Characteristics of Seedling Establishment and Yield of Platycodon grandiflorus by Ridge Width and Mulching Materials (이랑폭과 피복재에 따른 도라지의 입모율 및 수량특성)

  • This study was carried out to investigate the seedling establishment and yield of direct-sown Platycodon grandiflorus seeds cultured by three ridge width, 0.4, 0.8, and 1.2 m and four mulching materials, rice straw, rice bran, black plastic film, and no mulching in upland. Seedling establishment rate (70%) was the highest in rice straw mulched plot of 500 seeds/$m^2$ sowed and root yield after one year was also the highest in rice straw mulched treatment and followed by rice bran, no mulching, and black plastic film treatment. In this result, ridge width 120cm and rice straw mulching combined treatment was best for getting the highest seedling establishment ratio and seedling numbers per area. However, ridge width and mulching materials should be considered soil moisture content and weed population for saving labor cost.

  • Histone deacetylases (HDACs) play a pivotal role in epigenetic regulation, affecting the structure of chromatin and gene expression across different stages of plant development and in response to environmental stresses. Although the role of HDACs in Arabidopsis and rice has been focused on in extensive research, the role of the HDAC gene family in various medicinal plants remains unclear. In the genome of the balloon flower (Platycodon grandiflorus), we identified 10 putative P. grandiflorus HDAC (PlgHDAC) proteins, which were classified into the three families (RPD3/HDA1, SIR2, and HD2 HDAC families) based on their domain compositions. These HDACs were predicted to be localized in various cellular compartments, indicating that they have diverse functions. In addition, the tissue-specific expression profiles of PlgHDACs differed across different plant tissues, indicating that they are involved in various developmental processes. Furthermore, the expression levels of all PlgHDACs were upregulated in leaves after waterlogging treatment, implying their potential role in coping with waterlogging-induced stress. Overall, our findings provide a comprehensive foundation for further research into the epigenetic regulation of PlgHDACs, and particularly, on their functions in response to environmental stresses such as waterlogging. Understanding the roles of these HDACs in the development and stress responses of balloon flower could have significant implications for improving crop yield and the quality of this important medicinal plant.

  • This study was carried out to investigate the characteristics of Rhizome rot incidence of Platycodon grandiflorus by cultivation period and ridge width and furrow depth. The three types of ridge width 0.8, 1.0, and 1.2 m and several levels of furrow depth and 6 levels cultivation period. This experiment was done in farmer's farm and the treatments were also classified in the same spot. In this investigation, ridge width, soil water content, soil hardness, and cultivation period were positively related with Rhizome rot incidence, however, furrow depth was negatively related with that. So this experiment could draw a conclusion : excess water damage and soil hardness could directly or indirectly effect on the Rhizome rot incidence, so cultivation method should be developed such as making underground ditch or cultivation in well draining soil for escaping excess waster damage.

Studies on the Volatile Components of Platycodon grandiflorus (jacquin) A.De Candolle (도라지의 휘발성분에 관한 연구)

  • To identify the volatiles of Platycodon roots, the three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were extracted with ethyl ether by SDE method. 54.71mg% and 80.01mg% of volatile components were collected in the three-year-old and in 24-year-old Platycodon roots, respectively. As a result of GC/MS(TIC) analysis, 24 and 28 compounds were identified, respectively; The amounts of volatile components such as hexanol, trans-2-hexenol and nonanal in 24-year-old roots were detected above ten times than those in 3-year-old roots. The Volatiles of 24-year-old roots were fractionated by SCC and identified by GC/MS to be 10 hydrocarbons, 8 alcohols, 10 aldehydes, 5 esters, 6 acids and 1 unknown compound as the major components. The species and total amounts of volatile component in 24-year-old Platycodon roots were considered to be higher than those in 3-year-old Platycodon roots. *Abbreviations 1. Cytoprotective Effects of Platycodon grandiflorus (Jacq.) A.DC on C6 Glioma Cell Apoptosis by Oxidative Stress (산화스트레스로 인한 C6 Glioma Cell Apoptosis에 대한 길경의 세포보호효과)

