Optimization of Cookie Preparation by Addition of Polygonum multiflorum Radix Powder using Response Surface Methodology (반응표면분석법을 이용한 하수오 가루를 첨가한 쿠키의 제조조건 최적화)

  • The purpose of this study was to determine the optimal mixing ratios of three different ingredients of Polygonum multiflorum Radix powder, butter, and sugar for the development of recipe for cookies with Polygonum multiflorum Radix powder. Response surface methodology based on a five level and three variables by central composite design was employed to obtain the best possible combination for the ratios of Polygonum multiflorum Radix powder ($X_1$), butter ($X_2$), and sugar ($X_3$). The analytical results of the physical and mechanical properties for each sample including color L (P<0.01), color a (P<0.01), color b (P<0.01), spread ratio (P<0.001), and hardness (P<0.001) showed significant differences. The sensory measurements were significantly different in color (P<0.05), appearance (P<0.05), texture (P<0.001), flavor (P<0.01), taste (P<0.01), and overall quality (P<0.001). The optimal compositional ratios were determined to be 4.00 g for the Polygonum multiflorum Radix powder, 75.42 g for the butter, and 45.67 g for the sugar.

  • To examine the effects of Polygonum multiflorum on white rats with deteriorated immunity caused by methotrexate, first of all, methotrexate was fed to the rats once a day for 4 days. After the immune response of the rats are dereriorated, dried extracts of Polygonum multiflorum mixed in water was fed to the white rats once a day for 144days. The next conclusion was made by examining the rates of B-cells and T-cells of the peripheral blood and the changes in rates of CD4+ T-cells and CD8+ T-cell of the blood sampled from the spleen and peripheral region . Polygonum multiflorum has an effect of increasing immune responses on white rats with deteriorated immunity caused by MTX. Especially the count of CD4+ T-cells of the peripheral blood and the count of CD4+ T-cell of the spleen proved the significant effect o( increasing immune responses statistically . Verification of the effects of the Polygonum multiflorum should be made through comparitive .studies using various immune response indexes. Also additional studies for a modern and practicible application of Polygonum multiflorum seems to be needed.

  • Polygoni Multiflori Radix, the root of Polygonum multiflorum Thunb., has been used as a tonic, and in the inflammation and constipation in Korea. In order to evaluate the quality of Polygoni Multiflori Radix, the method of isolation and quantitative determination of emodin as a reference standard compound has been developed. Different specimens of Polygoni Multiflori Radix were collected from twenty Korean markets and were analyzed with HPLC using the mobile phase of MeOH-water (64:36). The average content of emodin from Polygoni Multiflori Radix in Korean markets was 0.021%.

Quantitative analysis of anthraquinones in Polygonum multiflorum Thunberg (하수오(何首烏)의 안트라퀴논 함량분석)

  • Objective: Polygonum multiflorum Thunberg (Polygonaceae) has been traditionally used as a tonic and the purgative in China and Korea. The genus Polygonum is a source of a wide range of phenolic compound, flavanoids, anthraquinones, stilbenes and tannins. In this study, three anthraquinones were isolated and quantitative determination of anthraquinones from P multiflorum has been developed for quality standardization. Methods : Three anthraquinone derivatives were isolated from a methanol extract of the radix of P. multiflorum by the chromatographic separation. Their structures were identified as emodin, physcion and ${\omega}$-hydroxyemodin on the basis of spectral data (MS, lH-NMR, 13C-NMR) and chemical analysis. HPLC analysis was performed to determine the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin in P. multiflorum from different specimens were collected from twenty Korean markets. Results: According to the results, the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin were 0.145%, 0.434%, 0.016%, 0.026%, 0.030% by HPLC, respectively. Conclusions : In these results, we have determined the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin in P. multiflorum, respectively. We hope that this study will contribute to the standardization and quality control of herbal medicine.

