Effect of Pretreatment and Holding Solution on Vase Life and Quality of Cut Polygonatum odoratum var. pluriflorum Stems (전처리와 보존용액이 무늬둥굴레 절지의 수명과 품질에 미치는 영향)

  • This study was conducted to examine the effect of several pretreatments and holding solutions on the vase life and quality of cut Polygonatum odoratum var. pluriflorum for. variegatum stems native to Korea. Postharvest foliar spray of 200 $mg{\cdot}L^{-1}$ $GA_3$ or 200 $mg{\cdot}L^{-1}$ BA + 200 $mg{\cdot}L^{-1}$ $GA_3$ markedly extended the vase life of cut stems of Polygonatum odoratum var. pluriflorum for. variegatum. These treatments maintained high chlorophyll contents as compared with the control (distilled water) during senescence of cut stems. Postharvest pretreatment (dipping) with 100 $mg{\cdot}L^{-1}$ $GA_3$ or 3% sucrose + 200 $mg{\cdot}L^{-1}$ 8-hydroxyquinoline sulfate (HQS) + 100 $mg{\cdot}L^{-1}$ $GA_3$ for 16 hours extended vase life of cut stems 2.5 times as long as nontreated control. Holding solution of 2% sucrose + 200 $mg{\cdot}L^{-1}$ HQS + 100 $mg{\cdot}L^{-1}$ $GA_3$ significantly extended vase life by 3.5 times and increased fresh weight as compared with control. $GA_3$ were very effective on preventing leaf yellowing of cut Polygonatum odoratum var. pluriflorum for. variegatum stems. Postharvest foliar spray of 200 $mg{\cdot}L^{-1}$ $GA_3$ or holding solution of 2% sucrose + 200 $mg{\cdot}L^{-1}$ HQS + 100 $mg{\cdot}L^{-1}$ $GA_3$ markedly extended vase life and improved quality of cut stems of Polygonatum odoratum var. pluriflorum for. variegatum.

Antioxidative Effect of Ethanolic Extracts of Some Tea Materials on Red Pepper Seed Oil (고추씨 기름에 대한 다류 에탄올 추출물의 항산화 효과)

  • Antioxidative effect of ethanolic extracts of various tea materials(Camellia sinensis, Cassia tora, Lyc ium chinense, Polygonatum odoratum, Schizandrae chinensis) on red pepper seed oil was investigated. Ethanolic extracts were added to red pepper seed oil at a concentration of 0.05%(w/v). Two experimental conditions were employed : 50$\pm$0.1oC for 45 days and 150$\pm$3oC for 24 hours. Oxidation of red pepper seed oil was determined by measuring peroxide value and acid value. Electron donating ability(EDA) and total phenolic contents of each extract were also determined. The result showed that the extracts possess an antioxidative activities. The effectiveness of them was in the following order: C. sinensis>C. tora>P. odoratum>S. chinensis >L. chinense. Ethanolic extracts of C. sinensis showed substantially higher EDA value and total phenol contents than other tea materials. These results indicate that the antioxidative effect was strongly related with EDA and total phenol contents.

  • Solomon's seal (Polygonatum odoratum) seedlings raised through its seeds can replace the rhizomes impelling higher cost for transplanting, This experiment was done to determine the seed characteristics and the germinating processes to give some information on bulk production of seedlings using the seeds. The external or internal morphology of seeds or seedlings grown in lab. or greenhouse was examined mainly with stereomicroscope. The external shape of Solomon's seal seed was hard seed-coat and orthotropous ovule with linear type embryo stretching to the center of seed. Germination proceeded through the several steps. The lower part of seed embryo having the primordia of bulbil and roots first grew before the bulbil and roots was developed from the primordia. The lower part of embryo was enlarged toward the endosperm of seed as soon as seed germinated. Then epicotyl was formed on the apex of bulbil. The epicotyl was elongated after at least 6-week chilling treatment for breaking its dormancy and the first leaf shape was affected by light intensity given during seedling emergence. The bulbil was the first organ of the rhizome used as tea or herb medicine.

