Micropropagation of Plants and Mass Production of Adventitious Roots from Culture of Seedling Explants of Polygonatum odoratum

  • When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino,especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.

Effect of Pretreatment and Holding Solution on Vase Life and Quality of Cut Polygonatum odoratum var. pluriflorum Stems (전처리와 보존용액이 무늬둥굴레 절지의 수명과 품질에 미치는 영향)

  • This study was conducted to examine the effect of several pretreatments and holding solutions on the vase life and quality of cut Polygonatum odoratum var. pluriflorum for. variegatum stems native to Korea. Postharvest foliar spray of 200 $mg{\cdot}L^{-1}$ $GA_3$ or 200 $mg{\cdot}L^{-1}$ BA + 200 $mg{\cdot}L^{-1}$ $GA_3$ markedly extended the vase life of cut stems of Polygonatum odoratum var. pluriflorum for. variegatum. These treatments maintained high chlorophyll contents as compared with the control (distilled water) during senescence of cut stems. Postharvest pretreatment (dipping) with 100 $mg{\cdot}L^{-1}$ $GA_3$ or 3% sucrose + 200 $mg{\cdot}L^{-1}$ 8-hydroxyquinoline sulfate (HQS) + 100 $mg{\cdot}L^{-1}$ $GA_3$ for 16 hours extended vase life of cut stems 2.5 times as long as nontreated control. Holding solution of 2% sucrose + 200 $mg{\cdot}L^{-1}$ HQS + 100 $mg{\cdot}L^{-1}$ $GA_3$ significantly extended vase life by 3.5 times and increased fresh weight as compared with control. $GA_3$ were very effective on preventing leaf yellowing of cut Polygonatum odoratum var. pluriflorum for. variegatum stems. Postharvest foliar spray of 200 $mg{\cdot}L^{-1}$ $GA_3$ or holding solution of 2% sucrose + 200 $mg{\cdot}L^{-1}$ HQS + 100 $mg{\cdot}L^{-1}$ $GA_3$ markedly extended vase life and improved quality of cut stems of Polygonatum odoratum var. pluriflorum for. variegatum.

Classification of Polygonatum spp. Collections Based on Multivariate Analysis (다변량 분석에 의한 둥굴레속 식물의 분류)

  • This study was conducted to obtain the basic data for practical use of the Polygonatum genetic resources. The 20 collections were analyzed by principal component analysis of 8 characters and cluster analysis. In the principal analysis, the first, the second and the third components contributed 54.10%, 18.95% and 11.62% of the variations, respectively. The cumulative contribution from the first to the third principal components was 84.68%. The first principal component was related to shape and size of plant, and assimilatory, reserve and reproductive organs. The second principal component was related to growth and development of plant, and reserve organ. And the third principal component was related to growth and development of plant. Based on cluster analysis, the 20 collections were classified into 4 distinct groups with the average distance greater than 0.7 between groups. Group I was Polygonatum sibiricum $D_{ELAR}$ and Group II included P. odoratum var. pluriflorum $O_{HWI}$, P. odoratum var. pluriflorum $O_{HWI}$ for 'Variegatum' Y. Lee, for. nov., P. odoratum var. thunbergii $H_{ARA}$ and P. odoratum var. maximowiczii $K_{OIDZ}$. GroupIII was P. involucratum $M_{AXIM}$, P. desoulavyi $K_{OMAROV}$ and P. humile $F_{ISHER}$ ex. $M_{AXIM}$. And GroupIV included P. lasianthum var. coreanum $N_{AKAI}$ and P. inflatum $K_{OMAROV}$.

