식물체의 면역반응 기작 (Molecular Mechanism of Plant Immune Response)

  • Disease resistance in plants is often controlled by gene-for-gene mechanism in which avirulence (avr) gene products encoding by pathogens are specifically recognized, either directly or indirectly by plant disease resistance (R) gene products and sequential signal transduction pathways activating defense responses are rapidly triggered. As a results, not only exhibit a resistance against invading pathogens but also plants maintain the systemic acquired resistance (SAR) to various other pathogens. This molecular interaction between pathogen and plant is commonly compared to innate immune system of animal. Recent studies arising from molecular characterization of a number of R genes from various plant species that confer resistance to different pathogens and corresponding avr genes from various pathogens resulted in the accumulation of a wealth of knowledge on molecular mechanism of gene-for-gene interaction. Furthermore, new technologies of genomics and proteomics make it possible to monitor the genome-wide gene regulation and protein modification during activation of disease resistance, expanding our ability to understand the plant immune response and develop new crops resistant to biotic stress.

A Novel Protein Elicitor PeBL2, from Brevibacillus laterosporus A60, Induces Systemic Resistance against Botrytis cinerea in Tobacco Plant

  • Here, we reported a novel secreted protein elicitor PeBL2 from Brevibacillus laterosporus A60, which can induce hypersensitive response in tobacco (Nicotiana benthamiana). The ion-exchange chromatography, high-performance liquid chromatography (HPLC) and mass spectrometry were performed for identification of protein elicitor. The 471 bp PeBL2 gene produces a 17.22 kDa protein with 156 amino acids containing an 84-residue signal peptide. Consistent with endogenous protein, the recombinant protein expressed in Escherichia coli induced the typical hypersensitive response (HR) and necrosis in tobacco leaves. Additionally, PeBL2 also triggered early defensive response of generation of reactive oxygen species ($H_2O_2$ and $O_2{^-}$) and systemic resistance against of B. cinerea. Our findings shed new light on a novel strategy for biocontrol using B. laterosporus A60.

  • The result of recent seismic hazard analysis indicates that the ground motion response spectra for Korean nuclear power plant site have relatively large high frequency acceleration contents. In the ordinary seismic fragility analysis of nuclear power plant structures and equipments, the safety margin of design ground response spectrum is directly used as a response spectrum shape factor. The effects of input response spectrum shape on the floor response spectrum were investigated by performing the direct generation of floor response spectrum from the ground response spectrum. The safety margin included in the design ground response spectrum should be considered as a floor response spectrum shape factor for the seismic fragility analysis of the equipments located in a building.

Characterizing of Rice Blast Lesion Mimic

  • When plants are infected by plant pathogens, typical disease symptom termed lesion, appears in compatible interaction. Whereas, in incompatible interactions, only small speck of lesions are visible on the leaf surfaces. Hypersensitive response (HR) of plant which is the result of infection by incompatible pathogens, is a well known defense response inducing rapid cell death resulting in complete resistance. However, some rice mutants show spontaneous disease symptoms during the growth stages without interaction with pathogens. We investigated the spontaneous cell death mutant called Blast Lesion Mimic(BLM) generated by EMS mutation, on the relationship with the hypersensitive response as well as resistant characteristics. Accumulation of phenolic compounds were detected around the lesions as lesions develop on leaf surface. Activation of PR gene was detected before the lesion appeared, and that result indicates the defense-related response are started earlier than lesion formation. The BLM mutant showed resistant response to inoculation of Magnaporthe grisea KJ201 with which the wild type Hwacheong is totally susceptible. Informations on the formation of spontaneous lesions and detail analysis of lesion mimic mutants and related genes are very limited to date. It is really important to understand the phenomenon of the defense-related lesion formation for developing resistant cultivar for rice blast pathogens

Development of Stress-tolerant Crop Plants

  • Adverse environmental conditions such as drought, high salt and cold/freezing are major factors that reduces crop productivity worldwide. According to a survey, 50-80% of the maximum potential yield is lost by these "environmental or abiotic stresses", which is approximately ten times higher than the loss by biotic stresses. Thus, improving stress-tolerance of crop plants is an important way to improve agricultural productivity, In order to develop such stress-tolerant crop plants, we set out to identify key stress signaling components that can be used to develop commercially viable crop varieties with enhanced stress tolerance. Our primary focus so far has been on the identification of transcription factors that regulate stress responsive gene expression, especially those involved in ABA-mediated stress response. Be sessile, plants have the unique capability to adapt themselves to the abiotic stresses. This adaptive capability is largely dependent on the plant hormone abscisic acid (ABA), whose level increases under various stress conditions, triggering adaptive response. Central to the response is ABA-regulated gene expression, which ultimately leads to physiological changes at the whole plant level. Thus, once identified, it would be possible to enhance stress tolerance of crop plants by manipulating the expression of the factors that mediate ABA-dependent stress response. Here, we present our work on the isolation and functional characterization of the transcription factors.n factors.

