Antibacterial Compound against Pasturella multiocida and Actinobacillus pleuropneumoniae Causing Porcine Pneumonia

  • Porcine pneumonia is caused by Pasturella multiocida and Actinobacillus pleuropneumoniae. To identify a potent drug for antipneumonia therapy, several herbal compounds showing antibacterial effects were screened, and it was found that a methanol extract of Coptidis rhizoma root stem exhibited activity against both pneumonia-causing bacteria. Using an activity-guided fragmentation procedure, an isoquinoline alkaloid was isolated which would be responsible for the antibacterial activities against P. multocida and A. pleuropneumoniae.

  • In order to find therapeutic agents for porcine pneumonia, we screened far antibacterial activities of methanol extracts of 81 higher plants against four pathogenic microorganisms of Heamophilus parasuis, Pasteurella multocida, Actinobacillus pleuropneumonia, and Bordetella bronchiseptica, and found the bark of Cinnamomi cortex showed potent activities. Since this was inexpensive, we purified active compounds from it. The structures of the final active fractions were obtained through an activity-guided fractionation and their antibacterial activities are reported here.

전북 익산 양돈단지 이유 후 위축자돈 질병 조사 (Investigation of post - weaning atrophic pig diseases in swine breeding complex in Jeonbuk - Iksan)

  • The purpose of this study was to investigate the infection situation of several diseases (post-weaning atrophic pigs) such as porcine reproductive and respiratory syndrome (PRRS) in swine breeding complex in Jeonbuk-Iksan. From February to October in 2006, a total of 28 swine samples (6-10 week old) were collected from 6 farms and examined by polymerase chain reaction(PCR) and clinical signs. In the rate of single infection, pneumonia was top (32.1%), followed by salmonellosis (14.2%)and Glasser's disease (10.7%) and double infection pneumonia/Glasser's disease (17.8%) was detected. PCR was detected of PCV 2 from 28 (100.0%) and PPV 6 (21.4%), PRRS PORF6 10 (35.7%) and POR7 11 (39.2%), but HC and AD was not detected. The results suggest that PCV 2 is complex infection PRRS, PPV and bacterial disease.

  • Proliferative and necrotizing pneumonia (PNP) is a form of interstitial pneumonia that occurs in post-weaning pigs. In this study, we investigated the presence of swine influenza virus (SIV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2) and Aujeszky's disease virus (ADV) in PNP lesions in Jeju pigs. Based on the histopathologic criteria for PNP, a total of 50 cases were selected in Jeju pigs between 2008 and 2010. Coupled with histopathological examinations, the presence of ADV and SIV by polymerase chain reaction (PCR) and reverse transcription PCR (RT-PCR) and PRRSV and PCV2 by immunohistochemical (IHC) methods were investigated. Based on the PCR and RT-PCR methods, ADV and SIV nucleic acids were not detected in all cases. According to IHC, PRRSV was detected in 38 of the 50 cases examined (76%) and PCV2 in 25 cases (50%). PRRSV or PCV2 were detected in 19 (38%) or 6 (12%) cases, respectively. Both PRRSV and PCV2 were identified in other 19 cases (38%). Antigens of PRRSV and PCV2 were commonly observed in the cytoplasm of macrophages and clusters of necrotic cells in alveolar cavities. The results of the present study demonstrate that PRRSV is predominantly associated with PNP in Jeju pigs. Co-infection with PRRSV and PCV2 may enhance the severity of PNP lesions in affected pigs.

  • The present studies was to seroepidemiological investigation with the related pneumonia due to a combination of both viral and bacterial agents. In selected herd in Jeonbuk-Iksan area of swine farms in 2006. The seroprevalence of antibody titers of PRRS, PCV-2 and mycoplasmal pneumonia were 87.2%, 69.8% and 47.4%, respectively. High seroprevalence of antibody in sow indicate that the infected sow major source of the diseases, and play an important role in circulation between production stage.

  • Enzootic pneumonia caused by Mycoplasma hyopneumoniae is responsible for major economic losses in pig herds of world wide. Mycoplasma hyopneumoniae can also act as a primary pathogen of porcine respiratory disease complex followed by bacterial or viral infection. This study was carried out to investigate the prevalence of mycoplasmal pneumonia of slaughtered pigs in Jeju for two years. The lungs and sera of 214 cases were examined for gross and microscopic lesions of the lungs, immunohistichemistry test for Mycoplasma hyopneumoniae antigen and enzyme-linked immunohistichemistry assay (ELISA) for serum antibody titer. Pulmonary consolidation was observed in the lungs of 163 pigs $(76.1\%)$ with average gross lesion score of $6.0\%$., Bronchointerstitial pneumonia was most frequently observed $(78.5\%)$. The incidence of pulmonary consolidation was decreased in vaccinated pigs compared to that of non-vaccinated pigs. The rate of consolidation in the lungs was significantly decreased in the vaccinated pigs (P<0.05). Antigen of Mycoplasma hyopneumoniae was identified by immunohistichemistry test in the lungs of 174 pigs $(81.3\%)$. ELISh antibodies to Mycoplasma hyopneumoniae were detected in 154 pigs $(72.0\%)$. These results showed the prevalence of swine pneumonia and the incidence of Mycoplasma hyopneumoniae in slaughtered pigs of Jeiu province. We expect that these results would be helpful for the control of swine mycoplasmal pneumonia and porcine respiratory disease complex in Jeju.

