Axenic Culture Production and Growth of a Dinoflagellate, Cochlodinium polykrikoides (적조 와편모조류, Cochlodinium polykrikoides의 순수분리 및 성장)

  • To know the antibiotic specificity of a Dinoflagellate, Cochlodinium polykrikoides, we investigated the survival time of C. polykrikoides against several concentrations of antibiotics and judged the selective specificity of antibiotics based on the $LT_50$ ($50\%$ of lethal time). The result showed that C. polykrikoides was sensitive to tetracycline and chloramphenicol, and resistant to polymixin-B, ampicillin, penicillin-G, dihydrostreptomycin, and neomycin. In the case of sensitive antibiotics to C. polykrikoides, tetracycline and chloramphenicol, the safety concentrations of both antibiotics were determined and the antibiotic specificity based or the plotted survival curve was analyzed. Before antibiotic treatment, we tested the antibiotic susceptibility of the contaminated bacterial population in tile culture of C. polykrikoides, and decided the proper kinds of antibiotics and concentrations before percoll-centrifugation. By percoll-centrifugation, we reduced bacteria, removed fungi, collected the algal pellet, and made axonic culture by antibiotic cascade procedure based on the result of antibiotic susceptibility test. We observed that axonic C. polykrikoides culture entered the logarthmic phase of growth when cell density was over 740 cells/ml and propagated to 5,800 cells/ml maximally. Divisions per day, k value of C. polykrikoides represented a good index for growth at the low density of cells. There was a highest k value shift before reaching to the logarithmic phase. We suggested that the preceeding highest k value shift stage is a good indicator for accurate broadcasting for red. tide blooming in the field, and the stage is also a good time for controlling red tide blooming in the filed, either.

  • This study was performed to find differences in phototaxis response of the larva, adult emergence, sex ratio and gall formation rate of Thecodiplosis japonensis Uchida et Inouye between two areas, north and south. Larvae from these two areas, Hweng-sung in north and Moo-an in south, both considered to be the sities of severe outbreak of the insect recently, were collected from the floor and transported to the entomology larboratory, Won-Kwang University in Iri, then overwintered. These larvae were used for various experimental purposes on various dates. The following results were obtained. 1. The sizes of larvae reached 2.45 in length, 0.70 in width for north, and 2.50 in length, 0.72 in width for sourth. It seems that the differents were not significant. 2. It is known that Leaving gall of larvae to the ground for the hibernation usually occurs on rainy days. For the larvae dispersion, the phototaxis response of the larvae was hypothesized. To check this, the author manipulated some different illumination intensities adjusting the distance between the glass tube in which 100 larvae were contained and electric bulb. The glass tubes were blackened all but except a small portion. The phototaxis responses between two areas seemed to be different particularly at low light intensities, 45, 145 and 1000 lux, The larvae from north assembled to the clear portion of the glass tube were more than 30 out of 100, however less than 30 in south sample regardless of the time passage. In either cases, the saturation points came after about 8 to 10 hours. If temperature units were used, the phototaxis curve= after 1 hour illumination between two areas provided significant differences. 3. The adult emergence of south area was lagged 10 days later than that of north area. The accumulated effective day-degrees of temperature for adult emergence were $934^{\circ}C$ for north area and $1180^{\circ}C$ for south area. The emerging duration of north area appeared to be in late May through the early of July and the peak emergence occurred in middle June that was approximately 15days earlier than that of south area. The sex ratio of female to male, regardless of area, was approximately 3 : 1. 4. The rates of gall formation of the red pine(Pinus densiflora Siebold et Zuccarini), 6-year-old seedlings, were as low as 9.94% for north area and 8.87% for south area. Through the close observation, the author presumed that the population reduction was greatly affected during the prepupa stage by relative moisture content and predators, such as spiders and ants presented on the ground.

