The protective effect of CXC chemokine receptor 2 antagonist on experimental bronchopulmonary dysplasia induced by postnatal systemic inflammation

  • Background: Animal studies have shown that a leukocyte influx precedes the development of bronchopulmonary dysplasia (BPD) in premature sheep. The CXC chemokine receptor 2 (CXCR2) pathway has been implicated in the pathogenesis of BPD because of the predominance of CXCR2 ligands in tracheal aspirates of preterm infants who later developed BPD. Purpose: To test the effect of CXCR2 antagonist on postnatal systemic and pulmonary inflammation and alveolarization in a newborn Sprague-Dawley rat model of BPD. Methods: Lipopolysaccharide (LPS) was injected intraperitoneally (i.p.) into the newborn rats on postnatal day 1 (P1), P3, and P5 to induce systemic inflammation and inhibit alveolarization. In the same time with LPS administration, CXCR2 antagonist (SB-265610) or vehicle was injected i.p. to investigate whether CXCR2 antagonist can alleviate the detrimental effect of LPS on alveolarization by attenuating inflammation. On P7 and P14, bronchoalveolar lavage fluid (BALF) and peripheral blood (PB) were collected from the pups. To assess alveolarization, mean cord length and alveolar surface area were measured on 4 random nonoverlapping fields per animal in 2 distal lung sections at ×100 magnification. Results: Early postnatal LPS administration significantly increased neutrophil counts in BALF and PB and inhibited alveolarization, which was indicated by a greater mean cord length and lesser alveolar surface area. CXCR2 antagonist significantly attenuated the increase of neutrophil counts in BALF and PB and restored alveolarization as indicated by a decreased mean cord length and increased alveolar surface area in rat pups exposed to early postnatal systemic LPS. Conclusion: CXCR2 antagonist preserved alveolarization by alleviating pulmonary and systemic inflammation induced by early postnatal systemic LPS administration. These results suggest that CXCR2 antagonist can be considered a potential therapeutic agent for BPD that results from disrupted alveolarization induced by inflammation.

Effects of a Postnatal Care Program on Self-efficacy, Self-management, and Glycemic Control in Women with Gestational Diabetes Mellitus (임신성 당뇨병 산모를 위한 산후관리 프로그램이 자기효능감, 자가 관리 및 혈당 조절에 미치는 영향)

  • Purpose: To examine effects of a postnatal care program on self-efficacy, self-management, and glycemic control in women with gestational diabetes mellitus (GDM). Methods: A non-equivalent control group non-synchronized quasi-experimental design was used. Sixty-two women with GDM were enrolled and assigned to either an experimental group (n=30) or a control group (n=32). The experimental group received an intervention which was postnatal care program for women with GDM. The postnatal care program for GDM included an individual education with leaflet and mobile web-based video with three times of telephone counseling. Effects of the intervention were measured with self-efficacy, self-management questionnaire, and a 75 g oral glucose tolerance test (75g OGTT). Statistical significance was examined using independent t-test and $x^2-test$. Results: Although there was no significant difference in 75g OGTT ($x^2=.11$, p=.748) or self-management (t=-1.28, p=.206), there was a statistically significant increase in self-efficacy (t=-2.02, p=.048) in the experimental group compared to that in the control group. Conclusion: A postnatal care program is needed for women with GDM to improve their self-efficacy. Further studies are warranted to improve self-management and glycemic control through tailored education for GDM postpartum women.

Assessment of Adipocyte Differentiation and Maturation-related Gene Expression in the Epididymal Fat of Estrogen Receptor α Knockout (ERαKO) Mouse during Postnatal Development Period

  • The absence of functional estrogen receptor α (Esr1) results in an overgrowth of the epididymal fat, as observed in estrogen receptor α knockout (ERαKO) mouse. The present research was aimed to evaluate expression of various molecules associated with adipocyte differentiation and maturation in the epididymal fat of ERαKO mouse at several postnatal ages by using quantitative real-time polymerase chain reaction. The highest transcript levels of all molecules were detected at 12 months of postnatal age, except leptin which the mRNA level was increased at 5 months of age and was unchanged until 12 months of age. The expression levels of CCAAT enhancer binding protein (Cebp) alpha, androgen receptor, and lipoprotein lipase were decreased at 5 months of age but increased at about 8 months of age. The mRNA levels of Cebp gamma and sterol regulatory element binding transcription factor 1 remained steady until 8 months of age. Continuous increases of transcript levels during postnatal period were found in Cebp beta, estrogen receptor (ER) beta, fatty acid binding protein 4, and delta like non-canonical Notch ligand 1. The increases of peroxisome proliferator-activated receptor gamma and adiponectin mRNA levels were detected as early as 8 months of age. The levels of fatty acid synthase and resistin transcript at 5 and 8 months of age were lower than that at 2 months of age. These findings show the aberrant expression patterns of genes related to adipocyte differentiation and maturation in the postnatal epididymal fat pad by the disruption of ER alpha function.