  • This study is designed to investigate cytoprotective effects of Platycodon grandiflorus (Jacq.) A.DC on C6 glioma cell apoptosis by oxidative stress. Experimental C6 glioma cells were classified into four groups as follows: normal group, PGE group, chemical groups, PGE+chemical groups. Oxidative stress that caused by chemicals in the C6 glioma cell, check the impact to Chemical group was administered normal group. Apoptotic effect protecting in order to observe the chemical group was administered PGE. We to observe effects of PGE on SOD inhibition, total glutathione production in C6 glioma cells were administered PGE. In case of administration PGE, apoptosis induced by Paraquat was significantly decreased. In case of administration PGE, apoptosis induced by SNP was significantly decreased. In case of administration PGE, apoptosis induced by $H_2O_2$ was significantly decreased. In case of administration PGE, apoptosis induced by Rotenone was decreased, but the statistical significance was not. In case of administration PGE, SOD inhibition activities significantly decreased. In case of administration PGE, Total glutathione did not affect the content. These results suggest that PGE is able to treat a disease caused by oxidative stress and prevent a aging. These results suggest that PGE is a disease caused by oxidative stress and aging, the prevention and treatment of food shall be able to be applied.

Anti-inflammatory Effects, Skin Wound Healing, and Stability of Bluish-purple Color Extracted from Platycodon grandiflorus (Jacq.) A.DC. Flower Extract (도라지꽃 추출물의 항염증, 피부재생 효과 및 색소 안정성 연구)

  • Platycodon grandiflorus (P. grandiflorus) flower is a perennial plant belonging to the family Campanulaceae and has many excellent pharmacological effects, so it has been used as a medicinal ingredient since ancient times. In addition, anthocyanin is a purple or blue natural pigment contained in plant flowers and fruits, and is known as a powerful antioxidant. The purpose of this study was to confirm the dermatological functionality of P. grandiflorus flower extract and the value of the bluish anthocyanin contained in flowers as a cosmetic material as a natural pigment. Firstly, 50% ethanol and 80% ethanol were added to the P. grandiflorus flower and extracted under reflux for 4 h at 25, 60, and 80 ℃, and the pH of each treatment group was similar. Based on the anthocyanin content and chromaticity (E*ab), 50% ethanol 60 ℃ extraction conditions showing the color development most similar to the natural color of the P. grandifloras flower were selected, and a sample was prepared by concentrating and lyophilizing. The analysis results showed that the total phenol, total flavonoid, and total anthocyanin contents were in the ranges of 23 ㎍/mL, 16 ㎍/mL, and 0.17 ㎍/mL, respectively. The P. grandiflorus flower extract suppressed the production of nitric oxide (NO) and interleukin-6 (IL-6) in lipopolysaccharide (LPS) induced RAW264.7 cells. Furthermore, the P. grandiflorus flower extract showed wound healing effects through the promotion of skin cell migration in TNF-α stimulated human keratinocytes. The stability of anthocyanin and extract color was studied during a storage period of 50 days at various temperatures (4 ℃, 25 ℃, and 45 ℃). Color values (L, a, and b) of the P. grandiflorus flower extract changed over 50 days, whereas the bluish-purple color of the extract was stabilized using 5% maltodextrin. These results suggest that P. grandiflorus flower extract may be useful as a natural cosmetic pigment.