Pharmacognostical Studies on Ha-Su-O(Polygoni Radix) (하수오류(何首烏類)의 생약학적 연구)

  • Experiments were carried out to evaluate the pharmacognostical aspects of Polygoni Radix which consists of the roots of Polygonum multiflorum $T_{HUNBERG}$(Polygonaceae), Cynanchum wilfordii $H_{EMSLEY}$ (Asclepiadaceae) and Polygonum ciliinerve $(N_{AKAI})O_{WI}$ (Polygonaceae), since it was known to be effective as tonic, nutrient, etc. in oriental medicine. The histological features of the above three plants of Polygoni Radix were compared in view point of external and internal morphologies and their habitats. Also their constituents were compared on the basis of anthraquinone group. From the methanol extract of Polygonum ciliinerve $(N_{AKAI})O_{WI}$, furthermore, a specific product of this country, two anthraquinone derivatives were isolated. One of the substances was identified as emodin $(C_{15}H_{10}O_5,\;m.p.\;268-269^{\circ})$ through comparison of the authentic compound and spectroscopy.

A study on external and internal morphology in 4 kinds of Uie-Suel Radix (4종 우슬(牛膝)의 외내부형태 연구)

  • Objectives : This study was designed to establish a characteristic discrimination of internal and external morphological standard of original plants and herbal states in Polygoni Multiflori and Cynanchi Wilfordii Radix. Methods : In this studies, the external-internal morphological standards were determined by using stereoscope and butanol series. Results: 1. The external characteristics of original plants: Polygonum Multiflorum has alternate leaves, and its flower with white color blooms at the shoot apex or leaf axil. In the other hand, Cynanchum wilfordii has opposite leaves, and its flower with yellowish green color blooms at the leaf axil. 2. The physical characteristics of herbal slates: Polygoni Multiflori Radix is red-brown in outer surface pink-brown in section. In the other hand, Cynanchi Wilfordii Radix is earthly-brown in outer surface greyish white in section. 3. The physical characteristics according to the place of production: Demonstrable difference according to the place of production is not seen. Polygoni Multiflori Radix is brown or dark brown in outer surface, soft $Yellow{\sim}dark$ brown in section. In the other hand, Cynanchi Wilfordii Radix is white in outer surface, $white{\sim}weak$ red in section, and has power type. 4. The internal characteristics: Polygonum multiflorum has hetero-vascular bundle and lignification of cork layer is progressive. In the other hand, Cynanchum wilfordii has not hetero-vascular bundle, and lignification of cork layer is weak. Conclusion: In the future, many fundamental study such as how to discriminate between Polygoni Multiflori Radix and Cynanchi Wilfordii Radix in origin and efficacy will be necessary. Also the standard of discrimination must be specific and distinct in that several kinds of fo-ti has been traded. It is considered the results of this study will be furnish κ I the basis Lo succeeding studies and it is needed to extensive comparative study for the same genus-degree of relatedness.

Rapid molecular authentication of three medicinal plant species, Cynanchum wilfordii, Cynanchum auriculatum, and Polygonum multiflorum (Fallopia multiflorum), by the development of RAPD-derived SCAR markers and multiplex-PCR

  • Definitive identification of original plant species is important for standardizing herbal medicine. The herbal medicines Cynanchi Wilfordii Radix (Baekshuoh in Korean and Beishuwu in Chinese) and Polygoni Multiflori Radix (Hashuoh in Korean and Heshuwu in Chinese) are often misidentified in the Korean herbal market due to morphological similarities and similar names. Therefore, we developed a reliable molecular marker for the identification of Cynanchi Wilfordii Radix and Polygoni Multiflori Radix. We used random amplified polymorphic DNA (RAPD) analysis of three plant species, Polygoni multiflorum, Cynanchum wilfordii, and Cynanchum auriculatum, to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed six sequence characterized amplification region (SCAR) markers for distinguishing Polygoni Multiflori Radix and Cynanchi Wilfordii Radix. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. These SCAR markers can be used for efficient and rapid authentication of these closely related species, and will be useful for preventing the distribution of adulterants.

