Callus Induction and Plant Regeneration form seeds of Zoysia japonica Steud. (들잔디 종자로부터 캘러스 유도 및 식물체 재분화)

  • conditions for callus induction and plant regeneratin from seeds of lawngrass (Zoysia japonica Steud.) were confirmed in this study. MS (Murashige and Skoog) medium containg 2,4-D 3 or 5mg/l was used for callus induction, and MS medium with different volumes of BA, NAA and kinetin hormones was used to regenerate the plants from induced calli. MS basic medium containing agar with no hormones or kinetin 1.0mg/l and MS basic medium containing gelite and NAA 1.0mg/l were higher for green callus induction. MS medium containing agar and kinetin 1.0mg/l ws highest degree of efficiency for plant regeneration.

Chromosome Aberration in Suspension Culture of Ginseng(Panax ginseng C. A. Meyer) Callus (인삼 캘러스 현탁배양에 있어서의 염색체 이상)

  • This study was to examine the variations of chromosome number and the ranges of variety in the suspension culture of ginseng (Panax ginseng C. A. Meyer) callus cell, and the effect of plant hormones for the chromosome aberration. Plant hormones added with MS medium in the suspension culture were 2,4-D, kinetin, and 2,4-D+kinetin and concentration of the plant hormones were $1000{\mu}M$ and $0.1\;{\mu}M$ respectively. As a result of these experiment the following conclusion has been obtained. Media contained with 2,4-D+kinetin in $10{\mu}M$ concentration was very effective in the suspension culture result from 26.4% mitosis frequency, and found the various variation of chromosome number. Variety of chromosome number was diversed ($9\sim110$), espicially frequency of hypohaploid and hyperhaploid cells were very higher than hyperdiploid cells. In this experiments, it is suggested that $10{\mu}M$ 2,4-D+kinetin added with medium in the suspension culture of ginseng callus was effect in the variations of chromosome number.

Comprehensive Analysis of the Expression of Twenty-Seven β-1, 3-Glucanase Genes in Rice (Oryza sativa L.)

  • lant ${\beta}$-1, 3-glucanases are involved in plant defense and in development. Very little data are available on the expression of rice glucanases both in developmental tissues and under various stresses. In this study, we cloned and characterized twenty-seven rice ${\beta}$-1, 3-glucanases (OsGlu) from at total of 71 putative glucanases. The OsGlu genes were obtained by PCR from a cDNA library and were classified into seven groups (Group I to VII) according to their DNA or amino acid sequence homology. Analysis of the expression of the twenty-seven OsGlu genes by Northern blotting revealed that they were differentially expressed in different developmental tissues as well as in response to plant hormones, biotic stress, high salt etc. OsGlu11 and 27 in Group IV were clearly expressed only in stem and leaf and were also induced strongly by SA (5 mM), ABA ($200{\mu}M$), and M. grisea. OsGlu1, 10, 11, and 14 were induced earlier and to higher levels in incompatible M. grisea interaction than in compatible one. Taken together, our findings suggest that the twenty-seven rice OsGlu gene products play diverse roles not only in plant defense but also in hormonal responses and in development.

  • The fat deposition is an important factor affecting chicken meat quality, which is closely related to lipid metabolism of chickens. Therefore, it is important to regulate the lipid metabolism of chickens to improve the chicken meat quality. Plant extracts have special regulatory effects on animal's growth and health and have been widely used in chicken breeding. Some plant extracts have been reported to have functions of changing the fatty acid composition, reducing abdominal fat percentage, and enhancing the intramuscular fat content of chickens by improving the antioxidant capacity, regulating the expression of genes, enzymes, and signaling pathways related to lipid metabolism, modulating intestinal microbiota, affecting hormones level, and regulating DNA methylation. This paper reviewed the application and mechanism of plant extracts on regulating lipid metabolism of chickens to provide a reference for the further application of plant extracts in chicken breeding.

  • Ethylene gas classified as one of five major plant hormones plays an important role in various plant metabolism. The precise analysis of ethylene production of plants or plant parts is a valuable research procedure because knowledge of ethylene production facilitates measures of the physiological activity within the tissue. This paper describes procedures for analyzing ethylene from plant tissues by gas chromatography and discusses problems associated with extracting gas samples either by introducing a vacuum to plant samples or by using a hypodermic syringe. Introduced are a continuous flow system for efficient analysis and an automated system for sampling, analyzing, calculating and recording ethylene production data.

Effect of GA3 and BA on Plant Growth of Ranunculus Cultivars

  • Ranunculus asiaticus characterizes colorful and attractive flower shapes that are related with the ornamental value of bulbous plants. Improving ornamental value of bulbous flowers has been the general goal of floricultural market. Gibberellic acid ($GA_3$) and benzyladenine (BA) play an important role in growth and developmental processes in floriculture. Combinational treatments of these two hormones have been used in floriculture to improve flower quality. We assessed the effects of combined $GA_3$ and BA, as well as the individual effects of each hormone, on growth characteristics using soil drench application to eight R. asiaticus cultivars, 'Giallo Millepetali', 'Bianco Millepetali', 'Arancio Millepetali', 'Rosa SC', 'Arancio Pratolino', 'Giallo Pratolino', 'Bianco Pratolino', and 'Rosa Ch Pratolino'. $GA_3$ treatments increased plant height and first flower size of R. asiaticus cultivars. Moreover, about 5 to 9 days to flowering were averagely shortened by $GA_3$ treatments compared to controls. On the other hand, the opposites, including first flower size and days to flowering, were observed for cultivars treated with BA, compared with controls. Treatments of $GA_3$ + BA generally affected growth traits, such as plant height, flower size, and the timing of flowering on some R. asiaticus cultivars. In particular, about 5 to 6 days to flowering were reduced on average by Treatments of $GA_3$ + BA. Our results showed positive growth effects, including plant height, days to flowering, first flower height, number of flowers from the application of individual and combined hormones to R. asiaticus cultivars and demonstrate a role for these hormones in future bulbous floriculture.

Effect of Plant Hormones on Rooting of Pinus spp. Cutting (식물 Hormones 처리가 소나무류 삽목발근에 미치는 영향)

  • In order to find out the basic factors governing the rooting ability of pine cutting, the rooting of cuttings of Pinus thunbergii were examined under various conditions. A total of 5 different solutions of growth hormones made up of 3-indole butyric acid, naphthalene acetic acide and rootone were employed at three different periods of year, the middle of June, the end of September and the early of December. Cuttings prepared in length of 4cm gave better results than those of control ones. Cuttings taken from the 1-0 seedlings were superior to those from the 7-year-old trees. Seasonal variation, i.e., time of collecting cutting materials, played great important role on rooting. The majority of roots were originated from the callus lamp formed at the base of cut surface.