In Vitro Micropropagation of Polygonatum odoratum cv. Gungangbeaksea (둥굴레 '건강백세'의 기내 대량번식)

  • The Polygonatum odoratum cv. Gungangbeaksea, bred in Gyeongsangnam-Do Agricutural Research & Extension Service, was cultured in vitro for micropropagate rapidly through the culture of rhizome explants ($5{\times}5mm$). The $7{\times}7mm$ explants of adventitious multi-bud clusters (AMC), obtained through the culture of rhizome explants (MS + 3.0 mg/L BA) were cultured on MS media with BA and TDZ. The shoot multiplication was favorable on the MS medium containing 3.0 mg/L TDZ with 2.8 in shoot number. But the formation of AMC was low in all media tested. The explants of AMC were cultured on MS media containing 1.0~5.0 mg/L TDZ and NAA to multiplicate AMC more. The formation of AMC was a little more stimulated on combined MS media of TDZ and NAA, than that with TDZ alone. The multiplication of shoots and AMC was favorable on MS media with 3.0 mg/L TDZ and 5.0 mg/L NAA, and 5.0 mg/L TDZ and 3.0 mg/L NAA. As the concentration of MS salts increased, the formation of AMC was decreased. But the formation of AMC was more stimulated, as the concentration of sucrose increased to 7%. Therefore, the multiplication of shoots and AMC was suitable on media containing 3.0~5.0 mg/L TDZ and NAA, and 7% of sucrose. The explants of AMC were rooted on media with 3.0 mg/L IBA, or 2.0 mg/L NAA with more than 80% in rooting ratio. The plantlets were treated at $5^{\circ}C$ for 8 weeks, and cultured ex vitro for 8 weeks. The survival ratio of plantlets were 100% in vermiculite, and the mixed soil with perlite 1 volumn and vermiculite 1 volumn.

Antioxidative and Nitrite Scavenging Activities of Polygonatum odoratum Root Extracts with Different Steaming and Roasting Conditions (둥굴레 근경의 증자 및 볶음조건에 따른 추출물의 항산화성 및 아질산염 소거능 변화)

  • Response surface methodology (RSM) was applied to monitor the effects of steaming and roasting conditions of Polygonatum odoratum roots an total phenolics content, electron donating ability (EDA) and nitrite-scavenging ability (NSA) of the extract. In steaming and roasting processes based on the central composite design. with variations in steaming time $(60\~180\;min)$, roasting temperature $(110\~150\;min)$ and roasting time $(10\~50\;min)$, coefficients of determinations $(R^2)$ were 0.9356 (p<0.01) in total phenolics, 0.9578 (p<0.01) in EDA and 0.9436 (p<0.01) in NSA (pH 3.0). The maximum value of total phenolics was $2847.67\;mg\%$ at 135.59 min of steaming time, $143.84^{\circ}C$ of roasting temperature and 43.47 min of roasting time. The maximum value of EDA was $75.00\%$ in 108.98 min, $135.56^{\circ}C$ and 48.86 min. The maximum value of NSA (pH 3.0) was $87.38\%$ in 162.80 min, $143.88^{\circ}C$ and 31.97 min, respectively. Total phenolics content of the extract was influenced by heating conditions in the order of roasting temperature, steaming time and roasting time. While EDA and NSA were appreciably influenced by roasting time, followed by roasting temperature and steaming time.

Brown Leaf Spot of Doonggulle (Polygonatum odoratum) Caused by Phyllosticta cruenta (Phyllosticta cruenta에 의한 둥굴레 갈색점무늬병)

  • Brown leaf spot caused by Phyllosticta cruenta from 2007 to 2009 occurred severely in an herbs exhibition on Doonggulle (Polygonatum odoratum) at Gangju pond in Jeongchon-myon, Jinju city, Gyeongnam province, Korea. The typical symptoms of this disease were appeared on the brown leaf spot of leaves. Lesions on the leaves were appeared elliptical or irregular with 2~8 mm in size. Infected leaves were wilted, blighted and eventually died. Pycnidiospores and pycnidia of the fungus were abundantly formed on PDA. The optimum temperature for mycelial growth was $20{\sim}{25^{\circ}C}$. Pycnidiospores were not readily formed on PDA, colorless in color, globose~subglobose in shape, $10{\sim}18{\times}5{\sim}12{\mu}m$ in size and appendage hanged $4{\sim}15{\mu}m$ in size. Pycnidia were globose~subglobose in shape, dark brown in color and $60{\sim}162{\times}70{\sim}174{\mu}m$ in size. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Phyllosticta cruenta (Kunze: Fries) Kickx. This is the first report on the brown leaf spot of Doonggulle caused by P. cruenta in Korea.

