Ascorbic acid (AA) is a multifunctional metabolite in plants that is essential for plant development and growth. We examined the effect of AA, an antioxidant, on the gravitropic response of primary roots in maize. The application of $10^{-3}$ M AA to the elongation zone did not affect the gravitropic response and slightly inhibited the root growth. However, treatment with both $10^{-5}$ M and $10^{-3}$ M AA at the root tip increased the gravitropic response and inhibited root growth. Differences in indole-3- acetic acid (IAA) activity between the upper and lower hemispheres of the root resulted in differential elongation along the horizontal root. Roots are extremely sensitive to IAA, and increasing the amount of IAA in the lower hemisphere of the root inhibited elongation. Therefore, we examined the effect of IAA in the presence of AA. The inhibitory effect of AA on the gravitropic response was greater in combination with IAA. To understand the role of AA in the regulation of root growth and the gravitropic response, we measured ethylene production in the presence of AA in the primary roots of maize. AA stimulated ethylene production via the activation of the 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase gene, which regulates the conversion of ACC to ethylene. These results suggest that AA alters the gravitropic response of maize roots through modification of the action of ethylene.
An actinomycetes isolate G-0299 obtained from a forest soil showed strong phytotoxic activity to Digitaria ciliaris. For the foliar application study, the culture filtrate of the isolate G-0299 showed strong herbicidal activity only to D. ciliaris among the 12 monocot and 5 dicot weed species. And herbicidal activity at a concentration of 500, 250, 125 and $62.5{\mu}gmL^{-1}$ of culture filtrate was 100%, 98%, 70% and 40%, respectively. Phytotoxic symptoms of the culture filtrate by foliar application were desiccation and burn-down or bleaching of leaves and finally plant death. And then the herbicidal activity was exhibited only under the light condition. Also, chlorophyll loss of D. ciliaris leaf tissues in the light condition was much higher than in the dark condition and then chlorophyll content decreased 82%, 5%, respectively. In conclusion, our results suggest that soil bacteria, isolate G-0299 could be a good candidate for new bio-herbicide and provide a new lead molecule for a more unique herbicide.
Developmental micropropagation method and somatic embryogenesis for hybrid poplars, Populns ehrarnericana Eco28, P. nigra ${\times}$ P. moximowiczii 62-9, were established using nodule culture system. Calli of Eco28 and 62-9 clone were initiated from leaf explant on the medium with 0.5mg/l and 2.0mg/l 2, 4-D, respectively. Cell suspension culture was established from callus derived from leaf explant culture. When suspended on MS medium with optimal combination of BA and NAA fine nodules were obtained after 2 weeks of culture. For shoot regeneration, nodules were transferred into liquid and agar solidified medium. Numerous shoots were regenerated from nodules of 62-9 on liquid media. Organogenesis was effectively achieved on agar solidified regeneration media containing different concentrations of BA and adenine sulfate. Average numbers of 27 and 24 shoots per nodule were induced from 62-1 and Eco28 clones after 8 weeks of culture, respectively. In addition, somatic embryogenesis also occurred in the same regeneration medium. This procedure can be applied to vegetative propagation, utilization of somaclonal variation, production of secondary metabolite and materials of biotechnology research.
The influenza neuraminidase (NA, E.C. 3.2.1.18), an antiviral, has been the target of high pharmaceutical companies due to its essential role in viral replication cycle. Perilla frutescens (P. frutescens) is used in traditional Chinese medicine for various diseases, such as cold due to wind-cold, headache and cough. In this context, four major polyphenolic compounds including rosmarinic acid-3-O-glucoside (1), rosmarinic acid (2), luteolin (3), and apigenin (4) isolated from P. frutescens were evaluated for their inhibitory effect on recombinant virus H1N1 neuraminidase (rvH1N1 NA). Among the test compounds, rosmarinic acid and luteolin inhibited the rvH1N1 NA with an $IC_{50}$ of 46.7 and $8.4{\mu}M$, respectively. The inhibition kinetics analyzed by the Dixon plots indicated that rosmarinic acid and luteolin were noncompetitive inhibitors and that the inhibition constant, $K_I$, was established as 43.9 and $14.3{\mu}M$, respectively. In addition, 578 genetically diverse accessions and 39 cultivars of P. frutescens were analyzed using HPLC to characterize the diversity of polyphenolic composition and concentration. The individual and total compositions exhibited significant difference (P < 0.05), especially rosmarinic acid which was detected as the predominant metabolite in all accessions (58.8%) and cultivars (62.8%). Yeupsil and Sangback cultivars exhibited the highest rosmarinic acid ($3,393.5{\mu}g/g$) and luteolin ($383.3{\mu}g/g$) content respectively. YCPL177-2 with the high concentration ($889.8{\mu}g/g$) of luteolin may be used as a genetic resource for breeding elite cultivars.
