We used nine microsatellite DNA markers to estimate genetic variation among wild and cultured populations of the sea squirt Halocynthia roretzi. The loci were polymorphic, with 6-32 alleles, and allelic richness ranged from 6.0 to 26.1 in each population. The wild and the cultured populations had similar mean heterozygosities ($H_O$ and $H_E$), allele numbers, and allelic richness. One cultured population with softness syndrome had a lower mean in the observed heterozygosity ($H_O$ = 0.57) and higher mean inbreeding coefficient ($F_{IS}$ = 0.261) than any other populations. This suggests that the loss of genetic variation in the diseased population might be due to increased inbreeding. A neighbor-joining tree and pairwise population estimates of $F_{ST}$ showed moderate genetic differentiation between the wild and the cultured populations. Additionally, the softness syndrome population was genetically divergent from wild populations, but it was genetically close to the cultured populations.
In this paper we have suggested a family of chain estimators of the population mean $\bar{Y}$ of a study variate y using two auxiliary variates in two phase (double) sampling assuming that the coefficient of variation of the second auxiliary variable is known. It is well known that chain estimators are traditionally formulated when the population mean $\bar{X}_1$ of one of the two auxiliary variables, say $x_1$, is not known but the population mean $\bar{X}_2$ of the other auxiliary variate $x_2$ is available and $x_1$ has higher degree of positive correlation with the study variate y than $x_2$ has with y, $x_2$ being closely related to $x_1$. Here the classes are constructed when the population mean $\bar{X}_1\;of\;X_1$ is not known and the coefficient of variation $C_{x2}\;of\;X_2$ is known instead of population mean $\bar{X}_2$. Asymptotic expressions for the bias and mean square error (MSE) of the suggested family have been obtained. An asymptotic optimum estimator (AOE) is also identified with its MSE formula. The optimum sample sizes of the preliminary and final samples have been derived under a linear cost function. An empirical study has been carried out to show the superiority of the constructed estimator over others.
Population is important as a fundamental element of local industry and economy, and census data is essential to regional planning and policy making. Although there have been many researches on population and regional planning, there are few studies on population considering spatial unit. In this study, the population of three spatial scales were compared in order to establish the spatial unit suitable for the rural planning. The study area is Gangwon, Chungcheong-Nam, Chungcheong-Buk, Jeolla-Nam, Jeolla-Buk, Gyeonsang-Nam, Gyeonsang-Buk and Jeju province. Population were compared using statistical data analysis, GIS visualization, and spatial statistics. The mean, maximum, minimum, and variance of population were calculated and the coefficient of variation according to spatial unit was compared. As the results, the census output area unit is difficult to interpret spatial analysis results. Administrative district unit has the limit that includes areas where the population does not live. The grid unit is well suited to the geographical characteristics but has many disadvantages of the grid with small population. Cornus controversa is a long-lived woody species mostly distributed in East Asia. Random amplified polymorphic DNA (RAPD) markers were used to investigate the genetic diversity and population structure of Korean populations of this species. A high level of genetic variation was found in seven populations of C. controversa. The mean genetic diversity (H) was 0.222 across populations, varying from 0.200 to 0.238. Eighty of the 93 loci (86.0%) showed detectable polymorphism in at least one population. Total genetic diversity values ($H_T$) varied between 0.192 and 0.231, giving an average overall polymorphic loci of 0.212. The interlocus variation of genetic diversity within populations ($H_S$) was high (0.167). Mean of genetic diversity in C. controversa was higher than average values for species with similar life history traits. The sexual reproduction, perennial habitat, and longevity are proposed as possible factors contributing to high genetic diversity. On a per locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) ranged from 0.169 to 0.278 with a mean of 0.216, indicating that about 21.6% of the total genetic variation was among populations. An indirect estimate of the number of migrants per generation (Nm=1.893) indicated that gene flow was extensive among Korean populations of C. controversa.
An approximate distribution of the plug-in estimator of the squared coefficient of variation ($CV^2$) is derived by using Edgeworth expansions under general population models. Also bias of the estimator is investigated for several important distributions. Under the normal distribution, we proposed the new estimator for $CV^2$ based on median of the sampling distribution of plug-in estimator.
Genomic DNAs (gDNAs) were isolated from the hard clam ($Meretrix$$lusoria$, Roding, 1798) populations of Gunsan located in the Yellow Sea of the Korean peninsula. Genetic distances among different individuals of the LSCP (light shell color population) population of the hard clam (lane 1-11), GSCP (grey shell color population) population of the hard clam (lane 12-22) and DSCP (dark shell color population) population of the hard clam (lane 23-33), respectively, were generated using Systat version 10 according to the bandsharing values and similarity matrix. The dendrogram, generated by seven reliable oligonucleotides primers, indicates 3 genetic clusters. LSCP population could be evidently discriminated with the other two populations among three populations. The longest genetic distance (0.801) was found to exist between individuals in the two populations, between individuals' no. 33 of the DSCP population and no. 06 of the LSCP population. The higher fragment sizes (>2,000 bp) are much more observed in the GSCP population. Three hard clam populations can be clearly distinguished, especially, by their morphological characters and PCR-based approach.