Promotive Effect of Polygonum multiflorum radix Ethanol Extract on Melanogenesis (적하수오 에탄올 추출물의 melanin 합성 촉진효과)

  • Hair color is determined by kind and amount of melanin. Melanocyte mainly synthesizes melanin from L-tyrosine by stimulation of ultra violet. Reactive oxygen species (ROS) play an important role in greying hair. Polygonum multiflorum radix has been reported to inhibit the aging process that black color of hair is turned into grey color. The aim of this study is to investigate the effect of Polygoni multiflorium radix ethanol extract (PMEE) on melanin synthesis related to black hair growth. In anti-oxidant experiment, PMEE decreased DPPH radical and increased reducing power, indicating that PMEE could eliminate ROS involved in greying hair. PMEE decreased cell viability in a dose-dependent manner. Furthermore, the effect of PMEE on the production of melanin was determined by DOPA assay and tyrosinase activity. PMEE increased tyrosinase activity and promoted melanin synthesis. In addition, the expression levels of tyrosinase, tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2) and microphthalmia-associated transcription factor (MITF), as well as anti-oxidant enzymes such as superoxide dismutase (SOD-3) and catalase were examined using western blot analysis. The expression levels of SOD-3 and catalase were decreased due to the enhanced antioxidant activity of PMEE. In particular, PMEE increased the expression levels of tyrosinase and TRP-2. These results suggest that PMEE could promote melanin synthesis that involved in tuning gray hair into black hair.

Antioxidant and antimicrobial activities of Jeok Hasuo (Polygonum multiflorum Thunb.) and Baek Hasuo (Cynanchi wilfordii Radix) root extracts (적하수오 및 백하수오 추출물의 항산화 및 항균 활성)

  • The objective of this research was to investigate the antioxidant and antimicrobial activities of Jeok Hasuo (Polygonum multiflorum Thunb., PM) and Baek Hasuo (Cynanchi wilfordii Radix, CW) root extracts. Total phenolic contents of PM and CW root extracts were determined and the antioxidant activities of the root extracts were determined by scavenging activity of diphenylpicrylhydrazyl (DPPH) radicals. The antimicrobial activities against Staphylococcus aureus and Propionibacterium acnes were determined and expressed as the minimum inhibitory concentration (MIC). The disc diffusion method was also used to determine the zone of inhibition. The butanol extracts of PM and CW roots exhibited greater total phenolic contents (1,212.6 and 1,454.5 mg/g GAE, respectively) than those of ethanol and water extracts. The ethanol (89.0%) and butanol extracts (88.9%) of PM exhibited significantly greater DPPH radical scavenging activities than that of water extracts (73.1%) (p<0.05). Only ethanol extract exhibited an MIC of 0.8 mg/mL against both bacteria. Zones of inhibition started to form when the concentration of extract was greater than 5 mg/disc. The diameters of the zone of inhibition of PM and CW were measured to be 8.9 and 9.2 mm against S. aureus and P. acnes, respectively, exhibiting the greatest antimicrobial activities among the extracts. This research demonstrated that the PM and CW root extracts possessed not only antioxidant activity but also strong antimicrobial activity against skin-related bacteria.

Active Compounds from The Aerial Parts of Polygonum multiflorum Thunberg (하수오 지상부의 활성 성분)

  • Polygoni Radix (Polygonaceae) has been used as renal disoder hematopoietic, menstrual irregularity and antiaging in Korean folk medicine. Specially aerial parts has been used for insomnia and sedative agent. In order to investigate the efficacy of antioxidant activity on these aerial parts, the bio-activity guided fraction and isolation of physiologically active substance were performed. H$_2$O, 30%, 60%, 100% MeOH were examined antioxidant activity by DPPH method. It was revealed that 30%, 60% MeOH fractions have significantly antioxidant activity. From 60% MeOH fraction, six known flavonoid glycosides (quercetin, quercetin 3-O-${\beta}$-D-galactopyranosyl-4'-O-glucopyrano side-3'-methyl ether, quercetin 3-O-${\alpha}$-L-rhannopyranoside, quercetin 3-O- ${\alpha}$-L-rhamnopyranoside-4'-methylethor, quorcetin 3-O-${\beta}$-D-galactopyranoside, quercetin 3-O-${\beta}$-D-galactopyranoside) and 3,4-Oihydrocinnarnic acid were isolated. To investigate the antioxidant activities of each com-pounds, we measured radical scavening activity with DPPH method and anti-lipid peroxidative efficacy on low density lipo-protein (LDL) with TBARS assay and anti-acetylcholinestrase activity with Ellman's Test. Four compounds (ll, IV Vl, Vll) of quercetin glycosides showed significant activity.
