The response of plants growing in a landfill in the Philippines towards cadmium and chromium and its implications for future remediation of metal-contaminated soils

  • During several visits to the Cebu City landfill in the Philippines, plants were observed growing within the area, including on top of the garbage piles. Studying the response of these plants is important in assessing which can be used in remediating metal contaminated soils. This study aimed to determine whether the plants in the Cebu City landfill excluded or accumulated cadmium (Cd) and chromium (Cr) in the plant tissues. The floristic composition of the landfill was analyzed prior to the sample collection. The samples were acid-digested before the desired elements were measured using atomic absorption spectrophotometry (AAS). The Cd and Cr concentrations in the plant root-zone soil were also measured using AAS. The results indicated that the landfill substrate was generally acidic based on the results of the pH measurement. Of the 32 plant species sampled, Cyperus odoratus showed potential for Cd uptake and internal transfer; Cenchrus echinatus, Vernonia cinerea and Terminalia catappa for Cr uptake, and Cynodon dactylon for Cr internal transfer. The plants in the landfill differed in their response towards the heavy metals. To confirm the behavior of C. odoratus towards Cd, and C. echinatus, C. dactylon, V. cinerea, and T. catappa towards Cr, controlled experiments are recommended, as the plant samples analyzed were collected from the field.

  • Sugar uptake is accompanied with H+-substrate-symport generally. Both H+/sugar-and H+/K+ stoichiometries during the sugar-uptake have been reported to be exactly 1 : 1. This paper reports that the stoichiometries were enhanced dramatically by the addition of CaCl2 into the medium and by the high cell density of 200 $\mu$L pc/mL. The concentration of free Ca2+ ions in the cells increased significantly with cell density. It is suggested that the free Ca2+ ions are responsible for the change of stoichiometry of sugar transport system by regulation of H+ ion level of biomembrane.

Rice Iron Metabolism: from Source to Solution

  • Iron is an important micronutrient for plants. Iron metabolism is a complex mechanism under a delicate balance. Iron metabolism represents two major problems for plants: deficiency as a consequence of solubility problems and toxicity due to excess solubility in anaerobic conditions. In the last few years, new genes have been discovered that influence iron uptake, transport and storage. Irrigated rice is exposed to high levels of $Fe^{II}$, normally rare in aerobic soil conditions. The implications of altering iron uptake rates and the effects of newly discovered genes are discussed.

Isolation and characterization of three maize aquaporin genes, ZmNIP2;1, ZmNIP2;4 and ZmTIP4;4 involved in urea transport

  • Urea-based nitrogen fertilizer was widely utilized in maize production, but transporters involved in urea uptake, translocation and cellular homeostasis have not been identified. Here, we isolated three maize aquapoin genes, ZmNIP2;1, ZmNIP2;4 and ZmTIP4;4, from a cDNA library by heterogous complementation of a urea uptake-defective yeast. ZmNIP2;1 and ZmNIP2;4 belonged to the nodulin 26-like intrinsic proteins (NIPs) localized at plasma membrane, and ZmTIP4;4 belonged to the tonoplast intrinsic protein (TIPs) at vacuolar membrane. Quantitative RT-PCR revealed that ZmNIP2;1 was expressed constitutively in various organs while ZmNIP2;4 and ZmTIP4;4 transcripts were abundant in reproductive organs and roots. Expression of ZmTIP4;4 was significantly increased in roots and expanded leaves under nitrogen starvation, while those of ZmNIP2;1 and ZmNIP2;4 remained unaffected. Functions of maize aquapoin genes in urea transport together with their distinct expression manners suggested that they might play diverse roles on urea uptake and translocation, or equilibrating urea concentration across tonoplast.

Fenobucarb와 Imidacloprid의 벼 뿌리 침지시간에 따른 벼멸구의 살충활성과 섭식행동비교 (Comparison of Insecticidal Activity and Feeding Behavior of Nilaparvata lugens by Root Uptake Times against Fenobucarb and Imidacloprid)

  • 벼멸구에 대한 Fenoburcarb(36, 12 ppm)와 Imidacloprid(0.7, 0.4 ppm)의 벼유묘 뿌리 침지 시간(1, 4, 8, 12, 16, 20, 24시간)에 따른 살충활성과 섭식행동의 상관관계를 조사하였다. EPG (electrical penetration graph)의 파형은 신호개시 후 4시간 동안 기록하여 분석하였다. 침투이행성과 살충력과의 상관관계는, 두 약제 모두 농도가 높을수록 높은 보정살충율을 보였고, 침지시간이 길수록 살충활성이 높았다. 파형에 있어서는 두 약제 모두 농도가 높아질수록 상대적으로 non-probe time은 길어졌고 체관을 찌르는 시간은 짧아졌다. 침지시간이 길어질때에도 같은 경향을 보였다. 따라서 침지시간에 따른 살충활성과 두 파형간에 상관관계가 있는 것으로 나타났다.

Comparison of Soil Testing Methods for Plant Available Phosphate

  • Most test methods for plant available soil phosphate are based on the extraction with a chemical solution. The objective of this study is to evaluate available phosphate of various tests at different soil phosphate levels. Two experiments were conducted as follows: i) Extracting capacities of soil phosphate tests - Mehlich III, Mehlich II, Bray I, Olsen, Kelowna, and Modified Lancaster(Mod. Lancaster) - were compared with that of Lancaster test for the soils collected from 32 paddy and 27 upland fields with various soil chemical properties. ii) Field trials on comparing to phosphate uptake by plant were accomplished by cultivating rice and corn plants in the pots filled with the soils. Available phosphate of Lancaster test was significantly correlated with those of Mehlich III, Mehlich II, Bray I, Olsen, Kelowna, and Mod. Lancaster. In upland soils, available phosphates of all the tests were curvilinearly regressed with phosphate uptake by corn. The determination coefficients ($R^2$) of the regression equation between available phosphate in soils and phosphate uptake by plants were ranged from 0.861 (Mehlich III) to 0.741 (Olsen). In paddy soils, the available phosphate measured by Mehlich III and Lancaster was significantly correlated with phosphate uptake by rice. In conclusion, Lancaster and Mehlich III tests could be used for predicting available phosphate in upland and paddy soils.