  • This paper deals with design method of the model following servo system in which optimal regulator probelm is used to design the controllers that make the step/ramp response of the plant be keptclose to a specified ideal step/ramp response of the model. The characteristics of this system is robust in the presence of the specified disturbances or the partameter perturbations of the plant. Especially, by direct feedforward compensator from the reference input the steady state offset of plant output response is excluded and the transient response is improved. Examples are give and the results of the design of the model follwing servo systems are verified by the computer simulation.

월성원전 구조물의 지진응답 특성 분석 (Analysis of Seismic Response Characteristics for Wolsong Nuclear Power Plant Structures)

  • The purpose of this study is to evaluate the seismic response characteristics of Wolsong nuclear power plant (NPP) structures for the Kyeongju earthquake(ML=4.3) occurred on June 26, 1997. The seismograms are obtained from five accelerographs of nuclear power plant at Wolsong, Kyeongbuk. The distance from the epicenter is about 25km. The peak acceleration (PA) due to the earthquake is 0.0235g, which is far lower value than that of design basis earthquake(DBE). The PA at the containment wall is about twice as large as that at free field. Also, the higher the accelerograph is located in, the larger the PA is measured to be From the response spectrum analysis, the dominant frequency of the response is close to 4 Hz, which is similar to the free field is poor because of contamination by high frequency waves as a result of reflection and diffraction between ground and NPP structure. We are of opinion that the accelerograph at the free field should be moved approximately twice the building dimension away from the containment structure.

Oomycetes RXLR Effectors Function as Both Activator and Suppressor of Plant Immunity

  • Plant pathogenic oomycetes, such as Phytophthora spp., are the causal agent of the most devastating plant diseases. During infection, these pathogens accomplish parasitic colonization of plants by modulating host defenses through an array of disease effector proteins. These effectors are classified in two classes based on their target sites in the host plant. Apoplastic effectors are secreted into the plant extracellular space, and cytoplasmic effectors are translocated inside the plant cell, through the haustoria that enter inside living host cell. Recent characterization of some oomycete Avr genes showed that they encode effector protein with general modular structure including N-terminal conserved RXLR-DEER motif. More detailed evidences suggest that these AVR effectors are secreted by the pathogenic oomycetes and then translocated into the host plant cell during infection. Recent findings indicated that one of the P. infestans effector, Avrblb2, specifically induces hypersensitive response (HR) in the presence of Solanum bulbocastanum late blight resistance genes Rpi-blb2. On the other hand, another secreted RXLR protein PexRD8 originated from P. infestans suppressed the HCD triggered by the elicitin INF1. In this review, we described recent progress in characterized RXLR effectors in Phytophthora spp. and their dual functions as modulators of host plant immunity.

Functional Conservation and Divergence of FVE Genes that Control Flowering Time and Cold Response in Rice and Arabidopsis

  • Recent molecular and genetic studies in rice, a short-day plant, have elucidated both conservation and divergence of photoperiod pathway genes and their regulators. However, the biological roles of rice genes that act within the autonomous pathway are still largely unknown. In order to better understand the function of the autonomous pathway genes in rice, we conducted molecular genetic analyses of OsFVE, a rice gene homologous to Arabidopsis FVE. OsFVE was found to be ubiquitously expressed in vegetative and reproductive organs. Overexpression of OsFVE could rescue the flowering time phenotype of the Arabidopsis fve mutants by up-regulating expression of the SUPPRESSOR OF OVEREXPRESSION OF CO1 (SOC1) and down-regulating FLOWERING LOCUS C (FLC) expression. These results suggest that there may be a conserved function between OsFVE and FVE in the control of flowering time. However, OsFVE overexpression in the fve mutants did not rescue the flowering time phenotype in in relation to the response to intermittent cold treatment.
