Lactobacillus plantarum 299v Surface-Bound GAPDH: A New Insight Into Enzyme Cell Walls Location

  • The aim of this study was to provide new insight into the mechanism whereby the housekeeping enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH) locates to cell walls of Lactobacillus plantarum 299v. After purification, cytosolic and cell wall GAPDH (cw-GAPDH) forms were characterized and shown to be identical homotetrameric active enzymes. GAPDH concentration on cell walls was growth-time dependent. Free GAPDH was not observed on the culture supernatant at any time during growth, and provoked cell lysis was not concomitant with any reassociation of GAPDH onto the cell surface. Hence, with the possibility of cw-GAPDH resulting from autolysis being unlikely, entrapment of intracellular GAPDH on the cell wall after a passive efflux through altered plasma membrane was investigated. Flow cytometry was used to assess L. plantarum 299v membrane permeabilization after labeling with propidium iodide (PI). By combining PI uptake and cw-GAPDH activity measurements, we demonstrate here that the increase in cw-GAPDH concentration from the early exponential phase to the late stationary phase is closely related to an increase in plasma membrane permeability during growth. Moreover, we observed that increases in both plasma membrane permeability and cw-GAPDH activity were delayed when glucose was added during L. plantarum 299v growth. Using a double labeling of L. plantarum 299v cells with anti-GAPDH antibodies and propidium iodide, we established unambiguously that cells with impaired membrane manifest five times more cw-GAPDH than unaltered cells. Our results show that plasma membrane permeability appears to be closely related to the efflux of GAPDH on the bacterial cell surface, offering new insight into the understanding of the cell wall location of this enzyme.

The Cryoprotective Effect on Frozen-thawed Boar Semen of Egg Yolk Low Density Lipoproteins

  • In order to protect the spermatozoa against cold shock, hen egg yolk is widely used as a cryoprotective agent in semen freezing extenders for domestic animals. The protective action of yolk is largely presumed to be due to low density lipoproteins (LDL). The effects of LDL on sperm quality of bull and northern pike (Esox lucius) after freezing-thawing have been reported, but no study has been made to evaluate the effect of LDL on boar sperm motility and other characteristics. The experiment was carried out to investigate the effect of LDL on the freezing of boar sperm in 0.25 ml straws. The aim was to evaluate the quality of boar spermatozoa cryopreserved in the presence of LDL. Motility of semen cryopreserved in LDL was analyzed and compared to semen cryopreserved with Tris-citric acid-glucose (TCG) and Tris-citric acid-fructose (TCF), two basic freezing extenders containing egg yolk. Similarly, acrosome and plasma membrane integrity were also evaluated and compared to semen cryopreserved with TCG and TCF. Analysis of sperm quality after freeze-thaw showed that the motility, acrosome and plasma membrane integrity were improved with LDL in the extender, as compared to the TCG and TCF. The highest post-thaw integrity of acrosome and plasma membrane and motility were obtained with 9% LDL (w/v). Consequently, the optimum LDL concentration in the extender was 9%. It is also suggested that the concentration of LDL addition is important for the effect on boar sperm protection during freezing and thawing. The percentage of motile spermatozoa was significantly higher after freezing in 9% LDL than in TCG and TCF 54.4% versus 30.4% and 30.1% (p<0.05), respectively. The integrity of acrosome and plasma membrane were also significantly higher at 70.3% and 50.5% respectively with semen frozen in 9% LDL extender compared to TCG at 37.8% and 30.3% and TCF at 36.4% and 29.9%, respectively (p<0.05),. In conclusion, we propose that extender containing LDL extracted from hen egg yolk could be used as a cryoprotective media with a better efficiency than TCG and TCF. LDL improved boar semen quality, allowing better spermatozoa motility, acrosome and plasma membrane integrity after the freeze-thaw process. Furthermore, we found out that the extender with 9% LDL concentration significantly enhanced motility, acrosome and plasma membrane integrity of boar sperm after freezing and thawing.

