가르시니아캄보지아 추출물 (HCA)과 L-Carnitine의 섭취 및 운동이 흰쥐의 체중에 미치는 영향 (Effect of Feeding Garcinia Cambogia Extract (HCA) and/or L-Carnitine and Exorcise on Body Weight in Rats)

  • This study was performed to investigate the effect of feeding garcinia cambogia extract (HCA) and/or L-carnitine and exercise (swimming) on body weight in rats. Forty-eight male rats (Charles River CD (SD) IGS) of eight weeks old and weighing $323.5\pm2.4g$ were raised for two months with high fat diet ($40\%$ fat as calorie) to induce obesity. After induction of obesity, rats weighing $552.8\pm5.8g$ were blocked into eight groups according to body weight and raised for six weeks with diet containing HCA and/or L-carnitine. Plasma aspartate aminotransferase, alanine aminotransferase, total protein and albumin levels were in normal ranges. Food and calorie intakes of H + C and H + C/H groups were highest among non-exercise (m) groups and exercise (I) groups. Food and calorie intakes of m groups were higher than those of I groups. E groups showed significantly lower body weight gain and calorie efficiency ratio than m groups and H/E group was the lowest among all groups. Perirenal fat pad weights of E groups were lower than m groups, and epididymal fat pad weights of H and H + C groups were lower than CO and C groups among m groups regardless of exercise treatment. Brown adipose tissue weights of I groups were higher than fn groups and highest in H/E group among all groups. Liver citrate lyase activities of H and C groups were lower than CO and H + C groups regardless of exercise treatment. The differences between NE groups and E groups were not found to be significantly. Liver carnitine acyltranferase activity was not significantly different among all groups. Fecal total lipid, triglyceride and total cholesterol excretions were found to be higher in H and H + C uoups compared to CO and C groups both in NE and E animals, those of lipid excretions of NE groups were higher than E groups. In conclusion, HCA was more effective in reduction of body weight and body fat than L-carnitine or HCA + L-carnitine. HCA ingestion with exercise was far more effective than without exercise treatment in reducing body weight and body fat.

  • This study was peformed to investigate effect of feeding experimental mixture containing chitosan, hibiscus extract and L-carnitine on body weight and lipid metabolism in rats. Forty-eight male rats(Charles River CD) of eight weeks old and weighing $336.5{\pm}2.3g$ were raised for five weeks with high fat diet(40% fat as calorie) to induce obesity. After induction of obesity, rats weighing $560.4{\pm}5.6g$ were blocked into four groups according to body weight and raised for eight weeks with diet containing either 0.09%(+1D group), 0.9%(+10D group) or 4.5%(+50D group) of experimental mixture. Aspartate aminotransferase(AST) and alanine aminotransferase(ALT), total protein and albumin were normal levels in plasma. Body weight gain and epididymal fat pad weight were lower in experimental mixture groups than control group However, weights of perirenal fat pad and brown adipose tissue were not significantly different among all groups. There was no significant difference in plasma and hepatic lipid levels among all groups. Liver citrate lyase and camitine acyltransferase activities were not significantly different among all groups, however, citrate lyase activity was tended to be decreased with increasing experimental mixture level in diet. Fecal total lipid and total cholesterol excretions were highest in +50D group, and triglyceride excretion was highest in +1D group. in conclusion, intake of experimental mixture containing chitosan, hibiscus extract and L-camitine was effective in reducing body weight and body fat, and its inhibitory effects might lead to obesity improvement.

The antioxidant roles of L-carnitine and N-acetyl cysteine against oxidative stress on human sperm functional parameters during vitrification

  • Objective: Amino acids can protect sperm structure in cryopreservation due to their antioxidant properties. Therefore, the present study aimed to investigate the protective effect of L-carnitine (LC) and N-acetyl cysteine (NAC) on motility parameters, plasma membrane integrity (PMI), mitochondrial membrane potential (MMP), DNA damage, and human sperm intracellular reactive oxygen species (ROS) during vitrification. Methods: Twenty normal human sperm samples were examined. Each sample was divided into six equal groups: LC (1 and 10 mM), NAC (5 and 10 mM), and cryopreserved and fresh control groups. Results: The groups treated with LC and NAC showed favorable findings in terms of motility parameters, DNA damage, and MMP. Significantly higher levels of intracellular ROS were observed in all cryopreserved groups than in the fresh group (p≤0.05). The presence of LC and NAC at both concentrations caused an increase in PMI, MMP, and progressive motility parameters, as well as a significant reduction in intracellular ROS compared to the control group (p≤0.05). The concentrations of the amino acids did not show any significant effect. Conclusion: LC and NAC are promising as potential additives in sperm cryopreservation.

