High Throughput-compatible Screening of Anti-oxidative Substances by Insect Extract Library (약용곤충추출물 라이브러리를 이용한 항산화 활성의 초고속 검색)

  • Oxidant stress is well-known for a pivotal parameter related to neuro-inflammatory diseases including Alzheimer's disease, Parkinson's disease, and ALS (Lou Gehrig's disease). In order to effectively screen for anti-inflammatory agents, we first established the infrastructure of high throughput screening for anti-oxidant agents from medicinal insect library extracted with water, methanol, ethanol, and dimethyl sulfoxide. By the screening system, we found that Tenodera angustipennis Saussure, Pyrocoela rupa Olivier and Papilio maackii Mntris had strong anti-oxidant activity. Moreover, Tenodera angustipennis Saussure and Tenodera aridifolia (Stoll) exhibited protection effects of cellular damage by treatment of an oxidant hydrogen peroxide. Together, the results suggest that some selected hits could be a potential agent against neuro-inflammation, although the in vivo studies should be clearly tested.

Effects of Shading Degree and Substrate Component on Early Growth and Physiological Activity of Panax ginseng Grown under Plastic House (배지조성과 차광정도가 시설하우스 재배 인삼의 초기 생육 및 생리활성에 미치는 영향)

  • Plastic house and laboratory experiments were conducted to determine the effects of substrate components and shade degrees on plant growth and physiologically-active substance content and antioxidant activity of Panax ginseng. No significant difference in growth characteristics among substrates was observed. However, shade treatment also significantly reduced leaf area, chlorophyll content, root length, and shoot and root fresh weight (P<0.05) with increasing of the degree. Total phenolics level [mg ferulic acid equivalents (FAE) $kg^{-1}$ DW] was higher in root parts (25.8 to 28.4 mg $kg^{-1}$) than shoot parts (20.7 to 23.8 mg $kg^{-1}$) of Panax ginseng, and with shade (23.7 to 28.4 mg $kg^{-1}$) than without shade (20.7 to 25.8 mg $kg^{-1}$). However, total flavonoids [mg naringin equivalents $kg^{-1}$ DW] was more detected in the shoot part (17.4 to 26.8 mg $kg^{-1}$) than in the root part (6.7 to 7.0 mg $kg^{-1}$) of Panax ginseng. Methanol extracts from the Panax ginseng plants dose-dependently increased DPPH free radical scavenging activity as a antioxidant potential, and the activity was higher in shoot parts (31.1 to 36.5%) than in root parts (24.9 to 28.7%), and without shade than with shade. In conclusion, shade affects plant growth as well as physiological activity of Panax ginseng under plastic house condition.

Mineralogical Transformation of Gold-silver Bearing Sulfide Concentrate by Mechanochemical Activation, and their Gold-silver Leaching with Non-cyanide Solution (기계적-화학적 활성화에 따른 금-은-정광의 광물학적 상변화와 비-시안 용매에 의한 금-은 용출 향상)

  • In order to leach Au and Ag from gold-silver bearing sulfide concentrate, the sulfide concentrate was ground in a ball mill for a dry pre-treatment and a wet pre-treatment process. Mineralogical studies and thiourea leaching experiments were carried out with the pre-treated sulfide concentrate. The results of the pre-treatment with the concentrate samples showed the mean particle size and iso-electrical potential was smaller in the dry pre-treatment sample than in the concentrate sample, and the contents was lower in the wet pre-treatment sample than in the dry pre-treatment sample. In XRD analysis, amorphous properties were only shown in the wet pretreatment sample. The results of the concentrate sample leaching experiments showed that the best Au, Ag leaching parameters were when the addition of thiourea was at a 1.0 g concentration, ferric sulfate was 1.0 M, sulfuric acid was 2.0 M and the leaching temperature was at $60^{\circ}C$. The Au, Ag leaching rate was always much greater and faster with the wet pre-treatment samples than with the dry pre-treatment samples. Accordingly, it is expected that more Au, Ag can be leached in an eco-friendly methodology using wet pre-treatment. The pre-treatment could be improved with an optimized grinding additive reagent and through researching grinding time in future non-cyanide processes.

Gleditsia Spina Extract Protects Hepatocytes from Oxidative Stress through Nrf2 Activation (皂角刺 추출물의 Nrf2 활성화를 통한 간세포 보호 효과)

