SDS-polyacrylamide Gel Electrophoresis on Partially Purified Ginseng Proteins (인삼단백질분획에 대한 SDS-폴리 아크릴아미드 전기영동)

  • Ginseng proteins have been extracted and partially purified by the methods of ammonium sulfate fractionation, heat inactivation and Sephadex G-75 column chromatography. The final three fractions obtained (GI, GII and GIII) were subjected to paper chromatography and SDS-polyacrylamide gel electrophoresis. Molecular weights of polypeptide chains from each fraction were est-mated by the standard line obtained from the plot of electrophoretic mobilities against the logarithm of molecular weights of standard proteins. Results are as follows: 1) GI showed three protein spots and four polypeptides of which M.W. were 63,000, 60,000, 56,000, 51,000 and 34,200. 2) GII showed four protein spots and five polypeptides with their M.W. of 64,600, 56,000, 45,400, 35,800, end 25,200, 3) GIII showed three protein spots and four polypeptides with their M.W. of 66,000, 63,000, 56,000 and 22,000.

Cloning and Sequencing Analysis of cadC Encoding Transcriptional Activator CadC from Salmonella typhimurium

  • Salmonella typhimurium possesses a cad operon, which contributes to an adaptive response against an acidifying environment. In Escherichia coli, the activation of the cad operon is dependent on cadC, which is located upstream of the operon. However, the activator of cad operon in S. typhimurium has not been known until now. In this study, we selected a putative cadC mutant by trasposon mutagenesis and cloned the cadc of S. typhimurium. Moreover, the cadC mutant was complemented by cadC clone. The cadC gene from S. typhimurium LT-2 consists of 1539 bp encoding a polypeptide ob 512 amino acids, and shows sequence similarity to cadC of E. coli with 53% identity and 67% similarity. The hydrophobicity profile of th S. typhimurim CadC sequence is very similar to E. coli CadC.

  • Cystocin is identified from the screening of Streptomyces sp GCA0001. It is isolated from the culture of streptomyces sp GCA0001. A white coloured cystocin is structurally similar to puromycin, an antibiotic produced by streptomyces albonrger. Like puromycin, cystocin possesses an antibacterial, antitumour and antiviral activity. It impairs the protein synthesis by inhibiting polypeptide chain elongation.

  • The orientation effect of galactose ligand on hepatocyte attachment was investigated. Poly(N-p-vinyl benzyl-o-${\beta}$-D-galactopyranosyl-D-gluconamide) (PVLA), a ${\beta}$-galactose-carrying styrene homo-polymer, was used as a model ligand for the asialoglycoprotein receptors on hepatocytes. PYVA was transferred onto the poly(${\gamma}$-benzyl L-glutamate)(PBLG) or PBLG/ poly(ethylene glycol)(PEG)/PBLG Langmuir-Blodgett (LB) films as the monolayer level. The dichroic fluorescence values of confocal microscope indicated that the PVLA transferred onto the LB films was located with a preferential orientation of its molecular axes with regard to the direction of the a-helix of polypeptide. Hepatocyte recognized well-oriented galactose moieties of the surface of PVLA through asialoglycoprotein receptors.

  • Transforming growth factor (TGF)-${\beta}$, a hormonally active polypeptide found in normal and transformed tissue, is a potent regulator of cell growth and differentiation. In this study, we examined the effect of TGF-${\beta}$ on invasion and motility of MCF10A human breast epithelial cells. TGF-${\beta}$ induced migration and invasive phenotype of the parental MCF10A cells in a dose-dependent manner.(omitted)

Bacillus cellulyticus K-12 Crystalline Cellulose-Degrading Avicelase Gene and Expression in Eschterichia coli

  • We have cloned the Bacillus cellulyticus K-12 avicelase (Avi, E.C.3.2.1.4) gene (ace A) In E. coli. This was accompanied by using the vector PT7T3U 19 and Hind W -Hind m libraries of Bacillus cellulyticus K-12 chromosomal inserts created in 5.cofi. The Libraries were screened for the expression of avicelase by monitoring the immunoreaction of the anti-avicelase (immunoscreening). Positive clones (Ac-3, Ac-5, and Ac-7) contained the identical 3.5kb Hind III fragment as determined by restriction mapping and Southern hybridization, and expressed avicelase efficiently and constituvely using its own promoter in the heterologous host. From the immunoblotting analysis, a polypeptide which showed a CMCase activity with an Mr of 54000 was detected.

Assignment of the Carbonyl Carbon Resonances in Anti-Dansyl Antibodies (항 단실 항체의 카르보닐탄소 유래 시그날의 귀속)

  • The anti-dansyl antibodies were specifically labeled with stable isotope by growing hybridoma cells in serum-free medium. Assignments of the observed carbonyl carbon resonances have been determined by using $^{13}C-{15}N$ double labeling method in order to assign the Leu resonances. However, when the identical dipeptide appears more than twice in the polypeptide sequences, we applied the proteolytic fragments in the fragment-specific method. Carboxypep-tidase B-treated antibody has also been used to assign the Lys-447 in C terminal amino acid. These unambiguously assigned carbonyl carbon resonances in antibodies are thought to be useful in elucidating not only the structure of antibodies but also the structure-function relationship in the antibody by $^{13}C$ neuclear magnetic resonance spectroscopy.

Molecular Conformation-Dependent Complexation between Acidic- and Basic-Polypeptides via Hydrogen Bonding in Solution

  • Interpolymer complex formation between basic polypeptide poly(L-proline) Form Ⅱ (PLP(Ⅱ)) and acidic polypeptides poly(L-glutamic acid) (PLGA) and poly(L-aspartic acid)(PLAA) has been studied in water-methanol (1:2 v/v) mixed-solvent by viscometry, potentiometry, light scattering and circular dichroism (CD) measurements. It has been found that polymer complexes between PLP(Ⅱ) and PLGA (or PLAA) are formed via hydrogen bonding with a stoichiometric ratio of PLP(Ⅱ)/PLGA (or PLAA)=1:2 (in unit mole ratio) and that PLP(Ⅱ) forms polymer complex more favorably with PLGA than with PLAA. In addition, the minimum (for pH 5.0) and the maximum (for pH 3.2) in reduced viscosity of dilute PLP(Ⅱ)-PLGA mixed solutions are observed at 0.67 unit mole fraction of PLGA (i.e., [PLP(Ⅱ)]/[PLGA]=1/2). These findings could be explained in terms of molecular structure (or conformation) of the complementary polymers associated with the complex formation.

Chain Dimensions and Intrinsic Viscosities of Polypeptides in the Helix-Coil Transition Region

  • An equation is derived which correlates the unperturbed dimensions $_0$ of polypeptides with the helical contents in the helix-coil transition region by using a simple model of a polypeptide chain. The model is a chain of connected balls which represent the repeating units, -CO-NH-CHR-, based on the fact that the repeating unit has a plane structure. The changing trend of the expansion factor ${\alpha}_{\eta}$ in the transition region is connected with the helical content $f_H$. The intrinsic viscosities [${\eta}$] of polypeptides are calculated from the unperturbed dimensions and the ${\alpha}_{\eta}$ factors. The above calculated results concerning $_0$ and [${\eta}$] are compared with other authors' theoretical and experimental results. From the comparison, we concluded that our theory explains better the chain dimensional behavior of polypeptides in the helix-coil transition region than others.
