  • An effective in vitro multiplication method was developed for clones of Populus davidiana Dode. Ten different media were tested for their effect on shoot multiplication. Both MS and LP medium with 0.2mg/l BAP appeared to be the best for the shoot multiplication with the rate of 9 shoots per explant. There were significant differences among the clones in both multiplication rate and shoot growth. While some clones did not require BAP to promote shoot formation, others did. More than 60% of in vitro shoots rooted on the half-strength GD medium containing 0.2mg/l IBA.

Anti-inflammatory Activity of Flavonoids from Populus davidiana

  • An in vitro bioassay-guide revealed that the methanol (MeOH) extract of the stem bark of Populus davidiana showed considerable inhibitory activity against cyclooxygenase (COX-1, COX-2). Continuous phytochemical study of the MeOH extract of this plant led to the isolation of ten flavonoids; sakuranetin (1), rhamnocitrin (2), 7-O-methylaromadendrin (3), naringenin (4), eriodictyol (5), aromadendrin (6), kaempferol (7), neosakuranin (8), sakuranin (9) and sakurenetin-5,4'-di-${\beta}$-D-glucopyranoside (10). Their structures were identified on the basis of their physicochemical and spectroscopic analyses. The isolated compounds, 1-10, were tested for their inhibitory activities against COX-1 and COX-2. Compound 7 was found to have potent inhibitory effect on COX-1 and a moderate effect on COX-2, meanwhile, compounds 1-6 showed moderate inhibition against COX-1 only. Moreover, compounds 5-8 exhibited suppressive effects on xanthine oxidase (XO). These results may explain, in part, the traditional uses of P. davidiana in ethnomedicine.

A Study on the Diameter Increment of Major Hardwood in Middle Area of Korea (중부지방(中部地方) 주요활엽수(主要闊葉樹)의 직경성장(直徑成長)에 관한 조사연구(調査硏究))

  • This study was carried out to compare the diameter increment of seven major selected tree species which grow wild in middle area of Korea. The results obtained were summarized as follows. 1) The difference in annual diameter increment among seven species according to D.B.H. classes showed to be the strongest in Juglang mandshurica, the second rank Corpus controversa, the third rank Betula platyphylla, the fourth rank Tilia amurensis, the second rank Fraxinus mendshurica, the sixth rank Populus davidiana, and the weakest in Ulmus davidiana. 2) Total diameter increment among seven species according to tree age classes were ranked as follows: Juglans mandshurica, Cornus controversa, Betula platyphylla, Tilia amurensis, Fraxinus mandshurica, Populus davidiana, and Ulmus davidiana. 3) Juglans mandshurica showed the highest diameter increment in comparison with other six species, while diameter increment in Ulmus davidiana and Populus davidiana decreased in earlier age than other species.

Variation in Aluminium Tolerance Among 5 Species of In Vitro Cultured Populus (기내(器內)에서 배양(培養)된 5수종(樹種)의 Populus류(類)에 대(對)한 Aluminium 내성비교(耐性比較))

  • The relative aluminium tolerance of 5 Populus species (Populus davidiana, P. maximowiczii, P. nigra, P. alba ${\times}$ P. glandulosa, and P. nigra ${\times}$ P. maximowicizii) was evaluated on Murashige and Skoog medium with 5 levels of aluminium-EDTA concentration (0, 0.1, 0.2, 0.5, 1.0mM). The number of root initiation of 5 Populus species was significantly affected by the medium containing various levels of aluminium concentration. The native poplar, P. davidiana showed the highest aluminium tolerance among 5 species and even stimulated shoot and root growth on the medium containing at the low levels aluminium, 0.1 to 0.2 mM concentrations. The medium containing 1.0 mM aluminium inhibited root initiation from plantlets of all species except P. davidiana. Plantlets cultured on the medium with more than 0.5 mM aluminium exhibited retarded growth and abnormal leaf shape. P. nigra ${\times}$ P. maximowiczii exhibited significantly higher aluminium tolerance than its parental species. Silvicultural aspects associated with aluminium tolerance of Populus are also discussed.

