식품을 통한 다이옥신류의 노출 평가 (Exposure assessment of dioxins through foods)

  • 4년 동안(2004-2007) 국내 유통중인 식품 총 221건을 구입하여 다이옥신류(PCDD/DF + 다이옥신유사 PCBs)를 동위원소희석법을 이용하여 분석하였고, 한국인의 다이옥신 1일 노출량을 산출하였다. 식품 중 다이옥신류의 검출량은 2004년과 2005년은 오염도 자료에 1998 WHO TEF값을, 2006년부터는 오염도 자료에 2005 WHO TEF값을 사용하여 다이옥신류 함량을 산출하였으며, 평균 검출량은 백미는 < 0.01 pg TEQ/g, 소고기 0.13 pg TEQ/g, 돼지고기 0.07 pg TEQ/g, 닭고기 0.04 pg TEQ/g, 우유 0.04 pg TEQ/g, 치즈 0.05 pg TEQ/g, 계란(노른자)는 0.13 pg TEQ/g, 수산물 중 고등어는 1.35 pg TEQ/g, 삼치 1.03 pg TEQ/g, 갈치 1.00 pg TEQ/g, 조기 0.16 pg TEQ/g, 명태 0.11 pg TEQ/g, 장어 0.52 pg TEQ/g, 굴 0.32 pg TEQ/g, 꽃게 0.14 pg TEQ/g 수준으로 조사되었다. 식품을 통한 한국인의 다이옥신류의 노출량은 체중 1 kg 당 1일 0.47 pg TEQ로 다이옥신류의 TDI (4 pg TEQ/kg bw/day) 대비 약 11%수준으로 조사되었으며, 이 수준은 건강상 위해가 없는 것으로 평가되었다.

항생제 대체제로서 Clostridium butyricum IDCC 9207 백미 발효물의 항균작용과 장관 면역증진효과 (White Rice Fermented by Clostridium butyricum IDCC 9207 as an Alternative to Antibiotic having Antibacterial and Immunostimulatory Activity)

  • 이 연구는 천연소재 probiotics 발효물을 대상으로 가축의 장내 유해세균으로 알려진 S. typhimurium KCTC 2054와 E. coli O157:H7의 생장을 억제시키는 유효 발효물을 선발하여 특성을 연구하는 데 있다. 실험 샘플 444개 중 유해 세균에 대한 항균력이 가장 우수한 것으로 C. butyricum IDCC 9207 백미 발효물(FWR)을 최종 선발하였다. C. butyricum IDCC 9207이 함유된 백미 발효물 (FWRCb9207)을 S. typhimurium KCTC 2054로 감염시킨 ICR-mice에 일정기간 투여하였을 때, salmonella의 생육이 억제됨을 확인하였으며, 장관에서 분비되는 secretory immunoglobulin A (sIgA)의 양이 증가됨을 확인하였다. FWRCb9207은 기존의 생균제 보다 빠른 정장효과를 나타낼 수 있으며, 발효물보다 생균보강을 통해 정장작용의 기간을 늘릴 수 있는 것이 장점이다. 또한 FWRCb9207의 급여를 통해 사료로부터 쉽게 감염되는 S. typhimurium, S. gallinarum, S. pullorum, S. choleraesuis ssp. choleraesuis 등의 유해세균 증식을 효과적으로 억제시키고 면역력을 증진시킴으로써 가축의 증체율을 높였다. 이러한 FWRCb9207의 synbiotic effects는 궁극적으로 기존 항생제를 대체함으로써 항생제 오남용으로 발생되는 문제점을 해소할 수 있을 것으로 기대된다.

Effects of Fe-soy Proteinate Chelate Supplementation to Diets of Periparturient Sows and Piglets on the Fe Level in the Blood of Piglets

