The inhibitory activities of a water extract of Sanghwang mushroom(Phellinus linteusau)(SWE) against ${\alpha}$-glucosidases were evaluated in this study. Inhibiting these enzymes involved in the absorption of disaccharides significantly decreases the postprandial increase in blood glucose level after a mixed carbohydrate diet. Oxygen radical absorbance capacity and 1,1-diphenyl-2-picryl hydrazyl scavenging activities of the SWE were evaluated to investigate the antioxidant activity of the SWE associated with complications of long-term diabetes. Furthermore, the postprandial blood glucose lowering effect of SWE was compared to a known type 2 diabetes drug($Acarbose^{(R)}$) in a Sprague-Dawley rat model. SWE significantly reduced the blood glucose increase after sucrose loading. These results suggest that SWE, which has high ${\alpha}$-glucosidase inhibitory activity and high antioxidant activities, has the potential to contribute to a useful dietary strategy for controlling postprandial hyperglycemia.
Objectives : To review to determine difference in the effect of age, gender, location of electrode, test meal and period of recording that influence Electrogastrography (EGG) parameters and to study its oriental medical application. Methods : We investigate the research results through internet search engines, Pub Med, Karger, Mary Ann Libert and Pro Quest. Results : Age and gender influence postprandial Dominant Frequency Instability Coefficient (DFIC). Body mass index (BMI) influence fasting and postprandial EGG parameters. Menstrual cycle phase influence fasting Dominant Frequency (DF). Test meal volume and composition influence postprandial EGG parameters. But reduced-calorie test meal have no effect on EGG parameters. Conclusions : Because the amplitude of surface-recorded SW(slow wave) is very weak, EGG is influenced by many factors. Age, gender, BMI, menstrual cycle phase, test meal volume and test meal composition influence EGG parameters. but reduced-calorie test meal have no effect on EGG parameters.
A feeding trial was conducted in tilapia to determine the growth performance, nutrient digestibility, digestive enzymes, and postprandial blood metabolites in response to different dietary amylose-amylopectin ratios. Five isonitrogenous and isolipidic diets containing an equal starch level with different amylose-amylopectin ratios of 0.11 (diet 1), 0.24 (diet 2), 0.47 (diet 3), 0.76 (diet 4) and 0.98 (diet 5) were formulated using high-amylose corn starch (as the amylose source) and waxy rice (as the amylopectin source). Each diet was hand-fed to six tanks of 15 fish each, three times a day over a 6-wk period. After the growth trial, a postprandial blood metabolic test was carried out. Fish fed diet 2 exhibited the highest percent weight gain and feed efficiency and protein efficiency ratio, whereas fish fed with diet 5 showed the lowest growth and feed utilization among treatments. The digestibility for starch in fish fed diet 1 and 2 was higher than those in fish fed with other diets (p<0.05). The highest activities for protease, lipase and amylase were found in fish fed the diet 2, diet 1, and diet 1 respectively among dietary treatments, while the lowest values for these indexes were observed in fish fed the diet 3, diet 5 and diet 4, respectively. The liver glycogen concentrations in fish fed diets 4 and 5 were found higher than in fish fed other diets (p<0.05). The feeding rate, hepatosomatic index, condition factor, and plasma parmeters (glucose, triglyceride, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol) did not differ across treatments. In terms of postprandial blood responses, peak blood glucose and triglycerides were lower after 3 or 6 h in the fish fed with diets 3-5 than in the fish fed diet 1, but delayed peak blood total amino acid time was observed in fish fed with the diets 1 or 2. The lowest peak values for each of the three blood metabolites were observed in fish fed diet 5. The results indicate that high-dietary amylose-amylopectin ratio could compromise growth, but help in reducing the blood glucose stress on fish caused by postprandial starch load.

