AN INVESTIGATION OF IMMUNIZATION AGAINST SOMATOSTATIN BY MEASURING ANTIBODY TITRES, SOMATOSTATIN AND SOMATOTROPIN PROFILES IN GILTS

  • The effect of active immunization against porcine somatostatin (SRIF-14) on somatostation and somatotropin secretion profile in 18 gilts was investigated. Gilts were assigned to the following treatments: control (sham injection, n = 6); bovine serum albumin (BSA) (injection of BSA with bacterial protein adjuvant, n = 6); SRIF (injection of BSA-SRIF-14 conjugate with bacterial protein adjuvant n = 6). Serum SRIF and pST were assayed from the blood samples taken on day 7 after the last immunization injection. Anti-SRIF antibody titres were assayed in weekly samples two weeks after the initial immunization to one week after the last immunization. Results revealed that the immunization protocol used in the present investigation failed to produce antibodies capable of neutralizing endogenous somatostatin. In addition, the porcine somatotropin assay revealed no significant differences in baseline pST concentration, mean peak amplitude and number of peaks during a 24 h secretory period among SRIF, BSA and control treatment. There were also no differences in SRIF baseline concentration, peak amplitude, and number of peaks during a 24 h secretory period among any of the three treatments. Circulating concentrations of pST and pSRIF were highly correlated (r = -0.09). Furthermore, anti-SRIF antibody titre was not detected in the serum of the gilts actively immunized against SRIF. These data, collectively, suggest that the protocol employed in the present investigation for active immunization against SRIF is not an effective method for changing SRIF and pST secretion profiles of the gilt and thus to enhance performance.

  • Safety of recombinant porcine somatotropin administration on pig was studied using 32 Landrace x Yorkshire crossbred pigs. The starting body weight ranged from 55.5kg to 65.3kg. Eight pigs were allotted to each low dose group of sustained releasing rPST(SL), high dose group of sustained releasing rPST(SH), daily injection group of rPST(DI), and control group(C). Pigs in SL group and SH group were injected subcutaneously twice in 3 week-interval with 1000$\mu\textrm{g}$ and 2000$\mu\textrm{g}$ of sustained releasing rPST per kg body weight, respectively. Pigs in DI group were injected intramuscularly with 100$\mu\textrm{g}$ of rPST everyday for 6 weeks. Blood was collected from anterior vena cava just before the first treatment, and at four weeks and six weeks of experiment. Hematological parameters and blood chemical parameters indicating liver function, kidney function, electrolyte metabolism, mineral metabolism and lipid metabolism were determined. Necropsy and urinalysis were performed after final blood collection. The results were summarized as follows, and it is concluded that rPST administration does not affect pig health negatively. 1. rPST administration did not affect kidney function as manisfested by BUN, creatinine and urinalysis. 2. rPST administration did not affect liver function as manisfested by total protein, albumin, serum AST activity serum ALT activity serum ALP activity, serum LDH activity, serum GGT activity and serum SDH activity. 3. rPST administration did not affect skeletal muscle, cardiac muscle and brain as manifasted by serum AST activity and serum LDH activity. 4. rPST administration increased blood glucose level within normal range. 5. rPST administration did not affect lipid metabolism as manisfested by triglyceride, cholesterol, and phospholipid concentrati on. 6. rPst administration dia not affect mineral metabolism as manisfested by calcium, phosphorus, magnesium and iron concentration. 7. rPST administration did not affect electrolyte metabolism as manisfested by Na, K, chloride concentration. 8. rPST administration did not affect erythrocyte count, leukocyte count, thrombocyte count, and plasma fibrinogen level.

  • 국내에서 제조된 서방출형 유전자재조합 돼지 성장호르몬 (porcine somatotropin; pST)이 돼지의 성장과 등지방두께에 미치는 영향을 구명하기 위해 평균체중이 78kg인 랜드레이스 거세돈 48두를 공시하여 대조구, 4:1제형 rpST구 (6회 투여), 1:1제형 rpST구 (4회 투여), 4:1제형 rpST구 (4회 투여)에 각각 12두씩 배치하였다. 대조구는 pST를 투여하지 않았고, rpST 처리구는 1주일에 1회, 목 근육에 서방형 제재를 주입하였는데, 6회 투여구는 개시부터 2주까지는 100mg을, 3주부터 6주까지는 125mg을, 4회 투여구는 개시부터 2주까지는 100mg을, 3주부터 4주까지는 125mg을 투여하였다. 시험사료는 라이신 함량이 0.9%인 시판 육성돈사료를 무제한 급여한 결과 (주) LG 화학에서 제조한 서방출형 유전자재조합 pST는 증체량 및 사료요구율을 개선시켜 생산성 향상과 등지방두께 감소로 육질이 향상되었다.