Co-infection of Toxoplasma gondii and porcine reproductive and respiratory syndrome virus in suckling piglets in Jeju, Korea

  • Two suckling piglets, 4 days and 10 days of age, showed lethargy and dyspnea after birth and mortality had been increased after incoming gilts from breeding farm. At necropsy, the lungs showed diffuse fail to collapse with rubbery consistency, edematous dilatation of interlobular septa, and lobular consolidation with purple red color. Heart was diffuse pale in color and had several irregular linear-shaped macules or patches. Histopathologically, diffuse interstitial pneumonia with the proliferation of type II pneumocytes was present in the lungs of 2 piglets. Alveolar lumens contained necrotic cellular debris derived from neutrophils and macrophages. Multifocal hemorrhage and necrotizing pneumonia with protozoan tachyzoites were observed in the lungs. Severe multifocal to confluent necrotic myocarditis, necrotic encephalitis, and necrotic adrenalitis with intralesional protozoan tachyzoites were observed in piglets. According to immunohistochemical analysis (IHC), Toxoplasma (T.) gondii tachyzoites antigens were confirmed in lung, heart, brain, and adrenal gland. And porcine reproductive and respiratory syndrome virus (PRRSV) antigens were also detected in the cytoplasm of macrophages in lungs using IHC. Based on the gross, histopathologic and immunohistochemical features, two suckling piglets were diagnosed as co-infection of T. gondii and PRRSV.

Studies for the Pathogenesis and Pathogenicity for the Porcine Circovirus Type 2 Korean Isolates in Weaned Pigs

Coagglutination 반응법에 의한 Actinobacillus pleuropneumoniae의 혈청형 조사 (Serotyping of Actinobacillus pleuropneumoniae by Coagglutination Test)

  • For the inspection of the occurrence situation of porcine pleuropneumonia and serotyping of Actinobacillus pleuropneumoniae strains isolated from lung lesions of pig in Korea, a series of experimentation have been carried out by the isolation and identification of A pleuropneumoniae, serotyping by coagglutination test, observation of lung lesion and clinical signs from 360 cases of porcine pneumonia in Clinical Pathology Laboratory, Bayer Veterinary Medical Research Institute. The results could be summarized as follows. The reaction of coagglutination between the reference antigens and the specific reagents of A pleuropneumoniae was strongly agglutinatied within 30 seconds without cross reaction. The 89 strains of A pleuropneumoniae were isolated from 360 cases of porcine pleuropneumonis and the biochemical properties of the isolates were same as the reference strains. The 89 isolated strains could be serotyped 39 strains as setotype 5, 34 strains as serotype 2, 8 strains as serotype 3, 2 strains as serotype 7 by coagglutination test, respectively. The clinical signs of pleuropneumonia were weakness, fever, anorexia, dyspnea and laboured breath in the later stages. The gross lesions of lung were haemorrhages, enlargement of interlobular septa, nodular formation and adhesion of the pleura.

국내 이유자돈의 써코바이러스 감염에 의한 이유후전신소모성 증후군 (Porcine Circovirus Infection in Weaned Pigs with Postweaning Multisystemic Wasting Syndrome in Korea)

  • Eight nursery to grower pigs exhibiting weight loss and sudden death were diagnosed as postweaning multisystemic wasting syndrome (PMWS) based on the results of gross findings, histopathology, immunohistochemistry, fluorescent antibody test, virus isolation, PCR, serology, and electron microscopy. Groosly, the pigs had a rough hair coats and were severely emaciated. And moot lymph nodes were pale and enlarged. Lungs were not fully collapsed and exhibited 10 to 40% pale red cranioventral consolidation. Histopathologically, typical lymphohistiocytic interstitial to bronchointerstitial pneumonia, chronic lymphadenitis, severe lymphoid depletion, and basophilic intracytoplasmic inclusions were noted in the most lymphoid tissues. Porcine circovirus panicles were observed in the inguinal lymph node of the pigs by electron microscopy. Porcine circovirus type 2 (PCV2) antigens or viral DNAs were detected in the lesions of all pigs using immunohistochemistry or PCR. Two PCV2 were isolated from a homogenate of pooled lung and lymph node in 2 of the 5 pigs. Additionally, antigens of porcine reproductive and respiratory syndrome virus (PRRSV) and Hemophilus (H.) parasuis were also detected by immunofluorescent antibody test. Serologically, 55% of randomly selected sows and fattening pigs was serum antibody positive to PCV2 by an indirect fluorescent antibody (IFA) test and approximately 18 % of animals in the herd were serologically pooitive by the ELISA kit for PRRSV. To our knowledge, this is the first report of PMWS co-infected with PCV-2, PRRS, and H. parasuis in Korea.
