Phylogenetic Relationships of Coprinoid Taxa and an Agaric-like Gastroid Taxon Based on the Sequences of Internal Transcribed Spacer (ITS) Regions (ITS 영역의 염기서열을 이용한 먹물버섯류 및 주름버섯 유사 복균류와의 계통학적 유연관계)

  • The phylogenetic relationships of black-spored basidioid taxa (mainly coprinoid taxa) and an agaric-like gastroid taxon were studied. The sequences of internal transcribed spacer regions (ITS) partially including 17S, 25S and 5.8S from 14 species (Coprinus comatus, C. atramentarius, C. micaceus, C. cinereus, C. disseminatus, C. rhizophorus, C. radians, C. echinosporus, Psathyrella candolleana, Podaxis pistillaris, Conocybe lactea, Bolbitius demangei, Agaricus balzei, and Stropharia rugosoannulata) were compared. The reciprocal homologies of ITS sequences among these species were in the range of $38.7{\sim}77.2%$. Black-spored taxa were classified into four clusters. Cluster I comprised C. micaceus, C. radians, and C. disseminata. Cluster II is consisted of C. cinereus, C. echinosporus, C. rhizophorus, and C. atramentarius. On the other hand, C. comatus is in cluster III with Agaricus balzei and Podaxis pistillaris even though this species is belonging to the section Coprinus in morphological aspect. Psathyrelloid taxon is included in cluster II. The question of the origin of secotioid (agaric-like) fungi has been taken, though largely on theoritical suggestions as to whether gastroid taxa give rise to agarics with secotioid taxa as intermediates or whether secotioid taxa are evolutionary novelities arising from many distinct groups of agarics. In this relationship, it was shown that secotioid taxon evolved from within agaric species.

Taxonomy of a Soil Bacteria YNB54 Strain Which Shows Specific Antagonistic Activities against Plant Pathogenic Phytophthora spp. (식물역병균 Phytophthora spp.에 특이 길항균인 YNB54 균주의 분류)

  • YNB54 strain which shows inhibitory activities specific to the plant pathogenic Phytophthora sp. on potato dextrose agar medium was screened among lots of strains isolated from Korean soils. To identify taxonomy of the Phytophthora specific antagonistic bacteria YNB54, 165 rDNA sequence, MIDI fatty acid composition, DNA-DNA hybridization, GC content, and commercial multitest systems such as API 20E and Biolog GN were performed. Results of commercial kits including lots of biochemical and physiological reactions showed that this strain was closely related to taxa including Enterobacter cloacae and Enterobacter cancerogenus species than other genera(Citerobacter Klebsiella, Leclercia). Also, analysis of its MIDI, G+C contents, and DNA-DNA hybridization suggests that this strain was more similiar to the Genus Enterobacter than other genera (Citerobacter Klebsiella, Leclercia). This strain was potentially identified as Enterobacter sp. by these results. But our 16S ribosomal DNA sequences (rDNA) analysis confirmed that it was more closely related to the cluster of Citerobacter freundii ATCC 29935 than any other Enterobacter species. In the absence of defined phylogenetic critia for delineating genera, the results observed with Citrobacter and Enterobacter species suggest that further studies are needed to clarify their relationships. This investigation demonstrates that YNB54 strain is genetically diverse and potentially more taxonomically complex than hitherto realized. Further study is necessary to confirm their taxonomic positions.

Phylogenetic Analysis of the Genus Gliocladium and its Related Taxa by Comparing the Sequences of Internal Transcribed Spacers and 5.8S r-DNA (Ribosomal DNA의 Internal Transcribed Spacer(ITS) 부위의 염기서열 분석에 의한 Gliocladium 속과 근연속에 관한 계통 분류학적 연구)