Optimization of Steaming and Roasting Conditions for Maximized Hypoglycemic Properties of Polygonatum odoratum Tea (둥굴레차의 혈당강하 성분을 극대화시킬 수 있는 증자 및 볶음조건의 최적화)

  • Response surface methodology (RSM) was applied in steaming and roasting processes of Polygonatum odoratum roots in order to monitor hypoglycemic components and sensory property. In steaming and roasting processes based on the central composite design with variations in steaming time $(60\~180\;min)$, roasting temperature $(110\~150^{\circ}C)$ and roasting time $(10\~50\;min)$, coefficients of determinations $(R^2)$ were 0.8691 (p<0.05), 0.8253 (p<0.l0), 0.8727 (p<0.05), 0.8706 (p<0.05) and 0.8316 (p<0.10) in soluble solid, stigmasterol, $\beta-sitosterol$, hypoglycemic component (total), and overall acceptability, respectively. The maximum value of soluble solid was $71.47\%$ in 65.24 min of steaming time, $126.93^{\circ}C$ of roasting temperature and 37.58 min of roasting time. The maximum value of hypoglycemic component (total) was $764.10\;{\mu}g/g$ in 107.76 min, $117.78^{\circ}C$ and 14.70 min. Meantime, the maximum value of overall acceptability was 6.89 in 126.04 min, $115.79^{\circ}C$ and 43.93 min. The predicted values in optimum conditions for hypoglycemic components and sensory property were in good agreement with experimental values.

Effects of Seed-coat Softening and Prechilling on Seed Germination and Bulbil Formation of Polygonatum odoratum Druce (둥굴레 종자의 발아(發芽) 및 소주아(小珠芽) 형성(形成)에 대한 저온(低溫)과 종피연화(種皮軟化) 처리(處理)의 영향(影響))

  • Solomon's seal (Polygonatum odoratum) was traditionally propagated by its rhizomes but seed propagation has not been practiced due to their dormancy and high price. To improve the seedling production through seed pretreatments, the experiment was conducted to determine the effect of seedcoat softening and seed prechilling on seed germination and bulbil formation of the seal. The prechilling was done for 4 to 16 weeks at $3^{\circ}C$ and the softening was forced with 10% KOH for 40 minutes. Seed germination, bulbil formation and length, no. of root and root length were measured at 6 and 12 weeks after sowing. Seed germination and bulbil formation were greater at 8 to 12 week prechilling than at shorter or longer treatment when only the prechilling was enforced. Prechilling had greater effect compared to seed coat softening. In addition, the softening treatment before prechilling showed higher germination and bulbil formation compared to single treatment of them. The softening treatment prior to prechilling was better than the reverse treatment in number of root except the above two characters. Duration of prechilling treatment was able to be shortened below 8 weeks when the softening treatment was applied before prechilling.

Effect of Gibberellin and Chilling Treatments on Breaking Epicotyl Dormancy and Seedling Growth of Polygonatum odoratum Druce (둥굴레의 상배축(上胚軸) 휴면타파(休眠打破)와 유묘생장(幼苗生長)에 대한 지베렐린 및 저온 처리효과)

  • Seed-propagation of Solomon's seal (Polygonatum odoratum) takes 2 years to shorten the period for becoming a practical method. The experiment was done to establish a proper method of breaking the epicotyl dormancy in bulk seed-propagation. Seedlings with a bulbil were treated with $GA_3$ every 2 days for 4 or 8 days and chilling treatments at $3^{\circ}C$ were enforced for 4, 6, 8 or 12 weeks. Emergence- and growth-related characteristics were examined immediately after the treatments, 3 and 6 weeks later. Rate of cotyledonary sheath rupture immediately after $GA_3$ treatment was greater in its 8-day treatment than in 4-day although its effect disappeared later. However, any epicotyl treated with $GA_3$ solution did not elongate so that new seedlings disemerged over the bed soil. That resulted from not breaking the epicotyl dormancy since $GA_3$ did not rupture all of the cotyledonary sheath formed with several sheets and consequently, the solution did not reach it. The $GA_3$ treatment for bulk seed-propagation, therefore, was impractical method. On the contrary, the chilling treatment was able to be applied to the seed-propagation because of getting the cotyledonary sheath rupture and the epicotyl elongation. Seedling emergence and its growth after chilling treatment were influenced by chilling period although required at least over 6-week treatment for satisfactory results.
