Improvement of Enzymatic Stability and Catalytic Efficiency of Recombinant Fusariumoxysporum Trypsin with Different N-Terminal Residues Produced by Pichiapastoris

  • Fusarium oxysporum trypsin (FOT) is a fungal serine protease similar to mammal trypsin. The FOT could be successfully expressed in Pichiapastoris by engineering the natural propeptide APQEIPN. In this study, we constructed two recombinant enzymes with engineered amino acid sequences added to the N-terminus of FOT and expressed in P. pastoris. The N-terminal residues had various effects on the structural and functional properties of trypsin. The FOT, and the recombinants TE (with peptide YVEF) and TS (with peptide YV) displayed the same optimum temperature ($40^{\circ}C$) and pH (8.0). However, the combinants TE and TS showed significantly increased thermal stability at $40^{\circ}C$ and $50^{\circ}C$. Moreover, the combinants TE and TS also showed enhanced tolerance of alkaline pH conditions. Compared with those of wild-type FOT, the intramolecular hydrogen bonds and the cation ${\pi}$-interactions of the recombinants TE and TS were significantly increased. The recombinants TE and TS also had significantly increased catalytic efficiencies (referring to the specificity constant, $k_{cat}/K_m$), 1.75-fold and 1.23-fold than wild-type FOT. In silico modeling analysis uncovered that the introduction of the peptides YVEF and YV resulted in shorter distances between the substrate binding pocket (D174, G198, and G208) and catalytic triad (His42, Asp102, and Ser180), which would improve the electron transfer rate and catalytic efficiency. In addition, N-terminal residues modification described here may be a useful approach for improving the catalytic efficiencies and characteristics of other target enzymes.

A Study on Male Disposable Product for Urinary Incontinence (남성 일회용 요실금 제품에 관한 연구)

  • Urinary Incontinence is any leakage of urine against their will. It is not a life threatening illness but it causes negative effects both socially and psychologically. Based on many previous studies, 20-40% of adult female and 30% of men in their sixties have experienced symptoms of urinary incontinence. As the number of active aged people experiencing the symptoms increases, demand for specialized product is growing. The purpose of the study is to research the types and features of male disposable products for urinary incontinence on the market and to provide the basic data for improvement and development of better product. Products from 6 brands (Tena, Depend, Carnation, Keepers, Molimed, Abrimen) with high brand awareness both in online and offline were selected and analyzed. And product evaluation was carried out by male in their sixties with light symptom of urinary incontinence. The results are as follows. There are three types of male products for urinary incontinence: panties, diaper and pad. Among the 6 brands, the subject of analysis, only Depend from Yuhan Kimberly produces male panties while others make ones for unisex. There is no diaper product only for male, and Tena, Molimed, Abrimen have pad type products for male. Male panties from Depend emphasizes their design considering male's physical characteristics. Male pad is differentiated with female one by having different designs like triangular or pocket type which is more comforting to men. As a result of product evaluation, wearable panties similar to normal underwear were highly appreciated and satisfactory. Among various shapes of pads, 25cm long pad with wide width of front and narrow back was valued to be comfortable.

Efficacy of non-surgical treatment accompanied by professional toothbrushing in the treatment of chronic periodontitis in patients with type 2 diabetes mellitus: a randomized controlled clinical trial

  • Purpose: The present study aimed to evaluate the clinical benefit of additional toothbrushing accompanying non-surgical periodontal treatment on oral and general health in patients with type 2 diabetes mellitus (T2DM). Methods: We conducted a doubled-blind randomized controlled trial in 60 T2DM patients between June 2013 and June 2014. The patients were randomly assigned to the scaling and root planing (SRP) group; the scaling and root planing with additional toothbrushing (SRPAT) group, in which additional toothbrushing was performed by toothpick methods; or the control group. Microbiological and oral examinations were performed for up to 12 weeks following treatment. Non-surgical treatment was conducted in the experimental groups. The SRP group received scaling and root planing and the SRPAT group received additional toothbrushing with the Watanabe method once a week from the first visit through the fifth visit. The primary outcomes were changes in haemoglobin A1c (or glycated haemoglobin; HbA1c) levels, serum endotoxin levels, and interleukin-1 beta levels. Periodontal health status was measured by periodontal pocket depth, the calculus index, and bleeding on probing (BOP). Results: Both the SRP and SRPAT groups showed improvements in periodontal health and HbA1c, but the SRPAT group showed significantly less BOP than the SRP group. Furthermore, only the SRPAT group showed a statistically significant decrease in serum endotoxin levels. Conclusions: Non-surgical periodontal treatment was effective in improving HbA1c and serum endotoxin levels in T2DM patients. Furthermore, non-surgical treatment with additional tooth brushing had a more favourable effect on gingival bleeding management. High-accuracy and High-speed Groove Die Set (고정도.고속 Groove Die Set)