A Inquiry of the Perception of Death in School Age (학령기 아동의 죽음인식에 관한 탐색적 연구)

  • Purpose: This paper aims to examine the subjective structures and types of school age children's perception of death through an investigative study on their perception of death in order to provide a basic material for them to understand death, and develop and carry out an effective death education program. Methods: The study method used the Q Methodology which can investigate the subjective structures and types of school age children's perception of death. For Q-population, 20 school age children were used as subjects for neutral interviews and open surveys, and through documentary research, a total of 132 statements were collected, For Q-samples, 23 statements (Q-samples) were derived through a non-structural method. P-samples were 31 school age children (8-13 year olds), Q-sorting was carried out using Q-cards, and the collected data was analyzed using the PC QUANL program. Results: As a result of the study, children's perception of death was divided into five types. The first type was functional type, characterized by prominent subjective perception regarding the elements of death, such as non-reversibility, universality, non-functionality, and causality. The second was after-life type, characterized by a strong, focus on life after death in one's perception of death, and it included children with Christian background and those who had experienced death in their immediate family. The third was religious type, characterized by a strong belief in being able to still watch over one's family and friends after one's death, resulting in a positive faith in the after-life. The fourth was fearful type, characterized by a deeper fear of death in comparison to other types. The fifth was realistic type, characterized by a strong and positive assent to the perception of good death. Conclusion: The significance of the results of this paper's study to Nursing is as follows. In terms of understanding the subjectivity of school age children's perception of death in nursing practice, and understanding the compositional elements of death presented with strong emphasis in existing literature and studies, the results will expand these understandings and allow us to understand the level of perception in school age children regarding the definition of death, after-life, and good death, be utilized as useful material in developing an effective death education program for them according to their type characteristics, and become the fertilizer for enabling the children to live a proper life and preventing the tendency to make light of death that occur in adolescence and the spread of suicides. In terms of nursing theory, the description and examination of the subjective structures and the characteristics of the different, types of school age children's perception of death can be utilized as useful material for building a model of school age children's perception of death, and be further used for teaching respect for life. In terms of nursing research, the results can contribute to research describing the effects of nursing intervention strategies and developing tools for providing psychosocial nursing in terms of giving school age children a positive perception of death according to their types as well respect for life.

  • This study is aimed to analyze and to evaluate the revegetation and soil conservation technology in desertification-affected sandy land, resulting from the project of "Studies on the desertification combating and sand industry development". Main native plants for combating desertification : The general characteristics of vegetation distribution in desertified regions are partially concentrated vegetation distribution types including the a) desert plants in low zone of desert or sanddune of depressed basin, b) salt-resistant plants around saline lakes, c) grouped vegetation with Poplar and Chinese Tamarix of freshwater-lakes, saline-lakes and river-banks, d) gobi vegetation of gravel desert and e) grassland and oasis-woods around the alluvial fan of rivers, etc. Generally, Tamarix ehinensis Lour., Haloxylon ammodendron Bunge., Calligonum spp., Populus euphratica Oliver., Elaeagnus angustifolia L., Ulmus pumila L., Salix spp., Hedysarum spp., Caragana spp., Xanthoceras sorbifolia Bunge., Nitraria tangutorum Bobr., Lespedeza bicolor, Alhagi sparsifolia Shap., Capparis spinosa L., Artemisia arenaria DC., etc. are widely distributed in desertified regions. It is necessary for conducting research in the native plants in desertified regions. Analysis of intensive revegetation technology system for combating desertification : In the wind erosion region, the experimental research projects of rational farming systems (regional planning, shelterbelts system, protection system of oasis, establishment of irrigation-channel networks and management technology of enormous farmlands, etc.), rational utilization technology of plant resources (fuelwood, medicinal plants, grazing and grassland management, etc.), utilization technology of water resources (management and planning of watershed, construction of channel and technology of water saving and irrigation, etc.), establishment of sheltetbelts, control of population increase and increased production technology of agricultural forest, fuelwood and feed, etc. are preponderantly being promoted. And in water erosion region, the experimental research projects of development of rational utilization technology of land and vegetation, engineering technology and protection technology of crops, etc. are being promoted in priority. And also, the experimental researches on the methods of utilization of water (irrigation, drainage, washing and rice cultivation, etc.), agricultural methods (reclamation of land, agronomy, fertilization, seeding, crop rotation, mixed-cultivation and soil dressing works, etc.) and biological methods (cultivation of salt-resistant crops and green manure and tree plantation, etc.) for improvement of saline soil and alkaline soil in desertified-lands are actively being promoted. And the international cooperations on the revegetation technology development projects of desertified-lands are sincerely being required.

