The Distribution of Planktonic Protists Along a Latitudinal Transect in the Northeast Pacific Ocean (북동 태평양수역에서 위도에 따른 부유 원생동물의 분포)

  • As a part of Korea Deep Ocean Study program, we investigated the distribution of planktonic protists in the upper 200 m of the northeast Pacific from $5^{\circ}N$ to $17^{\circ}N$, along $131^{\circ}30'W$. Area of divergence was formed at $9^{\circ}N$ which is boundaries of the north equatorial counter current (NECC) and the north equatorial current (NEC) during this cruise. Chlorophyll-a concentration was higher in NECC than in NEC area. Pico chl-a(<$2\;{\mu}m$) to total chl-a accounted for average 89% in the study area. The contribution of pico chl-a to total chl-a was relatively high in NEC area than in NECC area. Biomass of planktonic protists, ranging from 635.3 to $1077.3\;mgC\;m^{-2}$(average $810\;mgC\;m^{-2}$), was most enhanced in NECC area and showed distinct latitudinal variation. Biomass of HNF ranged from 88.7 to $208.3\;mgC\;m^{-2}$ and comprised 15% of planktonic protists. Biomass of ciliates ranged from 123.6 to $393.0\;mgC\;m^{-2}$ and comprised 25% of planktonic protists. Biomass of HDF ranged from 407.2 to $607.8\;mgC\;m^{-2}$ and comprised 60% of planktonic protists. HDF was the most dominant component in both NECC and NEC areas. Nano-protist biomass accounted for more than 50% of total protists in the both areas. The contribution of nanoprotist to total protists biomass was relatively higher in NEC area than in NECC. The biomass of planktonic protists was significantly correlated with phytoplankton biomass in this study area. The size structure of phytoplankton biomass coincided with that of planktonic protists. This suggested that the structure of the planktonic protists community and the microbial food web were dependent on the size structure of the phytoplankton biomass. However, biomass and size structure of planktonic protist communities might be significantly influenced by physical characteristics of the water column and food concentration in this study area.

Geochemical Properties of Deep Sea Sediment in the Benthic Environmental Impact Experiment Site (BIS) of Korea (심해 저층환경충격 시험지역의 퇴적물 지화학적 특성)

  • The benthic environmental impact experiment addresses environmental impacts at a specific site related to deep-sea mineral resource development. We have acquired several tens of multi- or box core samples at 31 sites within the Benthic environmental Impact Site (BIS) since 2010, aiming to examine the basic properties of surficial deep-sea sediment as a potential source for deep-water plumes. In this study, we present the geochemical properties such as major elements, rare earth elements (REEs), and heavy metal contents at the BIS. Such proxies vary distinctly according to the Facies association. The lithology of all core sediments in the BIS corresponds to both Association Ib and Association IIIb. The vertical profiles of some major elements ($SiO_2$, $Fe_2O_3$, CaO, $P_2O_5$, MgO, MnO) show noticeable differences between Association Ib and IIIb, while others ($Al_2O_3$, $TiO_2$, $Na_2O$, and $K_2O$) do not vary between Association Ib and IIIb. REEs are also distinctly different for Associations Ib and IIIb; in Association Ib, REY and HREE/LREE are uniform through the sediment section, while they increase downward in Association IIIb like the major elements; below a depth of 8 cm, REY is over 500 ppm. The metal enrichment factor (EF) evaluates the anthropogenic influences of some metals (Cu, Ni, Pb, Zn, and Cd) in marine sediments. In both Associations, the EF for Cu is over 1.5, the EF for Ni and Pb ranges from 0.5 to 1.5, and the EF for Zn and Cd are less than 0.5, indicating Cu is enriched but Zn and Cd are relatively depleted in the BIS. The vertical variations of geochemical properties between Association Ib and IIIb are shown to be clearly different, which seems to be related to the global climate changes such as the shift of Intertropical convergence zone (ITCZ).

