Effect of Working on Physiological, Biochemical and Haematological Parameters in Hariana Bullocks

  • An experiment was conducted on eight bullocks of Hariana cattle study draft efficiency using different appliances, changes in their physiological, biochemical and haematological parameters, and how the body tries to maintain these within normal physiological range. Blood collection was done at pre-exercise, three hours after exercise and two hours post exercise in summer and winter seasons of the year 1990-91. Average pulse rate and respiration rate per minute were found to be $63.09{\pm}0.78,\;25.60{\pm}0.54,\;97.21{\pm}2.51,\;63.37{\pm}2.70,\;64.05{\pm}1.90,\;25.33{\pm}1.20$, respectively, at pre-exercise, three hours exercise and two hours post exercise rest. The seasonal differences were found to significant and the values were higher in summer than in winter. After two hours post exercise rest Hariana bullocks regained their normal values for all the parameters, except that pulse rate, respiration rate and serum sodium during summer did not reach the pre - exercise levels.

  • The present paper analyzes performance of a variant of rate-of-return regulation called the ex-post adjustment regulation put in effect in the Korean petroleum refinery sector. Unlike the traditional rate-of-return regulation on a monopoly, the regulation is first for the oligopolistic industry as a whole and second of the ex post nature. Under the regulation, at the end of each year, each firm is responsible to pay a certain portion of the excess of the total realized profits in the industry over the allowed profits. It is shown that if the excess profits are completely collected(including the interests), the social optimum can be realized. When only a portion of the excess profits can be collected, the regulation generally increases consumer surplus by making the firms more competitive. Each individual firm's production under the regulation depends on whether the firm's output is regarded as a strategic substitute or complement of other firms'output.

Influence of Temperature on Microsporidian Multiplication and Spore Production in Various Tissues of Silkworm (Bombyx mori L.) During Larval Development

  • Multiplication and spore production of three microsporidia viz., Nosema bombycis, Nosema sp. 1 and Nosema sp. 2 in fifth instar larval tissues of silkworm, Bombyx mori L. in two seasons with distinct temperature regimes were studied. Nosema sp. 2 produced significantly (P < 0.01) higher number of spores in various tissues. Among the tissues, spore production was highest in silk gland, followed by fat body and gut. Spore production was significantly (P < 0.01) higher in season-II (Average temperature $29.4{\pm}1.1^{\circ}C$). Maximum spore production was observed 25 days post inoculation (p. i.) in season-I (Average temperature $18.9{\pm}1.1^{\circ}C$), whereas in season-II, it was 14 days p. i. In season-I, spore production was low up to 21 days p. i., then increased sharply. In season-II, there was a steady increase in spore production. The results indicate that the microsporidian multiplication is tissue specific and extremely sensitive to temperature at which the host is reared. It also reveals that, silk gland, fat body and gut are the most appropriate tissues for microscopic identification of microsporidia in the larval stage.

  • Multiplication and spore production of three microsporidia(Nosema bombycis, Nosema sp. 1 and Nosema sp. 2) in selected tissues of pupa and adult of silkworm, Bombyx mori L. were studied in two seasons (SI, SII) with distinct temperature (SI: $20.1{\pm}0.8^{\circ}C$ and SII: $25.1{\pm}0.7^{\circ}C$) regimes. Multiplication of the microsporidia followed a logistic pattern with a lag phase, an exponential phase and a stationary phase. In SII, spore production was significantly (P<0.01) higher in various tissues. Highest spore production was observed 30 days post inoculation (p.i.) in SI and in SII, it was $21{\sim}23$ days p.i. Spore production was significantly (P<0.01) higher in the gut tissues than other tissues. Nosema sp. 2 registered significantly (P<0.01) higher spore production in both the seasons compared to Nosema bombycis and Nosema sp. 1. Results indicate that the multiplication and spore production of microsporidia are tissue specific and extremely sensitive to the temperature at which the host is reared. Through this study, the precise day that the spore numbers of the microsporidia are maximized can be predicted in both pupa and adult in case the infection is initiated in the first instar.

