Alternaria mycotoxins and its incidence in fruits and vegetables

  • Alternaria is a ubiquitous fungal genus, widely distributed in the environment and a range of different habitats. It includes both plant pathogenic and saprophytic species, which can affect crops in the field or cause post-harvest spoilage of plant fruits and kernels. Numerous Alternaria species cause damage to agricultural products including cereal grains, fruits and vegetables, and are responsible for severe economic losses worldwide. Most Alternaria species have the ability to produce a variety of secondary metabolites, which may play important roles in plant pathology as well as food quality and safety. Alternariol (AOH), alternariol monomethyl ether (AME), tenuazonic acid (TeA), tentoxin (TEN) and altenuene (ALT) are considered the main Alternaria compounds thought to pose a risk to human health. However, food-borne Alternaria species are able to produce many additional metabolites, whose toxicity has been tested incompletely or not tested at all. Both alternariols are mutagenic and their presence in cereal grain has been associated with high levels of human esophageal cancer in China. TeA exerts cytotoxic and phytotoxic properties, and is acutely toxic in different animal species, causing hemorrhages in several organs. The possible involvement of TA in the etiology of onyalai, a human hematological disorder occurring in Africa, has been suggested. Altertoxins (ALXs) have been found to be more potent mutagens and acutely toxic to mice than AOH and AME. Other metabolites, such as TEN, are reported to be phytotoxins, and their toxicity on animals has not been demonstrated up to now. Vegetable foods infected by Alternaria rot are obviously not suitable for consumption. Thus, whole fresh fruits are not believed to contribute significantly with Alternaria toxins to human exposure. However, processed vegetable products may introduce considerable amounts of these toxins to the human diet if decayed or moldy fruit is not removed before processing. The taxonomy of the genus is not well defined yet, which makes it difficult to establish an accurate relationship between the contaminant species and their associated mycotoxins. Great efforts have been made to organize taxa into subgeneric taxonomic levels, especially for the small-spored, food associated species, which are closely related and constitute the most relevant food pathogens from this genus. Several crops of agricultural value are susceptible to infection by different Alternaria species and can contribute to the entry of Alternaria mycotoxins in the food chain. The distribution of Alternaria species was studied in different commodities grown in Argentina. These food populations were characterized through a polyphasic approach, with special interest in their secondary metabolite profiles, to understand their full chemical potential. Alternaria species associated with tomato, bell pepper, blueberry, apples and wheat cultivated in Argentina showed a surprisingly high metabolomic and mycotoxigenic potential. The natural occurrence of Alternaria toxins in these foods was also investigated. The results here presented will provide background for discussion on regulations for Alternaria toxins in foods.

Sterilization effect of electrolyzed water and chlorine dioxide on Rubus coreanus Miquel (전기분해수와 이산화염소수 처리 복분자(Rubus coreanus Miquel)의 살균효과)

  • This study was carried out in order to investigate sterilization effect and to extend storage periods of the Rubus coreanus by treating with tap water (TW), electrolyzed water (EW) and aqueous chlorine dioxide ($ClO_2$). After each treatment plot was soaked with 10, 50, 100, 200 ppm in each sterilizing solution within 30 sec, each treatment was compared during the storage time at room temperature and refrigerator temperature. As results of total plate count according to temperatures and periods, the microbial sterilizing power of each treatment plot was bigger at EW and $ClO_2$ treatment plots than the TW treatment plot; however, it sharply increased on the high concentration $ClO_2$ treatment plot. Futhermore, the cold storage treatment plot had more outstanding microbial sterilizing power than the room temperature treatment plot. As a result of observing the surface of the Rubus coreanus using scanning electron microscope (SEM), no microbe was seen in EW and $ClO_2$ treatment plot. The results of measuring enzyme activity showed a more significant decrease in EW and $ClO_2$ solutions treatment plot than TW treatment plot but gradually increased with time. The contents of total polyphenol revealed similar values on each treatment. The EW and $ClO_2$ treatment of the Rubus coreanus could be considered as good methods for inhibiting microbial growth in fresh vegetables and fruit, thereby contributing to quality maintenance.

