Preparation of PEGDA/PETEDA Dendrimer Membranes for $CO_2$ Separation ($CO_2$ 분리를 위한 PEGDA/PETEDA dendrimer 막의 제조)

  • PEGDA/PETEDA dendrimer composite membranes was prepared by UV photopolymerizing of poly ethylene glycol diacrylate (PEGDA) containing 5~15 wt% pentaerythrityl tetraethylenediamine (PETEDA) dendrimer. The prepared composite membrane was characterized by FT-IR, $^1H$-NMR and DSC. The glass transition temperature ($T_g$) of PEGDA/PETEDA dendrimer composite decreased with the increment of PETEDA dendrimer content. The $CO_2$ separation properties over $CH_4$ were investigated by changing the PETEDA dendrimer content and pressure. The composite membrane containing 10 wt% PETEDA dendrimer exhibited on excellent $CO_2/CH_4$ ideal selectivity of 31.8 and a $CO_2$ permeability of 162.2 barrer.

자기조합형 세리신-PEG 나노입자의 합성과 특성

  • 세리신은 누에고치의 고치실에서 추출되는 천연 단백질로서 많은 수산화기를 가지고 있어 뛰어난 보습성을 가진다. 이에 화장품에의 응용이 기대되나, 물에 잘 녹지 않는 특성을 가지고 있어서 화장품 원료로 사용하기에 어렵다는 단점이 있다. 본 연구에서는 친양쪽성 화합물인 poly(ethylene glycol)(PEG)을 세리신에 도입하여 수용성이며 자기조합형(self-assembled) 세리신-PEG 결합체를 합성하였다. 세리신내의 tyrosine 잔기의 방향 족 수산화기가 반응사이트임을 $^1$H-NMR 분석으로 알 수 있었으며, IR과 CD 측정으로 PEG 사슬의 도입으로 세리신의 구조가 불규칙한 coil구조에서 $\beta$-sheet구조로 구조적 변화가 일어남을 알 수 있었다. 또한 DSC 분석에서 세리신-PEG결합이후 각각의 녹는점이 떨어지는 것을 확인하여 상호 결정성에 영향을 주는 것을 확인하였다. 세리신-PEG 결합체는 자기조합하여 소수성 상호작용을 통해 200-400nm의 구형 나노입자를 이루며 투석방법을 통하여 얻을 수 있었다. 또한, 세리신-PEG 나노입자는 세리신자체보다 더 높은 보습력을 가지는 것을 확인하였다. 세리신-PEG 결합체의 cytotoxicity는 MTT assay에 의해 독성이 없음을 확인하였으며, 동물실험에 의한 독성도 전혀 나타내지 않음을 알 수있었다. 나노입자의 특성과 세리신의 보습성, 세리신-PEG의 친양쪽성 특성으로 세리신-PEG결합체 나노입자는 그 자체로도 원료로 사용 될 수 있을 뿐 아니라 생리 활성성분의 운반체로의 개발이 기대되어 진다.

Impact of carbon dioxide on the stability of the small-scale structures by trapping the material properties

  • The existence of active material in the environment causes the small-scale systems to be sensitive to the actual environment. Carbon dioxide is one of the active materials that exists a lot in the air conditions of the living environment. However, in some applications, the carbon dioxide-coated is used to improve the performance of systems against the destructive factors such as the corrosion; nevertheless, in the current research, the stability analysis of a carbon dioxide capture mechanism-coated beam is investigated according to the mathematical simulation of a rectangular composite beam utilizing the modified couple stress theory. The composite mechanism of carbon dioxide trapping is made of a polyacrylonitrile substrate that supports a cross-link polydimethylsiloxane gutter layer as the carbon dioxide mechanism trapping. Three novel types of carbon dioxide trapping mechanism involving methacrylate, poly (ethylene glycol) methyl ether methacrylate, and three pedant methacrylates are considered, which were introduced by Fu et al. (2016). Finally, according to introducing the methodology of carbon dioxide (CO2) trapping, the impact of various effective parameters on the stability of composite beams will be analyzed in detail.

Different Influences of Biotinylation and PEGylation on Cationic and Anionic Proteins for Spheroid Penetration and Intracellular Uptake to Cancer Cells

