Regulatory Mechanism of Radiation-induced Cancer Cell Death by the Change of Cell Cycle (세포주기 변화에 타른 방사선 유도 암세포 사망의 조절기전)

  • Purpose : In our Previous study, we have shown the main cel1 death pattern Induced by irradiation or protein tyrosine kinase (PTK) inhibitors in K562 human myeiogenous leukemic cell line. Death of the cells treated with irradiation alone was characterized by mitotic catastrophe and typical radiation-induced apoptosis was accelerated by herblmycin A (HMA). Both types of cell death were inhibited by genistein. In this study, we investigated the effects of HMA and genistein on cell cycle regulation and its correlation with the alterations of radiation-induced cell death. Materials and Methods: K562 cells In exponential growth phase were used for this study. The cells were Irradiated with 10 Gy using 6 MeV Linac (200-300 cGy/min). Immediately after irradiation, cells were treated with 250 nM of HMA or 25 $\mu$N of genistein. The distributions of cell cycle, the expressions of cell cycle-related protein, the activities of cyclin-dependent kinase, and the yield of senescence and differentiation were analyzed. Results: X-irradiated cells were arrested In the G2 phase of the cell cycle but unlike the p53-positive cells, they were not able to sustain the cell cycle arrest. An accumulation of cells in G2 phase of first ceil-cycle post-treatment and an increase of cyclin Bl were correlated with spontaneous, premature, chromosome condensation and mitotic catastrophe. HMA induced rapid G2 checkpoint abrogation and concomitant p53-independent Gl accumulation. HMA-induced cell cycle modifications correlated with the increase of CDK2 kinase activity, the decrease of the expressions of cyclins I and A and of CDK2 kinase activity, and the enhancement of radiation-induced apoptosis. Genistein maintained cells that were arrested in the G2-phase, decreased the expressions of cyclin Bl and cdc25c and cdc25C kinase activity, increased the expression of pl6, and sustained senescence and megakaryocytic differentiation. Conclusion: The effects of HMA and genistein on the radiation-induced cell death of KS62 cells were closely related to the cell cycle regulatory activities. In this study, we present a unique and reproducible model in which for investigating the mechanisms of various, radiation-induced, cancer cell death patterns. Further evaluation by using this model will provide a potent target for a new strategy of radiotherapy.

Physicochemical characteristics and antioxidant activity of pear vinegars using 'Wonhwang', 'Niitaka' and 'Chuhwangbae' fruits (원황, 신고 및 추황배를 이용한 배 식초의 이화학적 특성 및 항산화활성)

  • This study was to prepared vinegars using three kinds of pear cultivar with different maturities, 'Wonhwang', 'Niitaka', and 'Chuhwangbae', and investigate their physicochemical characteristics and antioxidant activities. Result showed that the firmness of 'Wonhwang' was much lower than other pear cultivars. No significant difference in soluble solid content was found among three pear cultivars. The total acidity of 'Chuhwangbae' pear vinegar was 4.89~7.98%, which was higher than that those of 'Wonhwang' and 'Niitaka' vinegars. The color of 'Niitaka' vinegar had a lower lightness and redness values, but a higher yellowness value, compared with the other two vinegars. The free sugar contents were mainly composed of fructose, glucose, and sorbitol, with the greatest content of sorbitol in all three peer vinegars. The total phenolics and flavonoids contents were 35.2~55.3 and 8.4~14.4 mg/100 mL, respectively. Antioxidant activity had a positive correlation with total phenolic contents. The DPPH free radical scavenging and $ABTS^+$ radical scavenging activity were 45.0~62.1% and 73.8~78.2%, respectively. From these results, we confirmed that immature pear cultivars such as 'Wonhwang', 'Niitaka' and 'Chuhwangbae' could be used as a raw materials to prepare vinegar regardless of maturity.

Effective correlation between coagulation efficiency and the sludge settling characteristic (슬러지 응집효율이 침강특성에 미치는 상관관계에 대한 연구)