비육우, 젖소 및 비육돈에서 이식형 소마토트로핀 지속성 제형의 유효성 시험 연구 (Efficacy test of sustained-release somatotropin by implantation in steer, cow and barrow)

  • Sixteen steers were used to investigate the efficacy of the sustained-release implant of bovine somatotropin (bST) in improving growth and feed:gain ratio during 12 weeks. Administration of the 400 mg bST implant resulted in a 16.1% increase in growth rate, and this increase was significant (p<.05). The use of the sustained-release implant did not alter (p>.05) feed intake and feed:gain ratio. Thirty-four cows were used to investigate the efficacy of the sustained-release implant of bST in milk production during 4 weeks. Administration of the 200 mg bST implant resulted in an 8.7% increase in milk production, and this increase was significant (p<.05). Twenty-four barrows were used to investigate the efficacy of the sustained-release implant of porcine somatotropin (pST) in improving growth, feed:gain ratio and backfat thickness during 6 weeks. Administration of the 120 mg pST implant resulted in a 11.4% increase in feed:gain ratio and a 60% decrease in backfat thickness, and these results were significant (p<.05). But the use of the sustained-release implant did not alter (p>.05) growth rate and feed intake.

Effects of Weekly Administration of Implant-type Recombinant Porcine Somatotropin on the Performance and Carcass Characteristics of Finishing Pigs

  • The present study was undertaken to investigate the effects of weekly administration of implant type recombinant porcine somatotropin (rpST) on the performance and carcass characteristics in finishing pigs. A total of 120 crossbred (Landrace${\times}$Yorkshire${\times}$Duroc) pigs were employed for 11 weeks in a growth trial in experiment. A rpST designed to implant every 7 d was used. Forty pigs, each weighing 75 kg, were allocated into three rpST treatments; control (CONT), implant of rpST from 75 kg (TRT1) or 90 kg (TRT2) of body weight. The CONT pig and pigs in TRT2 from 75 kg to 90 kg were treated without rpST but with placebo. In rpST-treated pigs, each 100 mg and 125 mg of the equivalent rpST was implanted from live weight of 75 kg to 90 kg and from 90 kg to market weight, respectively. Half of the pigs from each treatment were marketed at live weight of 110 kg and the rest at 130 kg. All pigs were allowed ad libitum access to a commercial feed containing 0.94% and 0.88% of lysine from 75 to 110 kg, 110 to 130 kg of body weights, respectively. rpST had no effect on daily gain, while feed efficiency was improved by 7 to 13% (p<0.05) in the rpST-treated groups compared with the CONT. Compared with the CONT, backfat thickness was decreased by 12% (p<0.05) in TRT1 at 110 kg of market weight, and by 23 to 32% (p<0.05) in the rpST-treated groups at 130 kg of market weight, respectively. Lean muscle rate tended to be higher in TRT1 at both 110 kg and 130 kg of market weight, and carcass fat percentage in the rpST-treated groups was decreased by 33 to 46% (p<0.05) compared with the CONT.

