Vesicular-Arbuscular Mycorrhizae in Some Plants (IV) (몇 가지 식물(植物) 중(中)의 Vesicular-Arbuscular Mycorrhizae 에 관(關)하여 (IV))

  • Of the 103 species (41 families) sampled from two limestone bites in Korea, 98 species (95.1%) contained VA mycorrhizae. No mycorrhizal structures were observed in the other 5 species. The nonmycorrhizal species were members of the following 4 families: Equisetaceae, Davalliaceae, Amaranthaceae and Ericacene. Of the 124 species (51 families) sampled from two different nonlimestone sites, 99 species (79.8%) were found to contain VA mycorrhizae and no mycorrhizal structures were observed in the other 25 species. The nonmycorrhizal species were members of the following 16 families: Equisetaceae, Pteridacaea, Davalliaceae, Cyperaceae, Commelinaceae, Polygonaceae, Chenopodiaceae, Phytolaceaceae, Amaranthaceae, Aizoaceae, Portulacaceae, Caryophyllaceae, Fumariaceae, Cruciferae, Ericaceae and Rubiaceae.

The Occurrence of Kranz Type Species Among the Noxious Weeds on Cultivated Land of Taiwan and their Biochemical Subdivision

  • One hundred and one noxious weeds on cultivated land of Taiwan were investigated for the occurrence of "Kranz" leaf anatomy and activities of PEP carboxylase and $C_4$ acid decarboxylating enzymes : NADP-malic enzyme, NAD-malic enzyme, PEP carboxykinase. Based on the leaf anatomy and a/b chlorophyll ratio, twenty-seven species exhibit "Kranz" type leaf anatomy, and seventy-four species were found without it. Among the species investigated, Digitaria radicosa (Presl) Miq., Leptochloa chinensis (L.) Nees, and Sporobolus fertilis (Steud.) W. D. Clayton in the Gramineae were first recorded as $C_4$ plants. Twenty-sven species of "Kranz" type leaf anatomy, include those of monocotyledon ; sixteen species in Gramineae, six species in Cyperaceae. Those of dicotyledon ; two species each in Euphorbiaceae and Amaranthaceae and one species in Portulacaceae. The subtype of fourteen previously uninvestigated species among twenty-seven species were further determined. The properties of the three decarboxylating enzyme from representative species were also characterized.

  • The repeated column chromatographic separation of the EtOH extract of Portulaca oleracea afforded seven compounds. The structures of these isolates were identified as bergapten (1), umbelliferone (2), daidzein (3), genistein (4), protocatechuic acid (5), ferulic acid (6), and gallic acid (7) by the analysis of physico-chemical and spectral data. Their antioxidant effect on free radical scavenging was evaluated in the DPPH assay.

Ecophysiological characteristcs of Plant Taxon-Specific Calcium Metabolism (식물 분류단위 특이적인 칼슘대사의 생리생태학적 특성)

  • In order to compare species-specific calcium metabolism, we collected 127 species belonging to 40 different families grown on various habitats including saline, limestone, wetland during the 1996 vegetation period, and analyzed their inorganic ion contents. Plants investigated were divided into 5 groups according to their physiological properties: 1) Chenopodiaceae, Aizoaceae, Caryophyllaceae, Portulacaceae and Phytolaccaceae of Centrospermales and Polygonaceae (Polygonales had a little water-soluble $Ca^{2+}$ but contained high contents of insoluble $Ca^{2+}$ particularly as Ca-oxalate (Chenopodiaceae type), 2) Some plant species such as Rosaceae produced oxalate in amounts insufficient to precipitate all incoming $Ca^{2+}$ and thus contained a surplus of dissolved $Ca^{2+}$ (Rosaceae type), 3) The contents of water-soluble $Ca^{2+}$ in plant species of Crassulaceae. Plantaginaceae, Asclepiadaceae, and Zygophyllaceae were equal to or greater than those of K ($K/Ca{\leq}1$; Crassulaceae type), and 4) K/Ca ratios of Compositae were significantly fluctuated depending on species and soil $Ca^{2+}$ level of their habitats (Compositae type). 5) Certain monocots (Gramineae, Cyperaceae, Juncaceae), in contrast to the dicotyledonous plant families mentioned above, showed a very distinct type of calcium metabolism, that is, the K/Ca ratios of 8~10 were maintained indifferently in the species and their habitat types (Graminae type). These results plants within the same taxon have similar physiological aspects as weel as morphological attributes. To understand calcium metabolism of certain plant species, therefore, it is desirable to approach on the basis of physiological concept (calciotroph or calciophobe) rather than the ecological one (calcicole or calcifuge).

