Chemical Contamination and Toxicity of Sediments from the Gunsan Coast, Korea

  • Polycyclic aromatic hydrocarbons (PAHs), butyltins (BTs), nonylphenol (NP), and fecal sterols concentrations in sediments were investigated from Gunsan coast of Korea to evaluate organic pollution from anthropogenic activities. Sediment toxicity was also examined by bacterial bioluminescence toxicity test (Vibrio fischeri). The concentrations of 16 PAHs in sediments ranged from 67.9 to 425 ng/g dry wt; BTs ranged from 2.79 to 14.1 ng Sn/g dry wt; NP ranged from 20.7 to 2171 ng/g dry wt; and coprostanol, a fecal sterol, ranged from 7.60 to 245 ng/g dry wt. Effective concentration 50% ($EC_{50}$) of sediments ranged from 0.38 to 23.8 mg/mL. Most of the chemicals were present at levels lower than or comparable to the previously reported values from Korea. However, NP levels in the present study were in the high range of levels reported from the Korean coast, and 40% of the measured samples exceeded screening and ecotoxicological values of NP suggested by the Netherlands and Canada. This suggests that an ongoing source of NP is a serious concern in the Gunsan coast. High levels of contaminants were found in the proximity of potential sources, such as the outfall of a wastewater treatment plant for NP, an anthracite-fired power plant for PAHs, and ports for BTs, fecal sterols, and sediment toxicity. This indicates that Gunsan coast has various potential sources of marine sediment contaminants.

A Single Oral Dose Toxicity Study of Plant Sterol Ester in Sprague-Dawley Rats

  • The present study was conducted to investigate the acute toxicity of plant sterol ester by a single oral dose in Sprague-Dawley rats. Ten males and 10 females aged 5 weeks were randomly assigned to two groups of 5 rats each and were administered by gavage at dose level of 0 or 20 ml/kg body weight. Parameters measured during the 14-day observation period were mortality, clinical signs, body weight changes, and gross findings. No mortality was observed in the present study. Treatment-related clinical signs, such as pasty stool and diarrhea, were observed on the day of treatment and these signs resulted in soiled fur on day 1 after the treatment. However, no clinical signs were observed on days 2-14 after the treatment. There was no significant difference in body weight changes between the control and treatment groups. At necropsy on day 14 after the treatment, no treatment-related gross findings were observed in the treatment group. Based on these results, it was concluded that a single oral dose of plant sterol ester induced pasty stool and diarrhea in Sprague-Dawley rats at dose level of 20 ml/kg and that the lethal doses were considered to be over 20 ml/kg for both sexes.

  • Angelica flaccida Kom. is a herbal plant growing wild in the marsh of valley in Korea. It has been used for sedative and analgesic as a folk medicine, but its constituents have not been clarified yet. Five known coumarins such as isoimperatorin, decursidin, (-) anomalin, decursin, decursinol and two sterols such as ${\beta}-sitosterol$ and stigmasterol were isolated from the root of this plant, Their structures were identified by the UV, IR, NMR, mass data and the physico-chemical properties.

Studies on the Components of Evodia daniellii $H_{EMSLEY}$ in Korea (한국산 오수유성분에 관한 연구)

  • Evodia daniellii $H_{EMSLEY}$, a plant belonging to Rutaceae, is cultivated in Korea and used as a folkmedicine for gastric inflammation, extermination of noxious insects, and headache. The seed oil of this plant also has been used in various diseases, for example, dermatitis, scabies and so forth. From the barks, fruit peels, and seed oil of Evodia daniellii $H_{EMSLEY}$, four crystalline compounds were isolated. Three compounds except one were characterized as methyl sinapate, bergapten, and evodiamine by m.p. determination, elemental analysis, UV, IR, NMR spectra and mass analysis. Even though methyl sinapate was a known synthetic compound, it was not yet reported as a natural product. From the seed oil, unsaponifiable fraction was separated and was analyzed. It was considered to be consisted with sterols, hydrocarbons and tocopherols.

  • Korean cabbage, a member of the Brassicaceae family which also includes cauliflower, mustard, radish, and turnip plants, is a crucial leafy vegetable crop. Korean cabbage is harvested after completion of the leaf heading process and is often prepared for use in "baechu kimchi", a traditional Korean food. Many of the components in Korean cabbage are essential for proper human nutrition; these components can be divided into two groups: primary metabolites, which include carbohydrates, amino acids, fatty acids, and organic acids, and secondary metabolites such as flavonoids, carotenoids, sterols, phenolic acids, alkaloids, and glucosinolates (GSLs). Using gas chromatography-mass spectrometry, this study examined the variety of volatile compounds (including isothiocyanates) contained in Korean cabbage and its seed, which resulted in the identification of 16 and 12 volatile compounds, respectively. The primary volatile compound found in the cabbage was ethyl linoleolate (~23%), while 4,5-epithiovaleronitrile (~46%) was the primary volatile component in the seed.