The concentrations of 4 kalosaponins from tissues of Kalopanax septemlobus (Thunb.)Koidz grown in 7 provenances in Korea were determined by HPLC. Kalosaponin contents in plant part were much higher in the inner bark(30.59 mg/g on the dry weight basis) than those of young leaves(22.74 mg/g on the dry weight basis) and root bark(18.02 mg/g on the dry weight basis). A considerable range of variation in the contents was observed among population. The kalosaponin contents in inner bark from each population were highest in the Mt. Barwang (30.37 mg/g on the dry weight basis) followed by Mt. Gariwang, Hanra II, Mangun, Paltan, and Hanra I population. A variation of kalosaponin contents among population may be affected by both environmental and genetic factors. Establishment of selection and propagation of high kalosaponin containing trees can be a good source for the development of valuable forest products.
Phenotypic variation among clones (individuals with identical genes, i.e. isogenic individuals) has been recognized both theoretically and experimentally. We investigate the effects of phenotypic variation on evolutionary dynamics of a population. In a population, the individuals are assumed to be haploid with two genotypes : one genotype shows phenotypic variation and the other does not. We use an individual-based Moran model in which the individuals reproduce according to their fitness values and die at random. The evolutionary dynamics of an individual-based model is formulated in terms of a master equation and is approximated as the Fokker-Planck equation (FPE) and the coupled non-linear stochastic differential equations (SDEs) with multiplicative noise. We first analyze the deterministic part of the SDEs to obtain the fixed points and determine the stability of each fixed point. We find that there is a discrete phase transition in the population distribution when the probability of reproducing the fitter individual is equal to the critical value determined by the stability of the fixed points. Next, we take demographic stochasticity into account and analyze the FPE by eliminating the fast variable to reduce the coupled two-variable FPE to the single-variable FPE. We derive a quasi-stationary distribution of the reduced FPE and predict the fixation probabilities and the mean fixation times to absorbing states. We also carry out numerical simulations in the form of the Gillespie algorithm and find that the results of simulations are consistent with the analytic predictions.

The phenetic relationships among six natural populations of Equisetum pratense in Korea were investigated at the population level by constructing a tree based on Random Amplified Polymorphic DNA (RAPD) markers. RAPD analysis was also conducted to estimate genetic diversity and the population structure of E. pratense. A mean of 26.7% at the six population levels indicated polymorphism. E. pratense was found to have fewer alleles per locus (1.267) and fewer effective alleles per locus (1.176). Genetic diversity (0.102) in E. pratense is lower than the average for species with similar life history traits. Total genetic diversity values (HT) varied between 0.112 (OPD-07) and 0.445 (OPD-16), for an average overall polymorphic locus of 0.141. Inter-locus variation in the within-population genetic diversity ($H_S$) was low (0.102). Asexual reproduction, small population size, and the colonization process are proposed as possible factors contributing to the observed low genetic diversity in E. pratense. On a per-locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) ranged from 0.129 for OPD-07 to 0.455 for OPD-09, with a mean of 0.277. This indicated that about 27.7% of the total variation was among populations. Thus, genetic variation (72.3%) resided within populations. This study contributes new information for research on the taxonomy and population genetics of E. pratense.
These studies were carried out to examine the anatomical, karyological characteristics, the variation of isozymes in needle and the frost damage of tissue for the purpose of investigating the relationships among the three natural Torreya nucifera populations of Cheju-do, Namhae, and Mt. Naejang in Korea. The results obtained can be summed up as follows : 1. The numbers of endodermal cells and inner layer cells of mucilage canal of needle conductive tissue were different in each population. The number of those of Cheju-do population was the largest. 2. In somatic chromosome, Namhae population showed one more secondary constriction. The values of $b^{arm}/a^{arm}$ were the same in all the three populations, but Cheju-do population particularly showed a different minimum value. And the karyotype formulas of each population showed difference. 3. During the meiosis, each population showed no significant difference in the irregular phenomena of chromosome. 4. In isoperoxidase and esterase variations of needle, each population showed its particular number and variation of bands. Cheju-do population showed the largest number and greatest variation of bands. 5. Under the same freeging conditions, the frost damage of Mt. Naejang population was comparatively slight, and Cheju-do population suffered from a greater frost damage than the others.