  • 쇠비름(Portulaca oleracea L.)은 쇠비름과(Portulacaceae)에 속하는 일년생 초본으로 우리나라 각지에 5~9월에 걸쳐 자생하고 있다. 한국보다는 중국에서 식용으로 애용되고 있으며, 해열, 이뇨, 산혈 등의 효능이 있어 임질, 요도염, 유종, 대하증, 임파선염 등에 약용으로 사용하고 있다. 쇠비름의 성분으로는 vitamin $B_1$ 및 C, saponin, tannin, flavonoid, dopamine, noradrenalin 등이 보고되었다. 쇠비름에 관한 연구로는 항산화효과, 항균효과 및 해독효과가 발표되었으나, 고지혈증에 관한 연구는 미흡한 실정이다. 따라서 본 연구에서는 고콜레스테를 유발식이에서 쇠비름 에탄을 추출물이 혈청 중 지질에 미치는 영향에 대해 검토하고자 하였다. 실험동물은 6주령의 Sprague Dawley 흰쥐 수컷을 각 실험군당 10마리씩, AIN-93M을 기본으로 하여 대조군(고지방식이), 쇠비름 에탄올 추출물 투여군의 3군(고지방식이+25mg/kg, 75mg/kg, 125mg/kg)으로 나누어 4주간 실험 식이를 급여하고 사육기간 중 사료 섭취량, 체중증가율, 식이효율을 측정하였다. 실험 종료 후, 혈액을 채취하여 혈액에서의 total lipid, total cholesterol, triglyceride, HDL-cholesterol은 효소법을 이용한 kit(아산제약)을 사용하여 측정하였고, 심혈관계 질환의 위험도에 이용되는 동맥경화지수는(atherogenic index, AI) 계산식을 이용하여 산출하였다. 실험결과, 최종체중, 체중증가율, 식이효율 및 장기무게는 실험군 간의 차이를 나타내지 않았다. 그러나 고지방식이에 쇠비름 에탄올 추출물을 125mg/kg의 농도로 병합 투여했을 때 혈중의 total lipid, total cholesterol 및 triglyceride는 대조군에 비해 유의적으로 감소하였으며, 고지방식이에 의해 증가된 동맥경화지수도 대조군에 비하여 유의적으로 저하되었다. 그리고 고지방식이로 증가된 ALT 및 AST 활성이 쇠비름 에탄올 추출물 투여로 유의적으로 감소하였다. 따라서 쇠비름 에탄올 추출물은 혈청 중 지질 개선 효과와 간 기능 개선효과가 있는 것으로 사료되나, 어떤 성분이 콜레스테롤 저하 효과를 나타내는지 보다 자세한 연구가 요구되어 진다.

Korean Red Ginseng and Portulaca oleracea Extracts Attenuate Lipopolysaccharide-induced Inflammation via Downregulation of Nuclear Factor Kappa-B and the Mitogen-activated Protein Kinase Signaling Pathway in Macrophage Cell Line RAW 264.7

