Inhibitory Effect of Moriniafungin Produced by Setosphaeria rostrata F3736 on the Development of Rhizopus Rot

  • Rhizopus rot is a serious postharvest disease of various crops caused by Rhizopus spp. and controlled mainly by synthetic fungicides. We detected the antifungal activity of a culture extract of Setosphaeria rostrata F3736 against Rhizopus oryzae. The active ingredient was identified as moriniafungin, a known sordarin derivative, which showed minimum inhibitory concentrations of 1-8 ㎍/ml against Colletotrichum spp. and 0.03-0.13 ㎍/ml against Rhizopus spp. in vitro. Moriniafungin showed protective control efficacies against Rhizopus rot on apple and peach fruits. Treatment with 25 ㎍/ml moriniafungin delimited the lesion diameter significantly by 100% on R. oryzae-inoculated apple fruits compared with the non-treated control. Treatment with 0.04 ㎍/ml of moriniafungin reduced the lesion diameter significantly by 56.45%, and treatment with higher concentrations of 0.2-25 ㎍/ml reduced the lesion diameter by 70-90% on Rhizopus stolonifer var. stolonifer-inoculated peach fruit. These results suggest moriniafungin has potential as a control agent of postharvest diseases caused by Rhizopus spp.

Control Effect of a Stored Grain Insect Pest, Tribolium castaneum, by 'CATTS' Postharvest Treatment (CATTS를 이용한 저곡해충 거짓쌀도둑거저리(Tribolium castaneum)의 소독 효과)

  • A postharvest treatment called CATTS (controlled atmosphere and temperature treatment system) has been used as an alternative nonchemical measure for methyl bromide fumigant treatment. This study applied CATTS to control the red flour beetle, Tribolium castaneum, infesting stored grains. Adults of T. castaneum were susceptible to $46^{\circ}C$ heat treatment. The susceptibility was further enhanced by addition of CA conditions (15% $CO_2$ and 1% $O_2$). When CATTS ($46^{\circ}C$, 15% $CO_2$, $16^{\circ}C/h$ treating rate) was applied to different developmental stages of T. castaneum, it showed 100% control efficacy by 120 min exposure. There was a variation in CATTS susceptibility among developmental stages, in which late instar larvae were most tolerant. Heat shock proteins of T. castaneum appeared to be implicated in the tolerance of CATTS.

  • Southeast Asia, a typical tropical region, plays an important role in exporting a variety of fruits worldwide. The market for fresh fruits has been growing consistently, and this is a chance for Southeast Asian countries to increase their national income. However, export of tropical fruits has limitations such as a short shelf life and difficulty in maintaining the quality because of tropical climate conditions and undeveloped postharvest technologies in Southeast Asia. An important objective for developing postharvest technologies is to extend the shelf life of fresh fruits without deterioration in fruit quality. Therefore, it is essential to determine factors that affect the shelf life of fruits. The shelf life of tropical fruits is significantly dependent on the inherent properties of the fruits, extrinsic conditions, postharvest treatment, and microbial contamination. Recently, Southeast Asian countries have supported agricultural research groups for developing new postharvest technologies and minimizing postharvest losses and maintaining export fruit quality so that the total sales of tropical fruit farms can increase. This review introduces how the primary factors for extending the shelf life of tropical fruits can be determined and discusses the development of postharvest technologies for tropical fruits in Southeast Asian countries.

Effect of Nutrient Solution Concentration in the Second Half of Growing Period on the Growth and Postharvest Quality of Leaf Lettuce (Lactuca sativa L.) in a Deep Flow Technique System (담액수경재배 시 재배후기 양액농도가 상추의 생장 및 수확후 품질에 미치는 영향)

  • We examined the effect of nutrient solution concentration in the second half of growing period on the pre- and postharvest characteristics of two leaf lettuce cultivars, 'Geokchima' and 'Cheongchima'. Plants were grown hydroponically in a deep flow technique (DFT) system at different concentrations of National Horticulture Research Institute hydroponic nutrient solution: 1/2 strength (S), 1S, 2S, and 4S. Lettuce leaf growth, number of leaves, and shoot fresh weight of both cultivars were greatest in plants grown in the 1S treatment. Compared to other treatments, pigment and nutrient ion contents were greater in the 4S treatment. Growth of lettuce was greatest in the 1S treatment, and decreased at higher or lower concentrations of nutrient solution. However, postharvest characteristics such as fresh weight loss, leaf chlorophyll level, and external appearance were better in both cultivars when grown in 2S solution. Variations in weight loss and SPAD values were smallest in the 2S treatment. These results show that the optimal nutrient concentration for growth does not necessarily provide the optimal postharvest storability.

Effects of Ionizing Radiation on Postharvest Fungal Pathogens

  • Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

Control of postharvest fungal spoilage of kiwifruit with TiO$_2$ photocatalytic ozonation.

  • TiO$_2$photocatalytic ozonation was attempted to disinfect fungal pathogens causing postharvest spoilage of kiwifruits and to decompose fungicide residuals on kiwifruits. TiO$_2$Photocatalytic ozonation process synergistically degraded organic compound and inhibited conidial germination of the fungal pathogen compared to single treatment of ozonation or photocatalysis. The efficient control of fungal spoilage and degradation of residual fungicide on kiwifruits indicate that TiO$_2$photocatalytic ozonation is a very attractive method for postharvest disease control of kiwifruits as an alternative to fungicides application.