On the Wintertime Wind-driven Circulation in the Yellow Sea and the East China Sea : Part I. Effect of Tide-induced Bottom Friction (황해.동중국해의 겨울철 취송 순환에 대하여: Part I. 조류에 의한 저면 마찰력의 영향)

  • The effect of bottom friction on the steady wind-driven circulation in the Yellow Sea and the East China Sea (YSECS) has been studied using a two-dimensional numerical model with and without tidal forcing. Upwind flow experiment in YSECS has also been carried out with a schematic time variation in the wind field. The surface water setup and circulation pattern due to steady wind forcing are found to be very sensitive to the bottom friction. When the effects of tidal currents are neglected, the overall current velocities are overestimated and eddies of various sizes appear, upwind flow is formed within the deep trough of the Yellow Sea, forming a part of the topographic gyre on the side of Korea. When tidal forcing is taken into account, the wind-induced surface elevations are smoothed out due to the strong tide-induced bottom friction, which is aligned almost normal to the wind stresses; weak upwind flow is farmed in the deep trough of the Yellow Sea, west and south of Jeju. Calculation with wind forcing only through a parameterized linear bottom friction produces almost same results from the calculation with $M_2$ tidal forcing and wind forcing using a quadratic bottom friction, supporting Hunter (1975)'s linearization of bottom friction which includes the effect of tidal current, can be applied to the simulation of wind-driven circulation in YSECS. The results show that steady wind forcing is not a dominant factor to the winter-time upwind flow in YSECS. Upwind flow experiment which considers the relaxation of pressure gradient (Huesh et al. 1986) shows that 1) a downwind flow is dominant over the whole YSECS when the northerly wind reaches a maximum speed; 2) a trend of upwind flow near the trough is found during relaxation when the wind abates; 3) a northward flow dominates over the YSECS after the wind stops. The results also show that the upwind flow in the trough of Yellow Sea is forced by a wind-induced longitudinal surface elevation gradient.

Solvent Filtration Performance of Thin Film Composite Membranes based on Polyethersulfone Support (폴리이터설폰 지지체를 활용한 박막복합막의 용매투과특성 연구)

  • Recently, the application range of organic solvent nanofiltration (OSN) technology has been expanding, requiring membranes with better performance. In this work, thin film composite (TFC) OSN membrane was fabricated. First, ultrafiltration support membrane was prepared via nonsolvent-induced phase separation (NIPS) technique using polysulfone (PSf) and polyethersulfone (PES). Then, the effect of pore forming additives such as polyvinylpyrrolidone (PVP) and pluronic F-127 were employed to improve the membrane permeance. The well-known interfacial polymerization technique was employed using MPD-TMC chemistry to form a thin film on top of the fabricated support, and its solvent permeance and nanofiltration performance was characterized. It was found that polyethersulfone support exhibited more reliable performance compared to polysulfone, and PVP additive was more effective compared to Pluronic F-127. As for the oSN performance, polar aprotic solvents like acetonitrile show significantly higher flux (986.5 L·m-2·h-1·bar-1) compared to water and EtOH (9.5 L·m-2·h-1·bar-1).

Seasonal Characteristics of Phytoplankton Distribution in Asan Bay (아산만 식물플랑크톤의 계절별 군집 분포 특성)

  • Samples were collected from five stations in February, May, July, and September 2004 to investigate seasonal variations in the phytoplankton community and the relationship between dominant genera and environmental factors in Asan Bay. In February, microphytoplankton contributed 80% to the total chlorophyll a. Diatom dominated the phytoplankton community, accounting for 85.9% of the total cell number, followed by dinoflagellates (6%). Dominant species were Skeletonema costatum and Thalassiosira spp. Abundant diatom, including S. costatum and Thalassiosira spp., may be affected by water temperature and silicate at Station 1 and 2 in February 2004. In May, the nanophytoplankton contribution to total phytoplankton was higher than in other seasons. However, abundance of S. costatum and Thalassiosira spp. decreased, since the growth of S. costatum and Thalassiosira spp. might be limited by phosphates (P) resulting from low P concentration and a high DIN:DIP ratio in the outer region. In July, dominant phytoplankton groups were diatom (39%), cryptophyceae (28%), and cyanophyceae (20%). Dominant genera were Oscillatoria spp. and phytoflagellate of a monad type in the inner region (Station 1 and 2), whereas S. costatum was dominant in the outer region (Station 4 and 5). In September, dominant phytoplankton were diatom (69%) and cryptophyceae (28%). Dominant genera were phytoflagellate of the monad type, S. costatum in the inner region, while Chaetoceros spp. was dominant in the outer region.

