Apparent digestibility coefficients of plant feed ingredients for olive flounder (Paralichthys olivaceus)

  • This study was designed to determine the apparent digestibility coefficients of soybean meal, soy protein concentrate (SPC), soy protein isolate (SPI), rapeseed meal (RSM), pea protein concentrate (PPC), wheat gluten meal (WGM) and wheat flour (WF) for olive flounder, Paralichthys olivaceus. A reference diet (RF) was formulated to meet the nutrient requirements of olive flounder with 1% chromic oxide (Cr2O3) as an inert indicator. Test diets were prepared to contain 70% RF and 30% of the test ingredient. Olive flounder, averaging 150 ± 8.0 g, was cultured in 400-L fiberglass tanks at a density of 25 fish per tank. Fecal collection columns were attached to each tank. Fecal samples were obtained from triplicate groups of fish for 4 weeks. Dry matter digestibility of SPC (75%) and WGM (76%) were significantly higher than the other test ingredients. Protein digestibility of SPC (85%), PPC (88%) and WGM (89%) were significantly higher than the other test ingredients, and protein digestibility of RSM (77%) and WF (76%) was lower than the other ingredients tested. Lipid digestibility of SPC (72%) and SPI (69%) were significantly higher than the other test ingredients. Energy digestibility of SPC (85%) and WGM (82%) were significantly higher than that of others tested ingredients. The availability of amino acids in WGM was generally higher than in other plant-feed ingredients. Therefore, SPC and WGM were seems to be efficient as potential protein sources for olive flounder compared to other tested ingredients. Overall, findings of the current study may assist in more efficient and economical formulation of diets using plant feed ingredients for olive flounder.

  • Digestibilities of nutrients and energy are among the most important parameters to be determined in feed evaluation research. The apparent digestibility coefficients (ADC) of protein, total carbohydrate (TCHO), fat, energy and amino acids were determined for 14 common feed ingredients using chromic oxide as external indicator with Israeli carp (Cyprinus carpio). The ingredients tested were; corn, corn starch, gelatinized starch, wheat middling, wheat grade inferior, corn gluten meal, rapeseed meal (solvent extracted), soybean meal (solvent extracted), blood meal (drum dried), feather meal(hydrolyzed), file fish meal (flame dried), sardine fish meal (steam dried), sardine fish meal (flame dried) and brewers yeast (dehydrated). The overall ADC values were high in Israeli carp showing high capacity to digest their feed ingredients irrespective of plant or animal sources. In addition the ADC of plant protein was high enough to support the successful supplementation of fish meal with other plant proteins.

Plant RNA Virus Sequences Identified in Kimchi by Microbial Metatranscriptome Analysis

  • Plant pathogenic RNA viruses are present in a variety of plant-based foods. When ingested by humans, these viruses can survive the passage through the digestive tract, and are frequently detected in human feces. Kimchi is a traditional fermented Korean food made from cabbage or vegetables, with a variety of other plant-based ingredients, including ground red pepper and garlic paste. We analyzed microbial metatranscriptome data from kimchi at five fermentation stages to identify plant RNA virus-derived sequences. We successfully identified a substantial amount of plant RNA virus sequences, especially during the early stages of fermentation: 23.47% and 16.45% of total clean reads on days 7 and 13, respectively. The most abundant plant RNA virus sequences were from pepper mild mottle virus, a major pathogen of red peppers; this constituted 95% of the total RNA virus sequences identified throughout the fermentation period. We observed distinct sequencing read-depth distributions for plant RNA virus genomes, possibly implying intrinsic and/or technical biases during the metatranscriptome generation procedure. We also identified RNA virus sequences in publicly available microbial metatranscriptome data sets. We propose that metatranscriptome data may serve as a valuable resource for RNA virus detection, and a systematic screening of the ingredients may help prevent the use of virus-infected low-quality materials for food production.

Screening of Inhibition Activity of LPS-induced NO Production by Ethanol Extracts from Jeju Island Native Plants and Algae

  • Herbal medicines have been used as a basic means of clinical trial throughout history, and traditional medicines are targeted to seek functional components. To discover new cosmetic or food ingredients among numerous natural resources from Jeju island, we screened for inhibition activity against nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Although NO formation plays an important role to relax vascular muscles or eliminate tumors, NO produced excessively in inflammatory condition can cause metabolic diseases or inflammatory dysfunctions. Among 52 natural resources ethanol extracts, 5 extracts inhibited NO production over 25% compared to only LPS-treated control at the concentration of $100{\mu}g/mL$. In further study, we try to investigate other bio-activities and the phytochemicals of 5 different extracts as useful ingredients for cosmetics or functional foods.

Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT. (고삼 수집종의 생육특성, 주요성분 및 항산화 활성)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.

  • The currently existing "Bonchojeonghwa (本草精華)" is a manuscript without the preface and the epilogue, composed of 2 books in 2 volumes. This book is a quintessence of knowledge on science of medicinal ingredients (medicinal phytology I herbal science) as well as an trial of new development in Chosun medical science. I.e. this book includes surprising change representing medical science in Chosun dynasty as a single publication on science of medicinal ingredients. It holds a value essential to clinician as a specialized book in medicinal ingredients, and Includes richer content on medicinal ingredients than any other books published before. In addition, it is away from boring list-up of superfluous knowledge as seen in "Bonchokangmok(本草綱目)" published in China, and well summarizes essential knowledge which can be used within a range of medicines available in Korea. This book has an outstanding structure that can be even used in today's textbook on science of medicinal ingredients, as it has clear theory, system and classification. Because it handles essential learning points prior to prescription to disease, it is possible to configure new prescription and adjustment of medicinal materials. Moreover, this book can play a good role for linguistic study at the time of publication, because it describes many drugs in Hangul in many parts of the book. "Bonchojeonghwa" includes a variety of animals, plants and mineral resources in Korea, like "Bonchokangmok" which was recently listed in UNESCO. As such, it has a significance in natural history as well as pharmacy in Korean Medicine. It has various academic relationships all in biologic & abiologic aspects. It has importance in sharing future biological resources, building up international potential, setting up the standard for biologic species under IMF system, and becoming a base for resource diplomacy. We should not only see it as a book on medicinal ingredients in terms of Oriental Medicine, but also make an prudent approach to it in terms of study strengthening Korea's national competitiveness. After bibliographical reviewing on the features & characteristics of the only existing copy of "Bonchojeonghwa" housed in Kyujanggak(奎章閣) of Seoul National University, the followings are noted. First, "Bonchojeonghwa" is a specialized book on medicinal ingredients voluntarily made by private hands to distribute knowledge on drugs in the desolate situation after Imjinoeran (Japanese Invasion in 1592), without waiting for governmental help. Second, it raised accessibility and practicality by new editing. Third, it classified 990 different kinds of drugs into plant, animal, and mineral at large, and dassified more in detail into 15 'Bu' and 48 'Ryu' at 258 pages. Fourth, the publication of this book is estimated to be around 1625~1633, at the time of Injo's reign in 17th century. Fifth, it contains the existing & up-to-date knowledge at the time of publication, and it is possible to see the supply-demand situation by Hangul descriptions in 149 places in the book. By the fact that there are many linguistic evidences of 17th century, explains well when the book was published.

Amino acid digestibility in plant protein sources fed to growing pigs

  • Objective: The objective was to determine standardized ileal digestibility (SID) of amino acids (AA) in 11 plant protein sources fed to growing pigs. Methods: Eleven feed ingredients used were sesame meal, two sources of soybean meal (SBM) produced in the Republic of Korea, a source of SBM produced in India, high-protein distillers dried grains (HPDDG), perilla meal, canola meal, copra meal, corn germ meal, palm kernel expeller, and tapioca distillers dried grains (TDDG). Experimental diets were prepared to contain each test ingredient as a sole source of AA, and a nitrogen-free diet was also prepared to estimate the basal ileal endogenous losses of AA. Twelve barrows surgically fitted with T-cannulas at the distal ileum with an initial body weight of 29.0 kg (standard deviation = 3.0) were individually housed in metabolism crates equipped with a feeder and a nipple drinker. A $12{\times}9$ incomplete Latin square design was employed with 12 experimental diets, 12 animals, and 9 periods. After a 5-d adaptation period, ileal digesta were collected on d 6 and 7 in each experimental period. Results: Values for apparent ileal digestibility of most indispensable AA in three sources of SBM were greater compared with other test ingredients except HPDDG and canola meal (p<0.05). Pigs fed diets containing SBM sources had also greater SID of most indispensable AA compared with those fed diets containing other test ingredients (p<0.05) except for HPDDG and canola meal. There was no difference in the apparent ileal digestibility and SID of AA among sources of SBM. The TDDG had the least value for the SID of methionine among test ingredients (p<0.05). Conclusion: The SID of most AA in SBM, HPDDG, and canola meal were greater than those in sesame meal, perilla meal, copra meal, and TDDG.