  • In these days, the importance of sludge treatment is emerging due to the London Convention, so this study was conducted to propose the alternatives for the improved sludge treatment on the organic wastewater and sewage sludge with JAR test and settling column equipped with stirrer. The minimum coagulant dosage to earn the optimum sludge settling efficiency resulted from 200mg/l and each critical sludge settling interface showed no distinct difference when PAC was dosed over 200mg/l. Accordingly, Clarification Rate(CR) with 200mg/l dosage was calculated to CR=(Ho-Ht) / Ho=1-0.4=0.6 because the critical sludge settling height stopped at 0.4. The settling velocity of sludge interface was decreased with the increase of MLSS concentration but rather increased with MLSS concentration over 1,000mg/l. This resulted from positive effect of interacted coagulation for floc formation by transfer to the zone of compressed settling when MLSS concentration increased over 1,000mg/l. The settling velocity of sludge interface showed $28.66{\times}10^{-3}/min$ for average settling velocity of sewage sludge which is 6.7 times higher than $4.25{\times}10^{-3}/min$ for average settling velocity of organic wastewater sludge. The increasing rate of CR for organic wastewater activated sludge was higher than that of settling velocity under 200mg/l of PAC dosage but settling velocity was higher than CR over 200mg/l of PAC dosage. However, in case of sewage sludge, the differential rate of CR was low when PAC dosage was increased but the settling velocity was suddenly increased with over 200mg/l dosage. Therefore coagulation effect was more efficient to MLSS settling velocity rather than SS removal effect in the supernatant.

Influences of Planting Density on Growth and Yield of Perilla frutescens BRITTON var. acuta KUDO (자소(紫蘇)의 재식밀도(栽植密度)가 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • In order to determine the optimum planting density of Perilla frutescens which is suitable for the southern part of Korea, agronomic characters, yield of fresh stem, leaf and seed, and yield components were investigated grown in 1985 and 1986 at Mokpo Branch Station of Crop Experiment Station. The heading date was Aug. $14{\sim}15$ and the blooming period was Aug. $19{\sim}21$ regardless of the difference of plant density, but the longest stem length was 135cm in the plant density of $70{\time}40cm$ and the stem length in the plant density of $80{\time}40cm$ was 134cm. The quantity of fresh weight of stem and leaf and fresh weight of seed was high as 531kg/l0a in the plant density of $80{\time}40cm$. The positive correlation such as $0.7315^*,\;0.9024^{**}\;and\;0.7425^*$ were found between stem length and fresh weight of stem and leaf, stem length and fresh weight of seed, fresh weight of stem and leaf and fresh weight of seed and so high significance was recongnized. In the verification of significance of row spacing, the disperse of the stem length was $55.67^{**}$, that of fresh weight of stem and leaf is $268.50^*$, theat of fresh weight of seed was $16.00^{**}$ and high significance was recognized. In the verification of significance of intrarow spacing, the diperse of stem length was $54.21^{**}$, that of fresh weight of stem and leaf was $2,582.00^{**}$, that of fresh weight of seed was $48.00^{**}$ and then high significance was recognized. Accordingly, the proper plant density of perilla frutescens was 80cm of row $spacing{\time}40cm$ of intrarow spacing.

Drumming bioacoustics of woodpeckers in South Korea (한국에 서식하는 딱다구리목 드러밍의 생물음향학적 특성 연구)

  • In this study, we conducted an analysis of the drumming bioacoustics of three woodpeckers typical to South Korea. The targeted species were, from largest to smallest in size, the Black Woodpecker(Dryocopus martius), the Great Spotted Woodpecker(Dendrocopos major) and the Japanese Pygmy Woodpecker(Dendrocopos Kizuki). The drumming sounds of these three species of woodpeckers were recorded and analyzed. Sound recordings were taken in the Guryongsa Valley, Chiaksan National Park and on the campus of Sangji University, Wonju City, South Korea. Drumming recordings were obtained in the following manner. As researchers walked along trails in the investigation areas, when woodpecker drumming sounds were indicated, positive identification of the species was made using binoculars then the recording of the drumming sound was started. The average time per drumming, in seconds, was Black Woodpecker(D. martius) 1.614 seconds, Great Spotted Woodpecker(D. major) 0.683 seconds and Japanese Pygmy woodpecker(D. Kizuki) 0.200 seconds. The average number of strikes for each drumming was Black Woodpecker(D. martius) 31.2 times, Great Spotted Woodpecker(D. major) 14.9 times and Japanese Pygmy Woodpecker(D. Kizuki) 6.7 times. The strike speed, in strikes/sec, for each species was the Black Woodpecker(D. martius)19.3strikes/sec, the Great Spotted Woodpecker(D. major) 21.8strikes/sec and Japanese Pygmy Woodpecker(D. Kizuki)33.3strikes/sec. The frequency of drumming, in Hertz, was Black Woodpecker(D. martius) 776.9Hz, Great Spotted Woodpecker (D. major) 1,213.8Hz and Japanese Pygmy Woodpecker(D. Kizuki) 826.0Hz. In interpreting this data, Analysis of Variance (ANOVA) was used and it was determined that there was a significant statistical difference between species in drumming duration, time and interval of striking. The findings support that the bigger the biomass of the woodpecker, the longer the drumming duration and striking time. However, the smaller the size of the woodpecker, the faster the strike speed. A correlation between body type size and drumming characteristics was clearly identified. As for strike frequency, measured in Hertz, the medium sized Great Spotted Woodpecker's (D. major) frequency was high whereas the Black Woodpecker(D. martius) and Japanese Pygmy Woodpecker's(D. Kizuki) frequency was similar. A clear trend in reference to body size on this measure does not exist.