Production, Cryopreservation and Transfer of Bovine Embryos Cultured in Serum-Free Medium

  • In vitro embryo culture techniques provide significant contributions not only for a basic research of fertilization and early embryogenesis, but also for a low cost mass production of bovine embryos for transfer, embryo diagnosis, nuclear cloning and the production of transgenic cows. This presentation introduces newly developed serum-free media (IVD101 and IVMD101) that are effective far high yields of transferable embryos of excellent quality from in vitro-matured and fertilized oocytes. Both serum-free media are superior to a conventional serum-containing medium on the increased rates of blastocyst formation, post-thaw embryo viability, and pregnancy after transfer. Furthermore, reduced risks of calf mortality and large calf syndrome are also observed for the serum-free-derived embryos. Serum-derived embryos contain a large number of lipid droplets and immature mitochondria in their cytoplasm that may account for the lower production of transferable embryos and poor embryo quality.

Ethylene Production of Packaged Apples under Vibration Stress in Simulated Transportation Environment

  • Fruits are subjected to a variety of vibration stress during the transportation from a production area to markets. Vibration inputs are transmitted from the transporting vehicle through the packaged fruit. And the steady state vibration input may cause serious internal damage of fruit. Product quality of fruits declines by various factors while they are stored right after harvesting and among the substance in charge of post ripening action, ethylene ($C_2H_4$) biosynthesis increases fruits' respiration process after harvesting and decreases storage expectancy. Ethylene production of apples rapidly increases while storage duration becomes longer. This tendency is much clearer for the apples with vibration stress at input acceleration level. When there was no vibration stress, change in ethylene production level of apples are not very large during storage. Ethylene production rates inside the gas collecting container increased significantly ($p{\leq}0.05$) after 24 hours storage, particularly for apples with vibration stress ($0.7{\mu}l/kg{\cdot}hr$ (1st stack), $0.78{\mu}l/kg{\cdot}hr$ (2nd stack), $0.96{\mu}l/kg{\cdot}hr$ (3rd stack)); whereas less ethylene was produced in control apples ($0.18{\mu}l/kg{\cdot}hr$ during storage. Also ethylene production rates of apples according to the stack position were significantly different ($p{\leq}0.05$). The vibration stress clearly accelerated the degradation of apple quality during storage, resulting in increased ethylene production.

Raw 264.7 세포에서 유해산소 생성에 미치는 Caffeic Acid의 영향 (Effect of Caffeic Acid on the Production of Reactive Oxygen Species in Raw 264.7 Cells)

  • To investigate effect of caffeic acid on the intracellular reactive oxygen species production, we used DHE for intracellular superoxide anion production, DCF for intracellular ${H_2}{O_2}$ production and DHR for intracellular hydroperoxide production in Raw 264.7 cells. DPPH assay showed that antioxidant activity of caffeic acid with 39.5 ${\mu}M$ of ${IC}_{50}$ values was similar to that of ascorbic acid with 41.3 ${\mu}M$ of ${IC}_{50}$ values. Caffeic acid dose-dependently inhibited silica-induced ${H_2}{O_2}$ and hydroperoxide production but did not affect superoxide anion production in Raw 264.7 cells, which suggest that antioxidant effect of caffeic acid acts on the post-step of superoxide anion. On the other hand, caffeic acid showed a potent antioxidant effect in $lCuSO_4$-induced lipid peroxidation. Furthermore, plasma superoxide dismutase activity (3.43${\pm}$0.23 U/ml) in 10 mg/kg caffeic acid-fed mice was significantly higher than that (2.32${\pm}$0.24 U/ml) of control. From the above results, it is referred that caffeic acid appears to have potent anti-oxidant activity in both cell system and in vivo system.