IMMUNOHISTOCHEMICAL STUDY ON THE POSTNATAL DEVELOPMENT OF SOMATOSTATIN IMMUNOREACTIVE NEURONS IN THE RAT CEREBRAL CORTEX (흰쥐 대뇌피질에서의 Somatostatin 신경세포의 생후발달에 관한 면역조직화학적 연구)

  • With immunohistochemical method, We have examined the distribution, the size, the number, and the morphology of somatostatin-immunoreactive neurons in the cerebral cortex of rats at the 1st. 2nd, 3rd, 4th postnatal week and the 2nd and 3rd months of age. The results are summarized as follows : 1) Strongly immunoreactive somatostatin neurons were found in the layer V and VI at the first postnatal week and then they were observed to be densely populated in the layer II and III from the 3rd postnatal week on as in the adult rat. 2) The size of the neuron was largest at the 2nd postnatal week while the number of the neurons was greatest in the 1st or 2nd postnatal week, depending on the different areas of the cortex. Overall the postnatal development of somatostatin neurons are well corresponds with the general principle of neuronal development of CNS.

  • The purpose of this study was to investigate the distribution of nerves in the rat circumvallate papilla by each developing stage. The distribution of nerves in the rat circumvallate papilla were investigated by means of immunohistochemistry for detection of calcitonin gene-related peptide(CGRP). The results were as follows : CGRP-immunoreactive(IR) nerve fibers entered the base of the papilla laterally to form a subepithelial plexus. On the 1st postnatal day, the bead-like appearance, typical appearance of CGRP-IR nerve fiber, was seen, but from the 5th postnatal day the bead-like appearance was seen less clearly and nerve fibers looking like a line were often observed. Mature taste buds having taste pores were first seen at the 10th postnatal day and then, increased markedly after this stage. CGRP-IR nerve fibers entering the epithelium were rarely seen on the 5th postnatal day but they increased in number on the 10th postnatal day when mature taste buds having taste pores were firstly observed and then on the 15th postnatal day, they were observed abundantly throughout entire epithelium. But from the 20th postnatal day, the nerve fibers in the lower two-third of the trench wall in which taste buds exist decreased but on the top surface and upper one-third of the trench wall the nerve fibers were observed limitedly.

Perinatal and Postnatal Study of KTC-1, a New Semisynthetic Rifamycin Derivative, in Rats (새로운 반합성 Rifamycin 유도체 KTC-1의 랫트 주산기 및 수유기 시험)

  • A perinatal and postnatal study of KTC-1, a new semisyntheitic rifamycin antituberculous drug, was conducted in Sprague-Dawley rats. Dosages of KTC-1 0, 12, 27.6, and 63.5 mg/kg/day were administered to dams orally by gavage from day 17 of gestation to day 21 of lactation. All pregnant rats were allowed to deliver naturally for postnatal examination of their offspring. At 63.5 mg/kg/day, weakness, dark-red discharge around eyes, a loss in body weight, and a decrease in food and water consumption were observed in dams. An increase in the weight of adrenal gland and spleen, and a decrease in the weight of kidney and heart were also found. An increase in neonatal deaths during the lactation period, a loss in body weight, a delay in physical development, a decrease in traction ability, an increase in the number of errors and the time required for the multiple T-maze trial were found in F1 offspring. In addition, an increase in the incidence of visceral variations and retarded ossification were observed in F1 4 day old rats. An increase in the incience of skeletal anomalies was seen in F2 fetuses. There were no sings of maternal toxicity or embryotoxicity at 12 and 27.6 mg/kg/day. From the results mentioned above, it can be concluded that the no-effect dose levels(NOELs)for dams, F1 offspring, and F2 fetuses are 27.6 mg/kg/day.