Organic Matter and Hydraulic Loading Effects on Nitrification Performance in Fixed Film Biofilters with Different Filter Media

  • Nitrification performance of fixed film biofilters using coarse sand, loess bead, or styrofoam beads in biofilter columns 1 meter high and 30cm in diameter were studied at different hydraulic and organic matter loading rates. Synthetic wastewater was supplied to the culture tank in order to maintain desired TAN concentrations in inlet water to biofilters. All the biofilters were conditioned 5 months before start of sampling. TAN and $NO_2-N$ conversion rates increased with an increase in the hydraulic loading rate (HLR). However, the improvement in biofilter performance was not linearly correlated to HLR in styrofoam bead filters. This is mainly due to the characteristics of the styrofoam beads used. TAN conversion rates of sand filters increased with the increase of HLR up to $200m^3/m^2$. per day. No increase in the TAN conversion rate was observed at the highest HLR since flooding on the media surface took place. HLR had a significant impact on the TAN conversion rates in loess bead filter up to the highest HLR tested (P<0.05). TAN conversion rates were much less at organic matter loading rates of 9 and 18kg $O_2/m^3$ per day than those without the addition of organic matter in styrofoam bead filters. The addition of glucose resulted in a reduction of the TAN conversion rate from 540 to 284g $TAN/m^3$ per day. No significant difference of TAN conversion rates between the two organic matter loading rates was found (p<0.05). This indicates that the impact of organic matter on nitrification becomes less and less sensitive with an increase in the COD/TAN ratio. At an organic matter loading rate of 9kg $O_2/m^3$. per day, a great reduction of TAN conversion rates was observed in sand filters and loess bead filters. Clearly, organic matter can be one of the most Important Impacting factors on nitrification. $NO_2-N$ conversion rates showed a similar trend for TAN. Based on the TAN and nitrite conversion rates, styrofoam beads showed the best performance among the three filter media tested. Also, the low gravity and price of styrofoam beads make the handling easier and more cost-effective for commercial application. The results obtained at the highest organic matter loading rates can be used in the biofilter design in recirculating aquaculture system.

A Comparative Study on the Lipid Components of Barley and Malt -I. Composition of Neutral Lipids- (보리와 맥아(麥芽)의 지방질(脂肪質) 성분(成分)에 관한 비교(比較) 연구 -제(第)1보(報) : 중성지질(中性脂質)의 조성(組成)-)

  • Neutral lipids of the total lipid extracted from 4 representative varieties of barley grown in Korea and their corresponding malt were studied. Total lipids of barley and malt were solvent extracted with chloroform:methanol:water (1.0 : 1.0 : 0.9, v/v), The total lipids were fractionated into neutral and polar lipids by silicic acid column chromatography, and neutral lipids fraction was separated by thin layer chromatography and quantitated by TLC scanner. The fatty acid compositions was determined by gas liquid chromatography. The average content of total lipid in the 4 barleys and their malts were 3.3 and 2.9%, and average of neutral lipids content in the barley and malt lipids were 73.8 and 68.5%, respectively. Among the neutral lipids, triglycerides and free fatty acids were the major components, and triglycerides content decreased and free fatty acids content increased during malting. Sterol esters, free sterols, 1,3-and 1,2-diglycerides were the minor components of the neutral lipids, and contents of those components showed increasing tendency during malting. The major fatty acid composition of the total lipids were linoleic, palmitic and oleic acids, and in general, the malts had lower amounts of unsautrated fatty acids and high amounts of saturated fatty acids. Fatty acid composition of neutral lipids was of almost the same pattern as that of the total lipids.