Effects of Aqueous Chlorine Dioxide against Escherichia coli O157:H7 and Listeria monocytogenes on Broccoli Served in Foodservice Institutions (급식소에서 제공되는 브로콜리에 있어 이산화염소 처리가 Escherichia coli O157:H7과 Listeria monocytogenes의 균수에 미치는 영향)

  • This study was undertaken to evaluate the effects of chlorine dioxide on reducing E. coli O157:H7 and L. monocytogenes on broccoli served in foodservice institutions. Broccoli samples inoculated with $10^6$ CFU/mL of E. coli O157:H7 and L. monocytogenes were treated with chlorine dioxide. Treatments with 5, 10, and 20 ppm for 1, 5, and 10 min were not sufficient in controlling E. coli O157:H7 on broccoli. L. monocytogenes were effectively reduced by $2.19{\sim}2.48log\;CFU/g\;and\;3.31{\sim}3.87log\;CFU/g$ with 10 and 20 ppm chlorine dioxide for 1, 5, and 10 min treatment, respectively, compared with the control. E. coli O157:H7 and L. monocytogenes population were significantly negatively correlated with concentration and treatment time of chlorine dioxide. These results show that the use of chlorine dioxide was effective in sanitizing L. monocytogenes on broccoli and the level of concentration was more associated with populations of E. coli O157:H7 and L. monocytogenes than treatment time of chlorine dioxide on broccoli.

The Influence of Job Stress and Job Satisfaction on Turnover Intention for Male Dental Hygienists (남자 치과위생사의 직무 스트레스와 직무 만족도가 이직의도에 미치는 영향)

  • The number of male dental hygienists has been continuously increasing in dental services. The purpose of this study aimed to identify the relationship between job stress, job satisfaction, and turnover intention in male dental hygienists and to provide basic data to improve their job satisfaction. The study population was 110 Korean male dental hygienists working for dental clinics or hospitals. The data were collected from November 10 to December 7, 2015. After an explanation about the objective of the study, 110 questionnaires were distributed via email, and 97 responses were analyzed using SPSS. Overall averages of job stress, job satisfaction and turnover intention in male dental hygienists were 3.05, 2.92, and 3.47 out of 5, respectively. Some sub-factors of job stress, including interpersonal relationships at the workplace and future prospects of dental clinics, were proven to have statistically significant negative influence on job satisfaction (p<0.001). Regression analysis was performed with job stress and job satisfaction as independent variables and turnover intention as a dependent variable. Results showed that job stress had a positive effect on turnover intention (p<0.01) while job satisfaction had a negative effect on turnover intention (p<0.001). Job satisfaction was revealed to have statistically significant negative influence on turnover intention (p<0.001). Some of sub-factors of job stress, including work environment and future prospects of dental clinics and professional position, had a positive effect on turnover intention (p<0.05). The study showed that higher job stress led to higher turnover intention, and higher job satisfaction led to lower turnover intention. Accordingly, job stress and job satisfaction are critical factors for turnover intention in male dental hygienists.

Antibacterial Activity of Ceftizoxime Against Gram Negative Enteric Bacteria in vitro and in vivo (Ceftizoxime의 장내세균에 대한 시험관내 및 생체내 항균효과)

  • Ceftizoxime sodium is a new synthetic ${\beta}$-lactam antibiotic combining potent antibacterial activity with high stability to a wide range of bacterial ${\beta}$-lactamase. This experiment was achieved to evaluate the antibacterial activities of ceftizoxime sodium againist Gram negative enteric bacteria isolated from in outpatient visiting Yeungnam university hospital and to study the emergence of drug induced bacterial varients which resist to ceftizoxime in vitro. The antibacterial activity of the ceftizoxime was compared with that of antibiotics and its effect on population of normal intestinal flora in mice was observed. The results are summarized as follows : 1. Highly effective antibacterial activity of ceftizoxime against Gram negative enteric bacilli was demonstrated and this antibacterial activity was superior to that of ampicillin. 2. Several test strains shows multiple antibiotic resistence. Among 15 strains of Escherichia coli, 1 strain was resistent to ampicillin, cefadroxyl, gentamicin, tetracycline, and 2 strains were resistent to ampicillin, cefadroxyl, tetracycline, five strains of Escherichia coli and Enterobacter cloacae was resistent to amplicillin, tetracycline and Shigella dysenteria was resistent to ampicillin, gentamicin, tetracycline. 3. The frequency of in vitro emergence of resistent varients among ceftizoxime sensitive bacteria in the presence of increasing concentrations of the compound was found to be low. 4. Plasmid was isolated in 6 of 9 strains (6 strains of Escherichia coli, Shigella dysenteriae, Enterobacter cloaceae and Salmonella typhi) That showed different antibiotic resistance. They were 5 strains of Escherichia coli and 1 strain of Shigella dysenteriae. However, plasmid could not be considered as a hallmark for antibiotic resistance by this. Further studies with curing experiment are to be accomplished for this purpose. 5. Changes in the bacterial count of normal intestinal flora following 25mg/kg/day administration of ceftizoxime over S consecutive days were not significant. In conclusion, ceftizoxime appeared to be a drug of choice in the treatment of Gram negative enteric bacilli infection.

