Altered Gene Expression and Intracellular Changes of the Viable But Nonculturable State in Ralstonia solanacearum by Copper Treatment

  • Environmental stresses induce several plant pathogenic bacteria into a viable but nonculturable (VBNC) state, but the basis for VBNC is largely uncharacterized. We investigated the physiology and morphology of the copper-induced VBNC state in the plant pathogen Ralstonia solanacearum in liquid microcosm. Supplementation of $200{\mu}M$ copper sulfate to the liquid microcosm completely suppressed bacterial colony formation on culture media; however, LIVE/DEAD BacLight bacterial viability staining showed that the bacterial cells maintained viability, and that the viable cells contain higher level of DNA. Based on electron microscopic observations, the bacterial cells in the VBNC state were unchanged in size, but heavily aggregated and surrounded by an unknown extracellular material. Cellular ribosome contents, however, were less, resulting in a reduction of the total RNA in VBNC cells. Proteome comparison and reverse transcription PCR analysis showed that the Dps protein production was up-regulated at the transcriptional level and that 2 catalases/peroxidases were present at lower level in VBNC cells. Cell aggregation and elevated levels of Dps protein are typical oxidative stress responses. $H_2O_2$ levels also increased in VBNC cells, which could result if catalase/peroxidase levels are reduced. Some of phenotypic changes in VBNC cells of R. solanacearum could be an oxidative stress response due to $H_2O_2$ accumulation. This report is the first of the distinct phenotypic changes in cells of R. solanacearum in the VBNC state.

Triterpenoids from the Fruits of Cornus kousa Burg. as Human Acyl-CoA: Cholesterol Acyltransferase Inhibitors

  • The fruits of Cornus kousa Burg. were extracted with 80% aqueous methanol (MeOH) and the concentrated extract was partitioned with ethyl acetate (EtOAc), n-butanol (n-BuOH), and $H_2O$. From the EtOAc traction, 5 triterpenoids were isolated through repeated silica gel ($SiO_2$), octadecyl silica gel (ODS), and Sephadex LH-20 column chromatography (c.c.). These compounds were determined to be ursolic acid (1), corosolic acid (2), taraxasterol (3), betulinic acid (4), and betulinic aldehyde (5) on the basis of their spectroscopic data including electronic ionization mass spectrometry, ultraviolet spectroscopy, infrared spectroscopy, and nuclear magnetic resonance. This is the first reported isolation of these compounds from this plant. Also, compounds 1, 3, 4, and 5 show a relatively high inhibitory activity against human acyl-CoA: cholesterol acyltransferase-1 (hACAT-1) with inhibition values of $52.8{\pm}0.7$, $91.1{\pm}0.4$, $93.0{\pm}0.7$, and $96.2{\pm}0.2%$ at a concentration of $100{\mu}M$, respectively.

  • The flower of Campsis grandiflora K. Schum. Was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and H$_2$O. From the EtO Ac fraction, seven triterpenoids were isolated through the repeated silica gel, ODS column chromatographies and preparative HPLC. From the result of physico- chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as 3${\beta}$-hydroxyolean-12-en-28-oic acid (oleanolic acid, 1), 3${\beta}$-hydroxyurs-12-en-28-oic acid (ursolic acid, 2), 3${\beta}$-hydroxyurs-12-en-28-al (ursolic aldehyde, 3), 2${\alpha}$,3${\beta}$-dihydroxyolean-12-en-28-oic acid (maslinic acid, 4), 2${\alpha}$,3${\beta}$-dihydroxyurs-12-en-28-oic acid (corosolic acid, 5), 3${\beta}$,23-dihydroxyurs-12- en-28-oic acid (23-hydroxyursolic acid ,6) and 2${\alpha}$,3${\beta}$,23-trihydroxyolean-12-en-28- oic acid (arjunolic acid, 7). These teriterpenoids were isolated for the first time from this plant. Also, compounds 4, 5, 6, and 7 revealed relatively high hACAT-1 inhibitory activity with the value of 46.2${\pm}$1.1, 46.7${\pm}$0.9, 41.5${\pm}$1.3 and 60.8${\pm}$1.1% at the concentration of 100${\mu}$g/mL, respectively.