  • The objective of the present study was to investigate the effects of Fe-soy proteinate chelate (Fe-SP) on sows milk, piglet blood parameters and performance. A total of 15 sows of 3 wk before parturition and pigs after births to 3 wk were assigned to three dietary treatments: control (sow-basal diet, piglets with Fe injection); Fe-SP 100 (Fe 100 ppm as Fe-SP in sow and piglet diet); Fe-SP 200 (Fe 200 ppm as Fe-SP in sow and piglet diet). Each treatment had 5 replicates (sows) of six piglets per sow randomly selected from the same offspring. For this experiment, Fe-SP was manufactured. There were no significant differences among treatments in number of pigs born in total or alive per litter, birth weight, number of pigs weaned per litter and weaning weight. However, weight gain, feed intake and feed conversion ratio significantly (p<0.05) decreased as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content at 3 wk before parturition in sow blood. However, Fe content at 2 wk before parturition in sow blood significantly (p<0.05) increased as the supplementation of Fe-SP. While there were no significant differences among treatments in Fe content at 1 wk before parturition in sow blood, it tended to increase as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content of sow milk. However, it tended to increase as the supplementation level of Fe-SP increased. Iron content in the blood of piglets was significantly (p<0.05) higher in control (Fe injected) than Fe-SP 100 and Fe-SP 200 treatments at $1^{st}$ and $2^{nd}$ wk but it was significantly higher in Fe-SP 200 than others in $3^{rd}$ wk. Zinc content in the blood also significantly (p<0.05) increased as the Fe-SP supplementation level increased in $3^{rd}$ wk. In conclusion, Fe-SP supplementation significantly affected Fe content in the blood of piglets. Iron injection was more effective at $1^{st}$ and $2^{nd}$ wk, while Fe-SP 200 supplementation was effective at $3^{rd}$ wk in improving blood Fe level in piglets.

Salmonella Gallinarum 감염닭의 대식세포에서 표고버섯 균사체 발효 미강생물전환소재에 의한 면역조절효과 (Immunomodulation by Bioprocessed Polysaccharides from Lentinus edodes Mycelia Cultures with Rice Bran in the Salmonella Gallinarum-infected Chicken Macrophages)

  • 본 연구에서는, 표고버섯 균사체 발효 생물전환공법으로 생산된 미강생물전환소재(BPP-RB)가 가금티푸스의 주요 원인균인 S. Gallinarum에 감염된 닭 유래 대식세포주 HD-11에 미치는 효과를 조사하였다. 그 결과, 미강생물전환소재 추출액은 S. Gallinarum 277에 대한 직접적인 성장억제 효과를 보여주지 않았으며, 총단백질 및 분비단밸질 발현 양상에 어떠한 변화도 유도하지 못하였다. 하지만, 미강생물전환소재 추출액은 (i) HD-11 대식세포의 탐식 능력(phagocytic activity)을 활성화하였고, (ii) Th1-type cytokines(tumor necrosis factor-${\alpha}$, interleukin $(IL)-1{\beta}$, iNOS)과 immunosuppressive cytokine IL-10의 발현 증가를 유도하였으며, (iii) Th2-type cytokines (IL-4, IL-6)의 발현은 감소시키는 것으로 확인되었다. 이러한 결과를 종합하면, 미강생물전환소재는 가금 농장에서 가금티푸스 및 다른 Salmonella종의 감염을 예방하기 위한 사료첨가제로서의 가능성을 가지고 있다고 사료된다.

조피볼락(Sebastes schligeli)에 있어 사료내 단백질 사료원으로서의 어분대체품 (Fish Meal Analog as a Dietary Protein Source in Koran Rockfish, Sebastes schlegeli)

  • 본 연구는 조피볼락 사료에 있어서 어분대체품을 결정하고, 상품어분대체품과 유인물질의 첨가 효과를 통하여 조피볼락용 어분대체품개발을 위한 기초자료도 사용하고자 수행되었다. 실험사료는 조단백질 52%로, 가용성 에너지 16.8 KJ/g으로 설계되었고, 주단백질원으로 북양어분, 어분대체품 및 상품어분대체품을 사용하였다. 사료의 성분조성은 다음과 같이 요약된다 : 사료1, 100%FM (control) ; 사료2, 60%FM : 40% CFMA ; 사료3, 60%FM : 40% CFMA+ATT ; 사료 4, 80%FM : 20% FMA ; 사료5, 80% FM: 20%FMA+ATT : 사료 6, ; 사료6, 60%FM : 40%FMA+ATT; 사료7, 40%FM : 60% FMA+ATT. 어분대체품은 혈분(BM), 오징어간분(SLP), 육골분(MBM), 수지박(LM), 가금부산물(PBP), 우모분(FM), 필수아미노산(Met, Lys, Ile) 등을 적절히 배합하여 제작 사용하였다. 성장률, 사료효율, 일간성장률 및 단백질효율에 있어서 사료 1은 사료 4, 사료 5 및 사료 6에 비해 유의적인 차이가 없었지만(P>0.05), 나머지 실험구들은 대조구에 비해 유의적으로 낮게 나타났다(P<0.05). 특히, 상품어분대체품(CFMA)인 사료 2와 사료 3은 실험사료로 제작된 동일한 수준의 사료 6과 비교하여 성장률, 사료효율, 단백질효율에서 유의적으로 낮은 경향을 보였다. 0.5% 유인물질을 첨가한 사료 3과 5는 사료 2와 4에 비해 유의적인 차이는 보이지 않았으며(P>0.05), 간중량지수, hemoglobin 및 비만도에 있어서는 유의적인 차이가 나타났다(P<0.05). 따라서, 조피볼락에 있어서 어분대체품(FMA)으로 어분단백질을 40%정도 대체 가능함을 알 수 있었다.

