BACKGROUND: Chemical fungicides not only may pollute the ecosystem but also can be environmentally hazardous, as the chemicals accumulate in soil. Biological control is a frequently-used environment-friendly alternative to chemical pesticides in phytopathogen management. However, the use of microbial products as fungicides has limitations. This study isolated and characterized a three-antifungal-enzyme (chitinase, cellulase, and ${\beta}$-1,3-glucanase)-producing bacterium, and examined the conditions required to optimize the production of the antifungal enzymes. METHOD AND RESULTS: The antifungal enzymes chitinase, cellulase, and ${\beta}$-1,3-glucanase were produced by bacteria isolated from an sawmill in Korea. Based on the 16S ribosomal DNA sequence analysis, the bacterial strain AM50 was identical to Streptomyces sp. And their antifungal activity was optimized when Streptomyces sp. AM50 was grown aerobically in a medium composed of 0.4% chitin, 0.4% starch, 0.2% ammonium sulfate, 0.11% $Na_2HPO_4$, 0.07% $KH_2PO_4$, 0.0001% $MgSO_4$, and 0.0001% $MnSO_4$ at $30^{\circ}C$. A culture broth of Streptomyces sp. AM50 showed antifungal activity towards the hyphae of plant pathogenic fungi, including hyphae swelling and lysis in P. capsici, factors that may contribute to its suppression of plant pathogenic fungi. CONCLUSION(S): This study demonstrated the multiantifungal enzyme production by Streptomyces sp. AM50 for the biological control of major plant pathogens. Further studies will investigate the synergistic effect, to the growth regulations by biogenic amines and antifungal enzyme gene promoter.

Bacterial leaf blight of carrots caused by Xanthomonas hortorum pv. carotae (Xhc) is an important worldwide seed-borne disease. In 2012 and 2013, symptoms similar to bacterial leaf blight were found in carrot farms in Jeju Island, Korea. The phenotypic characteristics of the Korean isolation strains were similar to the type strain of Xhc. Pathogenicity showed symptoms on the 14th day after inoculation on carrot plants. Identification by genetic method was multi-position sequencing of the isolated strain JJ2001 was performed using four genes (danK, gyrB, fyuA, and rpoD). The isolated strain was confirmed to be most similar to Xhc M081. Furthermore, in order to analyze the genetic characteristics of the isolated strain, whole genome analysis was performed through the next-generation sequencing method. The draft genome size of JJ2001 is 5,443,372 bp, which contains 63.57% of G + C and has 4,547 open reading frames. Specifically, the classification of pathovar can be confirmed to be similar to that of the host lineage. Plant pathogenic factors and determinants of the majority of the secretion system are conserved in strain JJ2001. This genetic information enables detailed comparative analysis in the pathovar stage of pathogenic bacteria. Furthermore, these findings provide basic data for the distribution and diagnosis of Xanthomonas hortorum pv. carotae, a major plant pathogen that infects carrots in Korea.
In 1991, the leaf brown spot of ivy-aureus (Scindapsus aureus) was found in Taejon and Seoul, Korea. The symptoms were appeared as dark-brown spots. The lesions were often surrounded by yellowish halos. These spots were enlarged to circular or elliptical in shape and dark-brown to black in color with slightly elevated in margin and sunken in center. The pathogenic bacteria were isolated from the diseased leaf of ivy-aureus were identified as Erwinia chrysanthemi on the basis of bacterial characteristics therefore, we would like to propose to the name of ivy-aureus disease caused by E. chrysanthemi as“bacterial brown rot of ivy-aureus”hereafter.
Xenorhabdus nematophila (Xn)와 Photorhabdus tempeerata subsp. temperata (Ptt)의 곤충병원세균을 배추좀나방(Plutella xylostella)의 혈강에 주입할 경우 높은 병원력을 보였다. 본 연구는 이들 세균 배양액의 섭식 처리에 따른 배추좀나방에 대한 병원성 유기를 조사하였다. 세균 배양액만을 이용하여 배추좀나방 3령충에 섭식 처리한 결과 뚜렷한 병원성을 유발하지 못하였으나, Bacillus thurigiensis(Bt) 와 혼합 처리하였을 때 높은 Bt 병원성 제고 효과를 나타냈다. 물질 추적을 위해서 이 세균 배양액을 유기 용매를 이용하여 헥산, 에틸아세테이트 및 수용액 추출 분획구로 분리하였다. 대부분이 Bt 상승효과는 에틸아세테이트 추출 분획구에서 나타났다. Thin layer chromatography 분석 결과는 에틸아세테이트 분획구가 대사물질을 포함하고 있으며, 이들이 헥산 또는 수용액 추출 분획구에 포함된 물질과는 상이하다는 것을 나타냈다. 이러한 결과는 이들 곤충병원세균이 Bt 병원성을 제고시키는 물질을 생산하고 배양액으로 분비한다고 제시하고 있다.
