Compositions of free-sugars and sugar-alcohols in three cultivated mushrooms, Pleurotus ostreatus, Lentinus edodes and Agaricus bisporus were analyzed by gas-liquid chromatography and compositional differences of these components depending on species, sizes of fruiting body and portions(pileus and stipe) were investigated. Glucose, fructose, trehalose, glycerol, arabitol and mannitol were identified in each of the three mushrooms. The major components were mannitol and arabitol in Lentinus edodes, mannitol in Agaricus bisporus, trehalose and mannitol in Pleurotus ostreatus. The total free-sugars and sugar-alcohols content were in the range of 12.7-18.9% in Lentinus edodes, 9.2-16.1% in Agaricus bisporus and 8.2-10.6% in Pleurotus ostreatus. Total contents of these components in Pleurotus ostreatus and Agaricus bisporus were higher in pileus than those in stipe and it was decreased as pileus size development in all of three edible mushrooms.
We developed and characterized 36 polymorphic microsatellite markers for the oyster mushroom (Pleurotus ostreatus). In total, 169 alleles were identified with an average of 4.7 alleles per locus. Values for observed ($H_o$) and expected ($H_E$) heterozygosities ranged from 0.027 to 0.946 and from 0.027 to 0.810, respectively. Nineteen loci deviated from Hardy-Weinberg equilibrium. Significant (P<0.05) excess heterozygosity was observed at nine loci. Linkage disequilibrium (LD) was significant (P<0.05) between pairs of locus alleles. Cluster analysis revealed that five species of genus Pleurotus made a distinct group, and the individual cultivars were grouped into major five groups from G-1 to G-5. The diverse cultivars of P. ostreatus were discriminated and the other four species revealed a different section in the UPGMA tree. These microsatellite markers proved to be very useful tools for genetic studies, including assessment of the diversity and population structure of P. ostreatus.
This study aimed to investigate possible cause of slow and abnormal growth of oyster mushrooms, Pleurotus florida and P. ostreatus collected from bad crop farms. Spherical virus particles of 30 nm in diameter from P. florida, 23 nm particles from P. ostreatus, and both 23 and 30 nm particles were also found from interspecies mated culture between P. florida and P. osreatus. The virus particles might be associated with the bad crop of Pleurotus species.
This study was carried out to develop techniques for the log cultivation of Pleurotus pulmonarius. Soil landfill cultivation of the in plastic container boxes containing yield per log than but there was no difference in the yield from both spawn Hyangsan" variety. In the case of soil landfill cultivation in a shaded vinyl house, an Ailanthus tree gave a higher yield than that using poplar tree, and the yield of the Hosan" variety was higher than that of the Hyangsan" variety. With regard to proper tree species selection, willow and cherry trees were good for both the Hosan" and "Hyangsan" variet.
Some factors affecting mycelial growth of Pleurotus sajor-caju and P. ostreatus. were investigated. The optimal temperature and pH of the medium for mycelial growth of both the species were $25^{\circ}C$ to $30^{\circ}C$, and 5 to 6, respectively. The effect of carbon and nitrogen sources on mycelial growth of the species was studied using modified Waksman's special medium as the basal medium. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of P. sajor-caju, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth of P. ostreatus. Generally the organic nitrogen compounds showed good mycelial growth of both the species. The mycelial growth of the species were favored by the ammonium nitrogen more than by the nitrate nitrogen source. Asparagine was superior to the other kinds of amino acid. Six percentage of glucose and 0.6% ammonium tartarate was optimum for mycelial growth of P. sajor-caju whereas the optimum concentration for the P. ostreatut was 4% glucose and 0.4% ammonium tatarate.

The primary objective of the present study is the characterization of the hybrids of monokaryon- monokaryon (mono-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus species strains with 85 mono-mono intraspecific hybrids of 7 combinations between six Pleurotus ostreatus strains and one Pleurotus florida strain. In this study, the results of analysis on hybridization rate, nuclear DNA patterns, and colors and yields of fruit-bodies, are presented as follows. The crossability between mono-mono crossing ranges between 50 and 93.75%. The results of the analysis on the nuclear DNA patterns of 85 hybrid strains of mono-mono crosses share the nuclei of both parents, but their genetic similarities were predominated by either parent. The hybrid strain between P. florida and P. ostreatus showed patterns more similar to P. florida, while the hybrid strain between P. ostreatus and P. ostreatus either had patterns predominated by either parent strain. The fruiting body colors of the mono-mono crosses mostly had combined colors of both parents but showed the tendency of being more similar to that of either parent. 82% of the hybrid strain indicated similar fruiting body yields compared to both parent strains, while 0% was higher and 18% were lower than both parents. The present study was able to find out and suggest superior hybrid trains by identifying the nuclear DNA patterns of hybrids between Pleurotus species as well as the characteristics of their fruiting bodies. This study expects that the advantages of the mono-mono crossing are needs to be fully utilized in mushroom breeding and it is better to develop superior strains of Pleurotus species strains together with the mono-mono crossing.
Pleurotus ferulae is an edible mushroom found on a medicinal plant, Ferula assa-foetida, in centeral China. This study was carried out to investigate the cultural characteristics of P. ferulae strains. Characteristics of mycelial growth were investigated for 5 strains of P. ferulae. All the 5 strains showed the best mycelial growth at $25^{\circ}C$ and their growth rate was very low at $35^{\circ}C$. The colony diameter reached to 45~72 mm after 7 days at $25^{\circ}C$. Malt extract agar medium was the best for mycelial growth of the species both in hyphal length and density. Mycelial growth was not affected by various pH from 6.0 to 8.0. An optimal carbon source was arabinose and an optimal nitrogen one was arginine. And an effective substrate for the mycelial growth was 8 to 2 mixture of oak sawdust and cottonseed meal.
Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.

In this study, 81 commercial strains of Pleurotus species cultivated in South Korea were analyzed with randomly amplified polymorphic DNA (RAPD) technique. Sequence characterized amplified region (SCAR) markers were developed by designing from one RAPD polymorhic band specific to Suhan strain. The SCAR primer pair 'S-OPA13-1' amplified a 590-bp fragment in the varieties originated from Suhan strain. The Blast search of S-OPA13-1 showed high homology to the POMFBO1 P. ostreatus cDNA clone MFB02-A05 and Laccaria bicolor S238N-H82. The results showed that this SCAR marker can clearly distinguish Suhan strains from Pleurotus spp.
Isolates of the different species groups of Trichoderma from the mushroom culture beds were identified according to Rifai's classification and influence of antibiotics produced by them against the oyster mushroom was examined. Trichoderma islolates were identified as Trichoderma hamatum, Trichoderma viride and Trichoderma koningii. Among the Trichoderma isolates, fungistatic action of Trichoderma viride was found to be most remarkable. Pleurotus ostreatus and Pleurotus sajor-caju were the most susceptable of the edible mushrooms tested, followed by Lentinus edodes, Flammulina velutipes and Auricularia auricula. A needle-shaped crystal gained from the chloroform extract of the culture filtrate of Trichoderma viride repressed distinctively the mycelial growth of the oyster mushroom. The grade of repression of the crystal at 500ppm and 1/10 aequous solution of the chloroform extract against the oyster mushroom, seemed equal to that of cycloheximide at $100{\sim}200ppm$.
