Molecular Characterization of Brassica Pollen Allergen

  • Allergy to Brassica pollen has been reported in some countries. We have cloned a cDNA encoding a Brassica pollen allergen, Bra r 1. Bra r 1 belongs to a new family of $Ca^{2+}$-binding proteins, characterized by the presence of two EF-hand calcium-binding domains. Bra r 1 was detected in the tapetum, microspores, pollen coat and pollen tubes, indicating Bra r 1 is involved in pollen pistil interaction and pollen tube growth. We have engineered the hypoallergenic mutants of Bra r 1 for immunotherapy. Here we describe the review of molecular characterization of Bra r 1.

작약의 화분배양에 의한 캘러스 및 배발생 (Callus Induction and Embryogenesis Through Pollen Culture in Paeonia albiflora PALL)

  • 작약의 화분배양에 의한 반수체 식물을 유기시킬 목적으로 화분배양을 실시하여 배양중 소포자의 변화를 관찰하고 캘러스 및 배발생에 미치는 저온전처리(5$^{\circ}C$/10 days)의 효과와 배양화분의 생존율을 조사하였다. 작약의 화분배양시 캘러스 및 배발생에는 저온전처리의 효과가 다소 인정되었으나 약배양의 효과에는 미치지 못하였다. 화분를 첨가한 고체배지에 이식했을 때 구형배가 형성되었다. 화분의 생존율에는 저온전처리 및 배지조성간 의 영향이 미치지 않았으며 배양기간이 지남에 따라 화분의 생존율도 저하되었다.

송화분이 흰쥐의 간장에 미치는 영향(II) (Effect of Pine Pollen on Rat Liver(II))

  • This study was conducted to investigate the nutritional constituents of pine pollen and the effects of pine pollen on rat liver. Using the chemical components analysis method, the constituents of pine pollen are crude protein(13.97%). crude lipid (6.50%), crude ash (3.25%). Detectable in pine pollen are main fatty acids and non-essential amino acids such as glutamic acid and aspartic acid such as oleic and linoleic acid and 18kinds of amino acid including essential amino acids. The effects of pine pollen on rat liver fed with a high fat diet for 6 weeks showed significant decrease in total lipid and triglyceride as compared with the group fed with a fat diet only. Total cholesterol content decreased by 20.33%. No significant difference was found in total protein content of each group. Pine pollen Is efficacious in preventing geriatric diseases (arteriosclerosis, hyper-lipidemia and cardiovascular system-related diseases).

시판 천연화분 및 가공화분의 품질에 관한 비교 연구 (Comparison of Quality of Natural Pollen and Processed Pollen)

  • Dissolution rate , periduium state, nutrient component and fatty acid composition of 5 species of natural pollen and 3 species of processed pollen were carried out for evaluation of quality. Natural pollens had average 11.5% of moistures, and had native peridium may be responsible for allergy. The average dissolution rate of natural pollens were low as 66.3%. One among 3 species of processed pollen was observed to have not peridium, resulted in higher dissolution rate. Major fatty acid of pollen was palmitic acid but content of oleic acid was lower than that of other plant seeds.

화분관 In Vitro 생장의 굴수성에 대하여 (Tropisms of Pollen-Tubes In Vitro)

  • Zephyranthes candida, Narcissus pseudonarcissus and Crinum asiaticum pollen were placed near their pistil parts respectively on agar cultural media(microslides) containing 10% sucrose and 100mg/l botic acid plus 1% agar with or without calcium and some other calciumsupporting inorganic salts. If fresh pistils (100% moisture) were used pollen grew toward their pistil parts, showing "positive" tropism. This was also true when combinations among three different species were made. Pollen tubes grew away from the pistils if they were dried (below 10% moisture), showing "negative" tropism. Pollen could not show any tropic growth and thus grew at random of all directions if the pistil parts were incompletely dried (approximately 50% moisture). The similar tropic responses of pollen-tube growth with the three species could be demonstrated with etiehr wet or dried tooth-pick segments. Calcium jons in the basic medium merely promated pollen-tube growth and so either "positive" or "negative" tropism became rather distinctive, but they were not tropically active. Pollen tubes grow toward pistil parts with more moisture content and seem to be hydrotropically sensitive. This was assumed due to the cohesive force existing in water molecules.esive force existing in water molecules.

