Background: Mammalian orthoreovirus type 3 (MRV3), which is responsible for gastroenteritis in many mammalian species including pigs, has been isolated from piglets with severe diarrhea. However, the use of pig-derived cells as an infection model for swine-MRV3 has rarely been studied. Objectives: This study aims to establish porcine intestinal organoids (PIOs) and examine their susceptibility as an in vitro model for intestinal MRV3 infection. Methods: PIOs were isolated and established from the jejunum of a miniature pig. Established PIOs were characterized using polymerase chain reaction (PCR) and immunofluorescence assays (IFAs) to confirm the expression of small intestine-specific genes and proteins, such as Lgr5, LYZI, Mucin-2, ChgA, and Villin. The monolayered PIOs and three-dimensional (3D) PIOs, obtained through their distribution to expose the apical surface, were infected with MRV3 for 2 h, washed with Dulbecco's phosphate-buffered saline, and observed. Viral infection was confirmed using PCR and IFA. We performed quantitative real-time reverse transcription-PCR to assess changes in viral copy numbers and gene expressions linked to intestinal epithelial genes and antiviral activity. Results: The established PIOs have molecular characteristics of intestinal organoids. Infected PIOs showed delayed proliferation with disruption of structures. In addition, infection with MRV3 altered the gene expression linked to intestinal epithelial cells and antiviral activity, and these effects were observed in both 2D and 3D models. Furthermore, viral copy numbers in the supernatant of both models increased in a time-dependent manner. Conclusions: We suggest that PIOs can be an in vitro model to study the infection mechanism of MRV3 in detail, facilitating pharmaceutical development.
Integrated pest management is essential for controlling plant diseases that reduce crop yields. Rapid diagnosis is crucial for effective management in the event of an outbreak to identify the cause and minimize damage. Diagnosis methods range from indirect visual observation, which can be subjective and inaccurate, to machine learning and deep learning predictions that may suffer from biased data. Direct molecular-based methods, while accurate, are complex and time-consuming. However, the development of large multimodal models, like GPT-4, combines image recognition with natural language processing for more accurate diagnostic information. This study introduces GPT-4-based system for diagnosing plant diseases utilizing a detailed knowledge base with 1,420 host plants, 2,462 pathogens, and 37,467 pesticide instances from the official plant disease and pesticide registries of Korea. Crop yield is critically related to the physiological responses and disease resistance of the crop, which could be strongly affected by high temperature conditions. We observed the changes in the growth of barley under higher than ambient air-temperature conditions using a temperature gradient field chamber (TGFC) during winter and spring. Before the stem extension stage of barley growth, Cladosporium sp. spontaneously appeared in the TGFC. The severity of disease became serious under warmer temperature conditions. Further, the stomata closed as the severity of the disease increased; however, stomatal conductance at the initial stage of disease was higher than that of the normal leaves. This was likely due to the Iwanov effect, which explains that stressed plants rapidly and transiently open their stomata before longer-term closure. In this study, we tested three optical methods: soil-plant analysis development (SPAD) chlorophyll index, photochemical reflectance index (PRI), and maximum quantum yield (Fv/Fm). These rapid evaluation methods have not been used in studies focusing on disease stress, although some studies have used these methods to monitor other stresses. These three indicative parameters revealed that diseased barley exhibited lower values of these parameters than normal, and with the increase in disease severity, these values declined further. Our results will be useful in efficient monitoring and evaluation of crop diseases under future warming conditions.

A study was conducted to find out the relationship of physico-chemical properties (viz. organic carbon(OC), pH, electrical conductivity, nitrogen, phosphorus and potassium content) of ginger growing soil with incidence percentage of rhizome rot and wilt disease complex of ginger. Organic carbon content and pH of the ginger soil contributed significantly (93%) in the prediction of ginger rhizome rot and wilt disease complex incidence with negative correlation. Soil having weak acidic reaction with OC percent greater than 2.25 was observed to have the lower average incidence of the disease.

