광합성세균 Rhodobater capsulatus PS-2의 대량배양 최적화 및 대사산물 분석 (Mass Cultivation and Secondary Metabolite Analysis of Rhodobacter capsulatus PS-2)

  • Plant growth promoting (PGP) hormones, which are produced in a small quantity by bacteria, affect in plant growth and development. PGPs play an important role on the crop productivity in agricultural field. In this study, a photosynthetic bacterial strain producing the PGP was isolated from paddy soil. Bacterial isolate was gram negative, rod-shaped and motility positive. From the 16s rRNA gene sequence analysis, the isolate was identified as Rhodobacter capsulatus PS-2. The mass cultivation of R. capsulatus PS-2 was optimized by considering of the carbon, nitrogen and inorganic salt sources. Optimal medium composition was determined as Na-succinate 4.5 g, yeast extract 5 g, $K_2HPO_4$ 1 g, $MgSO_4$ 5 g, per liter. From the result of 500 L fermentation for 2 days using the optimal medium, the viable cells were $8.7{\times}10^9cfu/mL$. R. capsulatus PS-2 strain produced the carotenoid and indole-3-acetic acid (IAA). The carotenoid extraction and quantitative analysis were performed by HCl-assisting method. Total carotenoid contents from R. capsulatus PS-2 culture broth were measured as $7.02{\pm}0.04$ and $6.93{\pm}0.05mg/L$ under photoheterotrophic and chemoheterotrophic conditions, respectively. To measure the productivity of IAA, colorimetric method was employed using Salkowski reagent at optical density 535 nm. The results showed that the highest content of IAA was $197.44{\pm}5.92mg/L$ in the optimal medium supplemented with 0.3% tryptophan.

Saponarin content and biosynthesis-related gene expression in young barley (Hordeum vulgare L.) seedlings

  • Flavonoids are widely distributed secondary metabolites in plants that have a variety biological functions, as well as beneficial biological and pharmacological activities. In barley (Hordeum vulgare L.), for example, high levels of saponarin accumulate during primary leaf development. However, the effect of saponarin biosynthetic pathway genes on the accumulation of saponarin in barley is poorly understood. Accordingly, the aim of the present study was to examine the saponarin contents and expression levels of saponarin biosynthetic pathway genes [chalcone synthase (CHS), chalcone isomerase (CHI), and UDP-Glc:isovitexin 7-O-glucosyltransferase (OGT)] during early seedling developmental and under several abiotic stress conditions. Interestingly, the upregulation of HvCHS, HvCHI, and HvOGT during early development was associated with saponarin accumulation during later stages. In addition, exposure to abiotic stress conditions (e.g., light/dark transition, drought, and low or high temperature) significantly affected the expression of HvCHS and HvCHI but failed to affect either HvOGT expression or saponarin accumulation. These findings suggested that the expression of HvOGT, which encodes an enzyme that catalyzes the final step of saponarin biosynthesis, is required for saponarin accumulation. Taken together, the results of the present study provide a basis for metabolic engineering in barley plants, especially in regards to enhancing the contents of useful secondary metabolites, such as saponarin.

기초형식에 따른 회전기기의 가동중 진동특성 분석 (Evaluation of Vibration Characteristics of Operating Rotational Machines Depending of Types of Foundation)

  • 비상디젤발전기는 원전의 안전성에 큰 영향을 미치는 매우 중요한 기기이다. 본 연구에서는 세 종류의 비상디젤발전기를 대상으로 가동중 진동을 계측하여 기초시스템에 따른 가동중 진동의 저감효과를 비교하였다. 대상 원전은 영광 5호기, 울진 2호기 및 울진 3호기 원전의 비상디젤발전기로 하였다. 영광 5호기와 울진 3호기의 비상디젤발전기는 동일한 종류의 비상디젤발전기로서 기초형식만 앵커볼트 고정과 스프링-댐퍼시스템을 이용한 면진으로 구분된다. 울진 2호기 비상디젤발전기는 기초부분에 seismic block을 설치하고 코일스프링을 이용하여 진동저감효과를 고려하였으므로 좋은 비교가 될 것이다. 이러한 진동계측의 목적은 기초형태에 따른 진동저감 효과를 비교해 보기 위함이다. 결과적으로 스프링-댐퍼 시스템이 진동저감과 시스템의 안정적 거동측면에서 좋은 효과를 보이고 있음을 알 수 있었다.

Analysis of soyasaponin content and biosynthesis-related gene expression in young pea (Pisum sativum L.) sprouts

  • In legumes, soyasaponins, one of triterpenoid saponins, are major components of secondary metabolites with a more diverse array of bioactive chemicals. Although the biosynthetic pathway of soyasaponins has been largely studied in soybean, the study on the soyasaponin contents and biosynthesis-related gene expression in pea (Pisum sativum L.) is poorly understood. Here, we found the accumulation of only soyasaponin Bb component in the sprouts of two Korean domestic pea cultivars (Dachung and Sachul). This pattern was consistent with our observation that increased expression of PsUGT73P2 and PsUGT91H4 genes, but not PsCYP72A69, could be responsible for biosynthesis of only soyasaponin Bb in pea by examining their gene expression. However, gradual accumulation of soyasaponin Bb at developmental stages was not consistent with the expression of PsUGT73P2 and PsUGT91H4, suggesting that the changes of their protein activities may affect the accumulation patterns of soyasaponin Bb. We also revealed that the increased expression levels of PsUGT73P2 and PsUGT91H4 during light to dark transition led to increase of soyasaponin Bb contents. Collectively, our results provided a molecular basis of metabolic engineering for enhancing useful soyasaponin Bb metabolites in Korean domestic pea cultivars.