더덕의 花粉發芽에 관한 溫度와 培地條件 (Factors Influencing Pollen Germination in vitro of Codonopsis lanceolata)

  • The present study has been undertake to obtain the fundatmental data of optimum germination condition and to establish storage time for artificial pollinationin Codonopsis lancelata pollen. In vitro condition for germination of freshly collected and stored pollen were examined. The optium temperature for germination of fresh pollen was $25^{\circ}C$. The optium sucrose concentration in the medium ranged from 30 to 40 % and optium pH 6.0% for pollen germination. The rate of pollen germination accelerated considerably in the medium with 1% agar. 30% sucrose, and pH 6. C. lanceolata pollen remained viable for 15 days when stored at 5$^{\circ}C$ with silica gel as desiccant.

  • Pollen and spores as well as their morphological characteristics are biological stability materials in which contains historical information about vegetation and climate changes. As noted researcher Neishtadt in 1971, the lake and swamp deposits were developed only in Holocene period. The geological history of Darkhad depression which is situated in the Northern end of Mongolia from ancient time become under scientific interest of many researchers. Our investigation was focused on pollen analysis of lake sediment of Darkhad depression which caused bottom of paleolake. In Mongolia the palynology science is beginning to develop since 1990 and in 2000 the laboratory of palynology was established at the department of Biology, State Pedagogical University of Mongolia. Researchers from this laboratory working on pollen morphology of present flora in order to classify plants taxonomy. Another part of our investigation is mellitopalynology, which is studying of pollen grains in a honey. Although, this research has been extending by pollen analysis of sediment and lake deposits in past few years on the basis of high sensitive microscope and modem technology.

Diurnal and Nocturnal Behaviour of Airborne Cryptomeria japonica Pollen Grains and the Allergenic Species in Urban Atmosphere of Saitama, Japan

  • Japanese cedar (Cryptomeria japonica) pollinosis is the most popular pollinosis in Japan. It has been reported that Cryptomeria japonica pollen allergenic species are suspended as fine particles in the urban atmosphere. These allergenic fine particles are responsible for inducing asthma by breaking into the lower respiratory tract. It has also been found that pollinosis symptoms on the sufferers appear mainly at night-time by the results from epidemiological studies. However, the exact reason for these phenomena is not yet clarified. In this study, the diurnal and nocturnal behaviours of Cryptomeria japonica pollen grains and their allergenic species in the urban area of Saitama city of Kanto Plain were investigated. Airborne pollen grains and allergenic Cry j 1 concentrations in total suspended particulate matter (TSP) were investigated at two sampling sites, a heavy traffic road (roadside site) and at the balcony of the $10^{th}$ floor of the Building of Research and Project of Saitama University (general urban site). The latter sampling site where located about 300 m away from the roadside site was used as a general urban site unaffected by automobile traffic. The airborne pollen counts were measured with a real-time pollen monitor. Cry j 1 particles were collected with two high volume air samplers, and these concentrations were measured by surface plasmon resonance method with a Biacore J system. The diurnal variation of the airborne pollen counts was similar to the trends of temperature and wind speed during the day-time; whereas its tendency with wind speed trend was not observed during the night-time. Airborne pollen counts were lower with northern wind than with southern wind because the pollen comes from the mountainous areas, and the mountains in the south are closer, about half the distance to the northern mountains. It is suggested that the peaks of airborne pollen counts during night-time in the sampling site occurred by transport of pollen grains released during day-time in the mountainous forest areas, located c.a. 100 km away from the sampling site. On the roadside site the allergenic Cry j 1 concentrations were higher than at the general urban site, nevertheless pollen grains counts were lower. These results suggested that worsening of pollinosis symptoms during night-time in urban area was caused by transport of pollen grains during day-time in the mountainous forest areas. Moreover, pollen allergenic species become different morphology from pollen grain at roadside site, and the subsequent pollen grains re-suspension by automobile traffic.