다공질 세라믹지지 촉매 상에서의 플라즈마 방전을 이용한 휘발성유기화합물의 분해 (Electrical Discharge Plasma in a Porous Ceramic Membrane-supported Catalyst for the Decomposition of a Volatile Organic Compound)

  • 다공질 세라믹 막을 사용하는 플라즈마-촉매 반응기에서 휘발성유기화합물의 분해가 수행되었다. 저압차 촉매 지지체로 사용된 세라믹 막에 광촉매인 산화아연을 담지하여 휘발성유기화합물의 산화 성능을 개선하고자 하였다. 교류 고전압에 의해 구동되는 플라즈마가 다공질 세라믹 막 내에서 전개되면서 휘발성유기화합물의 분해에 이용되는 라디칼, 오존, 이온, 여기상태 분자 등 다양한 활성성분을 생성하게 된다. 반응기에 공급되는 고전압이 증가함에 따라 플라즈마가 점차 방사방향으로 전개되어, 일정 전압을 넘어서면 세라믹지지체 전체적으로 균일한 플라즈마가 생성되었다. 휘발성유기화합물 분해 성능 평가에는 에틸렌이 이용되었다. 전기에너지밀도, 반응기 입구 에틸렌 농도, 촉매 담지 여부, 기체 조성에 따른 에틸렌 분해효율이 조사되었다. 같은 에너지 밀도에서 비교하면 산화아연이 담지된 촉매에서의 에틸렌 분해 효율이 담지되지 않은 경우보다 더 높은 것으로 나타났으며, 기체 조성 변화 실험을 통해 폐가스의 주요 구성성분인 산소와 질소 모두 에틸렌의 분해를 개시하는데 중요한 역할을 함을 알 수 있었다. 일반적인 기상반응과 달리, 플라즈마 반응기에서의 에틸렌 분해 반응은 활성 성분의 양에 의해 지배되므로, 방전 전력이 동일할 경우 에틸렌 농도가 높아질수록 분해효율이 저하되었다.

  • Plasma processes developed in our laboratory, of interest for biomaterials, barrier coatings for food packaging and corrosion protection, are briefly reviewed in this contribution. Particular attention is devoted to diagnostics aimed to rationalize plasma-surface interactions and to identify parameters and correlations to be utilized for process control.

Establishment of Optimal Conditions for the Hypoosmotic Swelling Test to Evaluate the Integrity of Spermatozoal Plasma Membrane in Dog

  • Hypoosmotic swelling test (HOST) is used for evaluating the plasma membrane function and fertilizing ability in mammal spermatozoa. However, HOS solutions and experimental conditions have not been determined clearly for assessing canine spermatozoa. This study was conducted to examine the HOS solutions and assay conditions, including incubation time (30 to 120 min), storage temperature (4, 17 and $20^{\circ}C$), semen status (fresh and frozen). Maximum spermatozoal plasma membrane swelling was obtained in an 150 mOsm Na-citrate/Fructose solutions with an incubation time for 45 min. The storage temperature and semen status affected the percentage of HOS positive spermatozoa. The HOS test adapted to canine spermatozoa in this study was simple and highly consistent assay with good repeatability. The optimal condition of HOST in canine spermatozoa is an 150 mOsm Na-citrate/Fructose solutions with an incubation time for 45 min regardless of semen storage temperature and semen status.

  • Synaptosomal plasma membrane vesicles (SPMV) were isolated from fresh bovine cerebral cortex by several well-known procedures, and the best procedures was selected by enzymatic and morphological standards. The SPMV isolated by the modified procedure of Smith and Loh showed the highest purity. The specific activities of Na, K-ATPase, acetylcholinesterase and 5'-nucleotidase were about 5, 6-fold, 2, 5-fold and 3, 3-fold, respectively, enriched in the plasma membrane fraction as compared to those of the crude homogenate. Electron microscpic examination also showed that the membranes were in vesicular form.