  • This study has been made to find out the possibilty of successfully using the following $K_2O$ recommended equation $K_2O\;kg/10a=(Ko/\sqrt{Ca+Mg}-Ks/\sqrt{Ca+Mg})sqrt{Ca+Mg}.\;47.\;B\;D$. where $Ko/sqrt{Ca+Mg}=0.03518+0.0007658\;Sio_2/O.M$. $K_Ssqrt{Ca+Mg}$=Exchangeable K me/100g/$\sqrt{Total\;soluble(Ca+Mg)me/100g\;in\;Soil}$ B. D. =Bulk density of top soil, when the dose of Nitrogen for rice is estimated from the following equation: $N\;kg/10a=(4.2+0.096\;SiO_2/O.M).F$ where $F=0.907+0.263x-0.013x^2$ $SiO_2/O.M=(available\;SiO_2=ppm)/(organic\;matter\;%)$in soil For this. two field experiments. one in sandy and the other in clay paddy soil. have been conducted using 3 levels of wollastonite (0, 500, 100kg/10a) as main treatments; 3 levels of $K_2O$ application were used as sub-plots. These were as follows : (1) 8kg of $K_2O$/10a regardless of the K activity-$K/\sqrt{Ca+Mg}$; (2) kg of $K_2O$/10a estimated from the above equation. and (3) same as (2) above plus additional 30% of $K_2O$. The dose of N kg/ 10a was determined from the above equation based on the value of $SiO_2$/O.M. ratio in each treatment. There were three replications. The leading variety of rice in Chung Chong Nam Do area. Akibare (introduced from Japan) was used. The data obtained. through soil and plant analysis and growth and yield observations. have been throughly examined to attain the following summarized conclusions. 1. The nitrogen dose. estimated from the above equation. was in excess for optimum growth of the rice variety Akibare; indicating the necessity of modification onthe value of "F" or the constants in the equation. The concept of using $SiO_2$/O.M. in the equation was shown to be applicable. 2. The dose of potash. estimated from the respective equation given above. also was in excess of the rice requirements indicating the necessity of minor change in the estimation of $Ko/\sqrt{Ca+Mg}$ value and some great modification in the calculation of $Ks/\sqrt{Ca+Mg}$ value for the equation; however the concept of using $K/\sqrt{Ca+Mg}$ as a basis of $K_2O$ recommendation was shown to be quite reasonable. 3. It was found. from the correlation study using the data of paddy yield and amount of $K_2O$ absorbed by rice plants that the substitution of the value of $Ks/\sqrt{Ca+Mg}$ in the equation for the vaule $Ks/\sqrt{Ca+Mg}=0.037+0.78K\;me/100g$ soil was much more applicable than using the value calculated from the data of soil and wollastonite analysis.

Microstructure of Glass-ceramics Made from Bottom Ash Produced at a Thermal Power Plant (화력발전소 바닥재로 제조된 결정화 유리의 미세구조)

  • Glass ceramics were made from coal bottom ash by adding CaO and $Li_2O$ as glass modifiers and $TiO_2$ as a nucleating agent in a process of melting and quenching followed by a thermal treatment. The surface of the glass ceramics has 1.6 times more $Li_2O$ compared to the inner matrix. When $TiO_2$ was not added or when only 2 wt% was added, the surface parts of the glass ceramics were crystalline with a thickness close to $130{\mu}m$. In addition, the matrixes showed only the glass phase and not the crystalline phase. However, doping of $TiO_2$ from 4 wt% to 10 wt% began to create small crystalline phases in the matrix with an increase in the quantity of the crystalline. The matrix microstructure of glass ceramics containing $TiO_2$ in excess of 8 wt% was a mixture of dark-gray crystalline and white crystalline parts. These two parts had no considerable difference in terms of composition. It was thought that the crystallization mechanism affects the crystal growth, direction and shape and rather than the existence of two types of crystals.

Designing Modified Atmosphere Packaging for Persimmon (Diospyros kaki cv. Fuyu) Fruit Based on Respiration Modelling (단감의 최적 Modified Atmosphere포장 규격 설정)

  • A respiration rate analysed by enzyme kinetics-based respiration model and gas permeability data of LDPE film were applied to design the optical modified atmosphere (MA) packaging condition of persimmon (Diospyros kaki cv. Fuyu) fruits. The fruit quality rapidly decreases due to physiology disorder such as softening and peel blackening. $O_2$ permeance ($Q_{O2}$ in $ml{\cdot}hr^{-1}{\cdot}atm^{-1}{\cdot}m^{-2}$) and $CO_2$ performance ($Q_{CO2}$ in $ml{\cdot}hr^{-1}{\cdot}atm^{-1}{\cdot}m^{-2}$) of low density polyethylene (LDPE) film samples were measured at $0^{\circ}C$ and described as function of thickness (L in ${\mu}m$) as $Q_{O2}=(2540{\times}1/L)-16$, and $Q_{CO2}=(13742{\times}1/L)-70$, respectively. MA package containing single persimmon fruit of 225g was designed and tested experimentally at $0^{\circ}C$ by using LDPE films. Package atmospheres predicted from the relationship of $O_2$, $CO_2$ and $N_2$ balances on the packages was in good agreement with those obtained experimentally. Physiology disorder occurrence was the lowest at 52 ${\mu}m$ package that attained optimum gas condition ($O_2$ 2.8% and $CO_2$ 5.4%). The computer simulation was found to be effective to help to design the optimum MA packaging condition of individual persimmon fruit.

Growth and Yield Response of Chinese Cabbage and Radish on Application of Potassium Chloride Fertilizer (염화칼리 시용에 따른 배추와 무의 생육과 수량)

  • The application of potassium above the optimum level may cause the inhibition of plant growth, fertilizer loss, and environmental pollution. Therefore, application rate of K fertilizer should be recommended on the basis of soil test. In order to determine critical K content in soils causing growth inhibition of vegetables, $1m^2-pot$ experiments with Chinese cabbage and radish were accomplished with various K-application rates. The threshold concentrations of exchangeable potassium causing the inhibition of plant growth were $0.96cmol_c\;kg^{-1}$ for Chinese cabbage in spring, and $1.28cmol_c\;kg^{-1}$ for radish in autumn. Above those concentration levels, the yields of them were decreased with the increase of potassium levels in soils. Germination rate of Chinese cabbage in spring decreased with increase of the electrical conductivity (EC) of soils due to application of potassium fertilizer. In the harvesting stage, the potassium contents of plant were increased with the increase of K application rate while plant uptake of nutrients was decreased at the K adjustment level of over $2.0cmol_c\;kg^{-1}$.