  • Currently existing high-accuracy and high-speed die sets used in reciprocal press create scratches at the surface of guide posts, steel balls, and bushes due to vertical movement of balls with point-contacts between inner surface of bushes and guide posts. Consequently, accuracy of the die set and the life span of the metal mold are reduced. However, those scratches could reduce the pre-load of the steel ball. This research designed and developed a groove-type die set which improves life span of the die set by eliminating point-contacts of steel balls with guide posts. The guide post consisted of a steel-ball retainer, a steel-ball retainer stopper, a guide bush, a guide pin, a snap ring, and a spring. The steel-ball retainer has 72 holes with 8 columns of 9 holes in each column. The inner surface of the guide bush was grinded(surface roughness: $Ra\;\\;0.2{\mu}m$, accuracy: $0\;{\sim}\;-0.002mm$) after NC turning and heat treatment. Also, a line of small intermediate pocket was processed inside of the guide bush for lubrication and elimination of foreign materials. Guide grooves of steel balls were processed using a wire EDM(Electrical Discharge Machining) after heat treatment. With such a design of the guide post stated above, loads against steel balls could be dispersed greatly by the line contacts through the guide groove between the guide post and the guide bush, and the life span of the guide post could be expanded semi-permanently.

A Study on the Change of Shape of "Jagui" (Adze) Used in Korean Traditional Architecture (전통건축에 사용된 자귀의 형태 변화에 대한 고찰)

  • The Axes and Adzes are the oldest tools since the beginning of human history which is used to cut the tree and make part of architecture such as boards, square timber etc. Nowadays, these old woodworking tools especially "Jagui(자귀)" (adze) has been almost disappeared at the working site of residential and cultural properties. It is necessary to study Korean traditional woodworking tools to keep Korean traditional skill and technology. It has been reviewed the change of shape of "Jagui(자귀)" (adze) from before Samhan (삼한) period to Joseon Dynasty through excavated relics and paintings and summarize as follows: Based on excavation relics of Dahori, both plate type blade and pocket type blade are used for "Tokki(도끼)" (axe) and "Jagui(자귀)" (adze). The excavated "Jagui(자귀)" (adze) from Jeongbaikri 356 tomb, near Pyongyang is prototype of "Jagui(자귀)" (adze) used nowdays which has almost same shape as well as the insert method of blade and handle. The auxiliary handle is inserted to blade and the main handle is inserted to the auxiliary handle which is different from the method of Chinese and Japanese "Jagui(자귀)" (adze). The length of handle of "Jagui(자귀)" (adze) until late Joseon Dynasty is short by which we assume it is used for a sitting position as hand held Jagui and from that time long handle has been introduced for a standing position which is called "Sunjagui(선자귀)". "Jagui(자귀)" (adze) has been used since the beginning of human history but it is almost disappeared at the most of woodworking site which is the crisis of Korean traditional architecture and we have to do something to keep it.

Evaluation of control feel in opening and closing the doors of a virtual refrigerator in the CAVE system with an Arm-Master (CAVE 공간에서 Arm-Master를 이용한 가상냉장고 도어 개폐의 조작감 평가)

  • This study aims to validate the effectiveness of CAVE system with an Arm-Master in the evaluation of virtual prototypes. A virtual kitchen with a refrigerator was presented in the CAVE system. Subjects put in and pull out virtual objects by using the Arm-Master. Twelve subjects, six males and six females, participated in the six experimental conditions : three types of refrigerator door grips and two reaction forces. After each trial, subjects evaluated a door grip of the virtual refrigerator in terms of easiness of operation, similarity in force, presence etc. The results show the mean values of vortical-type door grips are greater than the horizontal pocket-type door grip. Also the subjects preferred 5N in the reaction force of the Arm-Master rather than 15 N when they open and close virtual door. Unexpectedly, female subjects significantly marked low scores in the evaluation terms compared ruth mali subjects. It explains the Arm-Master should be modified much more if it works effectively in design evaluation.