  • Objectives : Oxidative stress is one of the most causes of hepatocyte injury. Gleditsia spina, the thorns ofGleditsia sinensisLam., has been known for its anti-cancer and anti-inflammatory effects in Korean medicine. The present study investigated hepatoprotective effect of Gleditsia spina water extract (GSE) against oxidative stress induced by arachidonic acid (AA) + iron in HepG2 cells.Methods : To investigate cytoprotective effect of GSE, cells were pretreated with GSE and then subsequently exposed to 10 μM AA for 12 h, followed by 5 μM iron. Cell viability was monitored by MTT assay, and expression of apoptosis-related proteins was examined by immunoblot analysis. To identify responsible molecular mechanisms, reactive oxygen species (ROS) production, GSH contents, and mitochondrial membrane potential were measured. In addition, effect of GSE on nuclear factor erythroid 2-related factor 2 (Nrf2) activation was determined by immunoblot and antioxidant response element (ARE)-driven reporter gene assays.Results : GSE pretreatment prevented AA + iron-mediated cytotoxicity in concentration dependent manner. In addition, ROS production, glutathione depletion, and mitochondrial impairment by AA + iron were significantly inhibited by GSE. Furthermore, GSE promoted translocation of Nrf2 to nucleus, which acts as essential transcription factor for induction of antioxidant genes. Increased nuclear Nrf2 that caused by GSE treatment promoted transcriptional activity of ARE. Finally, GSE up-regulated sestrin-2 which was widely recognized as target gene of Nrf2.Conclusions : This study demonstrates that GSE protects hepatocytes from oxidative stress via activation of Nrf2 signaling pathway.

Electrochemical Characteristics of Pt/PEM/Pt-Ru MEA for Water Electrolysis (수전해용 Pt/PEM/Pt-Ru MEA의 전기화학적 특성)

  • The membrane electrode assembly(MEA) was prepared by a nonequilibrium impregnation- reduction (I-R) method. Nafion 117 and covalently cross-linked sulfonated polyetherether with tungsto- phosphoric acid (CL-SPEEK/TPA30) prepared by our laboratory, were chosen as polymer electrolyte membrane(PEM). $Pt(NH_3)_4Cl_2$, $RuCl_3$ and reducing agent $(NaBH_4)$ were used as electrocatalytic materials. Electrochemical activity surface area(ESA) and specific surface area(SSA) of Pt cathodic electrode with Nafion 117 were $22.48m^2/g$ and $23.50m^2/g$ respectively under the condition of 0.8 M $NaBH_4$. But Pt electrode prepared by CL-SPEEK/TPA30 membrane exhibited higher ESA $23.46m^2/g$ than that of Nafion 117. In case of Pt-Ru anodic electrode, the higher concentration of Ru was, the lower potential of oxygen reduction and region of hydrogen desorption was, and Pt-Ru electrode using 10 mM $RuCl_3$ showed best properties of SSA $34.09m^2/g$ with Nafion 117. In water electrolysis performance, the cell voltage of Pt/PEM/Pt-Ru MEA with Nafion 117 showed cell property of 1.75 V at $1A/cm^2$ and $80{\circ}C$. On the same condition, the cell voltage with CL-SPEEK/TPA30 was the best of 1.73 V at $1A/cm^2$.

Skin Anti-aging Effect of Forsythia viridissima L. Extract (연교추출물의 피부 항노화 효과)

  • Skin anti-aging effect of Forsythia viridissima L. extract was evaluated by using antioxidant assay, expression of type I procollagen, and UVA-induced matrix metalloproteinase-1 in human dermal fibroblasts. Matairesinol-rich Forsythia viridissima L. extract was showed the scavenging activity of radicals and reactive oxygen species with the $IC_{50}$ values of $4.50\;{\mu}m/ml$ against 1,1-diphenyl-2-picrylhydrazly radical and $542.43\;{mu}m/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. The type I procollagen was increased 33.76% by treatment with matairesinol-rich Forsythia viridissima L. extract, and UVA-induced MMP-1 was reduced 35.78% in a dose dependent manner. In the human skin irritation test, 2% matairesinol-rich Forsythia viridissima L. extract did not show any adverse effect. Also, the clinical study indicated that a cream group treated with 0.2% matairesinol-rich Forsythia viridissima L. extract significantly reduced skin wrinkles, as compared with a non-treated cream group (p < 0.05). These results suggest that Forsythia viridissima L. extract may be useful as a potential source of functional anti-aging cosmetics.

  • The potential candidates for IT-SOFCs cathode materials, $Ba_{0.5}Sr_{0.5}Co_{0.8}Fe_{0.2}O_{3-{\delta}}$ (BSCF) and $La_{0.6}Ba_{0.4}Co_{0.2}Fe_{0.8}O_{3-{\delta}}$ (LBCF) powders, were synthesized by a EDTA-citrate combined method from $Sr(NO_3)_2$, $Ba(NO_3)_2$, $La(NO_3)_3{\cdot}6H_2O$, $Co(NO_3)_2{\cdot}6H_2O$, $Fe(NO_3)_3{\cdot}9H_2O$, citric acid and $EDTA-NH_3$. The cathode performance of symmetrical electrochemical cells consisting of BSCF-GDC or LBCF-GDC composite electrodes and a GDC electrolyte was investigated using by AC impedance spectroscopy at the temperature range of 500 to $700^{\circ}C$. It was found that a single phase perovskite could be successfully synthesized when the precursor is heated at $850^{\circ}C$ for 2 h. Due to thermal expansion mismatch between BSCF and GDC, the composite cathodes with lower GDC content than 45 wt% were peeled off from the GDC electrolyte and their electrode polarization resistance was estimated to be high. The thermal expansion coefficient of BSCF-GDC composites was decreased with increasing the GDC content and the electrode peeling off did not occur in BSCF-45 and 55 wt% GDC composites. BSCF-45 wt% GDC composite electrode showed the lowest area specific resistances (ASR) of 0.15 and $0.04{\Omega}{\cdot}cm^2$ at 600 and $700^{\circ}C$, respectively. On the other hand, LBCF-GDC composite cathodes showed higher ASR than the BSCF-45 and 55 wt% GDC and their cathode performance were decreased with the GDC content.