다양한 콩 자원들의 기내 조직배양 효율 및 형질전환 (In Vitro Tissue Culture Frequency and Transformation of Various Cultivars of Soybean (Glycine max (L.) Merr.))

  • 유전자 가위 기술 등 생명공학 기술을 콩에 적용하여 새로운 품종을 개발하기 위해서는 효율적인 조직배양 기술이 필수적이다. 식물의 유전형은 조직배양 효율에 의존하는 형질전환 기술의 성공 여부를 결정짓는 중요한 요소로 알려져 있다. 본 연구에서는 우리나라 콩 핵심 집단 내 21개 자원들을 선발하여, 외래 품종 2종과 함께 조직배양 효율을 조사하였다. 그 결과, 근연 관계가 높은 Kwangan, Anpyeong, Seonam은 발아율과 재분화 효율이 높았으며, Daepung, Daewon 품종은 발아율과 재분화율 모두 낮게 관찰되었다. 또한 3종의 외래 품종에서는 표준 유전체 해독에 사용된 Williams82와 Jack, Maverick 모두 높은 조직배양 효율을 보였다. 조직배양 효율이 높은 자원들을 대상으로 Agrobacterium법에 의한 형질전환을 수행하여 PCR 및 bar-strip 분석한 결과 Kwangan, Pungwon, Seonam, 그리고 Maverick 품종에서 제초제 저항성 유전자의 삽입을 확인할 수 있었다. 이들 결과를 바탕으로 농업적 가치가 높은 다양한 콩 품종들의 형질전환을 통한 새로운 품종 개발이 가능할 것이다.

국내 맥류에 발생하는 바이러스병 동시진단 방법 (Simultaneous Detection of Barley Virus Diseases in Korea)

  • 최근 국내 맥류 재배지에서는 대부분 BaMMV, BaYMV, BYDV의 발생이 확인되고 있다. 본 연구에서는 multiplex reverse transcription polymerse chain reaction (mRT-PCR) 방법에 의해 이들 3종류의 바이러스를 동시에 진단하는 방법을 확립하였다. 이들 3종 바이러스의 외피단백질 유전자 정보를 활용하여 각 바이러스에 대한 primer를 제작하였다. mRT-PCR에 사용한 primer는 RT-PCR 반응의 민감도와 특이성에 의해 선발하여 primer 농도와 mRT-PCR의 조건을 설정하였다. 각 바이러스에 대하여 선발된 primer 사용에 의한 mRT-PCR 결과 BaMMV 594 bp, BaYMV 461 bp, BYDV 290 bp의 PCR 산물을 얻을 수 있었다. 본 연구에서 확립된 맥류 바이러스 동시진단방법은 신속하고 특이적인 진단 뿐 아니라 맥류 바이러스병의 전염 등 역학 연구에도 활용 될 것으로 기대된다.

  • Assessment of nitrogen and chlorophyll content from crop leaves can help growers adjust N fertilizer rates to meet the demands of the crop. Numerous researchers have presented their studies about spectral signature of plant leaves to characterize the plant features. However, interrelational review and summary were limited and a communication gap exists between the plant science and optical engineering. Understanding the mechanism of leaf interaction to electromagnetic radiation and factors affecting spectrophotometric measurements can enhance the foundation of optical remote sensing technologies. This paper provides extensive review of previous works in optical sensing and explains the basics of plant optics, spectral measurements for plant stress, factors that affect sensitivity to spectral analysis, and applications that deploy optical remote sensing technologies.

  • 본 논문에서는, 친환경 Green IT 기술과 융합된 식물공장 시스템의 이상적인 표준 설계 및 구축안을 요소기술별로 제시하였으며, 이의 실현 가능성과 문제점 분석을 위해 요소기술별 관련 하드웨어 및 소프트웨어를 종합한 콘테이너형 식물공장 테스트베드를 구축하여 운영하고, 운영상의 문제점을 파악하여 미래의 대형 식물공장 구축과 운영의 기반이 되는 시험연구를 수행하였다.

Inhibitory effects of Thalictrum rochebrunianum var. grandisepalum on tyrosinase in IBMX-induced B16F10 melanoma cells

  • Thalictrum rochebrunianum var. grandisepalum (TRG) is a Korean endemic plant, and it is widely used for edible, medicinal, landscape materials. In this study, we examined the protein and mRNA expression levels of MITF, tyrosinase, TRP-1 and TRP-2 by TRG extract (TRGE) in IBMX-treated melanocytes to evaluate the possibility of using TRG as a whitening material. IBMX were reported as melanin synthesis enhancers. It could increase intracellular melanin synthesis by activation of the microphthalmia-associated transcription factor (MITF) signaling pathway. TRGE did not show cytotoxicity at concentrations below $100{\mu}g/ml$ in B16F10 cells. TREG dose-dependently inhibited protein and mRNA levels of MITF, tyrosinase, TRP-1 and TRP-2. Therefore, we suggest that TRGE is an important natural resource for cosmetic raw materials for whitening function.

발전용 대형 2 행정 디젤 엔진 및 기초의 구조 진동해석 (Structural Vibration Analysis of a Large Two-Stroke Engine and Foundation System for Stationary Power Plants)

  • Structural vibration analysis of the stationary power plant system employing a large two-stroke low speed diesel engine is performed to verify that the vibration characteristics of the system meet design requirements, The system consists of the diesel engine generator and concrete foundation including pile and soil. The system is modeled in the form of a mass-elastic system of 5 degrees of freedom for vibration analysis. Excitation moments and dynamic parameters including engine body stiffness soil stiffness and damping are identified for the analysis, Results of structural vibration analysis of the system are presented and compared with measurements in this paper.
