한란 및 심비디움의 기내 증식에 미치는 생장조절물질의 효과 (Effects of Plant Growth Regulators on in vitro Propagation of Cymbidium kanran and Cymbidium hybrida)

  • 본 실험은 식물조직배양 기법을 통하여 한란과 심비디움의 효율적인 증식을 위한 식물 생장조절물질의 적정 처리방법을 구명하고자 수행하였다. 한란의 rhizome 형성은 무처리 및 10 ppm kinetin + 2 ppm NAA, 심비디움의 protocorm 형성은 무처리 및 10 ppm kinetin + 0.05 ppm NAA에서 아주 양호하였다. Callus 유도의 적정 식물 생장조절물질의 농도는 구명하지 못했다. 한란 rhizome으로부터의 shoot 분화에는 10 ppm BA + 2 ppm NAA, 5 ppm BA + 2ppm NAA, 심비디움 protocorm으로부터의 shoot 분화는 10 ppm Kinetin + 0.05 ppm NAA, 1 ppm Kinetin + 2 ppm NAA에서 비교적 효과적인 것으로 나타났다.

Effects of Plant Growth Regulators on in vitro Propagation of Echinosophora koreensis Nakai

  • To establish in vitro nodal culture conditions of Echinosophora koreensis Nakai, one of rare and endangered species famous for beautiful flowers in the Korean Peninsula, the influence of plant growth regulators (PGRs) on shooting and rooting from in vitro shoots was investigated. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the half-strength Driver and Kuniyuki's media in the range of 2.22 to 8.88 ${\mu}M $induced 2.5 to 2.7 shoots per axillary bud; and addition of 2.27 ${\mu}M $ thidiazuron (TDZ) produced 3.2 shoots, during 4 weeks of culture, while zeatin and isopentenyl adenine (2ip) were not effective on shoot multiplication as observed from several combination treatments of BA with other PGRs. Shoots established were smaller than 2 cm in length, in most of the treatments. while in BA 8.88 ${\mu}M $ treatment more than 30% of shoots were longer than 2 cm and shorter than 4 cm. In rooting, naphthalene acetic acid (NAA) from 5.37 to 21.48 ${\mu}M $ showed the rooting rate from 40.0 to 62.5%. Indole butyric acid (IBA) addition had little effect on rooting (<10%), although some roots in IBA-containing media were longer than those in NAA. Micropropagation from axillary buds of nodular explants was applicable and promising to multiplication and conservation of Echinosophora koreensis Nakai.

오미자 대량증식을 위한 삽목번식 효율증대에 영향을 미치는 요인 (Cuttings for Mass Propagation Affecting the Impact of Increasing Reproductive Efficiency of Schisandra chinensis)

  • This experiment was carried out to establish the optimum cutting time, plant growth regulator, and bed-soil for rooting by greenwood cutting of Schisandra chinensis. Approximately 7 cm long-shoots of greenwood cuttings were transplanted by various cutting times, plant growth regulators, and bed-soils in the plastic-film house. The rooting rate of greenwood cutting was 12.5% in the April 5th, 73.5% in the May 8th, and 75.5% in the August 5th. The number and length of primary root in greenwood cutting were more in the early May than those in the early August. For mass propagation of Schisandra chinensis. using greenwood cutting, shoots were treated with plant growth regulators on May to increase rooting rate. Rooting rate was 100% with IAA $50mg/{\ell}$, 92.9% with NAA $100mg/{\ell}$, and NAA $1,000mg/{\ell}$, for 60 min. To select effective media for rooting, various medias for bed-soil were treated by single and mixture form for 100 days after cutting. Rooting rate was 91.8% in the single treatment of peat moss or decomposition of granite soil, and this result was better than those in other treatment. The treatment by 1 : 1 mixture of peat moss and horticulture bed soil was rooting with 94.0% best rooting rate.

  • 발아율이 저조한 한지형 잔디인 Kentucky bluegrass 와 Tall fescue종자를 식물생장조절제인 $GA_3$와 BAP를 처리하여 발아율을 극대화시킬 수 있는 최적 처리조건을 구명하였다. Kentucky bluegrass 초종에서는 $GA_3$ 처리는 농도에 관계없이 BAP 단독 또는 및 $GA_3$와 BAP혼용처리보다 발아증진 효과가 높았으며, 처리기간은 6일 처리가 3일 처리보다 좋았다. 따라서 최적 처리조건은 $GA_3$ $1000{\mu}M$농도로 6일간 처리였으며, 무처리보다 발아율을 $10{\sim}38%$ 향상시킬 수 있었다. Tall fescue 종자는 생장조절제를 처리하면 발아율이 향상되는 경향이나 현저한 수준은 아니었다. 최적 생장조절제 처리조건은 $GA_3\;1000{\mu}M$용액으로 3일간 처리였는데, 무처리 종자에서 비해 발아율을 10%증진시켰다. $GA_3$ 처리된 Kentucky bluegrass와 Tall fescue 종자는 묘출현율이 무처리에 비해 $8{\sim}9%$ 향상되었고, 묘출현속도는 1.2일 빨랐다. 또한 건물 중, 초장 및 뿌리생장 등 초기생육이 무처리에 비해 향상되었으나, 유의성은 인정되지 않았다.

