Plant-growth-promoting rhizobacteria can affect plant growth by various direct and indirect mechanisms. This study was conducted to determine the ability of some rhizobacterial strains to enhance the seed germination of Lactuca sativa (lettuce) and Raphanus sativus (radish). Seeds were inoculated using a spore suspension ($1{\times}10^7cfumL^{-1}$) and incubated in a growth chamber at $28^{\circ}C$ under dark conditions and 65% RH. Azotobacter chroococcum and LAP mix inoculation increased the plumule length of L. sativa by 1.3, 0.8, and 0.7 cm, respectively, in comparison to the uninoculated control. Pseudomonas putida showed an increase of only 0.6 cm in plumule length when compared to the control. Inoculation of A. chroococcum, P. putida, and LAP mix enhanced the seed germination rate of R. sativus, by 10, 5, and 30%, respectively, in comparison with the uninoculated seeds. The results demonstrated that the inoculation of seeds by select rhizobacterial strains showed remarkable enhancement to the radicle length of lettuce and radish seedlings.
Immature zygotic embryos (up to 4mm in length) were cultured on MS medium supplemented with 0.5 to 8mg/L 2,4-D. Up to 87% of them formed somatic embryos on the plumule without producing an intervening callus. The site of somatic embryo formation was confirmed by culturing plumule explants, which consisted of shoot apical meristem domes with 1 or 2 leaf primordia excised from 2-week-old seedlings. When the concentration of 2,4-D was increased over 4 mg/L, the plumule explants produced nonembryogenic calli only, whereas the distal end of the cotyledons directly formed numerous somatic embryos at frequencies of up to 60%. Upon transfer onto MS basal medium,2 out of 15 somatic embryos converted into plantlets. The plantlets were potted to a soil mixture and grown to maturity in a phytotron.
Investigated were the relationships between plumule elongation characteristics and seedling emergence of 98 varieties including native, improved and red rice varieties of Korea, and varieties from U.S.A., Italy, Japan, etc. under proper and excess soil moisture regimes. Under excess soil moisture condition the seedling emergence and establishment rates were much lower in most of the varieties and the seedling emergence rates were delayed substantially in all varieties employed. The seedling emergence rates were lowest in dwarf and some semi-dwarf varieties showing poor plumule elongation under proper soil moisture condition. The seedling emergence and establishment rates were significantly correlated with the length of mesocotyl, coleoptile and incomplete leaf under both soil moisture regimes, and with the lower first and second internode length under excess soil moisture condition. Average emergence duration showed significant negative correlation with the length of mesocotyl, coleoptile and incomplete leaf under both soil moisture regimes and significant positive correlations with the first and third lower internode length. Based on the standardized partial regression analysis only the variations of mesocotyl and coleoptile length contributed significantly to the varietal difference in seedling establishment rate, and so did the variations of mesocotyl and incomplete leaf length to the varietal differences in emergence speed. In conclusion varieties with greater elongation characteristics of mesocotyl, coleoptile and incomplete leaf are desirable for higher and speedy seedling establishment in dry-direct seeding.
The seeds of three barley varieties of which initial seed vigor were different were used to measure seed vigor of accelerated aging techniques. A vigor index derived from the product of percent germination and plumule length was used to estimate seed vigor. The index was compared with the results of other tests. The results of warm germination test well suited to the measurements of seed vigor at the advanced stages of seed deterioration. Other vigor measurements except ATP and GADA values, showed close relationships with the vigor index. The measurements of plumule length in cold test and tetrazolium test were found to be appropriate for predicting seed quality.
An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.
Investigated were the relationships between plumule elongation characteristics and seedling emergence of 46 varieties including native, improved and red rice varieties of Korea, and varieties from U.S.A., Italy, India, Japan under 1, 3, and 5cm deep sowing with irrigated and non-irrigated condition. Experiments were carried out in paddy field of sandy loam. There was heavy shower of 19.2mm on the next day of seeding and thereafter, clear and dry weather continued during the experiment period. Soil temperature averaged over 30 days after seeding was $16.4^{\circ}C$ at 3cm depth. Soil hardness increased linearly up to 2.5kg /$cm^2$ on the 14th day after seeding, on which date irrigated plot was irrigated through furrow, and up to 4kg / $cm^2$ on the 28th day in non-irrigated plot. Soil hardness dropped near to 0kg /$cm^2$ after irrigation and developed up to 2.5kg /$cm^2$ again by 28 days after seeding. Seedling emergence was higher in irrigated plots than non-irrigated plots at all seeding depths. Korean improved varieties were substantially lower in seedling emergence under non-irrigated condition of 1 cm deep sowing than those under irrigated condition. This poor seedling emergence resulted mainly from delayed emergence by exposing them to greater soil strength. Percent seedling emergence under irrigated and non-irrigated condition showed signifi-cant correlations at 3 and 5 cm deep sowing. Korean improved varieties belonged to the group of poor seedling emergence, and I taliconaverneco, Chinsura Boro and Weld Pally to best group under both irrigation conditions at 3 and 5cm deep sowing. Seedling emergence showed highly signifi-cant positive correlation with the plumule length of mesocotyl + 1st internode + incomplete leaf and of mesocotyl+coleoptile. Among the characters constituting plumule length, incomplete leaf length showed greatest positive correlation followed by coleoptile and mesocotyl under irrigated condition at 3 and 5 cm deep sowing, and highest correlation with mesocotyllength followed by first internode and incomplete leaf under non-irrigated condition. Days to 50% seedling emergence at 1 cm deep sowing with irrigation showed great varietal variation of 10 to 30 days, and showed high significant negative correlations with percent seedling emergence under both irrigation conditions except for 1 cm deep sowing with irrigation, Days to seedling emergence revealed sig-nificant negative correlations with plumule characters except 2nd internode, showing highest cor-relation with incomplete leaf length.