Density and Viability of Sclerotia of Rice Sheath Blight Pathogen Overwintering in Field (벼잎집무늬마름병균(病菌) 월동균핵(越冬菌核)의 밀도(密度)와 활성(活性))

  • Three post-harvest fields each in four rice growing areas, Iri, Naju, Jinju and Taegu were randomly selected and surveyed during December 1986 to examine sclerotial density of Rhizoctonia solani overwintering in the field. Surface soil of $0.09m^2$ area was sampled in each field with three replications and sieved to collect sclerotia. Germiability and pathogenicity of collected sclerotia were examined in the laboratory. Number of sclerotia $({\times}10^6)/ha$ in Iri, Naju, Jinju, and Taegu was estimated from the sample as 2.7, 1.2, 0.7 and 0.6, respectively. Based on sample variance with simple random sampling in each area, number of sampling required for estimating average sclerotial density with the precision of 10% apart from a chance of 1 in 20 was calculated to 41, 132, 232, and 395 for Iri, Naju, Jinju and Taegu, respectively. Percentage of germination of sampled sclerotia on potato sucrose agar (PSA) ranged from 42 to 78% depending on the area, and averaged 60%. About 49% of the germinated sclerotia were pathogenic to a rice cultivar Jinheung that was used to test pathogenicity of the sclerotia. Proportion of viable sclerotia that have both germiability and pathogenicity was thus estimated to 0.29 of total sclerotia collected. R. solani cultures obtained from the sclerotia could be distinguished into three groups based on colony morphology on PSA. Size and number of sclerotia formed on PSA differed between group but were not associated with pathogenicity to Jinheung.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.

Optimization of Supercritical Fluid Extraction of Tocotrienol from Grape Seed (초임계유체 추출을 이용한 포도씨 tocotrienol 추출조건 최적화)

  • In this study, supercritical carbon dioxide extraction (SFE) was utilized for the extraction of tocotrienol from grape seeds. The optimal conditions for vitamin E and tocotrienol extraction were determined via response surface methodology (RSM). Central composite design was utilized to assess the effects of oven temperature (30-$50^{\circ}C$, X1), operating pressure (17-25 MPa, X2), and extraction time (1-5 hr, X3) of supercritical fluid extraction. Vitamin E and tocotrienol contents were 8.65 mg/100 g and 7.88 mg/100 g at $40^{\circ}C$, 20MPa and 5 hr, respectively. The predicted extraction condition was validated via actual experimentation. The predicted extraction conditions were $40^{\circ}C$, 3.8 hr, and 20.7MPa. The vitamin E and tocotrienol contents under these conditions were 8.20 mg/100 g and 7.42 mg/100 g, respectively. The vitamin E and tocotrienol contents of solvent extraction with hexane were 8.18 mg/100 g and 7.24 mg/100 g, respectively.

High-Level Expression of T4 Endonuclease V in Insect Cells as Biologically Active Form

  • T4 endonuclease V (T4 endo V) [EC 3. 1. 25. 1], found in bacteriophage T4, is responsible for excision repair of damaged DNA. The enzyme possesses two activities: a cyclobutane pyrimidine dimer DNA glycosylase (CPD glycosylase) and an apyrimidic/apurinic endonuclease (AP lyase). T4 denV (414 bp cDNA) encoding T4 en do V (138 amino acid) was synthesized and expressed using either an expression vector, pTriEx-4, in E. coli or a baculovirus AcNPV vector, pBacPAK8, in insect cells. The recombinant His-Tag/T4 endo V (rHis-Tag/T4 endo V) protein expressed from bacteria was purified using one-step affinity chromatography with a HiTrap Chelating HP column and used to make rabbit anti-His-Tag/T4 endo V polyclonal antibody for detection of recombinant T4 endo V (rT4 endo V) expressed in insect cells. In the meantime, the recombinant baculovirus was obtained by cotransfection of BacPAK6 viral DNA and pBP/T4 endo V in Spodoptera frugiperda (Sf21) insect cells, and used to infect Sf21 cells to overexpress T4 endo V protein. The level of rT4 endo V protein expressed in Sf21 cells was optimized by varying the virus titers and time course of infection. The optimal expression condition was set as follows; infection of the cells at a MOI of 10 and harvest at 96 h post-infection. Under these conditions, we estimated the amount of rT4 endo V produced in the baculovirus expression vector system to be 125 mg/l. The rT4 endo V was purified to homogeneity by a rapid procedure, consisting of ion-exchange, affinity, and reversed phase chromatographies, based on FPLC. The rT4 endo V positively reacted to an antiserum made against rHis-Tag/T4 endo V and showed a residual nicking activity against CPD-containing DNA caused by UV. This is the first report to have T4 endo V expressed in an insect system to exclude the toxic effect of a bacterial expression system, retaining enzymatic activity.

