This study was conducted to determine the optimum conditions of inducing callus, proliferating callus, forming somatic embryos, and regenerating plantlets via somatic embryogenesis, for the purpose of producing artificial seeds and substantially developing plant factory technologies that can be employed to all seasons production of Bupleurum plants. Callus was efficiently induced from leaf tissues at three leaf stage in the MS medium supplemented with 2, 4-D 2mg /1 and thidiazuron(TDZ) 0.lmg /1. Callus induction from leaf tissues at maturity was mostly effective in the mixture of 2,4- D 2mg /1 and TDZ 1.0mg /1 while that from flower bud tissues was fairly good in the MS medium containing 2,4-D 1 or 2mg /1.Callus was formed in 15 to 20 days after culture initiation in the MS media supplemented with 2, 4- D 1-2mg /1 and TDZ 0.l-1.0mg /1. Such hormones as kinetin 3mg /1, GA 1mg /1, and the mixture of GA 1mg /1 and TDZ 1mg /1 effected markedly to proliferate the callus cells.The optimum temperature and light intensity for callus culture were found to be $25^{\circ}C$ and 3000 Lux, respectively. Direct plant regeneration from cultured callus was fairly made on hormone-free MS or half-strength MS medium. Somatic embryogenesis was most frequently observed in hormone-free media:60 somatic embryos per 20ml in MS medium and 28 somatic embryos per 20ml in half -strength MS medium. There were three stages-globular, heart, and torpedo-in development of somatic embryos, among which globular stage was more frequently observed in MS medium rather than in half-strength MS medium. Somatic embryos induced from suspension culture fairly differentiated a number of shoots and roots on hormone-free and half-strength MS solid medium.
Differing in water conditions, the dry matter weight per plant was highest at 0 cm flooding depth, and was decreased at above 2 cm flooding depths. The shoot and spikes per pot developed best at flooding depths of 0 and 2 cm, but worst at -5cm or above 4cm flooding depths. The dry matter weight of shoots was linearly increased, but the weight of roots was sharply decreased according to high temperature after flower initation. The shoots and spikes per pot developed more effectively at 25$^{\circ}C$ than at 35 or 15$^{\circ}C$. The number of shoot and spike per pot were decreased according to higher shading. The effects of shading of 25-45% were not significantly small. The natural white and yellow spectra were the most effective to increase dry matter weight, shoots and spikes per pot. The dry matter weights of shoot and root per plant were not significantly different among at: 50 and 75% clay. The number of shoots were best and continuously increased at 75% clay, and the increments at 0 and 100% clays showed the lag period at early stage. The pattern of spikes was similar to shoots with less difference in various clay composition. The dry matter weight per plant in paddy field and upland field was basically similar. The numbers of shoot and spike were increased in response to increased fertilizer levels up to 20Kg per 10a of each component.
The rice variety, Kwanok, was reared in the water, land and salty beds and transplanted to the reclaimed soil area having an average of 0.48% salt concentration 0.67% at the end of April Two levels of $NH_3$-N and urea-N with 6 treatments were used. The effect of each treatment was observed. The plant height of the land bed seedlings at transplanting stage was short but the dry-weight/plant-height ratio was large, and the rooting ability vigorous remarkably after transplantation in the salty area. The total carbohydrate content of the stem part was markedly larger in the land bed seedlings than the others and the C/N ratio was accordingly greater. This tendency was observed through the last rooting stage. In the salty nutrient solution, the roots of salty bed seedlings showed high respiratory activity. The activity of the land bed seedlings did not decreased in the hypertonic solution as much as the water bed seedlings. There was no difference in the effect of fertilization on the rough rice yield between ammonium sulphate and urea. The cultural practices with the land and salty bed seedlings increased the rough rice yields by 33% and 22% respectively, compared with the yields of the water bed Seedlings. The number of panicles, panicle weight and the number of grains per panicle were much greater from the rice plant grown by the land bed seedlings than from the other bed seedlings.
This study was conducted to obtain the basic information to increase the practical use of soil survey data through the subdividing of valley shapes with soil sequences due to different parent rocks, and to study the relationship between the valley shapes and parent rock. The various rocks such as sedimentary(gray shale and sand stone) and igneous rocks(granite, granite gneiss and andesite porphyry) which are the major parent rocks in Yeongnam area were investigated. The characteristics of valleys formed and the kinds of soils derived from different rocks were analysed by using aerial photographs and topographical maps scaled 1:5,000. The rill density in igneous rock area was as high as 40. But the rill bifurcation ratio of first order stream was higher in the sedimentary than the igneous rocks except granite area. The mean slope of valleys in igneous areas was about 8%, which was higher than that of the sedimentary areas. The variability of valley width in the complexly metamorphosed rock, such as granite gneiss, and andesite porphyry, was greater than in sedimentary and in granite rocks. Based on the variability of valley widths and valley slopes, it was possible to classify the valleys into two types. The "Uterus-shaped valleys" had wide variability of valley width and were located in the areas of granite gneiss and andesite porphry rocks. while the "Roots-shaped valleys" had narrow variability of valley width and were located in the sedimentary areas. "Uterus-shaped valleys" were typified by having land forms of mountain foot slopes and alluvial fans, and the soil drainage sequences also had complexities. So that, we concluded that the variability of valley width and valley slopes was associated with kinds of parent rocks and metamorphism which influences soil sequence and characteristics.