장쇄 지방산 산화 장애와 치료적 접근법 (Long-chain Fatty Acid Oxidation Disorders and Therapeutic Approach)

  • Long-chain fatty acid oxidation disorders (LC-FAOD) are an autosomal recessive inherited rare disease group that result in an acute metabolic crisis and chronic energy deficiency owing to the deficiency in an enzyme that converts long-chain fatty acids into energy. LC-FAOD includes carnitine palmitoyltransferase type 1 (CPT1), carnitine-acylcarnitine translocase (CACT), carnitine palmitoyltransferase type 2 (CPT2), very long-chain acyl-CoA dehydrogenase (VLCAD), long-chain 3-hydroxyacyl-CoA dehydrogenase (LCHAD), and trifunctional protein (TFP) deficiencies. Common symptoms of LC-FAOD are hypoketotic hypoglycemia, cardiomyopathy, and myopathy. Depending on symptom onset, the disease can be divided as neonatal period, late infancy and early childhood, adolescence, or adult onset, but symptoms can appear at any time. The neonatal screening test (NBS) can be used to identify the characteristic plasma acylcarnitine profiles for each disease and confirmed by deficient enzyme analysis or molecular testing. Before introduction of NBS, the mortality rate of LC-FAOD was very high. With NBS implementation as routine neonatal care, the mortality rate was dramatically decreased, but severe symptoms such as rhabdomyolysis recur frequently and affect the quality of life. Triheptanoin (Dojolvi®), the first drug for pediatric and adult patients with molecularly confirmed LC-FAOD, has recently been approved by the US Food and Drug Administration in 2020. In this review, the diagnosis of LC-FAOD and treatment including triheptanoin are summarized.

Dietary Docosahexaenoic Acid Decreases Plasma Triglycerides with Mixed Effects on the Indices of $\beta$-oxidation

  • One known effect of long chain n-3 polyunsaturated fatty acids is their ability to decrease plasma triglycerides. However, identification of the specific n-3 fatty acids and the underlying mechanisms responsible for this change remains uncertain. This present study was designed to evaluate the effects of moderate levels of dietary docosahexaenoic acid (22 :6(n-3)) on modulating plasma triglyderides. Male CD-1 mice were maintained for 15 days on identical diets containing either docosahexahexaenoic acid ethyl ester(1.5%, w/w) or linoleic acid(18 : 2(n-6)) ethyl ester (1.5%, w/w) . Plasma triglycerides were 40% lower in the docosahexaenoic acid group than in the linoleic acid group. Hepatic carnitine palmitoyltransferase activity (a key regulatory enzyme for mitocondria $\beta$-oxidation) was not significantly different between the dietary groups. However, plasma acid soluble acylcarnitine levels (which increase with increasing $\beta$-oxidation )were significantly higher in the decosahexaenoic acid group. This data suggests that plasma triglyceride levels are lower in mice fed diets containing moderate levels of docosahexaenoic acid compared to linoleic acid, but this effect on plasma triglycerides is not modulated through an augmentation of mitochondrial $\beta$-oxidation.

What is the Key Step in Muscle Fatty Acid Oxidation after Change of Plasma Free Fatty Acids Level in Rats?

  • The purpose of this study was to discern the critical point in skeletal muscle fatty acid oxidation by changing plasma free fatty acids (FFA) level in rat. In the study, 3 key steps in lipid oxidation were examined after changing plasma FFA level by acipimox. The rates of both palmitate and palmitoylcarnitine oxidation were decreased by decrease of plasma FFA level, however, carnitine palmitoyl transferase (CPT) 1 activity was not changed, suggesting CPT1 activity may not be involved in the fatty acid oxidation at the early phase of plasma FFA change. In the fasted rats, ${\beta}-hydroxy$ acyl-CoA dehydrogenase (${\beta}$-HAD) activity was depressed to a similar extent as palmitate oxidation by a decrease of plasma FFA level. This suggested that ${\beta}-oxidation$ might be an important process to regulate fatty acid oxidation at the early period of plasma FFA change. Citrate synthase activity was not altered by the change of plasma FFA level. In conclusion, the critical step in fatty acids oxidation of skeletal muscles by the change of plasma FFA level by acipimox in fasting rats might be the ${\beta}-oxidation$ step rather than CPT1 and TCA cycle pathways.