  • A sports center fire in Jecheon caused 29 deaths and 40 injuries. This study focused only on the structural factors of the building and equipment to investigate the causes of casualties based on the fire investigation results at the sports center. The structural factors of the building and equipment were a piloti-type parking lot, lack of a fire compartment between the piloti-type parking lot and lobby, lack of an installed sprinkler system, lack of an installed fire door in the main stairs on the $1^{st}$ floor, lack of an installed fire water tank on the rooftop, an installed pocket fire door in the main entrance on the $2^{nd}$ floor, poor fire compartments in an EPS space and a pipe pit and on each floor, a leak in the joint of a drain pipe, plywood installed in the hoistway of the freight elevator and interior wall, illegal remodeling of a closed rooftop structure, which cannot discharge smoke and heat, installed styrofoam insulation in the inside of the parking lot ceiling, an installed tempered glass window, which cannot be opened in the ladies bathroom on the $2^{nd}$ floor, and a finished dryvit exterior wall.

Application of Temperature Gradient Gel Electrophoresis To cAMP Receptor Protein (온도 기울기 전기영동장치의 CAMP 수용성 단백질에 응용)

  • Cyclic AMP receptor protein (CRP) is involved in the transcriptional regulation of more than 100 genes in E. coli. CRP dimer is converted into active form via the sequential conformation change of cAMP binding pocket, hinge region and HTH DNA binding motif by binding of cAMP. The temperature gradient gel electrophoresis (TGGE) was applied to CRP protein to know whether it was an efficient technique to study the conformational transitions and the thermal stability. TGGE showed the unfolding process of wild-type and S83G CRP proteins with the temperature gradient set from 29 to 71$^{\circ}C$ on nondenaturing polyacrylamide gel. Melting temperature (Tm) was 57$\pm$1 and 55$\pm$1$^{\circ}C$ for wild-type and S83G CRP, respectively in acidic buffer[89.8 mM Glycine and 24 mM Boric acid (pH 5.8)].

The effect of Body Image on Appearance Management Behavior of University Students Participating in Physical Activity (신체활동 참여 대학생의 신체이미지가 외모관리행동에 미치는 영향)

  • This study was conducted to examine the effect of body image of university students participating in physical activity on appearance management behavior. Using stratified cluster random sampling, a total of 298 male students, 109 male and 189 female students were sampled. The results are as follow. First, male students showed high in appearance, physical strength, health factors and appearance improvement type, and female students showed high appearance management and interpersonal relationship type. It was found that the arts and physical education department had an effect on the body image and appearance management behavior of university students with a lot of pocket money in terms of appearance and physical strength. Second, Body image was found to affect appearance management behavior. Appeared to be the body image of university students through physical activity affects the appearance management behavior, and these behaviors will have a positive relationship to lead a happy daily life in the future.

ANALYSIS ON THE PREDICTOR OF DISEASE PROGRESSION IN REFRACTORY PERIODONTITIS (난치성 치주염의 질환진행 예견 인자에 관한 분석)

  • Refractory periodontitis manifest progressive attachment loss in a rapid and unrelenting manner regardless of the type or frequency of therapy applied. The purpose of this study was ta evaluate the relation between the level of cytokines in GCF and periodontopathic microflora with disease activity of refractory periodontitis. Selection of patients with refractory periodontitis (7 males, 3 females) were made by long term clinical observation including conventional clinical history and parameters. Teeth that showed pocket depth greater than 6mm were selected as sample teeth. Subjects were examined at baseline and after 3 months. Prior to baseline test, individual acrylic stent was fabricated. Reference grooves were made on each sample tooth site. Pocket depth and attachment loss were measured by Florida Probe. Gingival index was measured at 4 sites each sample teeth. Disease activity was defined as attachment loss of ${\ge}$ 2.1mm, as determined by sequential probing and tolerance method. The pattern and amount of alveolar bone resorption was observed with quantitative digital subtraction image processing radiography. Morphological analysis of subgingival bacteria was taken by phase contrast microscopy. Predominant cultivable bacterial distribution and frequency were compared between disease-active and disease-inactive site using immunofluorescence microscopy and selective microbial culturing. Levels of $interleukin-l{\beta}$, 2, 4, 6 and $TNF-{\alpha}$ in GCF and blood serum sample were quantified by ELISA. In active sites, P. intermedia was significantly increased to compare with inactive site. $IL-1{\beta}$, IL-2, IL-6 and $TNF-{\alpha}$ in GCF were increased in active sites and IL-2 in serum was increased in active patients significantly. Alveolar bone loss in active site was correlated with $IL-1{\beta}$, IL-2 in GCF. And loss of attachment in active site was correlated with IL-2 in GCF. These results demonstrate that IL-2 in serum, $IL-1{\beta}$, IL-2, IL-6 and $TNF-{\alpha}$ in GCF, P, intermedia might be used as possible predictors of disease activity in refractory periodontitis before it is clinically expressed as attachment loss and quantitative alveolar bone change.