Effect of Bisphosphonate on Osteoblastic Activity of the Human Periodontal Ligament Cells in Vitro (Bisphosphonate가 배양된 치주인대세포의 조골작용에 미치는 효과)

  • previous studies have demonstrated an increase in bone mass and density with use of bisphosphonate in osteoporosis. This agent acts as an inhibitor of osteoclastic activity and results in increase of net osteoblastic activity. The purpose of the present study was to examine the effect of the bisphosphonate on osteoblastic activity of the human periodontal ligament cells in vitro. Periodontal ligament cells were primarily obtained from extracted healthy third molars. Cells of 4th to 6th passage were cultured in Dulbecco's modified Eagle's medium containing alendronate sodium or etidronate disodium at the concentration of $10^{-12}{\sim}10^{-6}mol/L$ in 5% $Co_2$ incubator at $37^{\circ}C$. Cell count and MTT assay for cellular activity were done at 2 to 7 days of culture. Alkaline phosphatase activity at 4 to 7 days of culture and formation of mineralized nodules at 28 days of culture with addition of $50{\mu}g/m{\ell}$ ascorbic acid, 10 nM${\beta}-glycerophosphate$, $10^{-7}M$ dexamethasone were evaluated. 1. Alendronate sodium Compared to the control, the proliferation of periodontal ligament cells was generally increased and the cellular activity was maintained at 2 days of culture and generally decreased at 7 days of culture. Alkaline phosphatase activity of periodontal ligament cells was inceased and the formation of mineralized nodules by periodontal ligament cells was enhanced compared to the control. 2. Etidronate disodium The proliferation of periodontal ligament cells was increased at 2 days of culture and decreased or maintained at 7 days of culture. Compared to the control, the cellular activity of periodontal ligament cells was generally decreased. Alkaline phosphatase activity of peridontal ligament cells was increased and the formation of mineralized nodules by periodontal ligament cells was enhanced compared to the control. These results suggest that alendronate sodium and etidronate disodium may have a potential effect on osteoblastic lineage of periodontal ligament cells, distinct from their inhibitory action on osteoclasts and could contribute to enhance periodontal regeneration and alveolar bone regeneration.

In Vitro Screening for Anti-Dementia Activities of Seaweed Extracts (해조류 추출물의 In Vitro 항치매 활성)

  • We investigated that methanolic extracts of 20 kinds of seaweeds from Jeju Island for their antioxidant activities, acetylcholinesterase and ${\beta}$-secretase inhibitory activities, and neuronal survival in order to evaluate their potentials as anti-dementia agents. Ecklonia cava extracts had the highest total polyphenol content among the 20 seaweed extracts. The antioxidant activity of seaweed extracts was measured by using 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. It was found that Ecklonia kurome extracts had the highest ABTS scavenging activity ($IC_{50}=0.07{\pm}0.01mg/mL$). As a result, Ecklonia cava, Ecklonia kurome, and Myelophycus simplex extracts were found to be the most effective in terms of acetylcholinesterase inhibitory activity. In the ${\beta}$-secretase activity assay, Ecklonia cava and Ecklonia kurome extracts were effectively inhibited ($84.41{\pm}1.70%$ and $81.17{\pm}2.43%$, respectively). As expected, neuronal cell death induced by $H_2O_2$ in SH-SY5Y cells was diminished by Ecklonia cava, Ecklonia kurome, and Sargassum yezoense extracts. Taken together, these results showed that Ecklonia cava extract has potential anti-dementia activity, which suggests that it might provide an effective strategy for improving dementia.

  • Conventional methods of measuring ground resistance and ground resistance field measurement are used to measure voltage drop according to the resistance value of the site by applying current by installing a constant interval of measurement electrode. If the stratified structure of the site site is unique, errors in boundary conditions will occur in the event of back acid and the analysis of the critical ground resistance in the ground design will show much difference from simulation. This study utilizes the Arduino module and smart ground measurement technology in the convergent information and communication environment to develop a reliable smart land resistance measuring device even if the top layer of land is unique, to analyze the land resistance and accumulate data to predict the change in the age of the land. Considering the topographical characteristics of the site, we propose a ground resistance measuring device and its method of measuring ground resistance so that the auxiliary electrode can be installed by correctly positioning the angle and distance in measuring ground resistance. Not only is ground resistance value obtained through electrodes installed to allow accurate ground resistance values to be selected, but it can also be used as a useful material for installing electrical facilities in similar areas. Moreover, by utilizing reliable data and analyzing the large sections of the site, a precise analysis of the site, which is important in ground design as well as construction cost, is expected to be used much in ground facility design such as potential rise.