Comparison of Growth Characteristics and Active Ingredients in Atractylodes Inter-Specific Hybrid Cultivars (삽주 육성품종 간 생육특성 및 유효성분 분석)

  • Background: Atractylodes japonica Koidz. and Atractylodes macrocephala Koidz. belong to the family (Asteraceae). Their rhizomes, called white Atractylodes rhizomes, are used in traditional medicine. To address some issues with their cultivation, we recently developed eight hybrid cultivars by interspecific hybridization of A. japonica and A. macrocephala. This study was conducted to screen the hybrid cultivars that have high amounts of active ingredients and yield ability. Methods and Results: Experiments were conducted using the eight hybrid cultivars and A. macrocephala (control cultivar) in the experimental field of the department of Herbal Crop Research located in Eumseong, South Korea. We investigated the growth characteristics of the aerial and underground parts. Among the cultivars, 'Sanwon' had the highest rhizome dry weight (53.8 g/plant), followed by 'Dachul' (50.0 g/plant). In addition, the content of atractylenolide I, II, III and total active ingredients were investigated using high-performance liquid chromatography. Compared with A. macrocephala, most of the inter-specific hybrid cultivars had a higher content of active ingredients and yield ability. Conclusions: Through study, we established the superior quality of Atractylodes inter-specific hybrid cultivars. In particularly, it was found that 'Dachul' may be grown as a superior cultivar, with high amount of active ingredients as well as yield ability.

Meat analog as future food: a review

  • The definition of meat analog refers to the replacement of the main ingredient with other than meat. It also called a meat substitute, meat alternatives, fake or mock meat, and imitation meat. The increased importance of meat analog in the current trend is due to the health awareness among consumers in their diet and for a better future environment. The factors that lead to this shift is due to low fat and calorie foods intake, flexitarians, animal disease, natural resources depletion, and to reduce greenhouse gas emission. Currently, available marketed meat analog products are plant-based meat in which the quality (i.e., texture and taste) are similar to the conventional meat. The ingredients used are mainly soy proteins with novel ingredients added, such as mycoprotein and soy leghemoglobin. However, plant-based meat is sold primarily in Western countries. Asian countries also will become a potential market in the near future due to growing interest in this product. With the current advance technology, lab-grown meat with no livestock raising or known as cultured meat will be expected to boost the food market in the future. Also, insect-based products will be promising to be the next protein resource for human food. Nevertheless, other than acceptability, cost-effective, reliable production, and consistent quality towards those products, product safety is the top priority. Therefore, the regulatory frameworks need to be developed alongside.

Changes in Growth, Active Ingredients, and Rheological Properties of Greenhouse-cultivated Ginseng Sprout during its Growth Period (하우스에서 재배된 새싹인삼의 재배시기별 생육, 유효성분 및 물성의 변화)

  • Background: The ginseng ginsenosides, which have various physiological activities, are known to be more abundant in the leaves than in the roots, and the consumers' interest in ginseng sprout as a functional vegetable has been increasing. Methods and Results: The aim of this study was to investigate the effects of growth period on growth properties, active ingredients and rheology of ginseng sprouts cultivated in a non-heated greenhouse equipped with a shade net for 60 days, starting from the end of May to the middle of July. The chlorophyll content of the leaves decreased, but their length and width increased with increasing cultivation days. In particular, growth increased significantly until 40 days, but only slightly after 50 days. The stem length did not increase greatly from the 20 th to the 30 th day of cultivation, but increased significantly from the 30 th to the 40 th day, and then further increased gradually. The weight of the leaves, stems, and roots increased slightly, but not change significantly. After 40 days of cultivation, the total ginsenoside content increased by 1.07 times in the leaves and decreased by 0.80 times in the roots with increasing cultivation days. The leaf contents of ginsenosides $Rg_1$, Re, $Rb_1$, Rc, $F_3$ and $F_4$ increased with increasing cultivation days. The rheological properties of ginseng sprout showed the greatest influence on stem hardening with increasing cultivation days. Conclusions: Therefore, based on the growth characteristics, active ingredients and physical properties, 40 days after sowing was considered to be an appropriate harvesting time for ginseng sprouts.
