  • The aim of the present experiments was to investigate the effect of Yukmijihwangtang water extracts on the plasma renin activity and plasma levels of atrial natriuretic peptide and aldosterone in rats. The results of this study were as follows: 1. Plasma renin activity decreased significantly after the administration of Yukmijihwangtang water extracts 1.5ml/kg. 2. Plasma levels of atrial natriuretic peptide (ANP) decreased significantly after the administration of Yukmijihwangtang water extracts. 3. Plasma levels of aldosterone increased significantly after the administration of Yukmijihwangtang water extract.

  • The aim of the present experiments was to investigate the effect of Yukmijihwangtang water extracts on the plasma renin activity and plasma levels of atrial natriuretic peptide and aldosterone in rats. The results of this study were as follows: 1. Plasma renin activity decreased significantly after the administration of Yukmijihwangtang water extracts 1.5 ml/kg. 2. Plasma levels of atrial natriuretic peptide (ANP) decreased significantly after the administration of Yukmijihwangtang water extracts. 3. Plasma levels of aldosterone increased significantly after the administration of Yukmijihwangtang water extract.

  • The aim of this experiments was to investigate the effect of Lycii Fructus water extracts on the blood pressure, plasma renin activity and plasma levels of atrial natriuretic peptide in two-kidney one clip Goldblatt hypertensive rats. Systolic blood pressure was decreased significantly after administration of Lycii Fructus water extracts. Plasma levels of atrial natriuretic peptide was increased significantly after adminstration of Lycii Fructus water extracts. Plasma renin activity was not changed after adminstration of Lycii Fructus water extracts. Thease results suggested that the depressor response after adminstration of Lycii Fructus water extracts were related with the changes of the plasma levels of atrial natriuretic peptide.

  • The changes of plasma renin activity, aldosterone concentration, serum sodium-, and potassium levels were studied before and after the water loading followed by diuretics injection. The materials were: 13 non-, 11 normal-, and 11 toxemic pregnancy cases. The plasma renin activity and aldosterone concentration of the cord and postpartum blood were also measured. Following were the results: 1. The plasma renin activity was elevated significantly in normal pregnancy, and slightly in toxemic pregnancy. The serum sodium levels were decreased in pregnancy. 2. The plasma aldosterone concentration was slightly decreased in normal pregnancy, and slightly increased in toxemic pregnancy, however, statistically insignificant. 3. The plasma renin activity of the cord and postpartum blood were lower than those of pregnancy cases. 4. The changes of plasma renin activity after the diuretic administration showed an initial increase, which recovered within 2 hours. These changes were the least in normal pregnancy, and the most in toxemic pregnancy. 5. The changes of plasma aldosterone concentration after the diuretic administration were similar to those of plasma renin activity, although the variations were not so wide.

  • The aim of this experiments was to investigate the effecd of Radix Aconiti water extrac on the plasma renin activity and plasma levels of aldosterone and atrial natriuretic peptide in rats. The results of study were as follows; Plasma renin activity was decreased significantly after administration of Radix Aconiti water extract. Plasma levels of aldosterone was decreased significantly after administration of Radix Aconiti water extract. Plasma levels of atrial natriuretic peptide was not changed after administration of Radix Aconiti water extract.

  • Rehmannia Radix is a thick tuberous root about 3-6 cm diameter. The tuberous root is repared for medicine. The material in the fresh state is shengdihuang (生地黃). The material in the dried state is gandihuang (乾地黃). Shudihuang (熱地黃) is made by taking juicy roots, washing them in millet wine, steaming on a willow frame in a percelain vessel, drying, and resteaming and redrying nine times. The aim of this experiments was to investigate the effect of Rehmannia Radix Water extracts on the plasma renin activity and plasma levels of aldosterone and atrial natriuretic peptide (ANP) in rats. The results of study were as follows: Plasma renin activity was not changed after administration of Rehmannia Radix water extracts. Plasma levels of aldosterone was decreased significantly after administration of Shudihuang (熱地黃) water extracts. Plasma levels of atrial natriuretic peptide was increased significantly after administration of Shudihuang (熱地黃) water extracts.

  • The aim of this experiment was to investigate the effects of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts on the blood pressure, plasma renin activity, plasma levels of aldosterone and atrial natriuretic peptide (ANP) in spontaneously hypertensive rats (SHR). The results of this study were as follows: 1. Systolic blood pressure decreased significantly after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma, and Coptidis Rhizoma water extract 0.2 ml/200g. 2. Plasma renin activity and plasma levels of ANP were not changed after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts, 3. Plasma levels of aldosterone was increased significantly after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts 0.1 ml/200g.

Effects of Angiotensin Converting Enzyme Inhibition on Gene Expression of the Renin-Angiotensin System in Rats

  • To investigate interaction of angiotensin converting enzyme (ACE) inhibitor with local tissue renin- angiotensin system (RAS), changes in gene expression of the RAS components in various tissues in response to chronic administration of an ACE inhibitor, enalapril, were examined in Sprague-Dawley male rats. Enalapril was administered in their drinking water $(3{\sim}4\;mg/day)$ over 8 wk. Plasma and renal ACE activity increased significantly after 4 and 8 wk of enalapril treatment. Renin levels of the plasma and kidney of the enalapril-treated rats markedly increased after 4 wk and decreased thereafter, but still remained significantly higher than those of control rats. Kidney mRNA levels of renin markedly increased after 4 and 8 wk of enalapril treatment, but those of angiotensinogen and ANG II-receptor subtypes, $AT_{1A}$ and $AT_{1B}$, did not change significantly. The liver expressed genes for renin, angiotensinogen and $AT_{1A}$ receptor subtype, but $AT_{1B}$ receptor subtype mRNA was not detectable by RT-PCR. None of mRNA for these RAS components in the liver changed significantly by enalapril treatment. The hypothalamus showed mRNA expressions of renin, angiotensinogen, $AT_{1A}$ and $AT_{1B}$ receptor subtypes. $AT_{1A}$ receptor subtype mRNA was more abundant than $AT_{1B}$ receptor subtype in the hypothalamus as shown in the kidney. However, gene expression of the RAS components remained unchanged during 8-wk treatment of enalapril. In the present study, chronic ACE inhibition increased plasma and renal levels of ACE and renin, but did not affect mRNA levels of other RAS components such as angiotensinogen, ANG II receptor subtypes in the kidney. Gene levels of the RAS components in the liver and hypothalamus were not altered by chronic treatment of enalapril. These results suggest the differential expression of the RAS components in response to enalapril, and localized action and some degree of tissue specificity of enalapril.

  • The aim of the experiments was to investigate the effect of Semen Cuscutae aqua-acupuncture at the meridian point BL 20(脾兪) and BL 23(賢兪) on the blood pressure, plasma renin activity, plasma levels of aldosterone and atrial natriuretic peptide (ANP) in spontaneously hypertensive rats (SHR). The results of this study were as follows: 1. Systolic blood pressure was decreased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20, BL 23. 2. Plasma renin activity was increased significantly after Semen Cuscutae aqua-acupuncture at meridian point BL 23, BL 20. 3. Plasma levels of aldosterone was increased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20. 4. Plasma levels of atrial natriuretic peptide (ANP) was increased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 23, but it was decreased significantly after Semen Cuscutae aqua-acupuncture at the meridian point BL 20. These results suggest that the changes of the depressor response after Semen Cuscutae aqua-acupuncture at the meridian point BL 20, BL 23 are related to the changes of the plasma renin activity, plasma levels of atrial natriuretic peptide (ANP) and aldosterone.