To investigate the effect of inorganic ($GeO_2$) and organic (Ge-132) germanium treatment on Brasica juncea C. plant, growth characteristics and Ge contents were examined with various inorganic or organic germanium treatments (0, 5, 10, 25, 50, 75 and $100mg\;L^{-1}$), respectively. Brasica juncea C. growth did not much inhibited until Ge $10mg\;L^{-1}$ concentration under both Ge-132 and $GeO_2$ treatments as compared to control. On the other hand, at Ge concentration higher than $25mg\;L^{-1}$ concentration, Brasica juncea C. growth was inhibited under both Ge-132 and $GeO_2$ treatments. Under treatment of $GeO_2$, length of root and shoot slightly increased until $5mg\;L^{-1}$ concentration and dry weight slightly increased until $10mg\;L^{-1}$ concentration. Under treatment of Ge-132, length of root and shoot slightly increased until $10mg\;L^{-1}$ concentration and dry weight slightly increased until $25mg\;L^{-1}$ concentration. Total Ge contents in Brasica juncea C. early seedlings with $GeO_2$ treatment were a bit higher than those with Ge-132 treatment. Germanium was primarily accumulated in the roots (77%) with organic Ge (Ge-132) treatments, whereas Ge was primarily accumulated in the leaf (70%, respectively) with $GeO_2$ treatments. The Ge uptake rates in inorganic Ge treatments were slightly high than those in organic Ge treatments. Under inorganic Ge treatment with $2.5mg\;L^{-1}$, about 3% of Ge was accumulated into plant and distribution in leaf and root was 84.8% and 15.2%, respectively. Under organic Ge treatment with $2.5mg\;L^{-1}$, about 2.6% of Ge was accumulated into plant and distribution in leaf and root was 66.4% and 33.6%, respectively.
Angelica gigas Nakai (AGN) is a perennial plant belonging to the family Apiaceae. Its root has been utilized as a traditional medicine especially in Korea. This study was carried out to evaluate the potential use of extracts from AGN root parts as a cosmetic material. The dried AGN roots are divided into body (B), thick root (TkR), medium root(MR) and thin root (TnR) according to their diameter before cutting into medicine. B, TkR and MR of AGN are combined and used as medicinal herbs (MH). The extracts from AGN each root part (B, TkR, MR, TnR, MH) were used to test the effect on cell viability using MTS assay and to examine inhibitory effect on melanin accumulation in B16F10 melanoma cells. All extracts (50 - 200 ㎍/mL) from the each root part did not affect the cell viability. And inhibitory effect of all root extracts (200 ㎍/mL) on melanin accumulation was 12-19%. Especially, TnR showed similar inhibitory effect on melanin accumulation to MH. In addition, DPPH and ABTS free radical scavenging activity were higher in the TnR extract compared to MH. This study showed that the TnR extract exhibit high inhibitory effect on melanin accumulation and antioxidant activity compared to MH, suggesting that TnR extract has potential as a cosmetic ingredient.

This study was conducted to develop a mass production for a commercial use by cutting of 4 kinds of V. glabrifolia Kitag., V. pusanensis Y.N.Lee, V. glabrifolia Kitag. × V. Spicata 'Alba' and V. spicata 'Ulster Blue Dwarf' × V. longifolia. Veronica L. Effects of shade, media and concentrations of plant growth regulators on the rooting of Veronica L. were examined. The rooting rate of V. glabrifolia Kitag. was higher as a 60% than the V. pusanensis Y.N.Lee as a 20% in the control of commercial media. In shading treatment, the rooting rate was highest at 30% or 60% shading, and the 30% shading was the best in number of root and root length, but the 90% shading was lower than no shading treatment. For cutting media, the rock wool and 100% perlite increased the rooting rate by more than 10% compared to commercial media, and increased the number of roots by 2 or 3 times. However the Cocopeat of media was lower of rooting rate, root number, and root length compared to another treatment. In the plant growth regulator treatment, the rate of rooting increased in all treatment compared to control, and was highest at IBA 1,000mg·L-1 as a 82.2% and NAA 200 or 400mg·L-1 as a 82.2% or 84.4% respectively, in the V. glabrifolia Kitag. × V. Spicata 'Alba'. However, the root number and root length was decreased as the concentration of growth regulators increased.