ICR 마우스와 streptozothocin (STZ)으로 유도된 당뇨병 마우스에 수용성 전분(2 g/kg body weigh)으로 경구부하 후 C3G (10 mg/kg body weight) 또는 acarbose (10 mg/kg body weight)를 단독 또는 함께 투여하였다. 혈액 샘플은 꼬리에서 0, 30, 60, 120분 간격으로 채취하였다. ${\alpha}$-글루코시다아제와 ${\alpha}$-아밀라아제에 대한 C3G의 $IC_{50}$ 값은 각각 13.72와 $7.5\;{\mu}M$의 결과값을 나타내어, 양성대조군인 acarbose보다 더 효과적이었다. 본 연구에서는 동충하초, 메밀쌀, 인진쑥 등 천연소재 혼합물 섭취가 당뇨병 환자의 혈당수준 및 당화혈색소 변화에 미치는 영향을 측정하였다. 천연소재 혼합물을 섭취한 후 공복혈당과 식후혈당이 각각 118.4${\pm}$3.9mg/dl, 160.5${\pm}$4.0mg/dl로 섭취 전에 비해 36.5%와 45.0%로 유의적인 감소를 보였으며, 대상자들의 혈당수준은 4주 이내에 정상범위에 도달하였다. 그러나 천연소재 혼합물의 섭취를 중단한지 3주 후에는 168.4${\pm}$6.2mg/dl 와 245.9${\pm}$7.8mg/dl로 유의적으로 증가하였고, 위약제품으로 전환하여 섭취시킨 7주후에는 실험시작 전의 혈당수준으로 돌아갔다. 대상자들의 당화혈색소는 7주간의 천연소재 혼합물을 섭취한 후 유의적으로 감소하였다. 결론적으로 7주 동안의 천연소재 혼합물의 섭취가 당뇨병 환자의 당질 대사에 긍정적인 효과를 미치는 것으로 관찰되었으므로 동충하초, 메밀쌀, 인진쑥이 첨가된 기능성 식품이 당뇨병의 치료 및 예방에서 유용한 것으로 판단된다.
벼메뚜기는 메뚜기과에 속하는 불완전 변태 곤충으로, 국내에서 오랫동안 식용으로 이용되어 왔다. 현재까지 벼메뚜기의 항산화 및 항염증 효과 등의 기능들이 연구되었으나, 탄수화물 소화 효소나 식후 혈당 수치에 미치는 영향에 관한 연구는 부족한 실정이다. 이에 본 연구는 벼메뚜기 에탄올 추출물(OEE)와 물 추출물(OWE)이 탄수화물 소화 효소를 저해하고, streptozotocin (STZ)으로 유도된 당뇨병 마우스에서 식후 고혈당을 강하시키는 효과에 대해 조사하였다. 그 결과 OEE에 의한 α-glucosidase와 α-amylase 저해 효과가 OWE 보다 더 효과적이었다. 당뇨병 마우스에 전분(2 g/kg)을 투여한 후의 혈당 증가는 15, 30, 60, 120분에 각각 406.50, 431.50, 473.50 and 460.00 mg/dl로 나타났고, 전분(2 g/kg)과 OEE 추출물(300 mg/kg)을 투여한 후 혈당 증가는 15, 30, 60, 120분에 각각 327.00, 361.00, 358.67 and 291.00 mg/dl로 나타나, OEE 추출물 투여군이 대조군에 비해 식후 혈당 강하가 효과적으로 나타남을 알 수 있었다. 이러한 결과는 벼메뚜기 추출물이 α-glucosidase와 α-amylas를 저해함으로써 식후 고혈당을 완화시키고, 특히 벼메뚜기 에탄올 추출물(OEE)이 벼메뚜기 물 추출물(OWE) 보다 식후 고혈당을 완화시키는데 더욱 효과가 있는 것으로 나타났다. 따라서 OEE가 탄수화물 소화 효소 저해 효과로 식후 고혈당을 완화시키는 유용한 천연 기능성 식품이 될 것으로 사료된다.