The Metabolism of (2-$^{14}C$) Mevalonic Acid on Photoperiodic Induction in Grafted Solanum Andigena

  • The metabolism of sterol precurosor in leaves of Salanum andigena grafted between photoinduced and noninduced plant was investigated with the use of (2-$^{14}$ C) mevalonic acid. By the technique of the preparative gas-liquid chromatography, radioactive compounds of squalene, 4,4’-dimethylsterols and 4-demethylsterol were isolated and determined quantitatively. When labeled mevalonic acid n as applied to leaves radioactivity was extensively incorporated into non-saponifiable materials of lipid fraction and aqueous fraction (ethanol-water fraction). Radioactivity of 14C derived from (2-$^{14}$ C) mevalonic acid was transmissible from photoinduced plant to non-induced plant across the graft union, as tuberization hormone was, and incorporated into the sterols of the non-induced plant. Inhibitors of sterol biosynthesis, SK & F 7997 and nicotinic acid, are effective suppressors of tuber growing, if applied to leaves during photoinduction period. The experimental results suggest that certain substance containing isoprene unit, or sterol-like compound may participate in tuber growing.

Effect of Sterols on Phytophthora infestans and Oospore Production on detached Potato Plants (감자 역병균에 대한 스테롤류의 영향 및 감자절편에서의 난포자 형성)

  • The effects of media, cholesterol, $\beta$-sitosterol and lecithin on the growth and oospore production of the isolates KM10, U6, CDB6, MHB6, JD1 (A$^2$type) of Phytophthora infestans isolated in Korea and F8l7, DNC303 (A$^1$type), IB908, DN107 (A$^2$type) obtained from Japan were investigated. Mycelium of P. infestans grew better on V-8 juice agar and rye meal agar than on the other media. Oospores were produced most abundantly on V-8 juice agar. Mycelium extended more 16.6, 8.3, and 5.2% on V-8 juice agar supplemented with 5 $\mu\textrm{g}$/$m\ell$ of cholesterol, $\beta$-sitosterol and lecithin, respectively, and oospores are produced 76.0, 58.0, and 34.6 % on V-8juice agar supplemented with 5 $\mu\textrm{g}$/$m\ell$ of cholesterol, $\beta$-sitosterol and lecithin, respectively. Oospores more produced on detached potato plant disks when $A^1$ and $A^2$ type exist simultaneously which indicating that variation of population can occur in the field, but the rate of oospore formation and the number of oospores produced was low and small quantity.

  • Three stigmastane-type sterols and one ergostane-type sterol were isolated from the ethyl acetate soluble fraction of the aerial parts of Artemisia princeps Pampanini (Sajuarissuk). From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as $stigmasta-5,22-dien-3,{\beta}-ol (stigmasterol, 1),stigmast-5-en-3{\beta}-ol({\beta}-sitosterol,2), 5{\beta},8{\beta}-epidioxy-5{\beta},8{\beta}-ergosta-6,22-dien-3{\beta}-ol(ergosterol peroxide, 3),\;and\;{\beta}-sitosterol\;3-O-{\beta}D-glucopyranoside(daucosterol,4)$.

  • From the whole plants of Oldenlandia diffusa ursolic acid together with sterols, ${\beta}-sitosterol$ and its $3-0-{\beta}-D-glucoside$ were isolated and characterized mainly by means of spectroscopic methods. Based on the present findings, it may be considered that this plant may contribute to be a rich source for ursolic acid.

Ethnobotany, Phytochemistry, Pharmacology of the Korean Campanulaceae: A Comprehensive Review (한국산 초롱꽃과(Campanulaceae)의 민속식물, 화학성분, 약리작용에 대한 종합적 고찰)

  • The present study was carried out to identify traditional konwledges on Korean Campanulaceae plants and conduct a comprehensive review of them through analyzing phytochemistry and pharmacology of Korean Campanulaceae plants. According to the literature study, the ethnobotanical plants of Korean Campanulacae consisted of a total 18 taxa. Of them, 12 taxa including Platycodon grandiflorus, Adenophora triphylla var. japonica, Codonopsis lanceolata and others have been used as ethnomedicinal plants. These plants have been used for the treatment of 49 diseases such as cold, asthma and postnatal care. Phytochemical studies have identified the constituents present from Korean Campanulaceae (Adenophora, Codonopsis, Platycodon, Campanula and Asyneuma). A wide range of chemical compounds comprised 109 triterpenes, 8 sterols, 4 polyacetylenes, 21 alkaloids, 14 flavonoids, 14 phenolic acids, 11 phenolic glycosides, 8 phenylpropanoids and 22 other compounds. Pharmacological studies of these compounds have demonstrated immuno-stimulating, anti-inflammatory, anti-asthmatic, apophlegmatic and anti-allergic effects. They have also shown antioxidant, estrogenic, anti-diabetic, hepatoprotective, neuroprotective, antinociception and anti-tumor activities, as well as anti-obesity and cardiovascular effects. In light of traditional knowledge and phytochemical and pharmacological studies summarized, uses of Korean Campanulaceae based on traditional knowledge (for the treatment diseases and conditions of respiratory, pregnancy, childbirth, puerperium, genitourinary, circulatory, musculoskeletal and other systems) have been supported by phytochemical and pharmacological studies.