  • The phylogenetic position of Gliocladium and its related taxa were investigated, using the neighbor-joining method of the sequences from internal transcribed spacers (ITS1 and ITS2) and 5.8S ribosomal DNA (rDNA). It was focused especially on the generic concept by comparing with the related genera such as Trichoderma, Hypocrea, Verticillium, Penicillium and Talaromyces. Gliocladium species and its related genus were divided into three groups by the phylogenetic analysis using the neighbor-joining method. The first group includes Penicillium-like strains such as Penicillium, Tararomyces, Verticillium and one species of Gliocladium (G. cibotii JCM 9203 and JCM 9206). Especially, Gliocladium cibotii JCM 9203 is thought to be the similar species with Verticillium bulbillosum JCM 9214. Between these two species, Gliocladium cibotii and Verticillium bulbillosum, the intraspecies concept needs to examined with culture condition. and morphological properties. The second group includes two species Verticillium, Verticillium tricorpus and Verticillium albo-atrum which extracted from the GenBank database in NCBI (National Center for Biotechnology Information). Trichoderma-like strains, such as Trichoderma, Hypocrea and several species of Gliocladium are included in the third group. Also, Gliocladium penicillioides IFO 5869 and Gliocladium catenulatum ATCC 10523 formed the subgroup of Trichoderma-like strains. The species of Gliocladium were dispersed in Trichoderma-like and Penicillinum-like group, and only one species of Gliocladium cihotii used in our study was located in Penicillium-like genus group. The species of Verticillium appeared in all three groups and the species of Trichoderma formed the monophylogeny with Hypocrea (telemorph). Also, Gliocladium virens was grouped with Trichoderma harzianum with a high bootstrap value, supporting that Gliocladium virens is to be placed in Trichoderma. The results suggest that Gliocladium is polyphyletic, and is more Trichoderma-like than Penicillium-like.

Phylogenetic Relationships of Genera Coprinus and Psathyrella on the Basis of ITS Region Sequences (먹물버섯속(Coprinus)과 눈물버섯속(Psathyrella)의 ITS 영역 염기서열에 의한 계통학적 유연관계 분석)

  • The internal transcribed spacer regions(ITS) of the ribosomal DNA gene repeat from Coprinus and Psathyrella spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 11 species including Coprinus comatus, C. atramentarius, C. micaceus, C. cinereus, C. rhizophorus, C. radians, C. echinosporus, C. disseminatus, Psathyrella candolleana, P. spadiceogrisea and Stropharia rugosoannulata were compared. The spacer region I and II were $258{\sim}301\;bp\;and\;253{\sim}275\;bp$ in length respectively and partially contained 17S, 5.8S and 25S. The reciprocal homologies of ITS sequences among these strains were in the range of $43.9{\sim}96.0%$. According to the analysis of ITS sequences, Coprinus and Psathyrella spp. were classified into three clusters. Cluster I consisted of Coprinus lagopus, C. cinereus, C. echinosporus, C. rhizophorus, and C. atramentarius. Cluster II comprised C. micaceus, C. radians, C. disseminatus, Psathyrella candolleana, and P. spadiceogrisea. On the other hand C. comatus is in Cluster III with Stropharia rugosoannulata even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that taxonomic position of Psathyrella would better be inculded in genus Coprinus. Coprinus comatus, the type species of Coprinus, gives a doubt on monophyletic evolution and is assumed to be paraphyletic or polyphyletic.

Phylogenetic study of the section Adonanthe of genus Adonis L. (Ranunculaceae) based on ITS sequences (ITS 염기서열에 의한 복수초속 복수초절(미나리아재비과)의 계통분류학적 연구)

  • DNA sequences of nrITS regions obtained from 49 accessions representing 12 species and one variety within the section Adonanthe of the genus Adonis were analyzed to test the previous intra-sectional classification system and to determine their phylogenetic relationships. The results showed that the seven accessions of A. amurensis Regel et Radde included in the present study did not form a monophyletic group, as some of the accessions showed a close relationship with the Japanese endemic species A. ramosa Franch., which implies that the current species delimitation and identification of A. amurensis is problematic. Adonis pseudoamurensis W. T. Wang, which is frequently misidentified as A. ramosa in Korea, formed a segregated group, which suggests that they should not be considered as conspecific taxa. Accessions from A. shikokuensis Nishikawa et Koji Ito, A. multiflora Nishikawa et Koji Ito, and A. pseudoamurensis formed a clade, but monophyly of each species was not evident. The nrITS data did not support the classification system proposed by Wang, who classified sect. Adonanthe into four series, as most of these were found to be either polyphyletic or paraphyletic.