  • To better understand the effects of PEGylation and biotinylation on the delivery efficiency of proteins, the cationic protein lysozyme (LZ) and anionic protein bovine serum albumin (BSA) were chemically conjugated with poly(ethylene glycol) (PEG) and biotin-PEG to primary amine groups of proteins using N-hydroxysuccinimide reactions. Four types of protein conjugates were successfully prepared: PEGylated LZ (PEG-LZ), PEGylated BSA (PEG-BSA), biotin-PEG-conjugated LZ (Bio-PEG-LZ), and biotin-PEG-conjugated BSA (Bio-PEG-BSA). PEG-LZ and Bio-PEG-LZ exhibited a lower intracellular uptake than that of LZ in A549 human lung cancer cells (in a two-dimensional culture). However, Bio-PEG-BSA showed significantly improved intracellular delivery as compared to that of PEG-BSA and BSA, probably because of favorable interactions with cells via biotin receptors. For A549/fibroblast coculture spheroids, PEG-LZ and PEG-BSA exhibited significantly decreased tissue penetration as compared with that of unmodified proteins. However, Bio-PEG-BSA showed tissue penetration comparable to that of unmodified BSA. In addition, citraconlyated LZ (Cit-LZ) showed reduced spheroid penetration as compared to that of LZ, probably owing to a decrease in protein charge. Taken together, chemical conjugation of targeting ligands-PEG to anionic proteins could be a promising strategy to improve intracellular delivery and in vivo activity, whereas modifications of cationic proteins should be more delicately designed.

Evaluation of continuous cultivation of anaerobic ammonium oxidation bacteria immobilized on synthetic media and granular form (입상형태와 합성담체에 고정화된 혐기성 암모늄 산화균의 연속배양 특성 평가)

  • The activity of anaerobic ammonium oxidation (ANAMMOX) immobilized in synthetic media (Poly Ethylene Glycol, PEG) and granular form was evaluated comparatively to investigate the effect of influent nitrogen concentration and exposure of oxygen. In ANAMMOX granule reactor, when concentration of influent total nitrogen increased to 500mg/L, removal efficiency of ammonium, nitrite and nitrate were shown to 90.5±6.5, 96.6±4.9, and 93.2±6.1%, respectively. In the case of the PEG gel, it showed lower nitrogen removal performance, resulting in that the removal efficiency of ammonium, nitrite and nitrate were shown to 83.3±13.0, 96.4±6.1, and 90.3±7.5%, respectively. In second step, when exposed to oxygen, the nitrogen removal performance in the ANAMMOX granule reactor also remained stable, but the activity of PEG gel ANAMMOX was found to be inhibited. Consequently, the PEG gel ANAMMOX was a higher sensitivity than that of granular ANAMMOX with two variables applied in this study.

Partitioning of Recombinant Human Interleukin-2 in a Poly(ethylene glycol)-Dextran Aqueous Two-Phase System

  • The partitioning of recombinant human interleukin-2(rhII-2) in PEG 8000-dextran 38800 aqueous two-phase system has been investigated using three different sources of rhIL-2. In the case of pure rhIL-2, the solubility in a PEG-dextran two-phase system was low and most of rhIL-2 was partitioned into the bottom phase. For the recovery of rhIL-2 from insoluble protein aggregates, the inclusion bodies of recombinant E. coli were solubilized by the treatment with sodium dodecyl sulfate (SDS). The addition of SDS significantly enhanced not only the solubility of rhIL-2 but also the partitioning of rhIL-2 to the top phase. When the ratio of SDS to rhIL-2 was 2.0, the partition coefficient(K) and the recovery yield(Y) at the top phase were 4.5 and 88%, respectively, at pH 6.8. In order to reduce the recovery steps further, SDS was directly added to the intact recombinant E. coli cells and then partitioned into the PEG/dextran aqueous two-phase system. The observed partition coefficient ($K{\cong{3.0$) and recovery yield ($Y{\geq}80%$ )of this method were comparable to the rhIL-2 recovery from insoluble protein aggregates. The results obtained in this work indicate that PEG-dextran two-phase partitioning might provide a simple way for the recovery and partial purification of recombinant proteins which are produced as inclusion bodies.

Preparation and Application of Polyurethane-urea Microcapsules Containing Phase Change Materials

  • For thermal adaptable fabrics, the polyurethane-urea microcapsules containing phase-change materials (PCMs: hexadecane, octadecane and eicosane) were successfully synthesized by interfacial polycondensation using 2,4-toluene diisocyanate (TDI)/poly(ethylene glycol) (PEG400)/ethylene diamine (EDA) as shell monomers and nonionic surfactant NP-12 in an emulsion system under stirring rates of $3,000{\sim}13,000$ rpm. The mean particle size of microcapsule decreased significantly with increasing the stirring rate up to 11,000 rpm, and then leveled off. The mean particle size increased with increasing the content and molecular weight (eicosane > octadecane > hexadecane) of PCMs at the same stirring rate. The mean particle sizes of microcapsules were found to decrease with increasing the NP-12 content up to 1.5 wt%, and thereafter increased a little. It was found that the melting temperature ($T_m$) and crystallization temperature ($T_c$) of three kinds of encapsulated PCMs and their enthalpy changes (${\Delta}H_m,{\Delta}H_c$) increased with increasing PCM contents. The encapsulation efficiencies (Ee) of hexadecane microcapsule linearly increased with increasing the content of hexadecane. It was found that the stable microcapsule containing 50 wt% of hexadecane could be obtained in this study. However, Ee of octadecane and eicosane microcapsules increased with increasing PCM's contents up to 40 wt%, and then decreased a little. By considering the encapsulation efficiency, it was found that the maximum/optimum contents of octadecane and eicosane microcapsules were about 40 wt%. By the dynamic thermal performance test, it was found that the maximum buffering levels of Nylon fabrics coated with hexadecane, octadecane, and eicosane microcapsules were about $-2.4/+2.9^{\circ}C,\;-3.6/+3.6^{\circ}C\;and\;-4.0/+4.7^{\circ}C$, respectively.