  • Korean red ginseng (Panax ginseng Meyer) is a well-known traditional medicine, with numerous biological functions in the body. Portulaca oleracea (P. ole) belongs to the Portulacaceae family and has bioactive potential as a traditional medicine. This study aimed to determine the anti-inflammatory effects of Korean red ginseng extract (RGE) and P. ole extract on lipopolysaccharide (LPS)-treated RAW 264.7 cells. The combination of RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL) extracts significantly suppressed LPS-induced nitric oxide synthesis. The expression of proinflammatory mediators, including inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), and proinflammatory cytokines, including interleukin-1β, interleukin-6, and tumor necrosis factor-α, were markedly decreased by the combined treatment with RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL). Moreover, iNOS and COX-2 protein expression levels were also significantly reduced in the combined treatment compared to the LPS-stimulated group. In addition, the nuclear translocation of phosphorylated nuclear factor kappa-B was suppressed by the treatment with RGE and P. ole. Moreover, the mitogen-activated protein kinase pathway was also partially inhibited by the combination treatment with RGE and P. ole. Our results demonstrate that the treatment mixture with RGE and P. ole could be used as functional food and therapeutic herbal medicine in various inflammatory diseases.

A Philological Study on The Portulacea (마치현에 대(對)한 문헌적(文獻的) 고찰(考察))

  • Objectives : The aim of this study is to find the therapeutic meaning of the Portulacea in herbal medication. Methods : About the origin, the component, the processing the drug, the properties and tastes of drugs, the meridian tropism, the effects, the treating disease, the contraindication and the method of administration, I have researched thirty literatures to mention the Portulacea in time sequence. Results : 1. The Portulacea belongs to the Portulacaceae herbs and it consists of noradrenaline, potassium sometimes containing small amounts of dopamine, dopa, malic acid, citric acid, glutamine acid, asparagin acid, alanine, cane sugar, grape sugar, fruit sugar. 2. The processing of drug is wash of water clealy, and remove the foreign substance. Then the drug is cutting to use. The method of burnt to ashes is used, too. 3. The properties and tastes of drugs is acid, cold, nontoxic. The meridian tropism is mainly liver and the large intestine meridian. 4. From old times, Portulacea has come into general use to treat eczema, the rose erysipelas, an acne, hemorrhoids, discharge from the womb etc. because it is effective on neutralizing poison, reduce a swelling, a tumor, an abscess and stopping of bleeding 5. Portulacea must be stoped When person have a weak digestive organ becase it is cold herba. And don't take use with Fish and shellfish. 6. Portulacea is useful method to external care. To use the herba, pulverize amount of property and then apply to the the affected part Conclusions : This study showed that the Portulacea is useful herb to treat of skin disease and useful method to external care.

Portulaca oleracea Seed Oil Exerts Cytotoxic Effects on Human Liver Cancer (HepG2) and Human Lung Cancer (A-549) Cell Lines

  • Portulaca oleracea (Family: Portulacaceae), is well known for its anti-inflammatory, antioxidative, anti-bacterial, and anti-tumor activities. However, cytotoxic effects of seed oil of Portulaca oleracea against human liver cancer (HepG2) and human lung cancer (A-549) cell lines have not been studied previously. Therefore, the present study was designed to investigate the cytotoxic effects of Portulaca oleracea seed oil on HepG2 and A-549 cell lines. Both cell lines were exposed to various concentrations of Portulaca oleracea seed oil for 24h. After the exposure, percentage cell viability was studied by (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) (MTT), neutral red uptake (NRU) assays, and cellular morphology by phase contrast inverted microscopy. The results showed a concentration-dependent significant reduction in the percentage cell viability and an alteration in the cellular morphology of HepG2 and A-549 cells. The percentage cell viability was recorded as 73%, 63%, and 54% by MTT assay and 76%, 61%, and 50% by NRU assay at 250, 500, and $1000{\mu}g/ml$, respectively in HepG2 cells. Percentage cell viability was recorded as 82%, 72%, and 64% by MTT assay and 83%, 68%, and 56% by NRU assay at 250, 500, and $1000{\mu}g/ml$, respectively in A-549 cells. The 100 $100{\mu}g/ml$ and lower concentrations were found to be non cytotoxic to A-549 cells, whereas decrease of 14% and 12% were recorded by MTT and NRU assay, respectively in HepG2 cells. Both HepG2 and A-549 cell lines exposed to 250, 500, and $1000{\mu}g/ml$ of Portulaca oleracea seed oil lost their normal morphology, cell adhesion capacity, become rounded, and appeared smaller in size. The data from this study showed that exposure to seed oil of Portulaca oleracea resulted in significant cytotoxicity and inhibition of growth of the human liver cancer (HepG2) and human lung cancer (A-549) cell lines.

