수수 유전자원의 작물학적 특성 (Agronomic Characteristics of Sorghum bicolor (L.) Moench Germplasm)

  • 수년 동안 국내 전역에서 수집된 수수 179 유전자원을 공시하여 작물학적 특성조사를 통한 품종육성의 기초자료를 제공코자 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 수수의 수형은 산수형, 소경수수형, 반소경수수형, 극밀수형, 및 밀수형의 5가지 수형이 존재하였으며, 수형별 빈도를 보면 소경수수형이 38.0%(68자원)로 가장 많았으며, 극밀수형 및 밀수형이 각각 16.2%, 7.8%로 가장 적었다. 2. 수수 이삭의 까락 유무는 28.5%(51자원)가 까락을 갖고 있었으며, 나머지 71.5%(128자원)는 까락이 없는 것으로 분류되었다. 3. 종실형태는 43.0%(77자원)가 원형으로 가장 많이 차지하였으며, 편원형이 7.3%(13자원)로 가장 적었다. 종피색은 황갈색이 84.4%(151자원)로 가장 많았으며, 갈색 및 흰색이 똑같이 1.1%로 가장 적었다. 4. 수수 179자원의 출수소요일은 67~88일의 범위를 나타내어 변이폭이 컷으며, 평균은 77.4일이었다. 출수소요일의 분포는 179자원 중 76~80일이 37.4%(67자원)로 가장 많았으며, 86일 이상도 5.6%이었으며, 70일 이전에 출수한 자원도 10.0%를 나타내었다. 5. 간장은 92~360 cm의 범위를 보였으며, 평균은 253.8 cm로 변이의 폭이 컷다. 간장의 분포는 291~310 cm가 20.6% (37자원)로 가장 많았다. 6. 수장은 15~49 cm의 범위를 보였고, 평균 28.8 cm이었다. 수장의 분포는 31~35 cm가 26.3%(47자원)로 가장 많았으며, 15 cm 이하가 0.6%로 가장 적었다. 7. 생육일수는 110~146일의 범위를 나타내어 변이 폭이 상당히 큼을 알 수 있었고, 평균은 125.5일이었다. 생육기간의 빈도는 111~115일이 39.7%(71자원)로 가장 많았으며, 110일 이하와 141일 이상에 속하는 자원도 각각 2, 23자원이나 되었다. 8. 1수립수의 범위는 163~4,532개로 변이 폭이 상당히 컷으며, 평균은 2,068.6개 이었다. 1수립수 분포는 1,601~2,000개가 25.7%(46자원)로 가장 많았다. 9. 공시된 179 유전자원의 1000립중은 범위가 10.6~38.1 g으로 변이폭이 컷으며, 평균은 25.6 g이었다. 1000립중의 분포는 26.0~30.0 g가 전체의 44.1%(79자원)로 가장 많았다

High Resolution Melting Analysis을 이용한 고추 유전자원의 Cucumber mosaic virus 저항성 평가 (Evaluation of Resistance in Pepper Germplasm to Cucumber mosaic virus by High Resolution Melting Analysis)

  • 농업유전자원센터에 보존하고 있는 고추 유전자원 1,941점을 240H02sp6 SNP 마커를 이용하여 Cucumber mosaic virus 저항성 정도를 평가하였다. 1941점의 고추 유전자원에서 89점이 저항성, 162점이 이형접합성, 1270점이 이병성으로 분석되었다. 고추 종별로는 C. annuum은 총 839자원 중 38점이 저항성으로 나타났으며, C. baccatum은 277점 중 23%에 해당하는 자원만 분석이 되었고 분석된 자원 중 20점이 저항성을 나타났다. C. chinense는 21점이 저항성, C. frutescens는 8점이 저항성을 보였다. C. pubescens는 K157779 자원만 이병성을 나타내었으며, Capsicum sp. 자원들 중에는 저항성 자원이 없었다. 야생근연종인 C. chacoense는 15점이 분석되어 1점이 저항성, 4점이 이형접합성, 6점이 이병성을 보였고, C. galapagoense, C. tovarii는 각 한 점씩 분석되었고 모두 이병성으로 나타났다.

Comparison of Grain Quality Traits between Japonica Rice Cultivars from Korea and Yunnan Province of China

  • Improving eating quality is one of the most important objectives in japonica rice breeding programs in Yunnan Province of China. Eating quality and its relevant traits of nine Korean and 11 Yunnan rice cultivars were comparatively analyzed in this study. The grain shape of most Yunnan japonica rice cultivars have a relatively slender shape and are slightly larger than Korean rice cultivars. Palatability value of cooked rice of Yunnan rice cultivars was significantly lower, while the protein content of Yunnan rice cultivars was significantly higher than that of Korean cultivars. Peak viscosity and breakdown viscosity of the Yunnan rice cultivars were significantly lower, while setback viscosity of the Yunnan rice cultivars was significantly higher than in Korean rice cultivars. Palatability value of cooked rice was negatively correlated with protein content and setback viscosity but positively correlated with peak viscosity, breakdown viscosity, and cool paste viscosity. Through multiple linear regression analysis, an equation for estimating palatability value(PV) of cooked rice based on quality traits was generated as dependent only upon protein content(PC), PV=139.024-(10.865$\times$PC) with an $R^2$ value of 0.822. The results suggest that reducing protein contents should be the major target in improving eating quality of Yunnan japonica rice cultivars through integrated approaches of both cultivar development and appropriate cultural practices. Genetic similarities among cultivars based on DNA markers which had been identified as associated with grain quality seemed not to be directly related to PV.