Background: Biofilms were the third-dimensional structure in the solid surface of bacteria. Bacterial biofilms were difficult to control by host defenses and antibiotic therapies. Escherichia coli (E. coli) and Salmonella were popular pathogenic bacteria that live in human and animal intestines. Essential oils are aromatic oily liquids from plant materials and well known for their antibacterial activities. Method: This study was conducted to determine effect of essential oil on anti-biological biofilm formation of E. coli and Salmonella strains in in vitro experiment. Two kinds of bacterial strains were separated from 0.2 g pig feces. Bacterial strains were distributed in 24 plates per treatment and each plates as a replication. The sample was coated with a Bacterial biofilm formation was. Result: Photographic result, Escherichia coli (E. coli) and Salmonella bacteria colony surface were thick smooth surface in control. However, colony surface in blended and single essential oil treatment has shown crack surface layer compared with colony surfaces in control. Conclusion: In conclusion, this study could confirm that essential oils have some interesting effect on anti-biofilm formation of E. coli and Salmonella strains from pig feces.
The induction of growth promotion on numerous crops by rhizobacteria is a well documented phenomenon. In case of tomato (Lycopersicon esculentum), fruit yield is higher in replant soil than that in fresh soil. To investigate what kind of rhizobacterium is involved, microbial community in rhizosphere and on rhizoplane of tomato plants from each soil was analyzed by dilution plating on selective media. Many Gram-negative bacteria and actinomycetes were isolated from tomato in replant soil. One Gram-negative rhizobacterium isolated was identified as Pseudomonas putida based on its biochemical characteristics, fatty acid methyl ester analysis and 16S rDNA sequence. This bacterium designated strain 17 inhibited the growth of Pseudomonas corrugata, and increased growth of tomato seedlings. In addition, its culture filtrate inhibited conidial germination of plant-pathogenic fungi such as Fusarium oxysporum f. sp. radicis-lycopersici, F. oxysporum f. sp. cucumerinum, and Nectria radicicola. Scanning electron microscopy revealed strain 17 colonized and persisted on the epidermal surfaces of tomato radicles and roots. These results suggest that P. putida strain 17 may serve as a biological control agent to suppress multiple soil-borne diseases for tomato plants. Increased microbial populations that suppress deleterious microorganisms including pathogens could be one of the major factors in increased tomato yield in replant soil.
길항력이 있는 그람음성균들의 생물적 방제제로 성공적인 개발은 균의 건조와 저장시의 낮은 생존을 때문에 건조제형으로 만드는 것은 제한되어있다. 그람음성균의 배양시 높은 염(NaCl)의 첨가는 삼투압 조절제인 osmolyte의 생성을 유도하여 건조 시 생존율을 높인다고 알려져 있다. 그람음성의 길항력을 내는 비병원성 Erwinia carotovora subsp. carotovora(Ecc) 9-3의 건조와 저장시의 염의 영향에 관해 연구하였다. Ecc 9-3의 생장률은 0.5 M의 염이 첨가되어 있는 배지에서 큰 차이가 없었지만 0.7 M의 염이 첨가된 배지에서는 급격하게 낮아진 것을 확인하였다. 건조와 저장시의 생존율은 0.5 M의 염을 넣은 배지가 염을 넣지 않은 배지보다 상온건조와 동결건조에서 각각 2배 그리고 3배의 높은 생존율을 얻었다. 건조 후 저장에서도 역시 염이 높을수록 Ecc 9-3의 생존율이 더 높았다. 그리고 상온건조와 동결건조 모두 $4^{\circ}C$에 저장하였을 때 생존율에 큰 차이가 없었다. 그러나 생존율은 $27^{\circ}C$, $37^{\circ}C$에 저장하였을 때 염의 농도가 높을수록 생존율이 증가하였다. 첨가제 실험에서는 lactose가 동결건조와 상온건조 모두에 영향을 주었고 dextrin은 상온건조에 큰 영향을 주었다.