Evaluation and Management of Antenatal Hydronephrosis

  • Antenatal hydronephrosis (ANH) is one of the most common abnormal findings detected on prenatal ultrasound (US), and it has been reported in 1-5% of all pregnancies. The likelihood of significant postnatal pathologic abnormality in the urinary tract correlates with the degree of anterior-posterior diameter (APD) according to the gestational age. Detection of urologic anomalies prenatally permits fetal interventions that avoid complications in rare cases of bladder outlet obstruction with oligohydramnios even though their final benefits still remain controversial. There is no clear consensus on the extent and mode of postnatal imaging after a diagnosis of ANH. US is the mainstay of the postnatal evaluation and helps guide further testing with voiding cystourethrography (VCUG) and diuretic renography. Although most algorithms continue to recommend generous VCUG for identification of lower urinary tract anomalies, VCUG may be safely reserved for high grade ANH cases or any grade of ANH with dilated distal ureter without increasing the risk of urinary tract infection (UTI). There are conflicting studies about efficacy of postnatal prophylactic antibiotics. It still seems reasonable to consider use of a prophylactic antibiotic to prevent infant UTIs in high-risk populations, such as females and uncircumcised males with high grades of hydronephrosis, hydroureteronephrosis, or vesicouretral reflux.

Examination of the Presence and Differential Expression of Connexin Isoforms in the Urinary Bladder of the Male Rat During Postnatal Period

  • The coordinated action of the detrusor muscle cells in the urinary bladder is governed by cell-cell communication through gap junction, consisted of connexin (Cx) molecules. Even though a number of researches have been mostly focused on expressional changes of a few Cx isoforms in clinically dysfunctional condition of the bladder, less attention has been paid for investigation of Cx isoforms present in the bladder. Using real-time PCR analysis, the present study examined Cx isoforms expressing in the male rat bladder during postnatal period. Also, expressional patterns of Cx isoforms were evaluated in the bladder at different postnatal ages. Of a total of 13 Cx isoforms tested in the present study, we were able to detect mRNAs of 6 Cx isoforms in the rat urinary bladder, including Cxs 31, 31.1, 32, 37, 40, and 45. The transcript levels of Cxs 31, 31.1, 37, 40, and 45 were gradually increased from 1 week of age until 25 days of age, followed by transient decreases at 45 days of age. However, abundance of Cx32 transcript was drastically increased at 15 days of age, followed by a sharp drop at 45 days of age. These results indicate that differential expression of Cx isoforms in the bladder during postnatal development would be necessary for maintaining proper function of the bladder. A question remains to be answered if significant decreases of transcript levels of some Cx isoforms at the elderly are associated with age-dependent dysfunction of the bladder.

The Effects of Preschooler Temperament and Maternal Postnatal Depression, Depression, and Parenting Stress on Preschooler Externalizing Problem Behavior (유아의 기질, 어머니의 산후우울, 우울 및 양육스트레스가 유아의 외현화 문제행동에 미치는 영향)

  • Objective: The main purpose of this study was to investigate the effects of preschooler temperament and maternal postnatal depression, depression, and parenting stress on preschooler externalizing problem behavior. Methods: The participants consisted of 98 preschoolers (ages 4-5 years) and their mothers. The subjects completed the following questionnaires: Emotionality, Activity, and Sociability (EAS), Edinburgh Postnatal Depression Scale (EPDS), Center of Epidemiological Studies Depression Scale (CES-D), Parenting Stress Scale, Korean Child Behavior Checklist for Ages 1.5-5 (K-CBCL 1.5-5), and Social Competence and Behavior Evaluation Inventory Short Form (SCBE-30). The data were analyzed by t/F tests, Pearson's correlation analysis, and multiple regression analysis. Results and Conclusion: First, preschooler emotionality temperament had positive correlations with attention problems and aggression. Second, maternal depression and parenting stress had a positive correlation with preschooler externalizing problem behaviors. Third, maternal parenting stress had an effect on preschooler attention problems. Forth, preschooler emotional temperament and maternal parenting stress had an effect on preschooler aggression.

Research on Biological and Sociobehavioral Factors Affecting Determinants of Breast Feeding Duration (모유영양의 실시 시간에 영향을 주는 생리적.사회행동적 요인 분석)

  • The process of breast feeding was investigated from the 36th week of pregnancy to 12 weeks postnatal, in order to analyze the factors affecting breast feeding duration and provide the basic data on an education program for successful breast feeding. The general characteristics of study group had no marked effects on the breastfeeding duration. The young and highly condition during pregnancy and the degree of morning sickness affected the practice of breastfeeding. In the study of prenatal sociobehavioral factors, intended duration affected the breastfeeding duration, and the maternal perception that breastfeeding is good for weight loss after delivery as well as infant formula milk is not as good as breast milk, led to successful breast feeding. Among the postnatal biological factors, the duration of gestation affected the breast feeding. Among the postnatal biological factors, the duration of gestation affected the breastiffeding outcome. In the case of early parturition, breastfeeding rate was low. Among the postnatal biocultural factors, time of first feed, milk volume and maternal perception of infant's sucking ability affected the bresatfeeding duration. From these result, it is suggested that an education program such as participation of nursing class to the pregnant women should be provided for successful breastfeeding.