Evaluation of Barley and Wheat Germplasm for Resistance to Head Blight and Mycotoxin Production by Fusarium asiaticum and F. graminearum

  • Fusarium head blight (FHB) is one of the most serious diseases in barley and wheat, as it is usually accompanied by the production of harmful mycotoxins in the grains. To identify FHB-resistant breeding resources, we evaluated 60 elite germplasm accessions of barley (24) and wheat (36) for FHB and mycotoxin accumulation. Assessments were performed in a greenhouse and five heads per accession were inoculated with both Fusarium asiaticum (Fa73, nivalenol producer) and F. graminearum (Fg39, deoxynivalenol producer) strains. While the accessions varied in disease severity and mycotoxin production, four wheat and one barley showed <20% FHB severity repeatedly by both strains. Mycotoxin levels in these accessions ranged up to 3.9 mg/kg. FHB severity was generally higher in barley than in wheat, and Fa73 was more aggressive in both crops than Fg39. Fg39 itself, however, was more aggressive toward wheat and produced more mycotoxin in wheat than in barley. FHB severity by Fa73 and Fg39 were moderately correlated in both crops (r = 0.57/0.60 in barley and 0.42/0.58 in wheat). FHB severity and toxin production were also correlated in both crops, with a stronger correlation for Fa73 (r = 0.42/0.82 in barley, 0.70 in wheat) than for Fg39.

Pathogenic Diversity of Ascochyta rabiei Isolates and Identification of Resistance Sources in Core Collection of Chickpea Germplasm

  • Ascochyta blight caused by Ascochyta rabiei (Pass.) Lab. (Telomorph: Didymella rabiei) (Kov.) is one of the most important fungal diseases in chickpea worldwide. Knowledge about pathogen aggressiveness and identification resistance sources to different pathotypes is very useful for proper decisions in breeding programs. In this study, virulence of 32 A. rabiei isolates from different part of Iran were analyzed on seven chickpea differentials and grouped into six races based on 0-9 rating scale and susceptibility/resistant pattern of chickpea differentials. The least and most frequent races were race V and race I, respectively. Race V and VI showed highly virulence on most of differential, while race I showed least aggressiveness. Resistance pattern of 165 chickpea genotypes also were tested against six different A. rabiei races. ANOVA analysis showed high significant difference for isolate, chickpea genotypes and their interactions. Overall $chickpea{\times}isolate$ (race) interactions, 259 resistance responses (disease severity ${\leq}4$) were identified. Resistance spectra of chickpea genotypes showed more resistance rate to race I (49.70%) and race III (35.15%), while there were no resistance genotypes to race VI. Cluster analysis based on disease severity rate, grouped chickpea genotypes into four distinct clusters. Interactions between isolates or races used in this study, showed the lack of a genotype with complete resistance. Our finding for virulence pattern of A. rabiei and newly identified resistance sources could be considerably important for integration of ascochyta blight resistance genes into chickpea breeding programs and proper decision in future for germplasm conservation and diseases management.

Cereal Resources in National BioResource Project of Japan

  • The National BioResource Project of Japan is a governmental project to promote domestic/international research activities using biological resources. The project has 27 biological resources including three cereal resources. The core center and sub-center which historically collected the cereal resources were selected for each cereal program. These resources are categorized into several different types in the project; germplasm, genetic stocks, genome resources and database information. Contents of rice resources are wild species, local varieties in East and Southwest Asia & wild relatives, MNU-induced chemical mutant lines, marker tester lines, chromosome substitution lines and other experimental lines. Contents of wheat resources are wild strains, cultivated strains, experimental lines, rye wild and cultivated strains; EST clones and full-length cDNA clones. Contents of barley resources are cultivar and experimental lines, core collection, EST/cDNA clones, BAC clones, their filters and superpool DNA. Each resource is accessible from the online database to see the contents and information about the resources. Links to the genome information and genomic tools are also important function of each database. The major contents and some examples are presented here.

Cryopreservation of in vitro Grown Shoot Tips of Two Freesia hybrida Cultivars by Droplet-vitrification

  • The droplet-vitrification technique for cryopreservation has proven successful across a diverse range of germplasm, ensuring safe and effective long term preservation. In this study, we investigate an effective cryopreservation protocol using the droplet-vitrification technique for shoot tips of Freesia hybrida cultivars 'Sunny Gold' and 'Sweet Lemon'. To determine optimal conditions for Freesia cryopreservation, we employed a carefully selected standard procedure along with additional treatments and alternative solutions. For 'Sunny Gold', the highest regrowth rate of 24% was achieved when shoot tips underwent dehydration with PVS3 solution for 120 minutes before direct immersion in liquid nitrogen (LN) for 1 hour, coupled with a standard protocol involving a two-step preculture with 0.3 M - 0.5 M sucrose, loading with C4 for 40 minutes, and unloading with 0.8 M sucrose for 40 minutes. In the case of 'Sweet Lemon,' regrowth of cryopreserved shoot tips was observed with dehydration treatments, including PVS2 (A3) for 60 minutes and PVS3 (B1) for 60 minutes, as well as longer exposure. The results reflect the distinct sensitivity of shoot tips to chemical toxicity and osmotic stress in these two genotypes. This study provides valuable evidence to consistently enhance the effectiveness of cryopreservation methods for the long-term conservation of Freesia germplasm.