Effects of Chromium Picolinate (CrP) on Growth Performance and Carcass Characteristics of Fattening Pigs Treated With or Without Porcine Somatotropin (pST)

  • Objectives of this study was to investigate effects of supplemental chromium (Cr) as CrP in growing pigs treated with pST. Seventy two Landrace pigs weighing average 60 kg were alloted to the three treatments during the 52-d experimental period: control (corn-soybean basal diet); pST treatment (4 mg/head/day); pST + CrP treatment (4 mg and 200 ppb/head/day). Upon termination of feeding trial weighing average 105 kg, thirty-six pigs randomly selected from each treatment were slaughtered to compare carcass traits. For the study of lipid metabolism, eighteen pigs were alloted to the same treatments. Adipose tissue samples from eighteen pigs were collected to investigate lipid metabolism. All treated samples with pST and pST + CrP showed improvements in daily weight gain, regardless of sex. Feed/gain ratio significantly improved in pigs treated with pST and pST + CrP. Dressing percentages were higher in pigs treated with pST and pST + Crp. Carcass grades were significantly higher in pigs treated with pST and pST + CrP. Lipolysis of adipose tissue measured in vitro was significantly increased in pigs treated with pST, lipogenesis in vitro showed opposite tendency. Even though the current data does not show synergistic effects on the above parameters when CrP and pST were supplied at the same time, but CrP supplementation tended to improve growth performance and carcass traits of pigs treated with pST.

  • 1. 돼지 성장호르몬을 연간 6000Kg을 생산하는 공장의 공정 설계를 Macintosh PC를 사용한 Bio D Designer를 이용해서 공정모사를 해 보았다. 각 단 위공정의 계산자를 정하기 위한 계산을 가정과 플라 스크배양의 결과를 이용하여 up-stream을 설계하고, downstream에서는 25%의 분리정제 수율을 가 정하고 전체공정을 설계하였다. 2. 경제성 분석에 의하연 돼지 성장호르몬의 dose 당 $ 2.00로 할 때 투자상환율 (return on invest­m ment, ROI)이 일 년에 104%이었으며, 투자금액의 상환에 걸리는 시간은 약 1년(0.96\건)이 걸리는 것으로 나왔다. 3. Sensitivity분석에 의하면 투자상환율은 돼 지 성장호르몬의 수율, 가격, 사용량 및 시장잠식율 순서의 변수에 의해 결정되는 것을 알 수 있었다.

Porcine Somatotropin Improves the Efficiency of Digestible Protein Use for Protein Deposition by Growing Pigs

  • A study was conducted to clarify the impact of recombinant porcine somatotropin (pST) on the efficiency of absorbed nitrogen use for protein deposition in growing pigs. Three levels of dietary crude protein (9.0, 11.5, 14.0% CP) were used. Each had either a sub-optimum or near optimum lysine: CP concentration (Low-lysine, 3.8 g/100 g CP and High-lysine, 5.5 g/100g CP) in order to achieve different metabolic efficiencies for nitrogen deposition (ca. 45 vs. 60%). Twelve crossbred female pigs $(59{\pm}4kg\;BW)$ were placed in metabolism cages and fitted with bladder catheters. Each pig received an excipient injection daily for the first 10-d, a pST (5 mg/d) injection for the second 10-d, and then excipient for the last 10-d. Pigs were randomly assigned to one of six dietary treatments (2 pigs/diet) and fed 4 times per d at $92g/kg\;BW^{0.75}$ $(3{\times}maintenance)$. Means for the excipient period were compared to means for the pST period. Urinary nitrogen (N) output declined in pST-treated pigs (p<0.01) irrespective of dietary protein content or lysine level. Nitrogen retention increased by an average of 11% (p<0.01) with pST treatment (726 vs. $803mg\;N/kg^{0.75}\;BW/d$). Forty-eight percent of the absorbed N was retained with Low-lysine diets, but this increased to 53% with pST injection (+11%, p<0.01). Pigs fed High-lysine diets retained 62% of absorbed N which increased to 69% with pST (+11% p<0.01). the addition of lysine improved N use by 27% (High vs. Low, p<0.01), but the effect of lysine and pST was additive (+40%). Therefore, pST improves N retention and the efficiency of apparently absorbed N use in growing pigs (>60kg). It does so with diets having the potential for either low or high efficiencies of N use (48% and 62%). More work is needed to determine if the partial efficiency of N use improves in direct proportion to pST dose since the improvement in protein deposition is a function of pST dose.