Earthquake impacts on hydrology: a case study from the Canterbury, New Zealand earthquakes of 2010 and 2011

  • On 4 September 2010 an earthquake of magnitude 7.1 on the Richter scale occurred on the Canterbury Plains in the South Island of New Zealand. The Canterbury Plains are an area of extensive groundwater and spring fed surface water systems. Since the September earthquake there have been several thousand aftershocks (Fig. 1), the largest being a 6.3 magnitude quake which occurred close to the centre of Christchurch on 22February 2011. This second quake caused extensive damage to the city of Christchurch including the deaths of 189 people. Both of these quakes had marked hydrological impacts. Water is a vital natural resource for Canterburywith groundwater being extracted for potable supply and both ground and surface water being used extensively for agricultural and horticultural irrigation.The groundwater is of very high quality so that the city of Christchurch (population approx. 400,000) supplies untreated artesian water to the majority of households and businesses. Both earthquakes caused immediate hydrological effects, the most dramatic of which was the liquefaction of sediments and the release of shallow groundwater containing a fine grey silt-sand material. The liquefaction that occurred fitted within the empirical relationship between distance from epicentre and magnitude of quake described by Montgomery et al. (2003). . It appears that liquefaction resulted in development of discontinuities in confining layers. In some cases these appear to have been maintained by artesian pressure and continuing flow, and the springs are continuing to flow even now. In spring-fed streams there was an increase in flow that lasted for several days and in some cases flows remained high for several months afterwards although this could be linked to a very wet winter prior to the September earthquake. Analysis of the slope of baseflow recession for a spring-fed stream before and after the September earthquake shows no change, indicating no substantial change in the aquifer structure that feeds this stream.A complicating factor for consideration of river flows was that in some places the liquefaction of shallow sediments led to lateral spreading of river banks. The lateral spread lessened the channel cross section so water levels rose although the flow might not have risen accordingly. Groundwater level peaks moved both up and down, depending on the location of wells. Groundwater level changes for the two earthquakes were strongly related to the proximity to the epicentre. The February 2011 earthquake resulted in significantly larger groundwater level changes in eastern Christchurch than occurred in September 2010. In a well of similar distance from both epicentres the two events resulted in a similar sized increase in water level but the slightly slower rate of increase and the markedly slower recession recorded in the February event suggests that the well may have been partially blocked by sediment flowing into the well at depth. The effects of the February earthquake were more localised and in the area to the west of Christchurch it was the earlier earthquake that had greater impact. Many of the recorded responses have been compromised, or complicated, by damage or clogging and further inspections will need to be carried out to allow a more definitive interpretation. Nevertheless, it is reasonable to provisionally conclude that there is no clear evidence of significant change in aquifer pressures or properties. The different response of groundwater to earthquakes across the Canterbury Plains is the subject of a new research project about to start that uses the information to improve groundwater characterisation for the region. Mapping and Race Specific Reaction of the Resistance Gene Pi45(t) in Rice (벼 도열병 저항성 유전자 Pi45(t)의 균계 특이적 반응과 고밀도지도 작성)

  • QTL analysis for blast resistance was carried out using 140 $BC_3F_3$ lines derived from a cross between Ilpum as a recurrent parent and Moroberekan as a donor parent. 140 $BC_3F_3$ lines with the parents were inoculated with nine blast isolates. To identify QTLs for resistance to nine blast isolates, 134 SSR markers showing polymorphisms between the parents were genotyped for the 140 $BC_3F_3$ lines. A total of 17 resistance QTLs to nine isolates were detected on chromosomes 2, 3, 4, 6, 7, 9 and 10. The phenotypic variance explained by each QTL ranged from 8.2% to 26.4%. The Moroberekan alleles contributed the positive effect at these 17 QTL loci. In a previous study, the QTL, Pi45(t) for durable resistance to blast was identified using a sequential planting method. To know the relationship between Pi45(t) and the isolate-specific resistance gene, an $F_2$ population was developed from a cross between Ilpum and an introgression line harboring Pi45(t). $F_3$ lines segregating for the Pi45(t) were inoculated to three isolates. $F_3$ lines from the $F_2$ plants with the Moroberekan segment at the target region showed resistance to two isolates. This result seems to indicate that the Pi45(t) and the isolate-specific resistance gene are tightly linked or the resistance is controlled by the same gene(s). The markers linked to genes controlling blast resistance would be useful in developing blast resistance lines in the breeding program.