Survival of Bacillus cereus and Its Transfer from Agricultural Product-Contact Surfaces to Lettuce (Bacillus cereus의 농산물 접촉 표면 재질별 생존력 및 상추로의 교차오염도 조사)

  • This study was conducted to investigate survival of Bacillus cereus (B. cereus) on stainless steel and polyvinyl chloride (PVC) and its transfer from two material to lettuce. The stainless steel and PVC were innoculated with B. cereus and stored at 6 combination conditions (temperature : $20^{\circ}C$ and $30^{\circ}C$, relative humidity (RH) : 43%, 69%, and 100%). Although the total numbers of B. cereus at RH 43% and RH 69% were reduced by 3.53-4.00 log CFU/coupon within 24 h regardless of material type, the spore numbers of B. cereus was lasted at 3.0 log CFU/coupon. When two materials were stored at $30^{\circ}C$, RH 100%, the spore numbers of B. cereus was rapidly increased by 3.0 log CFU/coupon. In addition, the reduction rate of B. cereus was decreased in the presence of organic matter. Transfer rate of B. cereus from surface of stainless steel and PVC to lettuce was increased by 10 times in the presence of water on the lettuce surface. As a result of this study, the presence of B. cereus on produce contact surfaces can increase the risk of cross-contamination. Thus, it is important that the packing table and conveyer belt in post harvest facility should be properly washed and sanitized after working to prevent cross-contamination.

Technology of Good Quality Seed Production in Snap-bean (Phaseolus vulgaris L.) (협채용 강낭콩의 채종기술 확립)

  • Pod-edible bean or snap bean is a fairly new crop to domestic farmers but the national demand is steadily increasing in recent years along with the development of western food business and change in dietary patterns. At the same time, much efforts are being made to export it to foreign country, mainly to Japan. The amount of seeds introduced from outside is also continuously increasing along with the enlargement of area planted for the crop. Hybridization breeding for the crop has already been started to supply the cheaper and better seeds which will reduce the seed costs and foster the higher income to the farmers. In this experiment, several technologies related with the production of quality seeds are preliminary investigated. Some of the results obtained are summarized as follows; 1. Highly significant interaction was recognized between planting dates and no. of pods per plant and no. of branches but no interaction between planting dates and plant height and no. of nodes on main stem. Days to maturity was proportionally reduced to later planting dates. 2. Rate of viviparous pods and seeds was gradually increased in later planting dates but rate of germination was increased in earlier planting dates with lower germination rate in white seed coat grains than in colored seed ones. 3. Seed yield was higher in the earlier planting dates with a great deal of varietal difference. Early to mid April was considered to he the optimum planting dates for snap bean in Kyungbuk area. High correlation was recognized between seed yield and no. of pods per plant, no. of seeds per plant, and 100 seed weight. 4. Days to flowering was three and seven days longer in Cheongsong, high mountainous area than in Kunwi, somewhat prairie lowland. One hundred seed weight was also higher in Cheongsong than in Kunwi. Rate of viviparous grains, pods, and decayed seeds was higher in Cheongsong but, at the same time, the rate of germination and seed yield was also higher in Cheongsong. 5. One hundred seed weight of KLG5007 increased continuously up to 35days after flowering and decreased thereafter but that of KLG50027 increased to 40days after flowering and slowly reduced thereafter. The content of crude oil reached to maximum at 40 days after flowering and reduced thereafter. The rate of germination in Gangnangkong 1 was the highest, 89.3%, at 35 days after flowering and reduced thereafter while that in KLG50027 reached to maximum, 70.7%. at 40days after flowering and reduced thereafter. Thus, the optimum harvesting time for snap bean was considered to be 35~40days after flowering. 6. The snap bean pods at yellow bean stage easily became viviparous ones under saturated moisture conditions for 24 hours at $25{\sim}30^{\circ}C$. Therefore, it is recommended to harvest pods somewhat earlier than yellow-bean stage and let them do post maturing, especially when it is to be rained.