개화 시 기온 및 상대습도가 참외의 화분발아에 미치는 영향 (Effect of Air Temperature and Relative Humidity during Flowering on Pollen Germination of Oriental Melon (Cucumis melo L. var. makuwa Makino))

  • 본 연구는 참외 개화시의 기온과 상대습도가 화분의 발아에 미치는 영향을 검토하고자 '만리장성', '오복꿀' 및 '조은대'의 3 품종을 재료로 하여 개화 시 온도를 $15^{\circ}C$, $25^{\circ}C$$35^{\circ}C$, 상대습도를 30%, 60% 및 90%의 처리를 두고 실험하였다. 기온조건에 따른 화분 발아율은 $25^{\circ}C$에서 채취한 것이 가장 높았고 $15^{\circ}C$에서 채취한 것이 가장 낮은 경향이었으며 품종별 화분 발아율은 '만리장성'이 가장 높았고 다음이 '오복꿀'이었으며 '조은대'가 가장 낮은 경향이었다. 화분관 신장길이는 기온조건별로는 $35^{\circ}C$에서 채취한 것이 가장 길었고 다음이 $25^{\circ}C$ 에서 채취한 것이었으며 $15^{\circ}C$에서 채취한 것은 현저히 짧았으며, 품종에 따라서는 '오복꿀'이 가장 길었다. 상대습도 조건에 따른 화분 발아율은 습도조건에 따라 뚜렷한 차이는 없으나 습도가 높을수록 높은 경향이었으며, 품종간에도 큰 차이가 없으나 '만리장성'과 '조은대'에 비해 '오복꿀'에서 낮았다. 그러나 화분관 신장길이는 처리간에 뚜렷한 차이를 나타내어 60%의 상대습도에서 채취한 것이 가장 길었으며, 다음이 30%의 상대습도에서 채취한 것이며 90%의 상대습도에서 채취한 것은 현저히 짧았다. 품종별 화분관 신장길이는 '오복꿀'이 가장 길었고 '조은대'가 가장 짧았다.

저온기 시설수박 착과율 증진을 위한 품종 및 화분분리 유기용매 선발 (Selection of Cultivars and Organic Solvents to Improve Fruit Set of Greenhouse Watermelon during Cold Period)

  • 화분량은 '금천' 수박을 제외하고 응화 1개당 10mg 이상이었다. '복' 수박은 1화당 화분무게가 13.8mg이었고, '스피드꿀' 수박은, 12.1mg으로 겨울철 저온기 재배에 인공수정용 화분생산 품종으로 적당한 것으로 판단된다. 유기용매 침지 후 즉시 건조된 화분발아율은 '금천' 수박이 67%로 기장 낮았고 '아폴로꿀' 수박이 92%로 가장 높았다. 유기용매에 15일간 저장했을 경우 '복' 수박 화분의 발아력이 74%로 가장 높았다. 저장 24일 후에는 5품종 모두 25% 이하의 발아율을 나타내었다. '금천'과 '삼복꿀' 수박의 화분관 신장속도는 $50{\mu}m/hr$ 미만(저장 24일)으로 다른 품종보다 낮았으며, 벌이도중 화분관이 파열되는 경우도 발생되었다. 유기용매 저장 24시간 후 화분발아율은 pentane, ethyl ether, n-hexane, ethyl acetate, acetone에서 각각 45, 39, 34, 23, 19% 순이었고 유기용매 침지시간이 길어질수록 화분활력이 급격히 감소되었다. 화분 표피는 유기용매에 저장한 시간이 길어질수록 장해를 많이 받았는데 유기용매의 종류에 따라 장해정도가 달랐다. '복' 수박이 화분량, 화분활력, 저장성이 우수하였는데 저온기 착과율 증진을 위한 화분생산 품종으로 보여진다.