Studies on the Taxonomical Characters in Populus tomentiglandulosa and P. glandulosa (Populus tomentiglandulosa와 P. glandulosa의 분류학적(分類學的) 연구(硏究))

  • Populus glandulosa and Populus tomentiglandulosa, which were known to be natural hybrids, were examined for morphological, physiological and karyological traits to illucidate its hybridity and taxonomical importance. The results abtained were as follows; 1. Survival rate in rooting of cuttings and grafting was different between the hybrids and their rooting abilities showed incomplete dominance. 2. Their leaf openings showed incomplete dominance. The leaf longevities of P. alba ${\times}$ glandulosa and P. tomentiglandulosa were stronger than the other hybrids. 3. There were differences in resistance to toxicity of $KClO_3$ between the hybrids. 4. Many external leaf characters of the hybrids also showed incomplete dominance. P. tomentiglandulosa was similar in those characters to P. alba ${\times}$ glandulosa while P. glandulosa was similar to hybrids crossed, reciprocally crossed or back-crossed between P. davidiana and P. alba. 5. Their numbers of male flower showed incomplete dominance or hybrid vigor. The numbers of P. tomentiglandulosa were similar to thosa of P. alba ${\times}$ glandulosa while those of P. glandulosa to those of P. alba ${\times}$ davidiana or P. davidiana ${\times}$ alba. 6. Morphology and band color of male catkin bract showed incomplete dominance. Those of P. glandulosa were similar to those of P. alba ${\times}$ davidiana while those of P. tomentiglandulosa to those of. P. alba ${\times}$ glandulosa. 7. There were differences in vascular bundle number and arrangement of petiole between the hybrids. 8. Differences in the anatomical traits of stem did not exist between the hybrids but those in wood fiber size existed. 9. The chromosomes of artificial hybrids, P. glandulosa and P. tomentiglandulosa showed irregular behavior in metaphase I and II. 10. All hybrids including P. glandulosa and P. tomentiglandulosa showed small number of P.M.C. with 19 II but many univalent chromosomes were exhibited in metaphase I. 11. All hybrids including P. glandulosa and P. tomentiglandulosa showed a little abnormal nuclear plates as laggard chromosome and chromosome bridge in anaphase I and II. 12. The frequency of pollen tetrad and fertile pollen was low in most of the hybrids including P. glandulosa and P. tomentiglandulosa.

  • Genetic relationships of some poplars in the section Leuce, including 5 species and 11 clones of Populus alba${\times}$glandulosa, were investigated on the basis of RAPD marker analysis. Twenty-two of the 88 arbitrary 10-mer primers, showed reproducible amplification in the preliminary experiment with 6 samples, were used for PCR and generated a total of 181 RAPD markers. Genetic relationships among the analyzed samples were tested by two phenetic methods of the UPGMA and the neighbor-joining, which revealed the close genetic relationship between P. glandulosa and P. alba. And the close genetic relationship between P. glandulosa and P. davidiana was ascertained by the principal component analysis. Based on the observation of the close genetic relationship between them, it was deduced that P. glandulosa might be originated by the saltational speciation caused by the hybridization between P. alba and P. davidiana in nature.

Densified Fuels from Poplars (포플러로부터 고밀화연료의 제조)

  • Recently, densified fuels from biomass are widely used North America and Europe as a regenerable and clean bioenergy. Compaction of sawdust of poplars(Populus tomentiglandulosa and Populus davidiana) with starch glue for densified fuel were studied. Calorific and elemental analysis were carried out to assess these species as fuel. Hot-press process was used and compaction was performed under temperatures from 120 to $160^{\circ}C$ and at pressure of 50kgf/$cm^2$ for prescribed time. Densified fuels were evaluated by its specific gravities, compressive strengths and fines. In the case of with-glue system, when the press temperature is $160^{\circ}C$ and press time is above 7.5 minutes, densified fuels with fines less than 5% can be produced. On the contrary, in the case of without-glue system, densified fuels with fines less than 5% can be produced by controlling the press conditions and the moisture content of sawdust.