The ubiquitous plant metabolite p-coumaric acid (p-CA) has antioxidant and anti-inflammatory properties, but its anti-cancer activity has not been established in gastric cancer cell lines. In this study, we investigated the effects of p-CA on the proliferation and transcriptome profile of SNU16 gastric cancer cells. Treatment with p-CA induced apoptosis of the SNU-16 cells by regulating the expression of pro-apoptotic and anti-apoptotic proteins, such as Bcl-2, poly (ADP-ribose) polymerase (PARP), Bax, procaspase-3, and cleaved-caspase-3. The genes differentially expressed in response to p-CA treatment of the SNU-16 cells were identified by RNA sequencing analysis. Genes regulated by p-CA were involved mainly in the inflammatory response, apoptotic processes, cell cycle, and immune response. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that the phosphatidylinositol-3-kinase-Akt and cancer signaling pathways were altered by p-CA. Protein-protein interaction (PPI) network analysis also revealed that p-CA treatment was correlated with differential expression of genes associated with the inflammatory response and cancer. Collectively, these results suggest that p-CA has potential utility in gastric cancer prevention.
Objective: To improve the feeding value of rice straw (RS), this study evaluated the potential of rice straw fermented with Pleurotus sajor-caju (FRS) as dairy goat feed. Methods: Spent Pleurotus sajor-caju mushroom substrate was used as fungi inoculum to break the lignocellulose linkage of rice straw, which was solid-fermented at 25℃ to 30℃ for 8 weeks. The ruminal degradation of pangolagrass hay (PG), FRS, and RS were measured in situ for 96 hours in three dry Holstein cows, respectively. Effect of fungi fermented RS on milking performance was studied in feeding trials. A total of 21 Alpine goats a trial were divided into 3 groups: a control group in which PG accounted for 15% of the diet dry matter, and FRS or RS was used to replace the PG in the control group. Goats were fed twice a day under two 28-day trial in individual pens. Meanwhile, a 3×3 Latin square trial (14 days/period) was conducted to study the rumen digestion of three diets by using three fistulated dry goats. Rumen contents were collected for metabolite analyses every one to three hours on the last two days. Results: In situ study showed that fermentation could elevate the rumen degradable fraction and effective degradability of RS (p<0.05). Effective degradability of FRS dry matter was significantly increased from 29.5% of RS to 41.7%. Lactating trial results showed that dry matter intake and milk yield in the PG group and FRS group were similar and higher than those in RS group (p<0.05). The concentration of propionic acid and total volatile fatty acid in the RS group tended to be lower than those in PG group (p<0.10). There were no differences in rumen pH value and ammonia nitrogen level among the groups tested. Conclusion: Fermentation of rice straw by spent Pleurotus sajor-caju mushroom substrate could substantially enhance its feeding value to be equivalent to PG as an effective fiber source for dairy goat. The fermented rice straw is recommended to account for 15% in diet dry matter.

Over the past 100 years, the global average temperature has risen by 0.75 ℃. The Korean Peninsula has risen by 1.8 ℃, more than twice the global average. According to the RCP 8.5 scenario, the CO2 concentration in 2100 will be 940 ppm, about twice as high as current. The National Institute of Crop Science(NICS) is using the SPAR (Soil-Plant Atmosphere Research) facility that can precisely control the environment, such as temperature, humidity, and CO2. A Python-based colony photosynthesis algorithm has been developed, and the carbon and nitrogen absorption rate of rice is evaluated by setting climate change conditions. In this experiment, Oryza Sativa cv. Shindongjin were planted at the SPAR facility on June 10 and cultivated according to the standard cultivation method. The temperature and CO2 settings are high temperature and high CO2 (current temperature+4.7℃ temperature+4.7℃·CO2 800ppm), high temperature single condition (current temperature+4.7℃·CO2 400ppm) according to the RCP8.5 scenario, Current climate is set as (current temperature·CO2400ppm). For colony photosynthesis measurement, a LI-820 CO2 sensor was installed in each chamber for setting the CO2 concentration and for measuring photosynthesis, respectively. The colony photosynthetic rate in the booting stage was greatest in a high temperature and CO2 environment, and the higher the nitrogen fertilization level, the higher the colony photosynthetic rate tends to be. The amount of photosynthesis tended to decrease under high temperature. In the high temperature and high CO2 environment, seed yields, the number of an ear, and 1000 seed weights tended to decrease compared to the current climate. The number of an ear also decreased under the high temperature. But yield tended to increase a little bit under the high temperature and high CO2 condition than under the high temperature. In addition, In addition to this study, it seems necessary to comprehensively consider the relationship between colony photosynthetic ability, metabolite reaction, and rice yield according to climate change.