Qualitative Changes in Precooling and Storage Temperatures on the Post-harvest Management of the Fruits in Strawberry for Export (수출딸기의 수확 후 예냉 및 저장온도에 따른 품질변화)

  • A study was conducted to examine the effect of precooling and storage temperatures on the storage life of 'Maehyang' strawberry for export. Fruits at a 80% ripe stage were harvested from a commercial greenhouse in Gyeongsangnamdo Jinju on January 11, 2011. Fruits were precooled at 0, 2, $4^{\circ}C$ for 2-5 hours, and then transported for about 30 minutes. And stored in set chamber at 4, 8, $10^{\circ}C$, immediately. Fruits were examined for their changes in weight, hardness, Hunter color values, soluble solid content (SSC), acidity, and incidence of gray mold (Botrytis cinerea) during storage. The fresh weight decreased during storage at all temperatures. Especially, strawberry treated the lowest precooling and storage temperatures was the greatest in quality. The hardness was drastically decreased in precooled at $4^{\circ}C$ and stored at $10^{\circ}C$ after storage 10 days. SSC was the greatest when strawberry stored at the lowest temperature. Acidity increased during storage at all temperatures. But in the last day after storage, the strawberry of precooled at $4^{\circ}C$ and stored at $10^{\circ}C$ was the highest as 0.7%. Gray mold was the greatest at $10^{\circ}C$ storage temperature. The results indicate that effectiveness for keeping the freshness was best achieved by precooling at $0^{\circ}C$ and storage at $4^{\circ}C$.

Assessment of the Effects of Interactions between Climatic Conditions and Genetic Characteristics on the Agronomic Traits of Soybeans Grown in Six Different Experimental Fields

  • Soybean [Glycine max (L.) Merr.] is a species of legume native to East Asia. The interactions between climatic conditions and genetic characteristics are known to affect the agricultural performance of soybean. Therefore, the present investigation was conducted to identify the main elements affecting the agricultural performances of 11 soybean varieties/lines from China [Harbin ($45^{\circ}12^{\prime}N$), Yanji ($42^{\circ}53^{\prime}N$), Dalian ($39^{\circ}30^{\prime}N$), Qingdao ($36^{\circ}26^{\prime}N$)] and the Republic of Korea [Suwon ($37^{\circ}16^{\prime}N$), and Jeonju ($35^{\circ}49^{\prime}N$)]. The days to flowering (DTF) of soybeans with the e1-nf and e1-as alleles and the E1e2e3e4 genotype, except in 'Keumgangkong', 'Tawonkong', and 'Duyoukong', were relatively short compared to those of soybeans with other alleles. Although DTF of the soybeans was highly correlated with all climatic conditions [negative: precipitation, average temperature (AVT), accumulated temperature; positive: day-length (DL)], days to maturity and 100-seed weight of the soybeans showed no significant correlation with any climatic conditions. The soybeans with a dominant Dt1 allele, except 'Tawonkong', had the longest stem length (STL). Moreover, STL of the soybeans grown in the test fields showed a positive correlation with only DL; however, the results of our chamber test that was conducted to complement the field tests showed that STL of soybean was positively affected by AVT and DL. Although soybean yield (YLD) showed positive correlations with latitude and DL (except L62-667, OT89-5, and OT89-6), the response of YLD to the climatic conditions was cultivar-specific. Our results show that DTF and STL of soybeans grown in six different latitudes are highly affected by DL, and AVT and genetic characteristic also affect DTF and STL.