Effects of Polygonatum sibiricum rhizome extract on lipid and energy metabolism in high-fat diet-induced obese mice (고지방 식이 유도 비만 마우스 모델에서 황정 추출물의 지방질 및 에너지 대사 관련 유전자에 대한 효능 연구)

  • In this study, factors involved in lipid and energy metabolism following treatment with ethanolic extract of the Polygonatum sibiricum rhizome (ID1216) were evaluated in high-fat diet-induced obese mice. ID1216-treated mice showed a significant reduction in weight gain compared to non-treated mice. ID1216 treatment increased the protein levels of AMP-dependent protein kinase, sirtuin1, peroxisome proliferator-activated receptor ${\gamma}$ coactivator 1-${\alpha}$ ($PGC1{\alpha}$), peroxisome proliferator-activated receptor ${\alpha}$ ($PPAR{\alpha}$) and uncoupling proteins in the adipose tissue, liver and muscle compared to vehicle treatment. Analysis of downstream signals of the sirtuin1 $PGC1{\alpha}$-$PPAR{\alpha}$ pathway showed that ID1216 regulates the expression of ${\beta}$-oxidation related genes such as acyl-CoA oxidase, carnitine palmitoyltransferase1, acyl-CoA dehydrogenase and adipocyte protein 2. In addition, ID1216 increased the expression of adipose triglyceride lipase. These results suggest that ID1216 has anti-obesity effects by regulating the genes involved thermogenesis, ${\beta}$-oxidation and lipolysis in a diet-induced obesity model.

Characteristics of Seed Structure and Seedling Development in Polygonatum odoratum Druce (둥굴레의 종자(種子) 구조(構造)와 출아(出芽) 특성(特性))

  • Solomon's seal (Polygonatum odoratum) seedlings raised through its seeds can replace the rhizomes impelling higher cost for transplanting, This experiment was done to determine the seed characteristics and the germinating processes to give some information on bulk production of seedlings using the seeds. The external or internal morphology of seeds or seedlings grown in lab. or greenhouse was examined mainly with stereomicroscope. The external shape of Solomon's seal seed was hard seed-coat and orthotropous ovule with linear type embryo stretching to the center of seed. Germination proceeded through the several steps. The lower part of seed embryo having the primordia of bulbil and roots first grew before the bulbil and roots was developed from the primordia. The lower part of embryo was enlarged toward the endosperm of seed as soon as seed germinated. Then epicotyl was formed on the apex of bulbil. The epicotyl was elongated after at least 6-week chilling treatment for breaking its dormancy and the first leaf shape was affected by light intensity given during seedling emergence. The bulbil was the first organ of the rhizome used as tea or herb medicine.

Effects of ethanol extract of Polygonatum sibiricum rhizome on obesity-related genes (황정 에탄올 추출물의 비만 조절 유전자에 대한 효과)

  • In previous studies, we confirmed that the ethanol extract of Polygonatum sibiricum (ID1216) has anti-obesity effects on high-fat diet-fed mice. To identify the obesity-related genes affected by ID1216, we studied its effects both in vivo and in vitro. In mice, single administration of ID1216 increased the expression of obesity-related genes including sirtuin1 (SIRT1), peroxisome proliferator-activated receptor ${\gamma}$ coactivator $1{\alpha}$ ($PGC1{\alpha}$) and peroxisome proliferator-activated receptor ${\alpha}$ ($PPAR{\alpha}$) compared to that in mice administered the vehicle; their downstream genes (uncoupling proteins, acyl-CoA oxidase, adipocyte protein 2, and hormone-sensitive lipase) were also increased by ID1216. In fully differentiated 3T3-L1 adipocytes, ID1216 showed the same effects on anti-obesity genes as those in the animal model. Based on these results, we propose that ID1216 has anti-obesity effects by regulating the $SIRT1-PGC1{\alpha}-PPAR{\alpha}$ pathway and their downstream genes, thereby controlling energy and lipid metabolisms.

Changes in Flavor Compounds of Polygonatum odoratum Root during Roasting (볶음처리에 따른 둥굴레 근경의 향기성분 변화)