Cervical Cancer Gene Therapy by Gene Loaded PEG-PLA Nanomedicine

  • Background and Aims: Advances in the treatment of cervical cancer over the last decade have predominantly involved the development of genes directed at molecular targets. Gene therapy is recognized to be a novel method for the treatment of cervical cancer. Genes can be administered into target cells via nanocarriers. This study aimed to develop systemically administrable nano-vectors. Floate (Fa) containing gene loaded nanoparticles (NPs) could target HeLa human cervical cancer cells through combination with receptors on the cells to increase the nuclear uptake of genetic materials. Methods: Fa was linked onto Poly (ethylene glycol)-b-poly (D, L-lactide) (PEG-PLA) to form Fa-PEG-PLA, and the resulting material was used to load plasmids of enhanced green fluorescence protein (pEGFP) to obtain gene loaded nanoparticles (Fa-NPs/DNA). Physical-chemical characteristics, in vitro release and cytotoxicity of Fa-NPs/DNA were evaluated. The in vitro transfection efficiency of Fa-NPs/DNA was evaluated in HeLa cells and human umbilical vein endothelial cells (HUVEC). PEG-PLA without Fa was used to load pEGFP from NPs/DNA as a control. Results: Fa-NPs/DNA has a particle size of 183 nm and a gene loading quantity of 92%. After 72h of transfection, Fa-NPs/DNA displayed over 20% higher transfection efficiency than NPs/DNA and 40% higher than naked DNA in HeLa cells. However, in HUVECs, no significant difference appeared between Fa-NPs/DNA and NPs/DNA. Conclusions: Fa-PEG-PLA NPs could function as excellent materials for gene loading. This nano-approach could be used as tumor cell targeted medicine for the treatment of cervical cancer.

Preparation and Evaluation of Chrysin Encapsulated in PLGA-PEG Nanoparticles in the T47-D Breast Cancer Cell Line

  • Background: Polymeric nanoparticles are attractive materials that have been widely used in medicine for drug delivery, with therapeutic applications. In our study, polymeric nanoparticles and the anticancer drug, chrysin, were encapsulated into poly (D, L-lactic-co-glycolic acid) poly (ethylene glycol) (PLGA-PEG) nanoparticles for local treatment. Materials and Methods: PLGA: PEG triblock copolymers were synthesized by ring-opening polymerization of D, L-lactide and glycolide as an initiator. The bulk properties of these copolymers were characterized using 1H nuclear magnetic resonance spectroscopy and Fourier transform infrared spectroscopy. In addition, the resulting particles were characterized by scanning electron microscopy. Results: The chrysin encapsulation efficiency achieved for polymeric nanoparticles was 70% control of release kinetics. The cytotoxicity of different concentration of pure chrysin and chrysin loaded in PLGA-PEG ($5-640{\mu}M$) on T47-D breast cancer cell line was analyzed by MTT-assay. Conclusions: There is potential for use of these nanoparticles for biomedical applications. Future work should include in vivo investigation of the targeting capability and effectiveness of these nanoparticles in the treatment of breast cancer.

Structural Characterization of Branched Polyesters Using TOF-SIMS Combined with Transesterification

  • Mass spectrometry technique provides the molecular weight distribution, data on the sequence of repeat units, polymer additives, and impurities, and structural information. time-of-Flight secondary Ion Mass Spectrometry (TOF-SIMS) has been used for structural characterization of various polymers1-2. the masses of repeat units and terminal groups and molecular weight distributions of polymers have been determined from their TOF-SIMS spectra. TOF-SMIS provides good sensitivity and structural specificity for high mass ions so that intact oligomers and large polymer fragments are observed. In this study, we investigated the detailed structural information on the oligomers and fragment ions of branched poly(1,3-butylene adipate) and branched poly[di(ethylene glycol) adipate] and the transesterification products of branched polyesters with trifluoroacetic acid or chloro difluoroacetic acid. Branched polyesters were chosen because they are important polymers but difficult to characterize; thus branched polyesters provide challanging test for TOF-SIMS. TOF-SIMS spectra of polyesters are obtained from thin polymer films cast from solution on a silver substrate. A good solvent for a polumer solution disrupts intermolecular forces between polymer chains but leaves the polumer intact. Transesterification reactions are potentially useful for characterization of high molecular weight and intractable polyesters. Transesterification products of polyesters and trifluoroacetic acid or an integral number of polyester repeat units and an additional diol. The progress of such reactions was monitored using peak intensities of reactants and products in TOF-SIMS spectra. The increasing abundance of tagged ions indicates that the reaction has progressed with time.
