 123 nts, respectively, were made from the transcription vectors by in vitro transcription. And then, antisense RNA in the concentration of 2${\mu}m$ were added to COS cells transfected with pATSV-W which is a recombinant plasmid containing the SV40 origin of replication. The inhibitory extent of DNA replication was measured by DpnI resistance and was confirmed by measurement of transient RNA primer synthesis. The result shows that six combinations of antisense RNA (I, II, BS, SN, I+SN, and BS+SN) lead to the inhibition of SV40 DNA replication by up to 85%.

Correlation Between Expression of ER, PR and C-erbB-2 Protein as Histologic Grade of Breast Cancer (유방암의 조직학적 악성도에 따른 에스트로겐 수용체(ER)와 프로게스테론 수용체(PR), 그리고 C-erbB-2 종양단백질 발현과의 상관관계)

  • Overexpressions of the estrogen receptors(ER), progesterone receptors(PR) and C-erbB-2 protein are important determiners of the response to chemotherapy in the breast cancer. For detecting ER, PR and C-erbB-2, immunohistochemistry are currently regarded as standard method. The purposes of this study compared to histologic grade and expression of the ER, PR and C-erbB-2 in breast cancer. We examined overexpression of ER, PR and C-erbB-2 protein in 84 breast carcinomas by using immunohistochemical stains. The following results were obtained. For histologic grade, 10 cases(11.9%) showed carcinoma in situ, 16 cases(19%) showed grade I, 36 cases (42.9%) showed grade II, and 22 cases(26.2%) showed grade III among the 84 test samples. The average positive rate ER and PR was 63%, 46% showed carcinoma in situ, 80%, 60% showed grade I, 64%, 41% showed grade II, 34%, 23% showed grade III, respectively. The induction of PR increased when induction of ER increased, thus showing significant relationship(p<0.05). The expression of C-erbB-2 protein was 9 cases(10.7%) in one positive(1+), 9 cases(10.7%) in two positive(2+), and 9 cases(10.7%) in three positive(3+). C-erbB-2 protein expression showed no statistical significance. In conclusion, ER and PR positive rates were inversely associated with histologic grades significantly(p<0.05). C-erbB-2 showed no significant difference with histologic grade. However ER, PR and C-erbB-2 showed significant relationship with each other(p<0.05). Therefore, these findings might be an important prognostic factor and might be arranged as a regular pathological examination in cases of breast cancer.

  • Several bisphosphine- and bisphosphite-substituted Re-imido complexes have been prepared from Re(NPh)(PPh3)2Cl3, 1, and Re(N-C6H3-i-Pr2)2Cl3(py), 4. Compound 1 reacted with trimethyl phosphate (P(OMe)3) to give a mixture of two isomers,mer,trans-Re(NPh)(P(OMe)3)2Cl3, 2, and fac,cis-Re(NPh)(P(OMe)3)2Cl3, 2a. In this reaction, the mer,trans-isomer is a major product. Complex 1 also reacted with triethylphosphine (PEt3) to exclusively give mertrans-Re(NPh)(PEt3)2Cl3, 3. Compound 4 reacted with trimethylphosphine (PMe3) to give mer,trans-Re(N-C6H3-i-Pr2)(PMe3)2Cl3, 5, which was converted to mer-Re(N-C6H3-i-Pr2)(PMe)(OPMe3)Cl3, 6, on exposure to air. Crystallographic data for 2: monoclinic space group P21/n, a = 8.870(2) Å, b = 14.393(3) Å, c = 17.114(4) Å, β = 101.43(2)°, Z = 4, R(wR2) = 0.0521(0.1293).   • 대용량의 데이터를 다루는 지리정보 시스템은 I/O가 시스템의 병목현상을 발생시킨다. 따라서, I/O의 병렬화로 GIS의 데이터 요구를 만족시키려는 연구가 이루어져 왔다. PR(Parallel R-tree)[1]도 이러한 연구에 속한다. 한편, 지리 정보 시스템의 질의는 영역질의가 대부분이다. 영역질의는 특성상 인접한 객체들의 동시에 요구한다. 그러므로, 인접한 데이터를 물리적으로 인접하게 배치하면 I/O의 성능이 향상된다. 본 논문에서는 인접한 객체를 물리적으로 인접한 위치에 저장하는 방식인 EPR(Enhanced PR)기법과 단말노드 구조를 제안하고 그 성능을 평가하였다.

Bulk Polymerization of L-lactide with Mixed Aluminum Organometallic Catalysts (Al계 유기금속화합물 혼합촉매 시스템을 이용한 L-lactide 벌크중합 특성 연구)

  • The differences between single and mixed aluminium catalyst systems in the bulk polymerization of L-lactide were studied. $Al(O-i-Pr)_3$, TMA, TOA and TIBA were employed for the mixed-catalyst systems, and TIBA was chosen as a reference catalyst. For the $Al(O-i-Pr)_3$/TIBA catalyst system, the conversion of polymerization increased as the composition of $Al(O-i-Pr)_3$ in the mixed catalyst increased. The molecular weight of the resulting PLA reached to about 13000 g/mol, and the polydispersity index of the polymer from the $Al(O-i-Pr)_3$/TIBA catalyst was slightly increased than that of single catalyst. The higher molecular weight tail or shoulder was revealed in the GPC curve. The conversion of the TOA/TIBA catalyst system decreased as the composition of TOA in the mixed catalyst increased. The molecular weight of PLA prepared with TOA/TIBA catalysts increased up to 14000 g/mol. The Al compounds-mixed catalysts could produce a higher molecular weight tail or shoulder in the GPC curve, which may result in enhancement of mechanical properties of PLA.