Habitat Characteristics of Benthic Macroinvertebrates at a Headwater Stream in the Yeonyeopsan (Mt.) (연엽산 산지계류에 있어서 저서성 대형무척추동물의 서식특성)

  • A total of 24 families, 44 species, and 658 benthic macroinvertebrates were identified, and Ecdyonurus dracon Kluge (13%) was the dominant species in forested streams within the Yeonyeopsan (Mt.). A total of four habit categories (i.e., clingers (56%), burrowers (19%), swimmers (14%), and sprawlers (56%)) were identified, and clingers were the dominant habit at all survey points except point one (UP1). Habitat characteristics were depended on the hydraulic factors (e.g., flow velocity, depth, and substrates), water quality (e.g., DO and water temperature), and the habitat characteristics were differed in the riffle, which has a faster the flow velocity, compared by in the stagnant pool. In other words, in riffles, the clingers dominated in high flow velocity with the large maximum and median grain size for substrates in the habitats regardless of depth, but the burrowers and sprawlers were dominant in low flow velocity with the small maximum and median grain size for substrates in the habitats. Moreover, DO and flow velocity were in positive correlation (y = 0.6666x - 0.659, R2 = 0.0851), and the habitat for burrowers was wider than that for sprawlers or clingers. The water depth was negatively correlated with water temperature (y = -26.397x + 283.87, R2 = 0.1802) since the water temperature is more sensitive to insolation in shallow depth. pH was positively correlated with water temperature. The investigation of the habitat characteristics by separating the relations between pH and DO in upstream and downstream showed the low pH and high DO in the upstream with a high crown density of 68%, regardless of community composition. On the other hand, high pH and low DO in the downstream with a relatively low crown density of 51%. It was considered that the riparian forest played a role in suppressing the growth of attached algae and the controlling water temperature in headwater streams. Our findings identified the habitat characteristics of benthic macroinvertebrates in a headwater stream. We expected that the finding can provide reference data for suggesting conservation and management plans in a headwater stream and increasing academic value.

Damage analysis and Control threshold of Frankliniella occidentalis Pergande (Thysanoptera: Thripidae) on Greenhouse Eggplant and Sweet pepper (시설재배 가지, 피망에서 꽃노랑총채벌레 피해해석과 방제수준)

  • Cage experiments by artificial infestations with different initial densities of Frankliniella occidentalis were conducted to analyze damages and develop control thresholds of F. occidentalis on greenhouse eggplant in 2005 and on greenhouse sweet pepper in 2007. In the eggplant experiment, the infestations of F. occidentalis resulted in direct damage on fruit surface and non-marketable fruits which had several thin or thick lines or bleaching patches on the surface. F. occidentalis adults were frequently found on the flowers of eggplants, while nymphs were mainly observed on leaves. The fruit yield of eggplants was not significantly different among experimental plots with different initial density of F. occidentalis. Relationship between % non-marketable fruits among harvested fruits of eggplant and sticky trap catches of F. occidentalis (no. thrips/trap/week) at two weeks before the harvest showed a positive correlation. Using the estimated relationship, the control threshold of F. occidentalis on greenhouse eggplant was estimated at 10 adults per week at two weeks before the harvest when 5% of non-marketable fruit was applied for the gain threshold. In the experiment of sweet pepper, the direct damage by F. occidentalis was observed on the fruit surface and calyx, and the marketable grade of the damaged fruits decreased. The significant yield loss of marketable fruits was found in plots with high initial introduced-densities. There was a high relationship between thrips density and percentage of damaged fruits. Assuming 5% yield loss (non-marketable fruit) for the gain threshold, the control threshold of F. occidentalis on greenhouse sweet pepper was 4.8 adults per trap and 0.9 individuals per flower at two weeks before harvest.