Combined Effects of Postharvest Calcium Chloride and Heat Treatment on the Quality Characteristics of Fresh-Cut Tsugaru Apple

  • To produce fresh-cut fruit products of high quality, the combined effects of postharvest calcium and heat treatment on Tsugaru apple were examined. Whole fruits were treated for 20 min at three different conditions: dipping in 3% $CaCl_2$ solution at $45^{\circ}C$ or ambient temperature, or in heated water at $45^{\circ}C$. The calcium content of the apples dipped in $CaCl_2$ solution at $45^{\circ}C$ was higher than that of the control and the non-heated calcium dipping. The fruits with calcium and heat treatment showed the same pattern of respiration rates as the control did during storage. The browning degree of the apple slices increased to approximately 3% after one-day storage, but no significant difference was observed between the treated and untreated apples. The fruits with $CaCl_2$ solution dipping at $45^{\circ}C$ had higher firmness than those with calcium dipping or heat treatment alone.

Initial cooling conditions that extend the shelf-life of fresh oak mushrooms (Lentinula edodes) after harvest (신선 표고의 수확 후 유통수명 연장을 위한 예냉조건 구명)

  • To optimize initial cooling conditions, forced-air cooling was applied to freshly harvested oak mushrooms at 2 levels (0℃ for 30 minutes, at 0℃ for 1 hour) followed by room cooling at 3 levels (-3℃ for 1 day, 0℃ for 1 day, 3℃ for 1 day). After initial cooling, the oak mushrooms were packaged with PVC film, then held in a storage room at 1℃ for 6 weeks. Quality characteristics and percentage marketability were then investigated. As a control, Mushrooms were placed in storage with no initial cooling. The quality factors impacting marketability of fresh oak mushrooms were color change and appearance of decay. Off-odor did not occur or developed only slightly, so it did not affect oak mushroom quality within 6 weeks of low temperature storage. In all treatment groups, the shelf life in which 100% marketability was maintained was up to 3 weeks. At week 5, percent marketability of the 3 treatment groups 1 hour room cooling treatment at 0℃, 1 hour forced air cooling, and control was 100%. 80% In the group that underwent 30 min forced air cooling retained 80% marketability, and the group exposed to 1 day in room cooling at -3℃ retained 86.7% marketability. At week 6 of 1℃ storage, the marketability ratio was 80% in the 1 day room cooling at 0℃ group, 66.7% in the 1 day room cooling at 3℃ group, 46.7% in the 1 hour forced air cooling group, and 33% or less in all other treatment groups. Therefore, the most suitable initial cooling parameter to extend shelf-life of oak mushrooms is 1 day of in room cooling at 0℃ immediately after harvest.

Postharvest Quality Changes of Kimchi Cabbage 'Choongwang' Cultivar as Influenced by Postharvest Treatments (저장 전처리 방법에 따른 배추 '춘광' 품종의 품질변화)

  • Kimchi cabbage 'Choongwang' cultivar is mainly cultivated during summer in Gangneung area. 'Choongwang' cultivar was harvested in late July, applied with predrying, room cooling, and forced air cooling, and then packaged with/without 0.02 mm HDPE film to estimate the effect of postharvest treatment on quality characteristics (weight loss, trimming loss, firmness, SSC, color index, sensory evaluation) during 8 week storage at $2^{\circ}C$. Kimchi cabbage without 0.02 mm HDPE film showed high weight loss up to 13-20% while those of with liner were significantly lower. Also forced air cooling among the postharvest treatments was effective to reduce both weight loss and trimming loss. Appearance and freshness in sensory evaluation were the important factors in estimating good quality during storage. Liner treatment with forced air cooling showed highly significant for maintaining appearance and freshness ($P{\leq}0.01$). Color index was no differences between with/without 0.02 mm HDPE film and postharvest treatments. After 6 weeks storage in without 0.02 mm HDPE film with room cooling or control appearance was severely damaged and also internal browning was found. While in with 0.02 mm HDPE film internal browning was found after 8 weeks storage, just in room cooling or predrying treatment.

Effect of Postharvest Treatments on Storage Quality of Buckwheat Sprouts (메밀 새싹채소의 저장품질에 대한 수확 후 처리공정 효과)

  • The storage quality of fresh buckwheat sprouts, as influenced by pretreatment and packaging within processing steps, was investigated to establish appropriate postharvest handling treatment for the commodity. After harvest, the sprouts were dipped in chlorine water (100 ppm), rinsed twice with clean water, pre-cooled with iced water, de-watered, and packed in plastic trays. Sprout samples taken from each processing step were stored at $5^{\circ}C$ for 6 days to measure quality attributes. Viable cell counts of mesophilic aerobes and coliform bacteria were lower by about 1 log scale in the postharvest treated samples compared to an untreated control, although the initial microbial reduction due to the postharvest treatments was offset by cell growth during storage. All sprout samples showed a decrease of fresh weight by approximately 4% after 6 days of storage. However, moisture and soluble solid contents were maintained at the initial levels of the sprouts. No significant difference in surface color was observed among sample treatments. For sensory properties including discoloration, wilting, decay, and visual quality, there were no significant differences among sample treatments. The present results suggest that proper postharvest processing treatments can exert positive effects on extending the shelf-life of fresh buckwheat sprout.