Antioxidant and anti-inflammatory activities of water extracts and ethanol extracts from Portulaca oleracea L. (쇠비름 물, 에탄올 추출물의 항산화 및 항염증 활성)

  • Portulaca oleracea L., a species of Portulacaceae, is ubiquitous. It is a well-known traditional Chinese medicine for removing heat, counteracting toxicity, cooling blood, and maintaining hemostasia; it is also used as antidysentery agent. This study investigated the anti-oxidative and anti-inflammatory activities of water and ethanol extracts from P. oleracea. The total polyphenol content ($21.08{\pm}0.03mg\;GAE/g$) and total flavonoid content ($5.45{\pm}0.76mg\;QE/g$) of the ethanolic extracts were higher than those of the water extracts. The antioxidative activities were determined by evaluating the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activity and by the ferric reducing antioxidant potential (FRAP) assay. The ABTS radical scavenging activity of the water extract (75.53%) was higher in those of the water extract (67.03%) at concentration of $1,000{\mu}g/mL$. The DPPH radical scavenging activity and FRAP of the ethanol extract were higher than those of the water extract. We also investigated the anti-inflammatory activity of the P. oleracea extracts in LPS-stimulated Raw 264.7 cells. The production levels of nitric oxide (NO) and reactive oxygen species (ROS) significantly decreased with an increasing concentration of the extract. The expression levels of pro-inflammatory cytokines (tumor necrosis faction (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, and IL-6) were significantly lower in the ethanol extract than in the LPS alone treatment group. Based on these results, ethanolic extract from P. oleracea could be an effective antioxidant and anti-inflammatory agent.

Bioactive effects of a Herbal Formula KDC16-2 Consisting Portulaca oleracea L. Extracts (마치현 추출물 함유 제제 KDC16-2의 생리 활성 효과)

  • Portulaca oleracea L. (PL) has been used in traditional medicine herb for treatment of various diseases, such as diarrhea, dysentery, and skin inflammation. Previous studies have shown that the PL regulates the inflammation by inhibition of pro-inflammatory cytokines. Although PL might have improvement effects of intestinal function and bioactive effects, there are not enough studies to demonstrate. This study investigated the effects of KDC16-2 on the improvement of intestinal function and anti-inflammatory effects in vivo and in vitro. The improvement effect of intestinal function was measured fecal amount, water content and intestinal transit rate in KDC16-2 treated ICR mice. As results, compared with the control group, the KDC16-2 group showed a significant increase in wet fecal weight, dry fecal weight and fecal water content. The intestinal transit rate of KDC16-2 group was significantly increased. Based on the results, KDC16-2 is considered to have effects on improving intestinal function. The effect of anti-inflammatory demonstrated by using dextran sulfate sodium (DSS)-induced colitis mice. The mice were administered 3% DSS along with KDC16-2 (100, 300 mg/kg) for 14 days. DSS-induced colitis mice were significantly ameliorated in KDC16-2 treated group, including body weight loss, colon length shortening, tight junction protein of colon and histological colon injury. The levels of inflammatory mediators (IgG2a, IgA, C-reactive protein and Myeloperoxidase) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-${\alpha}$, Interleukin (IL)-6) which are involved in inflammatory responses were increased in the DSS-treated group as compared to those in the control group, and the levels were significantly decreased in the KDC16-2 groups. In addition, we investigated the impact of KDC16-2 on lipopolysaccharide (LPS)-induced inflammatory responses in J774A.1 cells. KDC16-2 inhibited production of prostaglandin E2 (PGE2) and reactive oxygen species (ROS). These results suggested that the KDC16-2 could effectively alleviate the dysfunction of intestinal and inflammatory mediators. Thus, these KDC16-2 can be potentially used as health functional food of intestinal.