Salinity stress is an important environmental problem that adversely affects crop production by reducing plant growth. The impacts of rhizobacterial strains to alleviate salinity stress on the germination of Lactuca sativa and Raphanus sativus seeds were assessed using different concentrations of NaCl. Plant growth-promoting rhizobacteria (PGPR) strains were also examined to improve the early germination of Chinese cabbage seeds under normal conditions. Lactobacillus sp. and P. putida inoculation showed higher radicle lengths compared with non-inoculated radish (Raphanus sativus) seeds. LAP mix inoculation increased the radicle length of lettuce (Lactuca sativa) seedlings by 2.0 and 0.5 cm at salinity stress of 50 and 100 mM NaCl concentration, respectively. Inoculation by Azotobacter chroococcum significantly increased the plumule and radicle lengths of germinated seeds compared with non-inoculated control. A. chroococcum increased the radicle length relative to the uninoculated seeds by 4.0, 1.0, and 1.5 cm at 50, 100, and 150 mM NaCl concentration, respectively. LAP mix inoculation significantly improved the radicle length in germinated radish seeds by 7.5, 1.3, 1.2, and 0.6 cm under salinity stress of 50, 100, 150, and 200 mM NaCl concentration, respectively. These results of this study showed that PGPR could be helpful to mitigate the salinity stress of different plants at the time of germination.
Salinity has been a threat to agriculture in some parts of the world; and recently, the threat has grown. Plant growth-promoting rhizobacteria (PGPR) may benefit plant growth, either by improving plant nutrition or producing plant growth hormones. The effects of rhizobacterial strains to attenuate the salinity stress on the germination of Chinese cabbage seeds were tested using four different concentrations of NaCl (50, 100, 150, and 200 mM). Also, PGPR strains were tested to enhance the early germination of Chinese cabbage seeds under normal conditions. Azotobacter chroococcum performed best with enhancing the radicle length of 4.0, 1.2, and 1.0 times at treatments of 50, 100, and 150 mM of NaCl, respectively. Additionally, significant differences were found in plumule length, A. chroococcum and Lactobacillus sp. showed remarkable activation either in normal or under stress conditions. Co-inoculation by three rhizobacterial strains (LAPmix) indicated synergistic effect to enhance the early germination of the seeds. The results of this study are promising for application of rhizobacterial strains that possess plant growth promoting traits to enhance the plant tolerance against salinity.
This experiment was conducted to study whether priming with deep sea water results in enhancement of seed germination and to identify the optimum concentration of the priming solution, and duration of priming using sweet pepper (Cv. California wonder), rice (Cv. Ilpum) and ginseng seed. Sweet pepper and rice seeds were primed with 5 various concentrations (5%, 10%, 15%, 20% and 30%) for deep sea water for 48 hours, 24 hours and 12 hours at $25^{\circ}C$ and ginseng seeds in 5%, 10%, 15%, 20%, 25% and 30%, and 2,4,6, and 8 electrical conductivity (EC) which were made by desalinating deep sea water. Priming in deep sea water (DSW) improved the early and final germination percentage, mean germinal on rate, emergence percentage and root and shoot length, compared with plain water, $KNO_3$ and without priming treatments. In sweet pepper, 24 hours priming with 5 percentage DSW significantly improved the early germination percentage and radical length. It has also improved the mean germination and emergence days and early emergence percentage, compared with $KNO_3$ and control. Whereas, in rice, 48 hours priming with 10 percent DSW significantly improved the early germination percentage, plumule emergence percentage, root length and shoot height. Hence the best seed priming treatment on sweet pepper and Rice are 24 hours with 5 percentage DSW and 48 hours with 10 percentage DSW, respectively, whereas in ginseng, priming with EC4, EC8 and 25% DSW had shown better germination.
This experiment was conducted on wild vegetables; Gondalbi (Cirsium setidens), Deoduck (Codonopsis lanceolata Trautv.), and Jandae (Adenophora triphylla var. Joponica Hara) seed to study whether priming with deep sea water results in enhancement of seed germination and identify the optimum concentration of the priming solution, and duration of priming. Seeds were primed with 5 various concentrations (5%, 10%, 15%, 20% and 30%) of deep sea water (DSW) in 12 hours, 24 hours, and 48 hours at $24^{\circ}C$. Since Jandae had seed dormancy, it was kept for four weeks in refrigerator at $2^{\circ}C$ after priming treatment. In Deoduck, 5 percentage DSW priming significantly improved the early germination percentage, radicle length, and plumule emergence percentage. Among the priming period of treatments, 24 hours priming showed better performance in this treatment whereas, in Jandae, 12 hours priming with 10 percentages DSW significantly improved the germination percentage and germination rate. This treatment had increased the final germination percentage by 54%, 15% and 40% compared with control, plain water and $KNO_3$ priming respectively. But in Gondalbi, priming did not improve the germination of seed. However, among the priming treatments, 12 hours priming with 3% $KNO_3$ and 20% DSW gave better performance. In both the wild vegetables; Deoduck and Jandae, priming in deep sea water had improved the germination percentage and germination rate as compare to plain water, $KNO_3$, and without priming treatment. Hence the best seed priming treatment on Deoduck and Jandae are 24 hours with 5% DSW and 12 hours with 10% DSW respectively.