Growth and Development of Platycodon grandiflorus under Sensor-based Soil Moisture Control on Open Farmland and Pot Conditions

  • Soil moisture control system including soil sensing and automatic water supply chain was constructed on open farmland and pot conditions. Soil moisture was controlled by the system showing over the soil moisture contents except 40% treatment. EC was gradually decreased by increasing cultivation days. On applying this system to control soil moisture, the growth and development characters of the bellflower were improved compared with control, cultivation without the automatic irrigation. Of the growth and development characters, plant height with water treatments was higher than that of control in 1st-year plants. Moreover, numbers of branch were increased by the increased soil moisture on farmland and pot condition. Capsule numbers for seed were best at 20%, 30% soil moisture treatment in 1st-year plants, and 20% to 50% treatment in 2nd-year plants. The construction of automatic soil moisture control system provide fundamental data for plant growth and development on open farmland soil condition.

Beneficial constituents and physiological activity of fermented Codonopsis lanceolata and Platycodon grandiflorus by Lentinula edodes mycelium (더덕, 도라지 표고균사발효물의 유용성분 및 생리활성)

  • This study was conducted to develop food and medicinal products containing useful components of Lentinula edodes in Codonopsis lanceolata and Platycodon grandiflorus for use as herbal medicine. We manufactured C. lanceolata (FCLM) and P. grandiflorus (FPLM) extract fermented with L. edodes mycelium. The effect of the two fermented products on proximate composition, free sugar, organic acid, 𝛽-glucan, ergothioneine, ergosterol, and vitamin D2 levels, and 3T3-L1 preadipocyte cell growth were studied. The proximate composition analysis results showed that the crude fiber and crude fat content in FCLM was higher than that in FPLM, and the crude protein and soluble nitrogen content in FPLM was higher than that in FCLM. Free sugar analysis detected arabinose, glucose, and sucrose in both FCLM and FPLM, and the total free sugar content was high in FPLM. The organic acid content was lower in FCLM and FPLM compared to C. lanceolata and P. grandiflorus before fermentation. The 𝛽-glucan content was higher than that of L. edodes used as a control in both fermented products, FCLM and FPLM. The content of ergothioneine, an antioxidant, was higher in FCLM than in FPLM. Ergosterol content was highest in L. edodes which was used as a control, and the two fermented products showed similar content. Vitamin D2 was detected only in FCLM and FPLM, and FPLM (0.58±0.01 mg%) showed a higher vitamin D2 content than FCLM (0.47±0.01). FCLM and FPLM showed a higher level of cell viability for 3T3-L1 pre-adipocytes compared to non-fermented C. lanceolata and P. grandiflorus. In addition, FCLM and FPLM inhibited 3T3-L1 preadipocyte differentiation more than C. lanceolata and P. grandiflorus before fermentation, which may exert an anti-obesity effect.

  • Platycodon root contains abundant pharmaceutical substances and is widely used as a food and a medicinal herb. The three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were analysed and compared with their pharmaceutical substances to identify the major components. Reverse column HPLC analysis of ethanol extracts from the 24-year-old roots showed some distintive peaks after the retention time of 50 min which were absent in the extracts from the three-year-old roots. This indicates that several pharmaceutical substances are present in the older roots. There were no differences in sterol composition and saponin content between the 24-year and three-year-old roots. The 15-year-old and the 23-year-old Platycodon roots, the 6-year-old Ginseng root, and the three-year-old Condonopsis root all did not show significant anticomplement activity.

  • Platycodon root contains abundant pharmaceutical substances and is widely used as a food and a medicinal herb. To identify the major components of the three-year-old and the 24-year-old platycodon roots cultivated in a local farm were analyzed and compared with their chemical compositions prior to their pharmaceutical substances, which were used as the fundamental data. There were no differences in chemical composition, inorganic compositions and fatty acid compositions between the 24-year-old and three-year-old roots. Total amino acid contents were 375.8 mg% and 891.6 mg% in the 24-year-old and three-year-old platycodcon roots, respectively. Sixteen species of amino acid were identified by amino acid autolyzer except cysteine both in the 24-year-old and three-year-old roots. Linoleic acid, which was the highest fatty acid contained both in the 24-year-old and three-year-old roots, was estimated to be 48.7% and 39.1% of total fatty acids, respectively.