Distribution and differential expression of microRNAs in the intestinal mucosal layer of necrotic enteritis induced Fayoumi chickens

  • Objective: Despite an increasing number of investigations into the pathophysiology of necrotic enteritis (NE) disease, etiology of NE-associated diseases, and gene expression profiling of NE-affected tissues, the microRNA (miRNA) profiles of NE-affected poultry have been poorly studied. The aim of this study was to induce NE disease in the genetically disparate Fayoumi chicken lines, and to perform non-coding RNA sequencing in the intestinal mucosal layer. Methods: NE disease was induced in the Fayoumi chicken lines (M5.1 and M15.2), and non-coding RNA sequencing was performed in the intestinal mucosal layer of both NE-affected and uninfected chickens to examine the differential expression of miRNAs. Next, quantitative real-time polymerase chain reaction (real-time qPCR) was performed to further examine four miRNAs that showed the highest fold differences. Finally, bioinformatics analyses were performed to examine the four miRNAs target genes involvement in the signaling pathways, and to examine their interaction. Results: According to non-coding RNA sequencing, total 50 upregulated miRNAs and 26 downregulated miRNAs were detected in the NE-induced M5.1 chickens. While 32 upregulated miRNAs and 11 downregulated miRNAs were detected in the NE-induced M15.2 chickens. Results of real-time qPCR analysis on the four miRNAs (gga-miR-9-5p, gga-miR-20b-5p, ggamiR-196-5p, and gga-let-7d) were mostly correlated with the results of RNAseq. Overall, ggamiR-20b-5p was significantly downregulated in the NE-induced M5.1 chickens and this was associated with the upregulation of its top-ranking target gene, mitogen-activated protein kinase, kinase 2. Further bioinformatics analyses revealed that 45 of the gene targets of gga-miR-20b-5p were involved in signal transduction and immune system-related pathways, and 35 of these targets were predicted to interact with each other. Conclusion: Our study is a novel report of miRNA expression in Fayoumi chickens, and could be very useful in understanding the role of differentially expressed miRNAs in a NE disease model.

Enrichment and verification of differentially expressed miRNAs in bursa of Fabricius in two breeds of duck

  • Objective: The bursa of Fabricius (BF) is a central humoral immune organ belonging specifically to avians. Recent studies had suggested that miRNAs were active regulators involved in the immune processes. This study was to investigate the possible differences of the BF at miRNA level between two genetically disparate duck breeds. Methods: Using Illumina next-generation sequencing, the miRNAs libraries of ducks were established. Results: The results showed that there were 66 differentially expressed miRNAs and 28 novel miRNAs in bursa. A set of abundant miRNAs (i.e., let-7, miR-146a-5p, miR-21-5p, miR-17~92) which are involved in immunity and disease were detected and the predicted target genes of the novel miRNAs were associated with duck high anti-adversity ability. By gene ontology analysis and enriching KEGG pathway, the targets of differential expressed miRNAs were mainly involved in immunity and disease, supporting that there were differences in the BF immune functions between the two duck breeds. In addition, the metabolic pathway had the maximum enriched target genes and some enriched pathways that were related to cell cycle, protein synthesis, cell proliferation and apoptosis. It indicted that the difference of metabolism may be one of the reasons leading the immune difference between the BF of two duck breeds. Conclusion: This data lists the main differences in the BF at miRNAs level between two genetically disparate duck breeds and lays a foundation to carry out molecular assisted breeding of poultry in the future.

17세기 이전 조선시대 찬물류(饌物類)의 문헌적 고찰 (An Investigation of Side-dishes found in Korean Literatures before the 17th Century)