Studies on the Effects of Several Physico-Chemical Properties of Soils on the Growth, Nodulation and Yield in Soybeans (토양(土壤)의 몇가지 이화학적(理化學的) 성질(性質)이 대두(大豆)의 생육(生育), 근류형성(根瘤形成) 및 수량(收量)에 미치는 영향(影響))

  • Studies were carried out to find the effect of various soil properties on the growth, nodulation and yield using a soybean variety, 'Tongbuktae', at various soils. The results of the study are summarized as follow: 1. The formation ratios of various characters in soybeans were different before and after flowering. The root length(89.3%), stem diameter(82.1%), plant height(77.8%), number of nodes(67.4%), and number of nodes(67.9%) gave the high formation ratio before flowering. However, the formation of number of nodules(66.3%), top dry weight(74.9%) and total dry weight(71.7%)was accomplished mainly after flowering. The ratio of the formation in root dry weight was appeared to be about same in the growth period before and after flowering. 2. Nodulation ratio after flowering shelved significantly positive correlation with dry matter accumulation. 3. T/R, top dry weight/plant height and plant height/root length were significantly associated with yield. The tested soils with higher ratio mentioned above, also showed higher yield. 4. pH, oraganic matter, phosphate, potassium, total nitrogen and molybdenum in the soils were significantly correlated with nodulation and yield. Content of calcium in the soils only showed significant association with yield. 5. Soil properties influenced yield were molybdenum, calcium, organic matter, pH, number of nodules, magnesium, phosphate, total nitrogen and potassium in the order of importance. This order was varied according to the soils employed in these studies.

A Study on the Relationships between the Oral Health Activities and Oral Health Conditions of the Elderly (노인의 구강보건행위와 구강건강상태와의 관련성 조사 연구)

  • This study set out to analyze the relationships between the senior citizens' dental health activities and dental health conditions, to provide basic data to develop a program for their better dental health, and finally to search for the ways to improve their life quality through dental health care. The subjects were 142 senior citizens(58 were male and 84 were female) who were 65 years old or older and visited a dental hospital or clinic in the Seoul metropolitan area from April 17 to April 28, 2006. With the cooperation from the dentists, the investigators examined their dental conditions and conducted one-on-one interviews to collect the information about their demographic characteristics, dental health activities, and subjective dental health conditions. The collected data were analyzed with T-test, ANOVA and Pearson's correlation coefficient using the SPSS WIN 11.5 program. The analysis results were summarized as follows: First, 58 men(40.8%) and 84 women(59.2%) consisted of the subject group, where percentage of the female participants was bigger Those who were aged from 65 to 69 made the biggest age group in the subjects, accounting for 58.5% with 83. Second, those who had three dental caries or less and then didn't treat them or treated them(F) accounted for the biggest percentage with 117(82.4%) and 72(50.7%) respectively. As for the remaining teeth, the biggest number of them(40 subjects, 28.2%) had three or less remaining. Third, it turned out the female subjects had a higher level of dental health activities than their male counterparts(P = 0.00). As for living expenses, those who were paid salary or earned income themselves carried out their dental health activities in a higher level than those who lived on the benefit from the government(p = 0.02). Fourth, the subjects' subjective dental health conditions had negative correlations with their dental caries not treated. And there were positive correlations between their dental caries not treated and dental caries lost and between their dental caries treated and remaining teeth. Fifth, those subjects who earned their living expenses themselves had the most dental caries treated at 5.4(p = 0.02), and there was statistically significant difference with the numbers. Sixth, those subjects who brushed their teeth in their own manner had 11.8 teeth lost(p = 0.05), which was more than the number of those who brushed their teeth in other methods. And there was statistically significant difference among them. The remaining teeth were found most at 17.3(p = 0.00) among those who brushed their upper and lower teeth separately. In addition, those who visited the public health center often had significantly more teeth treated(4.3) than others(p = 0.00).

Evaluation of Food Intake and Diet Quality in High School Students (고등학생의 식품섭취 실태 및 식사의 질 평가)

  • To assess the food intake and diet quality of high school students who live in Chuncheon area, a dietary survey using 24-hour recall method was conducted with 318 subjects. $71.4\%$ of total food intake was in the form of plant foods and the rest on the form of animal food. Diet quality was assessed by food group pattern, dietary diversity score(DDS), and dietary variety score(DVS). When counting the major food groups consumed, $37.1\%$ of subjects had a DDS of 3 and $48.4\%$ of subjects had a DDS of 4. When investigating the consumption pattern of the major five food groups, only $14.5\%$ of subjects consumed foods from all five groups. On average, subjects habitually consumed 26.7 different foods daily, with the mean score of diet variety for males (27.3) being significantly higher than for females (25.8) . Correlation coefficients between nutrient adequacy ratio (NAR) and DVS ranged from r=0.39 for vitamin $B_2$ to r=0.61 for phosphorus. NAR also improved as the number or foods or rna groups consumed increased (p<0.001). Associations between the NAR and high level of DVS was more positive than those between the NAR with DDS. When assessing the dietary quality of subjects using DDS and DVS, many people appeared not to have a desirable food intake. Therefore, to lead them to have nutritionally balanced diet, educating students and their parents on nutrients and suggesting guidelines for a desirable diet is considered to be essential so that they can intake from all of major food group and have various foods in their diet.