This study was carried out to determine the effect of mixture of hymexazole and metalaxyl(Tachigarace) on the growth and reducing low temperature injury in rice seedlings. The amount of mixture of hymexazole and metalaxyl (Tachigarace) applied to reduce low temperature injury was 0, 9, 18 and 36mgr per pot (5${\times}$15${\times}$10cm) by soil incorperation befor sowing. Aging seedling was investigated on the growth responses by the application of 0.36gr mixture of hymexazole and metalaxyl (Tachigarace) per tray (30${\times}$60${\times}$3cm) after cutting of leaf at 35 day-old seedling. Rice seedlings treated with mixture of hymexazole and metalaxyl (Tachigarace) were better in the growth of top and roots, dry weight, erecting and rooting abilities than untreated seedlings. The effective amount of mixture of hymexazole and metalaxyl (Tachigarace) to increase erecting and rooting abilities was 18 mgr per pot. Under low temperature treatment, 12$^{\circ}C$, the leaf discoloration and dead leaf percent of rice seedlings were markedly decreased in the seedlings treated with mixture of hymexazole and metalaxyl (Tachigarace) as compared with untreated seedlings. The more amount of mixture of hymexazole and metalaxyl (Tachigarace) upto 36mgr per pot, the more effective on the low temperature injury in the seedling. The effect of mixture of hymexazole and metalaxyl (Tachigarace) to reduce low temperature damage in rice seedlings was mainly due to decreasing less physiological activities on the expanding leaf area and rooting, transpiration, stomatal diffusion and chlorophyll content of the leaves and oxygen consumption of the roots. Plant height, top dry weight and leaf area of aging seedlings were increased with mixture of hymexazole and metalaxyl (Tachigarace) treatment and leaf cutting as compared with untreated seedling and intact leaf, respectively. The ratio of missing hill of aging seedling was increased with increasing the period of growing seedling and was decreased due to leaf cutting, whereas the effect of mixture of hymexazole and metalaxyl (Tachigarace) application was not observed. Rooting ability of aging seedlings was decreased with lengthening of the period of raised seedling and was increased with mixture of hymexazole and metalaxyl (Tachigarace) treatment and leaf cutting.
In this study the essential oil and pharmacologically active components of different valerian species (Valeriana fauriei var. dasycarpa Hara and V. officinalis L. from Korea and V. wallichii DC from Nepal) were analyzed to search for native plant resources that can be used for medicinal herb and tobacco flavoring. The oil contents in dry roots of V. fauriei, V. officinalis and V. walhchii were 1. 71%, 0. 82% and 0. 83 %, respectively. The composition of the essential oil among different valerian species was compared. In V. fauriei 47 compounds were identified, while 21 and 25 compounds were identified in V. officinalis and V. wallichii. The major compounds in the oil of V. fauriei were bornyl acetate (33.8%) and camphene (10.8%), cedrol (4. 1 %), -pinene (3. 2%) and unidentified sesquiterpene alcohol (3. 0%). The major compounds were borneol (62. 5%) and ${\beta}-sesquiphelandrene$(6. 8%) and spathulenol (2. 1%) in V. officinalis, and borneol (74. 3%) and ${\alpha}-elemene$ (2.7%) and -sesquiphellandrene (2. 3%) in V. wallichii. Among the components known as major pharmacologically active compounds in European or Japanese valerian roots, valeranone, valerenal and -kessyl acetate was detected in a small amount in V. fauriei, but kessoglycol diacetate was not detected in V. faudei and V. officinalis Among the valepotrate compounds, major pharmacologically active components in V. wallichii, valtrate was detected in a small amount in V. fauriei and V. officinalis., and detected 1. 42% in V. wallichii. Didrovaltrate was also detected in the three valerian species tested, but acevaltrate was not detected except V. wallichii. On the other hand, antioxidative activity was slightly higher in V. fauriei than those of V. officinalis.

This research was conducted to evaluate the influence of root restriction materials and media on the growth of runner plantlets of 'Seolhyang' strawberry in a nursery field. To achieve this, the influence of three kinds of root media on the growth of runner plantlets was monitored when polyethylene film was used as the root restriction material. In addition, the influence of various root restriction materials (RRS) such as transparent polyethylene film (PE), non-woven fabric (NF), perforated polyethylene film (PP), and root proofing sheet (RPS) on the changes in volumetric water content (VWC) and temperature of root media as well as growth of runner plantlet were investigated when expanded rice hull (ERH) was used as the root medium. In the comparison of root media, growth parameters such as leaf area and crown thickness at 20 d after fixation as well as crown thickness and fresh weights of root and above-ground tissue at 40 d after runner plantlet fixation were higher in the ERH treatment than in sandy loam and loamy sand. When the influence of RRS was compared, the VWC of ERH was 55% just after irrigation, but decreased to 26% at just before irrigation. Ranges of the VWC as influenced by irrigation cycle were 16 to 10% in the PP and less than 10% in the NF and RPS. The soil temperature in the PE treatment was around $1^{\circ}C$ lower than in NF, PP, and RPS. The differences between day and night temperatures were also smaller in the PE treatment rather than those in NF, PP, and RPS. The growths of runner plantlet 50 d after fixation showed that plant heights as well as fresh weights of root and above-ground tissue were higher in the PE treatment than in NF, PP, and RPS. NF and PP did not effectively restrict roots inside the medium and the roots of runner plantlets penetrated through the root restriction materials resulting in the formation of root system below the restriction materials. The above results indicate that ERH is more effective than sandy loam or loamy sand as root medium. PE rather than NF, PP, or RPS as root restriction material resulted in better growth of runner plantlets in propagation of 'Seolhyang' strawberry. The results of this research will be used for production of high quality runner plantlets in strawberry propagation.