A Literature Study about Comparison of Eastern-Western Medicine on the Acne (여드름의 동(東)·서의학적(西醫學的) 문헌(文獻) 고찰(考察))

  • Objective : The purpose of this study is to investigate about comparison of Eastern-Western medicine on the acne. Methods : We searched Eastern and Western medicine books for acne. We analyzed these books and examined category, definition, etiology, classification, internal and external methods of treatment of acne. Results : The results were as follows. 1. In Eastern medicine, Acne belongs to the category of the Bunja(粉刺), Jwachang(痤瘡), Pyepungbunja(肺風粉刺). In Western medicine, the other name of Acne is acne vulgaris. 2. In Eastern medicine, the definition of Acne includes manual extraction of comedones and skin appearance. In Western medicine, Acne is a common skin disease during adolescence and a chronic inflammatory disease of pilosebaceous unit of self localization. It is characterized by noninflammatory, open or closed comedones and by inflammatory papules, pustules, and nodules and it affects the areas of skin with the densest population of sebaceous follicles, these areas include the face, neck, back, and the upper part of the chest. 3. In Eastern medicine, the cause and mechanism of Acne arose from the state of internal dampness-heat and spleen-stomach internal qi deficiency due to dietary irregularities and then invaded external pathogen such as wind-dampness-heat-cold-fire in lung meridian lead to qi and blood heat depression stagnation. So it appears in skin. In Western medicine, the etiology and pathogenesis of Acne is clearly not identified, but there are most significant pathogenic factors of blood heat depression stagnation. So it appears in skin. In Western medicine, the etiology and pathogenesis of Acne is clearly not identified, but there are most significant pathogenic factors of Acne; Androgen-stimulated production of sebum, hyperkeratinization and obstruction of sebaceous follicles, proliferation of Propionibacterium acnes and inflammation, abnormaility of skin barrier function, genetic aspects, environmental factors etc. 4. In Eastern medicine, differentiation of syndromes classifies clinical aspects, and cause and mechanism of disease; the former is papular, pustular, cystic, nodular, atrophic, comprehensive type; the latter is lung blood heat, intestine-stomach dampness-heat, phlegm-stasis depression, thoroughfare-conception disharmony, heat toxin type. In Western medicine, it divides into an etiology and invasion period, and clinical aspects; Acne neonatorum, Acne infantum, Acne in puberty and adulthood, Acne venenata; Acne vulgaris, Acne conglobata, Acne fulminans, Acne keloidalis. 5. In Eastern medicine, Internal methods of treatment of Acne are divided into five treatments; general treatments, the treatments of single-medicine and experiential description, the treatments depending on the cause and mechanism of disease, and clinical differentiation of syndromes, dietary treatments. In Western medicine, it is a basic principles that regulation on production of sebum, correction on hyperkeratinization of sebaceous follicles, decrease of Propionibacterium acnes colony and control of inflammation reaction. Internal methods of treatment of Acne are antibiotics, retinoids, hormone preparations etc. 6. In Eastern medicine, external methods of treatment of Acne are wet compress method, paste preparation method, powder preparation method, pill preparation method, acupuncture and moxibustion therapy, ear acupuncture therapy, prevention and notice, and so on. In Western medicine, external method of treatments of Acne are divided into topical therapy and other surgical therapies. Topical therapy is used such as antibiotics, sebum regulators, topical vitamin A medicines etc and other surgical therapies are used such as surgical treatments, intralesional injection of corticosteroids, skin dermabrasion, phototherapy, photodynamic therapy, and so on. Conclusions : Until now, there is no perfect, effective single treatment. We think that Eastern medicine approach and treatment can be helpful to overcome the limitations of acne cure.