  • In order to evaluate innate flavor during the roasting process, the components responsible for the aroma of unroasted Polygonatum odoratum root and the volatile odor components released during the roasting process were evaluated using the solid phase-microextraction (SPME) headspace method. The raw P. odoratum root contained aldehyde, alcohol, hydrocarbon and acid components, which gave it a grass-like smell. 2-Methylpyrazine was not revealed from the roasted P. odoratum root at $110^{\circ}C$ and $130^{\circ}C$, but was extracted at $150^{\circ}C$. The 2,5-dimethylpyrazine component was the greatest at $130^{\circ}C$. The hexanal component was greater when the root was roasted at $110^{\circ}C$, compared with those at $130^{\circ}C$ and $150^{\circ}C$. The production of 2-acetyl-1-pyrroline component was the greatest when it was roasted at $130^{\circ}C$. Quantitative descriptive analysis of P. odoratum after roasting revealed a high concentration of 2,5-dimethylpyrazine at $130^{\circ}C$ and $150^{\circ}C$, but a low concentration at $110^{\circ}C$. Most of the 2-acetyl-1-pyrroline, which significantly affected its smell, was revealed at $130^{\circ}C$.

In Vitro Micropropagation of Polygonatum odoratum cv. Gungangbeaksea (둥굴레 '건강백세'의 기내 대량번식)

  • The Polygonatum odoratum cv. Gungangbeaksea, bred in Gyeongsangnam-Do Agricutural Research & Extension Service, was cultured in vitro for micropropagate rapidly through the culture of rhizome explants ($5{\times}5mm$). The $7{\times}7mm$ explants of adventitious multi-bud clusters (AMC), obtained through the culture of rhizome explants (MS + 3.0 mg/L BA) were cultured on MS media with BA and TDZ. The shoot multiplication was favorable on the MS medium containing 3.0 mg/L TDZ with 2.8 in shoot number. But the formation of AMC was low in all media tested. The explants of AMC were cultured on MS media containing 1.0~5.0 mg/L TDZ and NAA to multiplicate AMC more. The formation of AMC was a little more stimulated on combined MS media of TDZ and NAA, than that with TDZ alone. The multiplication of shoots and AMC was favorable on MS media with 3.0 mg/L TDZ and 5.0 mg/L NAA, and 5.0 mg/L TDZ and 3.0 mg/L NAA. As the concentration of MS salts increased, the formation of AMC was decreased. But the formation of AMC was more stimulated, as the concentration of sucrose increased to 7%. Therefore, the multiplication of shoots and AMC was suitable on media containing 3.0~5.0 mg/L TDZ and NAA, and 7% of sucrose. The explants of AMC were rooted on media with 3.0 mg/L IBA, or 2.0 mg/L NAA with more than 80% in rooting ratio. The plantlets were treated at $5^{\circ}C$ for 8 weeks, and cultured ex vitro for 8 weeks. The survival ratio of plantlets were 100% in vermiculite, and the mixed soil with perlite 1 volumn and vermiculite 1 volumn.

Antioxidative and Nitrite Scavenging Activities of Polygonatum odoratum Root Extracts with Different Steaming and Roasting Conditions (둥굴레 근경의 증자 및 볶음조건에 따른 추출물의 항산화성 및 아질산염 소거능 변화)

  • Response surface methodology (RSM) was applied to monitor the effects of steaming and roasting conditions of Polygonatum odoratum roots an total phenolics content, electron donating ability (EDA) and nitrite-scavenging ability (NSA) of the extract. In steaming and roasting processes based on the central composite design. with variations in steaming time $(60\~180\;min)$, roasting temperature $(110\~150\;min)$ and roasting time $(10\~50\;min)$, coefficients of determinations $(R^2)$ were 0.9356 (p<0.01) in total phenolics, 0.9578 (p<0.01) in EDA and 0.9436 (p<0.01) in NSA (pH 3.0). The maximum value of total phenolics was $2847.67\;mg\%$ at 135.59 min of steaming time, $143.84^{\circ}C$ of roasting temperature and 43.47 min of roasting time. The maximum value of EDA was $75.00\%$ in 108.98 min, $135.56^{\circ}C$ and 48.86 min. The maximum value of NSA (pH 3.0) was $87.38\%$ in 162.80 min, $143.88^{\circ}C$ and 31.97 min, respectively. Total phenolics content of the extract was influenced by heating conditions in the order of roasting temperature, steaming time and roasting time. While EDA and NSA were appreciably influenced by roasting time, followed by roasting temperature and steaming time.