Genetic differences between Korean-Japanese and Chinese-Taiwanese Dendrobium moniliforme (L.) Sw. (한국-일본과 중국-대만 석곡의 유전적 차이)

  • The nucleotide sequences of nuclear ribosomal ITS regions and chloroplast rbcL, matK and psbA-trnH regions of 30 individuals of Dendrobium moniliforme from several localities in four countries and 28 related species of Dendrobium were compared to investigate the genetic differences among Korean, Japanese, Taiwanese and Chinese D. moniliforme, and to verify the homogeneity of D. moniliforme, which is used as a traditional medicine in East Asia. A phylogenetic analysis showed that Korean D. moniliforme and Japanese D. moniliforme form a monophyletic group, with no significant differences between their nucleotide sequences. This confirms that they are the same species. However, the Chinese and Taiwanese D. moniliforme were polyphyletic. Various species related to D. moniliforme were located between the Korean-Japanese D. moniliforme and the Chinese-Taiwanese D. moniliforme, and other related species were found between individuals of Chinese-Taiwanese D. moniliforme. D. moniliforme is described in Japan, providing evidence that the Korean-Japanese D. moniliforme is the original species. In addition, our data suggest that the Chinese-Taiwanese D. moniliforme complex is a mixture of a range of other species. Further studies are required to understand the taxonomic identity of this species. In the Korean-Japanese D. moniliforme, there were almost no genetic differences among the localities, whereas the genetic heterogeneity was high among individuals of the Chinese-Taiwanese D. moniliforme.

  • In the 1990s the intellectuals in China were transferred into global capitalism, and faced with the changes of society deepened by capitalism, and were forced to sign on it. The New-left proposed a question to the society of modern China being accelerated in the capitalism. The controversy with the Neo-liberal became the top issue of the 1990s' world of thought. According to the New-left, 'reform' should be attained not by capitalism of laissez-faire bringing up for wealth concentration, but by extending the democracy of political affairs and economy along with fairness guarantee of social distribution and avoiding widening the gap between rich and poor. Additionally, 'opening' should be reevaluated as a problem of considering difference and polyphyletic matter. Opening is not the meaning of accepting capitalism unconditionally and transferring into global capitalism. Based on these beliefs, the New-left criticize the socialism after reform and opening. In addition, the New-left discuss how enlightenment and modernity were understood and how they should be understood. The New-left reflect that the enlightenment in China was considered as the same as one in the western societies and emphasize the efforts of having been overcoming the contradiction of modernity through the Chinese history. As a result, the New-left seek out a new perspective and an alternative proposal beyond the dichotomy between capitalism and socialism, western countries and China, and tradition and modern.

Change Process of the Zoo in the Seoul Children's Grand Park (서울 어린이대공원 내 동물원의 변화과정)

  • This study aims to analyze the change process in order to set the improvement strategies for the zoo in the Seoul Children's Grand Park. The zoo can be reviewed through three significant time periods with noticeable changes. As a framework to analyze the major changes that happened in the zoo, this study looks at the changes in terms of the planning aspect as well as the animal welfare and program operation aspect. The findings are as follows: first, the era of general theme park turned out to have focused on exhibiting animals to meet visitor demands by expanding the zoo area of the zoo without enlarging stockyards for the animals. Second, the environmental park era created a zoo having entertaining and educational functions by arranging animal houses with the concept of zoological taxonomy and introducing animal behavioral enrichment, animal welfare programs and visitor participatory programs. The era of the zoo as an Urban Cultural Park improved old animal houses and facilities for the welfare of the animals and increased educational programs to preserve species and provide environmental education. The current status of the zoo turns out not to meet the conditions for creating an ecological zoo, which is the overall goal for contemporary zoos. The improvement strategies based on the analysis through three different eras are three-fold. First, the zoo needs to improve the boundary conditions of the animals to showcase animal wildness through landscape immersion. Second, the zoo should provide a shared environment for animals from the same habitats by changing the classification methods from the existing polyphyletic taxon to a classification that considers ecological habitat. Third, the zoo needs to develop various ecological education programs by supplementing specialists in professional education.
