Analysis of Upstream Regulatory Region from Populus nigra × P. maximowiczii by Inverse PCR Technique (Inverse PCR 기법(技法)을 이용(利用)한 양황철 DNA의 Regulatory Region의 탐색(探索))

  • This research was conducted to identify plant regulatory regions by gene tagging method. A promoterless GUS coding sequence was introduced to Populus nigra ${\times}$ P. maximowiczii via Agrobacterium strains(LBA4404/EHA101), and putative transgenic poplars were selected by culturing on medium containing G418($60mg/{\ell}$) and by GUS assay. Among them one positive plant was to amplify the native sequences flanking to the introduced GUS gene in plant genome by inverse PCR method and from this 730 by DNA product was obtained. After subcloning and sequencing, it has 88% homology to the Eucalyptus gunnii CAD(cinnamyl alcohol dehydrogenase) gene. The GUS gene fused with the putative promoter reinserted into poplar leaves by particle bombardment method to test the funtional promoter activity. Upon staining with X-gluc, many blue spots appeared on the leaf segments bombarded by the chimeric gene 2-3 days, thus the isolated DNA fragment contain some possible coding region as well as a putative regulatory sequences of poplar CAD gene.

Selection of Superior Poplar and Willow Clones in Growth Performance and Adaptation Abilities at Sudokwon Landfill Site (수도권매립지에서 생장과 적응력이 우수한 포플러류 및 버드나무 클론 선발)

  • Poplars and willow were planted to identify suitable species and varieties for landfill reclamation at the Sudokwon Landfill Site in 1997. Survival rate, growth performance, vitality, visible foliar injury by pollutants, fungi, and leaf insects, and stem borer damage have been investigated for 10 clones of 4 poplar species and 2 clones of one willow species from 1997 to 2005. The average survival rates of poplar and willow clones were drastically decreased from 90% in 1997 to 53% in 2005. Among poplar species, Populus alba ${\times}$ P. glandulosa showed the highest mean survival rate of 66%, while Populus koreana ${\times}$ P. nigra var. italica and Populus euramericana were the lowest of 41%, respectively in 2005. Clivus, which is one of the clones from Populus alba ${\times}$ P. glandulosa, showed the highest survival rate of 73%. For mean height, Ec028 clone(P. euramericana) showed the highest of $11.2m{\pm}2.1m$ and followed by Clivus of $11.0m{\pm}2.0m$. Clone 131-27(Salix alba) was the lowest of $7.8m{\pm}1.6m$. Vitality, defoliation, visible foliar damage, and stem borer damage were significantly different among clones. Most of Populus alba ${\times}$ P. glandulosa and Salix alba clones seemed to have strong vitality and to be tolerant to various stresses at the site. However, Populus nigra ${\times}$ P. maximowiczii was sensitive to the stress. We have selected 5 clones in total: Clivus as the best clone for waste landfill reclamation, and additionally two Salix clones 131-25, 131-27 and two clones of Populus alba ${\times}$ P. glandulosa (72-9, 72-16) have been selected. These five clones could be supplied for planting at sites having an environment similar to the Sudokwon Landfill Site.

Effects of Slurry Composting and Biofiltration Liquid Fertilizer on Growth Characteristic of Poplar Clones in a Reclaimed Land Mounding Soil (간척지 성토지 식재 포플러의 SCB액비 처리에 따른 클론별 생장특성)

  • We studied on the effects of slurry composting and biofiltration liquid fertilizer (SCBLF) on growth of two-year-old poplar clones planted in a reclaimed land mounding soil. The soil on the experimental site had lower concentrations of both exchangeable cations and salinity than before reclamation. However, the content of organic matter was low compared with the most soils. We applied SCBLF to the poplars six times and 5 L in each time for 80 days. Ten clones of six poplar species or hybrids were tested in this study: Populus alba ${\times}$ P. glandulosa(Clivus, 72-30, 72-31, Bongwha1), P. deltoides ${\times}$ P. nigra(Dorskamp), P. deltoides(Lux) ${\times}$ P. deltoides(Harvard)(97-19), P. euramericana(Eco28, I-476), P. nigra ${\times}$ P. maximowiczii(62-2) and P. Koreana ${\times}$ P. nigra var. italica(Suwon). Growth performance varied more among clones than among species. Average height growth of treated plots was 18% greater than control, and clones Clivus, 97-19, Eco28 and Dorskamp were more vigorous than other clones. Diameter at breast height in treated plots was 41% greater than control, and 97-19, Dorskamp, Eco28 and Clivus were the four best clones in this respect. Mean leaf area of treated trees was 26% greater for control trees. Chlorophyll content was similar between treated and controlled trees. Total nitrogen values in leaves were much higher in treated trees 18%. SCBLF treatment on poplars planted in reclaimed land helped tree growth.

In Vitro Selection for Salty Tolerance of Populus nigra ${\times}$ P. maximowiczii (포푸라 내염성(耐塩性) 개체(個體)의 기내선발(器內選拔))

  • The concentration of 50 mM Nacl inhibited the shoot growth of P. nigra ${\times}$ maximowiczii in viro culture. The punctured leaves have produced so many individuals on MS basal medium with cytokinin. Especially MS basal medium with BAP 0.8mg/l showed the best shoot performance in which the average number of shoots were 127.6. For selection of NaCl tolerance shoot of poplar, punctured leaves were inoculated on MS basal medium with BAP 0.8mg/l and various concentrations of NaCl(from 10 mM to 100 mM graded by 10 mM). On the medium with over 50mM of NaCl, 13.7 to 15.7 shoots were obtained. Especially on the medium with go mM and 100mM. 10.7 and 8.3 shoots, respectively. The shoots derived from control medium (non-NaCl) were depressed growth, while the selected shoots from MS with NaCl showed good growth performance on MS basal medium with 50 mM of NaCl. From this results, we suggested that the possibility of in vitro selection to tolerance for inorganic salts in forest tree species.