돼지에서 서방형 성장호르몬의 약물동태 및 조직잔류성 (Pharmacokinetics and tissue levels of a sustained-release recombinant porcine somatotropin in pigs)

  • 서방형 돼지성장호르몬(sustained-release formulation of porcine somatotropin, PST-SR)을 1주 간격으로 6차례 피하 및 근육주사하고 혈액과 조직중의 돼지 성장호르몬(PST)과 insulin-like growth factor 1(IGF-1)의 농도를 측정하여 다음과 같은 결과를 얻었다. 대조군의 혈중 PST와 IGF-1의 농도는 각각 2.41과 95.2 ng/ml 이었다. 1. PST-SR을 투여한 후 PST의 혈중농도는 8시간만에 최대에 도달하여(30 ng/ml) 곧 감소하였다. 혈중농도 반감기(decay half life)는 91~227시간이었다. IGF-1의 혈중농도는 투여후 12시간에 최대에 도달하였으며(165 ng/ml), 이후 서서히 감소되었고 반감기는 77~99시간이었다. 2. 혈중 PST농도-시간의 자료는 제재에서 PST가 유리되는 과정에는 두단계 즉, 투여후 24시간까지의 유리속도가 빠른 단계와 그 이후의 유리속도가 느린 단계가 있음을 보여주었다. 3. 여섯번의 반복투여기간에는 PST의 혈중농도는 투여직후 증가하여 24시간 이후 다음 투여전까지 지속적으로 감소되는 패턴이 반복되었고, 최종투여후 1주일경에는 정상수준으로 회복되었다. 반면에 투여가 반복됨에 따라 매 투여직후의 PST의 혈중 최고치는 다소 증가되는 경향을 보였다(20~40 ng/ml). IGF-1의 혈중농도는 투여가 반복됨에 따라 누적적인 증가현상이 뚜렷하였으며, 이후 2주일후 까지도 정상농도보다 높게 유지되었다(200ng/ml). 임상용량 투여군에서 PST 및 IGF-1의 혈중농도는 투여경로에 따른 차이는 나타나지 않았다. 4. 최종(6번째) 투여후 6, 8, 10, 14일에 조사한 간장, 신장, 소장, 근육, 지방 및 주사부위의 조직중의 PST 농도는 6일째에 이미 대조군 수준으로 회복되었다. IGF-1의 경우 최종투여후 6일에는 간장, 신장, 소장, 지방조직에서 정상보다 높은 농도로 잔류하나 이후 14일까지 모두 대조군 수준으로 감소되었다. 5. 이상의 결과는 본 실험에서 사용된 서방형 PST제제는 최소 1주간 유효성이 유지되며, 동시에 PST는 투여 6일째에, IGF-1은 투여후 14일에 정상수준으로 회복됨을 보여주고 있다.

Feeding and Management System to Reduce Environmental Pollution in Swine Production - Review -

  • In this manuscript, several effective feeding and management systems to reduce environmental pollution in swine production have been briefly introduced. It is logical that reducing the excretion of nutrients in manure should be the first step to reduce the environmental impact of pig production. it is evident that the excretion of nitrogen and phosphorus can be reduced when more digestible or available feedstuffs are used. Also, it is well known that proper feed processing can reduce anti nutritional factors (ANF) and improve nutrient digestibilities. Supplementation of effective feed additives can reduce excretion of nitrogen and phosphorus due to efficient feed utilization. These include enzymes (e.g., phytase), antibiotics, probiotics, organic acids and growth hormones ($\beta$-agonists and porcine somatotropin). One of the most effective ways to reduce pollutants from swine manure is to use synthetic amino acids in feed manufacturing. Many studies showed that reduction of 2 to 4% unit (U) of dietary protein with supplemental amino acid (AA) could dramatically reduce (15 to 20%) nitrogen excretion. Regarding feeding strategies, it has been recognized that phase feeding regimen could be used to reduce nitrogen and phosphorous excretion by feeding pigs in better agreement with age and physiological state. Feeding barrows and gilts separately, known as split sex feeding, can also decrease excretion of nitrogen and phosphorus. With the increasing concerns on the negative impact of animal production systems on the environment, animal nutritionists and producers should be aware that sustainability of animal agriculture is as important as high production performance. Therefore, some feeding and management strategies described in this manuscript will help to reduce environmental pollution in swine production. Proper combination of feeding regimen and environment-friendly diet formulation through nutritional approach will be more effective to reduce nutrient excretion in swine production system compared to single approach to do so.