Several outbreaks of Getah virus (GETV) have been reported in horses and pigs, causing large economic losses. However, there have been no reports describing serological survey for GETV in South Korea pigs. The present study conducted serological survey of GETV in South Korean pigs. A total of 670 whole blood samples were collected from domestic pigs. The overall seropositive rate was 26.4%, higher than the rates observed in racehorses in 2013-2014. Preparations for epidemics of novel diseases caused by climate change should include regular serological survey for these diseases, including GETV, and the development of vaccines against novel pathogens.
This test checked jujube witches'-broom disease, sumac witches'-broom disease, paulonia witches'- broom disease, and mulberry dwarf disease whether or not they were infected by phytoplasma, using universal and specific primers. Upon treatment of DNA amplified by PCR of phytoplasma with Alu I , Hpa II and Sat I restricted enzymes, distinction of phytoplasmas was possible. Particularly, phytoplasma of each host was distinguishable by treatment of Hpa II restricted enzyme. Meanwhile, analysis of restricted enzymes of jujube witches'-broom disease showed a higher infectivity of phytoplasmas of two origins. There were a lot of relations between jujube witches'-broom disease and sumac witches'-broom disease, and between paulonia witches'-broom disease and mulberry dwarf disease.
Plants and microorganisms (microbes) use information from chemicals such as volatile compounds to understand their environments. Proficiency in sensing and responding to these infochemicals increases an organism's ecological competence and ability to survive in competitive environments, particularly with regard to plant-pathogen interactions. Plants and microbes acquired the ability to sense and respond to biogenic volatiles during their evolutionary history. However, these signals can only be interpreted by humans through the use of state-of the-art technologies. Newly-developed tools allow microbe-induced plant volatiles to be detected in a rapid, precise, and non-invasive manner to diagnose plant diseases. Beside disease diagnosis, volatile compounds may also be valuable in improving crop productivity in sustainable agriculture. Bacterial volatile compounds (BVCs) have potential for use as a novel plant growth stimulant or as improver of fertilizer efficiency. BVCs can also elicit plant innate immunity against insect pests and microbial pathogens. Research is needed to expand our knowledge of BVCs and to produce BVC-based formulations that can be used practically in the field. Formulation possibilities include encapsulation and sol-gel matrices, which can be used in attract and kill formulations, chemigation, and seed priming. Exploitation of biogenic volatiles will facilitate the development of smart integrated plant management systems for disease control and productivity improvement.
An equine herpesvirus-4 (EHV-4) was isolated in nasal swabs collected in a horse showing respiratory clinical signs. Equine dermis cells inoculated with the sample were observed with characteristic viral cytopathic effects after 3 days of postinoculation and the infected cells exhibited bright intracelluar fluorescence by indirect immunofluorescence assay. At the nucleotide level, the partial glycoprotein B gene of the Korean EHV-4 isolate (K001) had 99.9% identity to 1942 strain (GenBank No. M26171). To author's knowledge, the report describes the first isolation and partial characterization of EHV-4 in Korea. The virus can be used for further study of EHV-4.

On a farm with 6 Korean beef cattle, an adult cow and a calf suddenly died while eating. At that time, hair was pressed upon the stanchion facilities where the adult cow was fed. Two dead cattle were submitted to the Animal and Plant Quarantine Agency for disease diagnosis. Grossly, the mouth of the adult cow was full of feed that had been eaten before death, and there was a foamy fluid in the calf's trachea. Both hemorrhages were observed in the muscles, as well as in the internal organs such as the liver, lungs, and heart. Ruminal mucosa was easily peeled off. Histopathologically, both muscles and internal organs showed severe hemorrhages. In particular, affected muscles were shown segmental and band-like patterns of myonecrosis, disappeared some myocytes, burned hair follicles, and irregular surface of skin. Severe hemorrhages of muscles and internal organs were consistent with previous reports. Easily separated mucosa of rumen and muscle lesions were very characteristic. We determined electrocution according to the pathological findings, laboratory results, and traces of accidents. This report will help to understand of pathological lesions of electrocution. Furthermore, this case suggests that the electrical wires and devices must be installed and handled carefully during the construction of the cowsheds care.
Canine parvovirus (CPV) is a major diarrhea-causing agent in puppies. Since CPV type 2 (CPV-2) emerged in 1978, new antigenic variants including CPV-2a, CPV-2b, and CPV-2c have been identified in many countries. Two puppies died suddenly at a veterinary clinic in Gyeonggi province, South Korea. Two viruses were isolated in A72 cells, confirmed as CPV strains based on a CPV rapid kit and an indirect fluorescence test and designated QIACP1403 and QIACP1404. The nucleotide sequences of complete VP2 genes of QIACP1403 and QIACP1404 were determined, and the corresponding amino acid sequences were deduced. Molecular analyses revealed that the QIACP1403 and QIACP1404 isolates were type CPV-2b. Several mutated amino acids were detected on VP2 gene residues of the two isolates. Phylogenetic analyses showed that the two isolates were most closely related to strain CPV-BM11, which was isolated from Chinese dogs in 2011. Our results suggest that these isolates may be a candidate for a vaccine to prevent CPV infection in dogs after conducting passages of the isolates in an in vitro culture system.