Fine-scale Spatial Genetic Structure of a Small Natural Stand of Populus davidiana in South Korea using AFLP markers (AFLP 마커를 이용한 소규모 사시나무림의 공간적 유전구조 구명)

  • A locally adapted plant population under harsh environmental changes might survive for a long generation through maintaining proper level of genetic diversity. When it happens losing the genetic diversity too much fast, the population could be declining and probably become extinct. An isolated small population of Populus davidiana was investigated to study out the genetic diversity and the fine-scale spatial genetic structure. The estimated number of adult trees in the population of Mt. Worak, South Korea, was 350 in the total area of $14,000m^2$. The number of adults in a study plot ($70m{\times}70m$) was 123. The average age was 16-year-old and a 32-year-old tree was the oldest. The distribution of individuals was slightly aggregated in the plot. Sixty-one among the 123 individuals were randomly sampled to estimate genetic variation using AFLP markers. One hundred fifty-one (77%) of total 196 amplicons were polymorphic from six AFLP primer combinations. The average number of loci per primer combination was 32.7 (S.D.=7.2). Expected heterozygosity ($H_e$) and Shannon's diversity index (S.I.) were 0.154 and 0.254, respectively. These values were extremely lower than those of other P. davidiana populations in South Korea. Genetic patchiness was showed within 21 meters by spatial autocorrelation analysis and the isolated small size of population might be mainly attributed to the formation of such small patch size.

Identification of Leaf Characteristics from Various Crosses in Relation with Populus glandulosa U. (Populus glandulosa U.에 유사(類似)한 교잡종(交雜種)의 엽특성(葉特性))

  • This study was conducted to widen the range of characteristics of the hybrids, P.alba ${\times}$ P.glandulosa, i.e. aiming for gene population expansion P.glandulosa seemed to have the similar characteristics as the one segregated from the natural hybrids between P.alba and P.davidiana. Thus the main objectives of this study were to make many crosses among poplars and then to identify leaf characteristics of the crosses similar to P.glandulosa, the results obtained can be summerized as follows; 1. Leaf characteristics such as leaf margin, presence of glands at leaf base and pubescence density, of crosses made from P.alba.davidiana ${\times}$ P.datidiana, and P.davidiana.alba ${\times}$ P.davidina showed 44% and 90%, respectively, of similarity to P.glandulosa. 2. The ratio of leaf size, including leaf length, leaf width, length from leaf base to width line, and petiole length, of the above crosses was similar to P.glandulosa. 3. Pubescence density of the dorsal leaf surface in hybrids between P.alba and P.davidiana showed generally intermediate of the parental appearance. Frequency of pubescence appearance differed from depending upon the use of P.alba, either as a female or a male parent. The use of P.alba as a male parent increased frequency of Pubescence appearance. 4. The presence of glands at the leaf base in P.glandulosa may be inherited from P.davidiana which possesses gland although gland is not present in all P.davidiana rather from P.alba which has no gland.

Variation and Heredity of Stomatal Frequency, Stomatal Size and Transpiration in Populus alba × P. glandulosa and Its Parents (Populus alba × P. glandulosa 및 교배양친수(交配兩親樹)의 기공빈도(氣孔頻度), 기공(氣孔)크기 및 수분증산(水分蒸散)의 변이(變異)와 유전(遺傳))

  • The good growth of Populus alba ${\times}$ P. glandulosa is shown at the foothill of the mountain where good soil moisture is available, but its growth is poor at the upper part of the mountain where soil moisture is limitted. Stomatal frequency, stomatal size and transpiration rates that affect soil moisture demand were determined in order to know how this character is inherited from its parents. Number of stomata per $mm^2$, length and width of stomata from the abaxial leaf and transpiration rate ($g/cm^2$) per day in P. alba ${\times}$ P. glandulosa showed intermediate values between those of their parents, while P. alba showed the highest values and P. davidiana the lowest values. The values of stomatal frequency of P. alba ${\times}$ P. glandulosa were close to the expected mean values of parents in the $x^2$ test, and in the regression analysis the transpiration rates for the hybrid and its midparent had strong relationship with 0.97 of regression coefficient. The fact that these intermediate characteristics were observed in the hybrid suggests incomplete dominance in the hybrid. It is concluded that soil moisture demand in P. alba ${\times}$ P. glandulosa and its parents is closed related to stomatal frequency, stomatal size and transpiration rate.
