Charcoal rot disease, caused by the fungus Macrophomina phaseolina, is one of the most important diseases of Sesame (Sesamum indicum) all over the world. However, the population biology of M. phaseolina is poorly understood. In this study, M. phaseolina isolates from five different regions of Iran (Khuzestan, Fars, Bushehr, Hormozgan, and Kohgiluyeh & Boyer-Ahmad provinces) (n=200) were analyzed for genetic variation using inter simple sequence repeats marker. In total, 152 unique haplotypes were identified among the 200 M. phaseolina isolates, and gene diversity (H=0.46-0.84) and genotypic diversity were high in each of the regions. The structure analysis clustered five Iranian populations into two distinct groups, the individuals from group 1 were assigned to the Bushehr population and the individuals from Khuzestan, Fars, Hormozgan and Kohgiluyeh & Boyer-Ahmad were aggregated and formed group 2. The results matched with genetic differentiation and gene flow among regions. Analyses of the distribution of gene diversity within and among five Iranian populations were 61% and 39%, respectively. Our results showed that infected seeds are thought to be the dominant mechanism responsible for the spreading of the pathogen in southern parts of Iran. In summary, it is essential to have local quarantine and prevent seed exchanges between geographical populations to restrict the dispersal of pathogen over long distances and provide certified seeds in Iran.

Scab disease caused by Venturia nashicola is of agroeconomic importance in cultivation of Asian pear. However, little is known about the degree of genetic diversity in the populations of this pathogen. In this study, we collected 55 isolates from pear scab lesions in 13 major cultivation areas in Korea and examined the diversity using sequences of internal transcribed spacer (ITS) region, ${\beta}$-tubulin (TUB2), and translation elongation factor-$1{\alpha}$ ($TEF-1{\alpha}$) genes as molecular markers. Despite a low level of overall sequence variation, we found three distinctive subgroups from phylogenetic analysis of combined ITS, TUB2, and $TEF-1{\alpha}$ sequences. Among the three subgroups, subgroup 1 (60% of isolates collected) was predominant compared to subgroup 2 (23.6%) or subgroup 3 (16.4%) and was distributed throughout Korea. To understand the genetic diversity among the subgroups, RAPD analysis was performed. The isolates yielded highly diverse amplicon patterns and none of the defined subgroups within the dendrogram were supported by bootstrap values greater than 30%. Moreover, there is no significant correlation between the geographical distribution and the subgroups defined by molecular phylogeny. Our data suggest a low level of genetic diversification among the populations of V. nashicola in Korea.
A study on the dominance-diversity of forest vegetation in Mt. Seolag was conducted from May 1981 to Aug. 1983. Based on the field data, the dominance-diversity curves were for 16 sites including slopes and vallies. The curves are grouped in two types, lognormal distribution at the sites of mature vegetation and geometric series at the disturbed or rocky sites. It seems that the curves express the nature of their ecocline, by the hypotheses of some investigators, i.e. Random Niche hypothesis, Niche Pre-emption hypothesis, Lognormal distribution and Logarithmic series. The dominance concentration among the southern, northern and western slope, H'=1.282 at southern slope and H'=1.385 at western slope. Dominance-diversity curves of 16 sites showed Preston's lognormal distribution with small variations among them. It seems that the dominance diversity reflects the differences in the coenocline of their sites. The top 10 dominant species in species sequence of 113 tree species in whole the mountain, were noticed: Quercus mongolica, Pinus densiflora, Acer pseudo-siebold anum, Quercus serrata, Carpinus laxiflora, Styrax obassia, Fraxinus rhynchophylla, Tilia amurensis, Lindera obtusiloba and Abies holophylla in order.
We have determined the complete chloroplast genome of Erigeron Canadensis isolated in Korea. The circular chloroplast genome of E. canadensis is 152,767 bp long and has four subregions: 84,317 bp of large single-copy and 18,446 bp of small single-copy regions are separated by 25,004 bp of inverted repeat regions including 133 genes (88 protein-coding genes, eight rRNAs, and 37 tRNAs). The chloroplast genome isolated in Korea differs from the Chinese isolate by 103 single-nucleotide polymorphisms (SNPs) and 47 insertions and deletion (INDEL) regions, suggesting different invasion sources of E. canadensis in Korea and China. A nucleotide diversity analysis revealed that the trend of the nucleotide diversity of E. canadensis followed that of 11 Erigeron chloroplasts, except for three peaks. The phylogenetic tree showed that our E. canadensis chloroplast is clustered with E. canadensis reported from China. Erigeron canadensis can be a good target when attempting to understand genetic diversity of invasive species.