현대인의 기호식품인 차를 이용하여 비 발효차인 녹차와 3가지 발효차로 구분하여 식후 고혈당의 지표가 되는 알파글루코시데이스(α-glucosidase) 활성과 고혈압활성의 지표로 사용되는 안지오텐신전환효소(angiotensin converting enzyme, ACE)의 억제작용을 확인하였다. α-glucosidase 억제 활성의 상승효과를 알아보기 위해 녹차추출물에 인슐린과 유사기능이 있다고 밝혀진 바나듐을 미량(50 ㎍/mL)으로 혼합하여 α-glucosidase 활성 억제를 위한 상승효과 여부를 조사하였다. 또한 녹차의 기능성물질로 알려진 에피갈로카테킨 갈레이트(epigallocatechin gallate, EGCG)의 농도와 카페인(caffeine)의 농도를 측정하여 녹차와 발효차 효능을 분석하고자 하였다. 녹차와 발효차 추출물로부터 식후 혈당을 증가시키는 α-glucosidase 활성이 크게 억제되어 차의 식후 혈당저해 효과가 있음을 확인하였으며 차 추출물 중에서도 20% 발효차가 가장 우수한 기능을 보였다. 그러나 차 추출물만을 단독으로 처리한 경우와 바나듐과 함께 처리한 경우 서로 다른 결과를 보였다. α-glucosidase 저해효과는 저 농도(50 ㎍/mL)의 녹차와 바나듐 혼합 처리에서 단독 처리에 비해 저해활성이 2배나 증가되어 녹차가 바나듐과 가장 큰 상승효과를 보임을 알 수 있었다. ACE 저해활성은 모든 차 추출물에서 확인되었으나 녹차에서 가장 높게 조사되어 녹차가 다른 차에 비해 가장 높게 측정된 EGCG 농도의 효과로 생각되며 발효차의 식후 혈당강하 및 ACE 저해효과는 EGCG 외에 또 다른 기능성물질이 관여한 것임을 알 수 있었다.
This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa on ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) and postprandial hyperglycemia. Methanol extract and organic solvent (n-hexane, ethyl acetate, butanol, aqueous) fractions from the crude drug were determined for the inhibitory activities against maltase, sucrase and ${\alpha}-amylase$. The methanol extract from the crude drug strongly inhibited maltase (72%) and sucrase (76%) at the concentration of $100\;{\mu}g/ml$. Among the fractions examined, the ethyl acetate fraction from the natural plant drug showed potent inhibitory effects on maltase (85%) and sucrase (81%) at the concentration of $100\;{\mu}g/ml$. The ethyl acetate fraction from root cortex of Paeonia suffruticosa also exhibited significant reductions (21%) of blood glucose elevation in mice loaded with maltose.
This study was designed to investigate whether Sargassum ringgoldianum extract may inhibit ${\alpha}$-glucosidase and ${\alpha}$-amylase activities, and alleviate postprandial hyperglycemia in streptozotocin-induced diabetic mice. The $IC_{50}$ values of Sargassum ringgoldianum extract against ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.12 mg/mL and 0.18 mg/mL, respectively, which evidenced higher activities than those of acarbose. The blood glucose levels of the Sargassum ringgoldianum extract administered group were significantly lower compared to the control group in the streptozotocin-induced diabetic mice. Moreover, the area under the two-hour blood glucose response curve was significantly reduced and the absorption of dietary carbohydrates was delayed after administration of Sargassum ringgoldianum extract in the diabetic mice. Therefore, these results indicated that Sargassum ringgoldianum extract may help decrease the postprandial blood glucose level via inhibiting ${\alpha}$-glucosidase.
Since diet is an essential component of the treatment for diabetic mellitus, a progressive educational plan was designed to educate diabetic children for the proper selection of their foods. Seven suspicious children were chosen according to the previous oral glucose tolerance test and present blood glucose levels (fasting and postprandial 2 hours). The education program includes the basic nutrition study, learning of the five basic food groups. familiarization with the food exchange lists. calorie calculation and menu planning, and follow-up evaluation by checking every day -food intake. The duration of the education was five weeks. The levels of fasting blood glucose and postprandial -2 hours blood sugar of the seven children were significantly reduced after the 5-weeks education:FBS:from 92-125mg% to 67-80mg%, pp-2hours BS: from 130-169mg% to 69-90mg%. The children have felt much better with profound self-confidence after to program . Their oral glucose tolerance test levels were also significantly decreased after the 5-weeks education in comparison with those of one year ago.