  • In this study, we investigated e kinds and names of side dishes along with their recipes and ingredients occuring in Korean cookbooks published before the 17th century. The side dishes were classified 79 kinds of Guk, 23 kinds of Jjim and Seon, 15 kinds of Gui, 3 kinds of Jeon, 7 kinds of Nureumi, 3 kinds of Bokkeum, 30 kinds of Chae, 11 kinds of Hoe, 7 kinds of Jwaban, 6 kinds of Mareunchan, 12 kinds of Pyeonyuk and 5 kinds of Jeonyak, Jokpyeon and Sundae. The earliest records were found on Guk, Jjim, Jwaban, Po and Pyeonyuk Gui, Namul and Hoe were recorded after the 1500's and Nureumi, Jeon, Jeonyak, Jokpyeon and Sundae were developed relatively late in the late 17th century. As to the kinds of side dishes, Guk was the most common. Guks cooked before the 17th century used different recipes and more types of ingredients than today, including some that are not used today. For Jjim, various seasonings were added to main ingredients such as poultry, meat, seafood and vegetable. Most of the records found for Jjim used chicken as the main ingredient. Gui was recorded as Jeok or Gui and there weren't many ingredients for Gui before the 17th century. Gui was usually seasoned with salt or soy bean sauce and broiled after applying oil. Vegetables were broiled after a applying flour-based sauce. The Jeon cooked at that time was different from the one that is cooked today in that cow organs or sparrows were soaked in oily soy bean sauce before being stewed. Nureumi, which was popular in the 17th century, but rarely made today, was a recipe consisting of adding a flour or starch-based sauce to stewed or broiled main ingredients. Chae was a side dish prepared with edible plants, tree sprouts or leaves. Chaes like Donga and Doraji were colored with Mandrami or Muroo. Hoe was a boiled Hoe and served after boiling seafood. Jwaban was cooked by applying oil to and then broiling sparrows, dudeok, and mushrooms that had been seasoned and dried. For dried Chans, beef or fish was thin-sliced, seasoned and dried or sea tangle was broiled with pine nuts juice. There are some recipes from the 17th century whose names are gone or the recipes or ingredients have changed. Thus we must to try to rebuild three recipes and develop recipes using our own foods of today.

Diclazuril을 주성분으로 하는 콕시멸-에스의 육계에 대한 실험실 및 야외적용 실험에서의 항콕시듐 효과 (Anticoccidial Efficacy of Coccimuel-S composed with Diclazuril on Experimental and Field Coccidiosis in Broiler Chickens)

  • 디클라쥬릴의 음수형 제제인 콕시멸-에스의 항콕시듐 효과를 Eimeria spp. 감염 육계를 이용하여 수행하였다. 본 실험은 실험실 및 야외적용시험으로 나누어 진행하였으며, 실험실 실험에서는 Eimeria spp.를 인공 감염시킨 육계를 대상으로 콕시멸-에스 0.5 ml/L을 음수로 투여한 군에서 대조군과 비교하여 콕시듐 치료, 콕시듐 충란수의 감소, 그리고 장상해도 및 분변 설사지수 등에 있어서 유의성 있는 효과를 나타내었다($p$ < 0.001). 또한, 야외적용실험에서는, 콕시듐증에 걸린 육계를 대상으로 콕시멸-에스를 0.25와 0.5 ml/L로 각각 음수로 투여한 결과, 대조군과 비교하여 모두 유의한 콕시듐 치료효과를 나타내었다($p$ < 0.001). 이상의 결과로부터, 콕시멸-에스 0.25와 0.5 ml/L를 음수로 콕시듐증에 걸린 육계에 투여할 경우, 콕시듐증 치료에 매우 효과적일 것으로 판단된다.

소, 돼지, 가금육류의 신속한 동정을 위한 TaqMan probe를 이용한 real-time PCR 개발 (Development of TaqMan probe-based real-time PCR for rapid identification of beef, pork and poultry meat)

  • Species-specific $TaqMan^{(R)}$ probe-based real-time PCR assays were developed for detection of beef, pork, chicken, duck, goose and turkey. The primer and probe sets used in this study were designed to be complementary to fibroblast growth factor (FGF) for cattle and pig, mitochondrial NADH dehydrogenase (ND) subunit 3 and ND2 for chicken and duck, 12S rRNA for goose and turkey, respectively. As internal positive control we used conserved region in the ribosomal 18S RNA gene to ensure the accuracy of the detection of target DNA by real-time PCR. We confirmed that real-time PCR assays with the primer and probe sets were positive for cattle, pig and chicken intended target animal species with no cross-reactivity with other non-target animal species. Only >50 ng DNA of beef show cross-reactivity in the determination of duck. Using species-specific primer and probe sets, it was possible to detect amounts of 0.1 ng DNA of cattle and pig, 1.0 pg DNA of chicken, duck and turkey, and 0.1 pg DNA of goose for raw samples, respectively. The detection limits were 0.1 ng DNA of cattle, 1.0 ng DNA of pig and 1.0 pg DNA of chicken for DNA mixtures (beef, pork and chicken) extracted from heat-treated ($121^{\circ}C$/5 min) meat samples. In conclusion, it can be suggested that the $TaqMan^{(R)}$ probe-based assay developed in this study might be a rapid and specific method for the identification of meat species in raw or cooked meat products.