Rapid Propagation through Tissue Culture of Cudrania tricuspidata, Medicinal Plant

  • An effective rapid propagation method was established through in vitro cultures of the medicinal plant, Cudrania tricuspidata. In vitro plantlets were obtained from in vitro germinated seeds. The various levels of cytokinins (BAP, Kinetin and TDZ) were tested on multiple shoot formation from plantlets. BAP (1.0 mg/l) treatment induced highest number of multiple shoots. Single shoot cultures gave higher initial shoot numbers than 5 shoots per culture. Among the various culture media, the shoot elongation was optimal on 2 MS basal medium without growth regulators. The IAA (2.0 mg/l) treatment induced highest number of roots. IBA (2.0 mg/l) treatment more promoted in vitro root growth than other concentrations. Rooted shoots were transferred directly to small pots with an artificial soil and successfully acclimatized.

Establishment of Efficient Regeneration System Through In Vitro Culture of Lettuce (Lactuca sativa)

  • An efficient regeneration system was established by using in vitro plantlets of germinated seedlings from different cultivars of lettuce (Lactuca sativa cv. Chongchima, Chongchuckmyun, Jeokchima, Jeokchuckmyun). Shoot formation were observed from all cultivars on MS medium supplemented with 0.1 mg/L NAA and 0.5 mg/L BA. In all cultivars, when cotyledon was cultured, the number of shoot per explant was more greater than that hypocotyl and leaf disc were cultured. Shoot formation rate (91.7%) was high in a cotyledon culture of cultivar, Chongchukmyun. The growth of multiple shoots derived from the cultivar, Chongchukmyun, was most effective on medium containing 0.5 mg/L BA and 1.0 mg/L GA$_3$. When shoots were transferred on MS medium without plant growth regulators, roots were effectively differentiated. Rooted plantlets were acclimated on pots for further propagation.

In Vitro Regeneration of Pongamia pinnata Pierre

  • Pongamia pinnata Pierre is a tree legume, having potential in production of raw material for biodiesel. A protocol for in wk propagation of this plant was standardized using seedling explants. Growth regulators (GR) including gibberellic acid $(GA_3),\;N^6-benzylaminopurine(BA)$, thidiazuron (TDZ), and Adenine sulphate (Ads) were tested for optimum germination of seeds. Removal of seed coat prior to germination, controlled fungal growth partially but enhanced bacterial growth. Antibiotic cefotaxime was ineffective in controlling bacterial contamination. Seedling derived nodal explants and cotyledon nodes with attached cotyledons were excised and cultured for induction of shoots. Optimum sprouting and multiplication of shoot buds were obtained in MS medium supplemented with $8.88{\mu}M$ BA. These buds differentiated and rooted on medium devoid of GR. Optimum growth of Pongamia seedling was obtained in cotton plugged culture vessels. Reculturing of the cotyledon node explants produced more shoots from the same site. This process of removing shoots and reculturing of cotyledon node was followed for eight passages yielding 4 to 8 shoots in each cycle. The shoots (75%) rooted on half strength MS basal medium supplemented with 0.22% charcoal. All plants survived on transfer to soil. This is the first report on in vitro regeneration of Pongamia pinnata. This report demonstrates the possibility of coupling more than one parameter in single experiment to hasten the process of standardization. The process of cycling the nodal explant repeatedly for production of large number of shoots from single meristem may find application in genetic transformation experiments wherein meristems are used for transformation.

Cultures of Ginkgo biloba, Effect of Nutritional and Hormonal Factors on the Growth of Cultured Cells Derived from Ginkgo biloba

  • Calli and suspension cultures were obtained following inoculation of the explant from leaves of Ginkgo biloba L on the supplemented MS basal medium. The obtained calli and suspension cultured cells were able to produce detectable amounts of ginkgolides which are known as natural specific PAF antagonists. The production of ginkgolides in the calli and suspension cultured celles were identified using GC/MS, GC and HPLC with authentic ocmpounds. Since the production of ginkgolides A and B the calli and suspension cultured cells had been confirmed, effects of types and concentration of plant growth regulators, media and illumination on the induction and growth of the callus were studied. The concentrations of growth regulators for optimal callus were studied. The concentrations of growth regulators for optimal callus induction were studied. The concentrations of growth regulators for optimal callus induction were 1.0 to 2.0 mg/L for NAA and o.1 mg/L for kinetin. The growth of the Callus seemed to be more simnultaed with the combination of NAA and kinetin than NAA and BA with illumination at all concentration ranges of 1.0 to 4.0 mg/l for NAA and o.1 to 1.0 mg/L for kinetin or BA studied. Amogn 8 different media used, the induction rate of callus on Anderson, Eriksson, and Shenk and Hildebrant at 4 weeks after the innoculation was almost the same as that of MS. However, callus was rarely induced on Heller or White medium. Suspension cultures were easily initiated with 3 g of callus (fresh weight) derived from ginkgo leaves on supplemented MS medium. A typical growth curve of suspension cultured cells could be obtained by measuring the fresh weight of the suspension cultured cells at every 3 days. To improve the growth of suspension cultured cells of ginkgo, effects of concentrations of NAA, sucrose, phosphate ions and molar ratio of $NH_{4}^+\;to\;NO_{3}^-$ ions in the culture medium were studied. The maximum growth of the cells was achieved when the culture medium contained 1.0 mg/L of NAA, 30 g/L sucrose, 1.75 mM phosphate ions and 1:5 molar ratio of $NH_{4}\;to\;NO_{3}^-$ ions.