Bacterial Spot Disease of Green Pumpkin by Pseudomonas syringae pv. syringae (Pseudomonas syringae pv. syringae에 의한 애호박 세균점무늬병)

  • A pathogen that causes a new disease on green pumpkin in the nursery and the field was characterized and identified. Symptoms of the disease on green pumpkin were water soaking lesions and spots with strong yellow halo on leaf, brown lesion on flower, and yellow spot on fruit. The bacterial isolates from the leaf spot were pathogenic on the 8 curcubitaceae crop plants, green pumpkin, figleaf gourd, wax gourd, young pumpkin, zucchini, cucumber, melon, and oriental melon, whereas they did not cause the disease on sweet pumpkin and watermelon. They were Gram-negative, rod shape with polar flagella, fluorescent on King's B agar and LOPAT group 1a by LOPAT test. Their Biolog substrate utilization patterns were similar to Pseudomonas syringae pv. syringae's in Biolog database. Phylogenetic trees with 16S rRNA gene sequences and multilocus sequence typing (MLST) with nucleotide sequences of 4 housekeeping genes, gapA, gltA, gyrB, rpoD and those of P. syringae complex strains in the Plant Associated and Environmental Microbes Database (PAMDB) showed that the green pumpkin isolates formed in the same clade with P. syringae pv. syringae strains. The clade in MLST tree was in the genomospecies 1 group. The phenotypic and genotypic characteristics suggested that the isolates from green pumpkin lesion were P. syringae pv. syringae.

Evolution of sea Urchin Strongylocentrotus intermedius Based on DNA Sequences of a Mitochondrial Gene, Cytochrome c Oxidase Subunit I (미토콘드리아 유전자, 치토그롬 옥시다제(subunit I)의 염기서열을 이용한 새치성게(Strongylocentrotus intermedius)의 진화과정 분석)

  • Sea urchin S. intermedius occurring in the Korean east coast is a cold water species that belongs to the family Strongylocentrotidae of Echinoidea. Although it is known that there are nine species in the family, species identification criteria, phylogenetic relationships, time and process of evolution of the family members have not been uncovered clearly. In the present study, I tried to find some clues to such problems for S. intermedius by means of DNA sequences. For this, cytochrome c oxidase subunit I (COI), one of the mitochondrial genes that evolve fast and follow maternal inheritance was analyzed. DNA was extracted from the female gonad of S. intermedius, a segment of COI gene amplified by polymerase chain reaction (PCR), and finally a total of 1077 base pair sequence of COI obtained by cloning and sequencing the PCR product. The sequence was compared with homologous genes of other sea urchins and echinoderm species. Phylogenetic trees of the COI gene segment revealed that S. intenedius is a sister species of S. purpuratus which lives along the east coast of the Paciflc. With reference to the fossil records of sea urchins and genetic distances in the molecular phylogenies, it is estimated that the two species were separated about 0.89 million years ago when the earth temperature fluctuated significantly. The current disjunct distribution patterns of the two species and the climate change of the earth at the time of separation suggest that speciation might have occurred by vicariance. The COI gene sequence obtained here now can be used as a molecular character which discerns S. intermedius from the other sea urchin species of Strongylocentrotidae.

Graphene Oxide/Polyimide Nanocomposites for Gas Barrier Applications (산화그래핀이 함유된 폴리이미드 나노복합막의 기체차단성 평가 및 활용)

  • Polymeric films for gas barrier applications such as food packaging and electronic devices have attracted great interest due to their cheap, light and easy processability among gas barrier materials. Especially in electronic devices, extremely low gas permeance is necessary for maintaining the device performance. However, current polymeric barrier films still suffer from relatively high gas permeance than other materials. Therefore, there have been strong needs to enhance the gas barrier performance of polymeric barrier films while keep their own advantages. Recently, graphene is highlighted as a 2D-layered material for gas barrier applications. However, owing to the poor workability and difficulty to produce in engineering scale, graphene oxide (GO) is on the rise. GO consists of oxygen-containing functional groups on surface with intrinsic 2D-layered structure and high aspect ratio, and it can be well-dispersed in aqueous polar solvents like water, resulting in scalable mass production. Here, we prepared GO incorporated polyimide (PI) nanocomposites. PI is widely used barrier polymer with high mechanical strength and thermal and chemical stability. We demonstrated that PI/GO nanocomposites could perform as a gas barrier. Furthermore, surfactants (Triton X-100 (TX) and Sodium deoxycholate (SDC)) are introduced to enhance the gas barrier performance by improving the degree of dispersion of GO in PI matrix. As a result, TX enhanced the gas barrier performance of PI/GO nanocomposites which is similar to predicted value. This finding will provide new insight to polymer nanocomposites for gas barrier applications.