Pyrrolnitrin, pyrrolomycin, and pyoluteorin are functional halogenated phenylpyrrole derivatives (HPDs) derived from microorganisms with diverse antimicrobial activities. Pyrrolnitrin is a secondary metabolite produced from L-tryptophan through four-step reactions in Pseudomonas fluorescens, Burkholderia cepacia, Serratia plymuthica, etc. It is currently used for the treatment of superficial dermatophytic fungal infections, has high antagonistic activities against soil-borne and foliar fungal infections, and has many industrial applications. Since pyrrolnitrin is easily decomposed by light, it is difficult to widely use it outdoors. As an alternative, fludioxonil, a synthetically produced non-systemic surface fungicide that is structurally similar and has excellent light stability, has been commercialized for seed and foliar treatment of plants. However, due to its high toxicity to aquatic organisms and adverse effects in human cell lines, many countries have established maximum residue levels and strictly control its levels. Pyrrolomycin and pyoluteorin, which have antibiotic/antibiofilm activity against Gram-positive bacteria and high anti-oomycete activity against the plant pathogen Pythium ultimum, respectively, were isolated and identified from microorganisms. This review summarizes the biosynthesis and production of natural pyrrolnitrin derived from bacteria and the characteristics of synthetic fludioxonil and other natural phenylpyrrole derivatives among the HPDs. We expect that a plethora of highly effective, novel HPDs that are safe for humans and environments will be developed through the generation of an HPD library by microbial biosynthesis and chemical synthesis.

Schisandra chinensis (S. chinensis) is a deciduous broad-leaved cave plant belonging to the Schisandraceae family and is widely distributed in East Asia including Korea, Japan, China, and Taiwan. It has been reported that the main components contained in S. chinensis include lignan compounds and triterpenoid compounds. To distinguish the characteristics of S. chinensis by production region of Korea, a discriminant was established by performing metabolite profiling and principal component analysis, a multivariate statistical analysis technique. As a result, 16 types of triterpenoids, 9 types of lignan, and 1 type each of flavonoid, phenylpropanoid, and fatty acid were identified. In addition, through multivariate statistical analysis, it was confirmed that the four groups in Danyang, Moongyeong, Geochang, and Pyeongchang were divided, by applying the s-plot model of orthogonal partial least squares discriminant analysis. Biomarkers were identified: lanostane, cycloartane, schiartane triterpenoid, and dibenzocyclo-octadiene lignan were identified as chemical markers, respectively.
This study aims to establish a system for the rapid discrimination of Zoysia species using metabolite fingerprinting of FT-IR spectroscopy combined with multivariate analysis. Whole cell extracts from leaves of 19 identified Zoysia japonica, 6 identified Zoysia sinica, and 38 different unidentified Zoysia species were subjected to Fourier transform infrared spectroscopy (FT-IR). PCA (principle component analysis) and PLS-DA (partial least square discriminant analysis) from FT-IR spectral data successfully divided the 25 identified turf grasses into two groups, representing good agreement with species identification using molecular markers. PC (principal component) loading values show that the $1,100{\sim}950cm^{-1}$ region of the FT-IR spectra are important for the discrimination of Zoysia species. A dendrogram based on hierarchical clustering analysis (HCA) from the PCA and PLS-DA data of turf grasses showed that turf grass samples were divided into Zoysia japonica and Zoysia sinica in a species-dependent manner. PCA and PLS-DA from FT-IR spectral data of Zoysia species identified and unidentified by molecular markers successfully divided the 49 turf grasses into Z. japonica and Z. sinica. In particular, PLS-DA and the HCA dendrogram could mostly discriminate the 47 Z. japonica grasses into two groups depending on their origins (mountainous areas and island area). Considering these results, we suggest that FT-IR fingerprinting combined with multivariate analysis could be applied to discriminate between Zoysia species as well as their geographical origins of various Zoysia species.