단기간 근귄 저온처리가 수경재배 상추의 질산태 질소 함량 미치는 영향 (Effect of Short Term Cold Treatment to Rhizosphere on Nitrate Concentration in Lettuce Plant under Hydroponic Culture System)

  • 질산태 질소의 일중 농도 변화와 단기간의 저온 처리가 질산태 질소의 흡수 및 농도에 미치는 영향을 살펴보고자 상추 (Lactuca sativa L.)를 공시 작물로 하여 온실에서 수경 재배하였다. 질산태 질소의 농도는 엽신에 비하여 주맥에서 일중 지속적으로 2배 이상 높았으며, 질산태 질소의 일중 변화는 14:00까지 지속적으로 감소하여 최저치 (8.7 mg-N/GDW)를 나타낸 후 다시 증가하였다. 질산태 질소의 일중 흡수량은 11:00$\sim$17:00에 평균 4.8 mg-N/GDW-Root/hr로 최대치를 나타내었다. 단기간의 저온 처리는 엽의 질산태 질소의 농도를 14$\sim$18%, 질산태 질소의 흡수량을 50$\sim$55%를 감소시켰다. 이러한 결과는 수확 전 단기간의 저온처리가 상추 잎의 질산태 질소함량을 낮출 수 있는 방법으로 응용될 수 있음을 보여준다.

Zn Acquisition by Extraradical Hyphae in Arbuscular Mycorrhizal Plant Depending on Zn Nutritional Status of Cucumber(Cucumis sativus cv. Baekdadagi)

  • The contribution of plant nutrition status in arbuscular mycorrhizal (AM) plant to the nutrient acquisition by extraradical hyphae of AM fungi was investigated using cucumber colonized with Glomus intraradicies (BEG 110) focusing on the Zn. Compartmentalized pots with separated Bones for hyphal growth were used to determine the contribution of extraradical AM hyphae to Zn uptake from hyphal zones. $0.5\;{\mu}M$ Zn was supplied into the hyphal zones as nutrient solution (10 mL/day) with a form of $ZnSO_4$. Zn foliar application was made two times for one week before harvest (8 mL/plant). The colonization rate by AM were high in all of Zn treatments. The dry weight of cucumber increased by AM colonization compared to those of non-mycorrhizal counterpart. However: Zn foliar application resulted in no significant difference in dry weight between mycorrhizal- and non-mycorrhizal plant. In addition, the enhancement of Zn content in cucumber shoot by AM colonization were also reduced by Zn foliar application. These results indicate that the interaction between host plant and AM fungus for nutrient uptake might be related to plant nutritional status and nutrient contents. In consequence, higher Zn contents in host plant by foliar application of Zn could restrict the role of extraradical hyphae of AM fungus on the Zn acquisition and transfer from fungus to host plant.

하수종말처리장 방류수와 비점오염원 처리를 위한 주암호 인공습지 2년 운영 사례 (Case Study: Operation of the Juam Constructed Wetland for Effluent from a Sewage Treatment Plant and Diffuse Pollution for Two Years)

  • In order to improve water quality of the Juam Lake, a constructed wetland was implemented and operated for 2 years with the effluent of sewage treatment plant and diffuse pollutant discharged from agricultural area. During the summer season, average removal efficiencies for BOD and SS were 15.8% and 39.4%, respectively. Due to the mixed effect of vegetation, soil microbes and sediments, the higher nutrient removal efficiencies were obtained: average T-N and T-P removal efficiencies were 64.2% and 71.7%, respectively. The concentration of sediment was increased initially, and maintained constant throughout monitoring period. The highest nitrogen and phosphorus uptake were observed in Phragmites japonica. The nitrogen uptake was estimated as 0.235 DW mg/g while phosphorus uptake was estimated as 2.059 DW mg/g.

Overexpression of Gene Encoding Tonoplast Intrinsic Aquaporin Promotes Urea Transport in Arabidopsis

  • Complementation assay of the urea uptake-defective yeast mutants led to the identification of the Arabidopsis AtTIP4;1 gene encoding the aquaporin. However, its physiological functions still remain elusive. In the present study, histochemical and genetic analyses were performed to understand the physiological roles of AtTIP4;1 in urea uptake. The AtTIP4;1 product was detectible in the roots, but not in the leaves, the stem, and the flower. Its promoter allowed the expression of the $\beta$-glucuronidase reporter gene in the roots and the apical meristem in Arabidopsis. The AtTIP4;1 products were induced under nitrogen-deficient conditions. To investigate the role of the tonoplast intrinsic protein in urea transport and developments, Arabidopsis with the loss- and the gain-of-function mutations by T-DNA insertion in AtTIP4;1 and 35S promoter-mediated overexpression of AtTIP4;1 were identified, respectively. The transfer DNA insertion and the AtTIP4;1-overexpressed plants showed normal growth and development under normal or abiotic stress growth conditions. The urea-uptake studies using $^{14}C$-labeled urea revealed higher accumulation of urea in the AtTIP4;1-overexpressed plants. These results provide evidence that overexpression of AtTIP4;1 leads to the increase in the urea-uptake rate in plants without detectable defects to the growth and development.