Effects of Magnetite(Fe3O4) as Electrical Conductor of Direct Interspecies Electron Transfer on Methane Yield of Food Wastewater (종간직접전자전달 전도체로서 Magnetite(Fe3O4)가 음폐수의 메탄생산에 미치는 영향)

  • Methane production by anaerobic digestion occurs through interspecies electron transfer (DIET), a synthetic metabolism between acetic and methanate bacteria through hydrolysis and acid production steps. In this study, to improve methane yield, the effect of addition of magnetite (Fe3O4), a conductor promoting DIET on methane production in food wastewater was investigated, and the effect on methane yield was assessed by methane potential (Bu) and maximum methane production rate [Rm(t0)] by the operation of batch type anaerobic reactor adding Fe3O4. The Bu and Rm(t0) of food wastewater without Fe3O4 were 0.496 Nm3/kg-VSadded and 38.24 mL/day, respectively. The t0 which reached to Rm appeared at 21.06 days during the operation of the anaerobic reactor. The Bu of food wastewater with Fe3O4 was 0.502, 0.498, 0.512, 0.510, 0.518, 0.523, 0.524, 0.540, and 0.549 Nm3/kg-VSadded in the treatment of 5, 10, 15, 20, 25, 30, 40, 70, and 100mM-Fe3O4, respectively, and the Bu significantly increased to 36.95% with the addition of magnetite in the addition of 15mM-Fe3O4. And, the addition of Fe3O4 shortened the duration to reach Rm from 21.06 days to the maximum of 14.67 days by the addition of Fe3O4. Therefore, the methane yield and production rate of food wastewater significantly improved with the addition of Fe3O4.

Investigation of Pellet-Clad Mechanical Interaction in Failed Spent PWR Fuel

  • A failed spent fuel rod with 53,000 MWd/tU from a nuclear power plant was characterized, and the fission products and oxygen layer in the pellet-clad mechanical interaction region were observed using an EPMA (Electron Probe Micro-Analyzer). A sound fuel rod burned under similar conditions was used to compare and analyze, the results of the failed fuel rod. In the failed fuel rod, the oxide layer represented $10{\mu}m$ of the boundary of the cladding, and $35{\mu}m$ of the region outside the cladding. By comparison, in the sound fuel rod, the oxide layer was $8{\mu}m$, observed in the cladding boundary region. The cladding inner surface corrosion and the resulting fuel-cladding bonding were investigated using an EPMA. Zirconium existed in the bonding layer of the (U, Zr)O compound beyond the pellet cladding interaction gap of $20{\mu}m$, and composition of UZr2O3 was observed in the failed fuel rod. This paper presents the results of the EPMA examination of a spent fuel specimen, and a technique to analyze fission products in the pellet-clad mechanical interaction region.

Isolation and Characterization of an Acyclic Isoprenoid from Semecarpus anacardium Linn. and its Antibacterial Potential in vitro - Antimicrobial Activity of Semecarpus anacardium Linn. Seeds -

  • Objectives: Semecarpus anacardium Linn. is a plant well-known for its antimicrobial, antidiabetic and anti-arthritic properties in the Ayurvedic and Siddha system of medicine. This has prompted the screening of this plant for antibacterial activity. The main aims of this study were to isolate compounds from the plant's seeds and to evaluate their antibacterial effects on clinical bacterial test strains. Methods: The n-butanolic concentrate of the seed extract was subjected to thin layer chromatography (TLC) and repeated silica gel column chromatography followed by elution with various solvents. The compound was identified based on observed spectral (IR, $^1H$ NMR, $^{13}C$ NMR and high-resolution mass spectrometry) data. The well diffusion method was employed to evaluate the antibacterial activities of the isolated acyclic isoprenoid compound (final concentration: $5-15{\mu}g/mL$) on four test bacterial strains, namely, Staphylococcus aureus (MTCC 96), Bacillus cereus (MTCC 430), Escherichia coli (MTCC 1689) and Acinetobacter baumannii (MTCC 9829). Results: Extensive spectroscopic studies showed the structure of the isolated compound to be an acyclic isoprenoid ($C_{21}H_{32}O$). Moreover, the isoprenoid showed a remarkable inhibition of bacterial growth at a concentration of $15{\mu}g/mL$ compared to the two other doses tested (5 and $10{\mu}g/mL$) and to tetracycline, a commercially available antibiotic that was used as a reference drug. Conclusion: The isolation of an antimicrobial compound from Semecarpus anacardium seeds validates the use of this plant in the treatment of infections. The isolated compound found to be active in this study could be useful for the development of new antimicrobial drugs.