L-카르니틴 섭취와 복합운동이 비만여고생의 신체조성, 혈중지질 및 아디포넥틴에 미치는 영향 (Combined Effects of L-Carnitine Supplementation and Exercise on the Body Composition, Serum Lipids and Adiponectin in the High School Obese Female Students)

  • 본 연구는 10주간 주 3회, 1회 운동시간 50분을 한 결과, 비만여고생을 대상으로 L-카르니틴 섭취와 복합운동이 신체조성, 혈중지질 및 아디포넥틴에 미치는 영향을 구명하기 위하여 체지방률 35% 이상 비만여고생을 대상으로 실험하였다. 신체조성에서는 운동과 카르니틴 섭취를 병행한 군에서 체중, 체지방량 및 체지방률이 유의하게 감소하였고, 제지방량이 유의하게 증가하였다. 혈중지질변화에서는 T-C는 대조군에서 집단 내 변화를 보였으며, TG는 복합운동과 카르니틴섭취를 병행한 군에서 유의하게 감소하였다. 아디포넥틴의 변화에서는 복합운동과 카르니틴섭취를 병행한 군이 유의하게 증가하였다. 이러한 연구결과를 토대로 보다 효과적인 운동프로그램과 적절한 식이요법으로 비만을 개선하는데 효과가 있을 것으로 사료된다.

운동(수영)여부에 따른 중년여성의 체지방 대사비교 (Commarison of Body Fat Metabolism in Middle-aged Women Depending Upon Swimming Practice)

  • The present study examined the effects of swimming on some plasma parameters of lipid metabolism in the middle-aged women. The control group(C) was the women who lived without any intentional physical exercise, and the first experimental group(S I) and the second experimental group(S II) had gone swimming everyday at least one hour for 3-6 months, and more than one year, respectively. The results obtained were summarized as follows: 1) Energy and nutrient intakes of the three groups were not different, but the body fat(%) of two experimental groups(S I, S II) was significantly lower than that of control. 2) Total cholesterol and HDL-cholesterol in plasma were not significantly changed by swimming and swimming period. However, total lipids and LDL-cholesterol were significantly lower in the two experimental groups than in control. 3) The level of plasma triglycerides of SI was not different from the control, but SII showed significantly lower triglycerides. 4) Acid-soluble acylcarnitine and $\beta$-hydroxybutyrate concentration in plasma were significantly greater in SII than in control. There results suggest that regular swimming in a longer period might help to protect the risk of obesity and cardiovascular disease by modulating lipid metabolism.

Dietary Niacin Supplementation Suppressed Hepatic Lipid Accumulation in Rabbits

  • An experiment was conducted to investigate the effect of niacin supplementation on hepatic lipid metabolism in rabbits. Rex Rabbits (90 d, n = 32) were allocated to two equal treatment groups: Fed basal diet (control) or fed basal diet with additional 200 mg/kg niacin supplementation (niacin). The results show that niacin significantly increased the levels of plasma adiponectin, hepatic apoprotein B and hepatic leptin receptors mRNA (p<0.05), but significantly decreased the hepatic fatty acid synthase activity and adiponectin receptor 2, insulin receptor and acetyl-CoA carboxylase mRNA levels (p<0.05). Plasma insulin had a decreasing tendency in the niacin treatment group compared with control (p = 0.067). Plasma very low density lipoproteins, leptin levels and the hepatic adiponectin receptor 1 and carnitine palmitoyl transferase 1 genes expression were not significantly altered with niacin addition to the diet (p>0.05). However, niacin treatment significantly inhibited the hepatocytes lipid accumulation compared with the control group (p<0.05). In conclusion, niacin treatment can decrease hepatic fatty acids synthesis, but does not alter fatty acids oxidation and triacylglycerol export. And this whole process attenuates lipid accumulation in liver. Besides, the hormones of insulin, leptin and adiponectin are associated with the regulation of niacin in hepatic lipid metabolism in rabbits.

Fenofibrate reduces adiposity in pregnant and virgin rats but through different mechanisms

  • Fenofibrate has been proven to reduce adiposity. Since gestation produces an increase in white adipose tissue (WAT) mass, we comparatively studied this drug-effect in virgin and pregnant rats. Fenofibrate reduced lumbar WAT weight in both pregnant and virgin rats. Fenofibrate treatment did not modify plasma free fatty acid (FFA) concentration in virgin rats, it greatly increased it in pregnant animals. Remarkable differences between the two groups were obtained for two proteins related to fatty acid oxidation and esterification and storing. Respectively, the mRNA levels of carnitine palmitoyltransferase I (CPT-I) were increased by the fenofibrate only in the virgin rats and a similar finding was observed for the expression of phosphoenolpyruvate carboxykinase (PEPCK). These findings indicate that fenofibrate reduces adiposity in pregnant and virgin rats through different mechanisms: a) in virgin rats, by promoting fatty acid oxidation; and b) in pregnant rats, by enhancing fatty acid output.