A COMPARATIVE STUDY ON THE PERFORMANCES OF F1 CROSSBRED COWS UNDER RURAL CONDITIONS

  • Data were collected on productive and reproductive performance of different $F_1$ crossbred cows (Deshi cows joined by A.I. to Sindhi, Sahiwal, Jersey or Holstein-Friesian Bull semen) reared by farmers in and around the Bangladesh Agricultural University campus at Mymensingh. The traits considered were birth weight, age at first heat, age at first calving, gestation length, post-partum heat period, milk yield, lactation length, dry period and calving interval. Data were analyzed statistically using a completely randomized design and Duncan's Multiple Range Test to compare and determine the significance of between individual group means. It was observed that the Holstein $\times$ Deshi had the highest birth weight (21.4 kg) followed by Sahiwal (17.6 kg), Jersey (16.7 kg) and Sindhi (16.1 kg). Shortest age at first heat (854 and 920 days) and age at first calving (1002 and 1201 days) were found for the Jersey $\times$ Deshi and Holstein $\times$ Deshi crosses respectively. The post-partum heat period were significantly different between breeds with Jersey and Holstein sired calves having the shortest intervals. There was no difference in the gestation length of the four crosses. Holstein $\times$ Deshi and Jersey $\times$ Deshi cows gave the highest milk yields due to longer lactation periods and higher daily milk production. Similarly, Holstein $\times$ Deshi and Jersey $\times$ Deshi crosses had significantly shorter dry periods and Jersey $\times$ Deshi had a shorter calving interval. From this study it may be concluded that exotic genotypes such as Holstein-Friesian and Jersey can perform very well under rural conditions in Bangladesh.

EFFECT OF FORCE MOLTING INDUCED CONVENTIONALLY OR BY HIGH DIETARY ALUMINUM ON EGG AND SHELL QUALITY OF LAYING HENS

  • Eggs used in this study were obtained from Saudi Arabian Baladi laying hens which were divided into four experimental groups and subjected to the following treatments: Commercial laying ration (17% CP, 3.6% Ca and 0.343%, available P) fed ad libitum as a control (C); Conventional force molting, feed removal for 10 days followed by 18 days full-feed of cracked corn (F); 15 days ad libitum intake of the control ration supplemented, to initiate forced-molt, with 0.35% aluminum as the sulfate (ALS) or the chloride (ALC). The hens were in production for 52 weeks and 17 months of age at the start of the trial and the post-treatment period lasted 36 weeks. During the treatment period F and AL treated groups had similar egg and shell weight, egg surface area, shell thickness and shell weight per unit of surface area but significantly (p<0.05) lower than the control. F had significantly (p<0.05) the highest and the control the lowest Haugh unit values whereas AL fed groups had significantly (p<0.05) lower meat spot incidence compared with the control which tended to have higher value than F group. ALC and F had significantly (p<0.05) the lowest yolk color grade whereas ALC had significantly (p<0.05) lower egg index than ALS and the control. During the post-treatment period the control had significantly (p<0.05) the highest egg index and blood spots incidence and ALS the lowest shell diensity compared with other groups. ALS had significantly (p<0.05) lower shell weight than ALC and the control whereas F and AL treated hens had significantly (p<0.05) the highest Haugh unit values and yolk color grades respectively. F had significantly (p<0.05) lower meat spots incidence than ALC and the control. The same results were observed for ALS compared with the control.

한국 전통 미생물발효차(청태전)의 미생물 군집분석 (The microbial diversity analysis of the Korea traditional post-fermented tea (Chungtaejeon))

  • 차는 세계적으로 인기있는 음료로 불발효차(녹차), 반발효차(우롱차), 완전발효차(홍차)와 흑차(미생물발효차 or 후발효차)를 포함하고 구분된다. 미생물발효차는 차나무(Camellia sinensis)의 잎을 미생물 발효과정을 통해 제조된다. 삼국시대부터 전해 내려온 청태전은 한국 남해안지역에서 제조되며 돈차 또는 떡차로 불리는 독창적인 한국의 미생물발효차이다. 본 연구에서는 청태전에 우점하는 미생물군집구조 분석을 위해 16S rRNA 유전자를 이용하였다. 청태전에 우점하는 미생물은 ${\gamma}$-proteobacteria에 속하는 Pantoea sp.와 Klebsiella oxytoca가 우점하였다. 미생물 군집크기 분석을 통해 청태전의 미생물 군집크기가 다른 미생물발효차와 비교해 가장 큰 것을 확인하였다.