Effect of bicarbonate and progesterone on plasma membrane integrity, acrosome reaction and proportion of fatty acids in boar sperm

  • This study investigated the influence of sodium bicarbonate (NaHCO3) and progesterone on acrosome reaction and proportion of polyunsaturated fatty acid (PUFA) composition boar sperm. The sperm were diluted with semen extender and incubated with NaHCO3 and progesterone at 38℃, 5% CO2 for 6 h. Plasma membrane integrity and acrosome reaction were analyzed using SYBR14/propidium iodide (PI) and FITC-PNA/PI doubling staining method, and proportion of PUFA was analyzed using gas chromatography. In results, Plasma membrane integrity was significantly decreased in 50 mM NaHCO3 group and acrosome reaction was significantly increased by over the 100 mM NaHCO3 group compared to control group (p < 0.05). In addition, progesterone significantly increased decreased plasma membrane integrity at 100 mM progesterone and acrosome reaction at over the 5.0 µM progesterone (p < 0.05), but there was no difference among the 5.0 to 100 µM groups. PUFAs were significantly decreased in 100 mM NaHCO3 and 50 µM progesterone treatments compared to control group. In summary NaHCO3 and progesterone induce acrosome reaction and reduce PUFA composition in boar sperm, therefore, the results maybe help to understand basically knowledge for the acrosome reaction and PUFA composition in boar sperm.

플라즈마 처리된 폴리스티렌 막을 통한 순수한 CO2 와 N2 기체의 선택·투과 특성 (Selectivity and Permeability Characteristics of Pure CO2 and N2 Gases through Plasma Treated Polystyrene Membrane)

  • 폴리스티렌 막(polystyrene membrane, PS)의 표면을 Ar, $O_2$ 플라즈마로 처리하고, 처리 전후의 변화를 관찰하였고, $CO_2$, $N_2$의 투과도와 $N_2$에 대한 $CO_2$의 선택도는 연속흐름 기체 투과 분석장치(GPA)를 이용하여 측정하였다. Ar플라즈마 처리의 경우 O/C비율이 0에서 0.179로 증가하고, 표면 거칠기가 $15.86{\AA}$에서 $71.64{\AA}$로 증가함으로써 접촉각은 처리전의 $89.16^{\circ}$에서 $18.1^{\circ}$로 감소하였다. 따라서 플라즈마 처리는 막표면을 높은 친수성을 갖도록 만들었다. $CO_2$의 투과도와 선택도에 대한 Ar플라즈마 처리최적조건은 60 W, 2 min, $70^{\circ}C$이며, 투과도와 선택도는 각각 $2.1{\times}10^{-12}[m^3(STP){\cdot}m/m^2{\cdot}sec{\cdot}atm]$와 4.51이었다. $O_2$플라즈마 처리의 경우에, 접촉각은 O/C비율(0.189)과 표면 거칠기($57.10{\AA}$)의 증가에 의해 $13.56^{\circ}$로 감소하였다. 최적의 처리조건은 90 W-2 min-$70^{\circ}C$이며, 값 $7.1{\times}10^{-12}[m^3(STP){\cdot}m/m^2{\cdot}sec{\cdot}atm]$와 값 11.5이었다. 플라즈마 처리 후 막 표면의 변화는 표면에서의 교차결합과 식각효과의 경쟁적인 관계에 의해 결정된다. 결국 플라즈마 처리된 막의 투과도와 선택도가 플라즈마 기체, 처리시간, 출력세기등과 같은 플라즈마 상태를 제어함으로써 향상되었음을 확인할 수 있었다.

Salt Repellent Behavior of Superhydrophobic Filtration Membrane

  • In this study, we present the salt repellent behavior of superhydrophobic filtration membrane. Bio-fouling or mineral-fouling is the main factor of decreasing the performance of filtration membrane. The superhydrophobic modification of filtration membrane using PECVD (Plasma enhanced chemical vapor deposition) is introduced. The oxygen plasma was introduced for developing nano hairy structures and subsequent HMDSO (Hexamethyldisiloxane) coating was used for enhancing low surface energy. Saline water evaporation test was carried out to evaluate the difference of contamination of salt on superhydrophobic and moderately hydrophobic filtration membrane. EDS and EPMA were used for visualizing the residue of salt crystal.