Several endophytic fungal strains were isolated from Taxus cuspidata and identified by molecular analysis of the internal transcribed spacer and RNA polymerase II second largest subunit. This study aimed to determine the relative abundance and compare the species diversity of endophytic fungal communities within needle leaves and twigs. We identified a total of 49 endophytic fungal species. Notably, two species, Trichoderma dingleyae and Xylaria cubensis, were discovered to be previously unrecorded in Korea. The fungal communities in both plant tissues demonstrated distinct species composition. Differences were observed in the relative abundance and species diversity index between needle leaves and twigs. Our findings suggest that the host plant tissues influence the species diversity of endophytic fungal communities.
Plant-parasitic nematodes are not only an important constraint on agricultural crop production, but also cause both direct and indirect damage to turfgrass, which is a ground cover plant. However, studies on plant-parasitic nematodes of turfgrass in Korea are scarce. A survey for plant-parasitic nematodes was carried out on 13 golf courses in Korea. The results yielded 28 species/taxa belonging to 16 genera and 12 families of plant-parasitic nematodes. Among the isolated species, Helicotylenchus microlobus, Mesocriconema nebraskense, Tylenchorhynchus claytoni, Mesocriconema sp., and Meloidogyne graminicola were the most prevalent species in all management zones. Twelve species were new records of plant-parasitic nematodes in Korea. Highest maximum densities were showed by T. claytoni, Paratylenchus nanus, M. nebraskense, M. graminicola, and H. microlobus. Diversity (H'), was significantly higher in fairways compared to tees and greens, though species evenness (J') and dominance (D) showed no statistically significant differences. This information is crucial in nematode problem diagnosis, and the subsequent formulation of management strategies.
Background : Plant breeding requires the collection of genetically diverse genetic resources. Studies on the characteristics of Platycodon grandiflorum resources have not been carried out so far. The present study was carried out to discriminate P. grandiflorum based on morphological characteristics and genetic diversity using simple sequence repeat (SSR) markers. Methods and Results :We collected 11 P. grandiflorum cultivars: Maries II, Hakone double white, Hakone double blue, Fuji white, Fuji pink, Fuji blue, Astra white, Astra pink, Astra blue, Astra semi-double blue and Jangbaek. Analyses of the morphological characteristics of the collection were conducted for aerial parts (flower, stem and leaf) and underground parts (root). Next, the genetic diversity of all P. grandiflorum resources was analyzed using SSR markers employing the DNA fragment analysis method. We determined that the 11 P. grandiflorum cultivars analyzed could be classified by plant length, leaf number and root characteristic. Based on the genetic diversity analysis, these cultivars were classified into four distinct groups. Conclusions : These findings could be used for further research on cultivar development using molecular breeding techniques and for conservation of the genetic diversity of P. grandiflorum. Moreover, the markers could be used for genetic mapping of the plant and marker-assisted selection for crop breeding.
The present study is an attempt to give an account of the inventory of plant species, phytosociological characteristics of vegetation and pedological characteristics of Rambi Beat Forest under Senchal East Forest Zone, Darjeeling, West Bengal, India. Its plant community were analyzed quantitatively and synthetically. The results reflect dominancy of dicotyledons over monocotyledons in the four studied sites The plant community comprising of 50 plant species belonging to 40 genera of 27 families. Ramhi beat reflected higher diversity of species. Maximum IVI value was recorded by Viola surpense (47.17) in Rambhi forest beat. The Berger parker index and evenness index were found to be highest for Viola surpense, Fragaria nubicola, Pilea umbrosa in Rambhi beat. The soil characteristics of the different pedons revealed alkaline nature of soil in Rambhi beat. Higher levels of soil organic carbon content reflect higher fertility of the soil of Rambhi beat. The response towards soil available nitrogen and phosphate were different among the ten pedons of Rambhi beat. Therefore, proper management and conservative measures needs to be implemented for conservation of bioresources in Senchel wildlife Sanctuary of West Bengal, India.