Transformation of Populus nigra × P. maximowiczii Using Agrobacterium tumefaciens vectors (Agrobacterium tumefaciens vector를 이용(利用)한 양황철의 형질전환(形質轉換))

  • This study was conducted to find the optimum transformation condition using Agrobacterium harboring promoterless GUS gene. The optimal medium for shoot induction from leaves of Populus nigra${\times}$P. maximowiczii was MS medium supplemented with $0.1mg/{\ell}$ NAA, $0.5mg/{\ell}$ BAP(94% regeneration frequency and 11.5 average number of shoot) According to the test using pBI121, the concentration of antibiotics for selection marker gene was $100mg/{\ell}$ kanamycin or $60mg/{\ell}$ geneticin in the SIM(shoot inducing medium) 3. Two weeks later, callus was induced in the SIM 3 and this callus grew up to 0.5-1cm shoots after 6 weeks in the new SIM 3. And the treatment with methylation inhibitor(5-azacytidine) led to a dramatic increase in foreign gene expression rate from 5.7% to 26.7%. The vector systems showed. different transformation efficiencies based on the fluorometric and histochemical GUS assay. In this study the vector systems used for transformation seemed to affect transformation frequency, in which pEHA101 yielded more transformants(35.9%) than LBA4404/pBI121 did(5.7%). This result indicated that pEHA101 was effective to insert the promoterless foreign gene into a poplar genome.

Growth Response and Adaptability of Poplar Species Treated with Liquid Pig Manure (양돈분뇨 처리에 대한 포플러류의 생장반응 및 적응능력)

  • This study was conducted to analyze growth characteristics and adaptability of poplar clones under the treatment of liquid pig manure. The average of survival rate was 33% better at control than the treatment under age of 5 years. But, tree height and DBH growth were higher at the treatment than control. Populus euramericana 'Eco28' showed the highest survival rate (97.9%) under the treatment. P. euramericana 'Eco28' and P. deltoides hybrid 'Dorskamp' could be selected as superior clones for height and DBH growth under the liquid pig manure treatment. The above-ground biomass production was also investigated when the poplar clones were 5 years old. The aboveground biomass under the liquid pig manure treatment was, on average. 52.6 ton/ha, which was 80% higher than control (29.1 ton/ha). P. euramericana 'Eco28' (73.6 ton/ha) and P. deltoides hybrid 'Dorskamp' (71.1 ton/ha) showed superior biomass production than other clones at the treatment of liquid pig manure. Based on survival, growth and demage traits, the adaptability of poplar clones to liquid pig manure treatment was estimated. P. deltoides hybrid 'Dorskamp' and P. euramericana 'Eco28' showed better adaptability to the treatment. P. nigra ${\times}$ P. maximowiczii '62-10' and P. koreana ${\times}$ P. nigra var. italic 'Suwon' were identified as poor adaptability clones.

Plant Regeneration of Hybrid Poplars Through Nodule Culture System (Nodule 배양방법(培養方法)을 이용(利用)한 잡종(雜種)포플러의 식물체(植物體) 재분화(再分化))

  • Developmental micropropagation method and somatic embryogenesis for hybrid poplars, Populns ehrarnericana Eco28, P. nigra ${\times}$ P. moximowiczii 62-9, were established using nodule culture system. Calli of Eco28 and 62-9 clone were initiated from leaf explant on the medium with 0.5mg/l and 2.0mg/l 2, 4-D, respectively. Cell suspension culture was established from callus derived from leaf explant culture. When suspended on MS medium with optimal combination of BA and NAA fine nodules were obtained after 2 weeks of culture. For shoot regeneration, nodules were transferred into liquid and agar solidified medium. Numerous shoots were regenerated from nodules of 62-9 on liquid media. Organogenesis was effectively achieved on agar solidified regeneration media containing different concentrations of BA and adenine sulfate. Average numbers of 27 and 24 shoots per nodule were induced from 62-1 and Eco28 clones after 8 weeks of culture, respectively. In addition, somatic embryogenesis also occurred in the same regeneration medium. This procedure can be applied to vegetative propagation, utilization of somaclonal variation, production of secondary metabolite and materials of biotechnology research.

Studies on the Properties of Populus Grown in Korea (포플러재(材)의 재질(材質)에 관(關)한 시험(試驗))

  • In Korea, this is the situation at moment that the total demand of timber in 1972 is more than 5 million cubic meters. On the other hand, however, the available domestic supply of timber at the same year is only about, 1 million cubic meters. A great unbalancing between demand and supply of timber has been prevailing. To solve this hard problem, it has been necessitiated to build up the forest stocks as early as possible with fast grown species such as poplar. Under circumstances, poplar plantations which have been carryed on government and private have reached to large area of 116,603 hectors from 1962 up to date. It has now be come a principal timber resources in this country, and required the basic study on various properties of wood for it's proper utilization, since it has not been made of any systematic study on the properties of Populus grown in Korea. In order to investigate the properties such as anatomical, physical and mechanical properties of nine different species (P. euramericana Guiner I-214. P. euramericana Guiner I-476, P. deltoides Marsh, P. nigra var. italica (Muchk) Koeme, P. alba L.,P. alba $\times$ glandulosa P. maximowiczii Henry, P. koreana Rehder, P. davidiana Dode) of poplar for their proper use and development of new ways of grading processing and quality improving, this study has been made by the Forest Research Institute.