여러 식물의 근권에서 세균 균주들을 분리하여 이들의 항진균성 물질 생성능을 조사하였다. 분리균주 중 Fusarium oxysproum을 포함한 5가지 주요 식물병원성 진균에 대한 생장저해가 우수한 7개 균주를 16S rRNA 유전자 염기서열로 동정하였다. 이들 중 Paenibacillus peoriae RhAn32, Pseudomonas otitidis TK1과 Bacillus cereus TK2가 가장 넓은 생장 저해범위를 나타내었으며 항진균성 물질 중 siderophore 생성능은 P. otitidis TK1이 3.21 mM/ml로 가장 높았고 HCN은 Pseudomonas koreensis Rh2와 Rh7 균주만이 생성하였으며 Brevibacterium frigoritolerans EnA9와 P. peoriae RhAn32는 각각 1558.9와 $1436.7{\mu}M$ glucose/min/mg protein의 높은 chitinase 활성을 나타내었다. P. otitidis TK1과 P. peoriae RhAn32의 항진균능을 진균과의 공동배양을 통해 정량분석한 결과 세 종류의 Fusarium oxysproum의 생장을 유의성 있게 저해하였다. P. otitidis TK1은 식물생장을 촉진하는 호르몬인 auxin의 생성능도 가장 높았는데 이 균주를 4주 자란 토마토 유묘에 F. oxysporum f. sp. lycopersici와 동시에 접종하여 8주간 재배하였을 때 줄기와 뿌리 길이 및 습윤 중량이 비접종 대조군에 비해 각각 45.7, 64.9와 118%로 유의성 있게 증가하였다. 따라서 이 균주들은 유기합성농약을 대체하면서 식물생장을 촉진하는 미생물제로서의 적용이 가능할 것으로 기대된다.
Stem rots caused by Rhizoctonia solani and Sclerotinia sclerotiorum have been known as devastating diseases in balloon flower plants. Antifungal activities of four fungicides, azoxystrobin, polyoxin B, trifloxystrobin and validamycin A were evaluated in vitro, showing effective suppression with mycelial growth of the fungal isolates on PDA media. Efficacies of the four fungicides were also demonstrated in stem tissues of balloon flower plants against R. solani and S. sclerotiorum. A commercially available Bacillus subtilis strain Y1336 was tested in terms of antagonistic biological control of stem rot disease of balloon flower plants. The bacterial strain revealed its antifungal activities against R. solani and S. sclerotiorum demonstrated by dual culture tests using paper discs and two plant pathogenic fungi on PDA media, as well as by plant inoculation assay, indicating that this antagonistic bacterial strain can be incorporated into disease management program for balloon flower stem rot diseases together with the four chemical fungicides.
목적: 본 연구는 육제품 제조 시 가열조건 및 육가공장 규모에 따른 육제품의 저장 중 이화학적 및 미생물학적 변화에 대해 알아보고자 실시하였다. 연구방법: 가열조건은 1차 및 2차 가열조건으로 하였고, 육가공장 규모는 대, 중, 소로 구분하여 이화학적 및 미생물학적 변화로 pH, VBN, 일반세균, 대장균군, 대장균 및 병원성 세균에 대해 7일 간격으로 측정하였다. 결과: 육제품의 가열조건에 따른 병원성 세균(Staphylococcus aureus, Listeria monocytogenes, Clostridium perfrengens 및 Escherichia coli)에 대해 측정한 결과 1차 가열 후 모두 음성으로 나타났다. pH는 초기 1차 및 2차 가열 후 각각 6.17-6.55 및 6.17-6.56의 범위로 가열조건에 따른 차이를 보이지 않았다. 그러나 저장 35일에는 1차 가열 제품은 5.13-6.00의 낮은 pH를 보인 반면 2차 가열 제품의 경우 5.42-6.19 범위로 1차 가열제품보다 높은 경향을 보였다. VBN 함량은 초기 3.89-7.77 mg%의 범위를 보였고 저장 35일에는 9.38-13.05 mg%로 점차 증가하는 경향을 보였다. 가열조건에 따라서는 2차 가열 후 감소하는 경향을 보였으나 육가공장 규모에 따라 다르게 나타났다. 일반세균수는 초기 1.39-2.96 log CFU/g이였고 저장 35일에 1차 및 2차 가열 후 각각 6.13-7.12 log CFU/g 및 3.46-6.92 log CFU/g으로 2차 가열 후 세균수가 적게 나타났다. 또한 육가공장 규모에 따라서는 이화학적 및 미생물학적으로 차이가 나타나지 않았다. 결론: 따라서 1차 가열로 병원성 미생물 제어는 가능하나 2차 가열로 인하여 저장기간을 연장시킬 수 있을 것으로 보이며 제품의 특성에 맞는 관리가 필요할 것으로 사료된다.
