Effect of Plant Growth Regulator 'Hoe 78784' on Lodging in Rice (식물생장 조절제 Hoe 78784가 벼 도복에 미치는 영향)

  • This experiment was carried out at International Rice Research Institute (IRRI) farm to investigate the effect of plant growth regulators on lodging in rice and to determine plant characteristics when plant growth regulator applied at different concentrations and different times. The experimental plant growth regulator Hoe 78784 when applied at booting prevented lodging and significantly increased grain of IR 21820-154-3-2-3. However, they did not affect significantly the crop vegetative characteristics and yield component, except by decreasing plant height and internode elongation, and increasing the number of vascular bundles, and the thickness and diameter of the third and fourth internodes. But when applied at 30 DT, it significantly decreased grain yield of all test varieties due to shortened panicle length and increased unfilled spikelet percentage. Hoe 78784 applied at heading did not increase yield nor did it prevent lodging.

Effect of Growth Regulators and Chilling Treatment on Bulblet Increment of Narcissus pseudo-narcissus 'King Alfred' in Vitro (수선의 기내 구 비대에 미치는 생장조절제와 저온처리의 영향)

  • This experiments were conducted to investigate growth regulator concentrations and a period of chilling treatment for increasing the size of bulblets of Narcissus pseudo-narcissus cv King Alfred. Combination of 2.5 mg/L BA and 2.5 mg/L NAA remarkedly increased the diameter and growth of regenerated bulblets. Chilling period favorable for the growth of regenerated bulblets was found to be 8 weeks, and addition of 1.0 mg/L NAA alone to MS medium was the most effective for increasing the size of bulblet for the culture after the chilling treatment.

Plant Regeneration and Bulblet Formation of Allium wakegi Araki

  • Allium wakegi was cultured shoot tip in the condition of light culture. The Allium wakegi added plant growth regulator was observed of plant regeneration and bulblet formation. Callus Induction and growing rate was the best of 78% when added alone 2,4-D 0.5mg/L. In the formation of shoot, its regeneration rate was 96% when added BA 0.5mg/L in the light culture condition. When BA 0.5mg/L and NAA 0.5mg/L mixed and BA 0.5 mg/L and NAA 1.0mg/L mixed, the rates were 99% and 97% respectively, and these conditions were suitable for forming shoot. In the formation of roots, when added NAA 2.0mg/L in the light culture condition, the regeneration rate was 90.6 % and the roots were abnormal. When added NAA 1.0mg/L, the rate was 82 % and the highest. In the formation of bulbs, when BA 05mg/L and NAA 1.0mg/L mixed, the root generantion and its size in the bulbs was the best compare to other treatment experiments.

High Frequency Shoot Regeneration from leaf Explants of Cucumber

  • Leaf explants of the cucumber (Cucumis sativus L.) were cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of $\alpha$-naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP). Direct shoot orgnogenesis as well as callus formation with somatic embryos and multiple shoots was observed from leaf explants of cvs. Shinhukjinju and Chungjang. The highest frequency of shoot formation 80% was observed on MS medium supplemented with NAA/BAP (5.0 ${\mu}{\textrm}{m}$/2.5 ${\mu}{\textrm}{m}$), with explants forming 3-7 shoots. Shoots formation occured within 3 to 4 weeks. Only one subculture of calli was required for plant regeneration on normal growth regulator-free medium. Plantlets transferred to soil developed into plants of normal appearance, which flowered and set fruits.

  • This Study was carried out to select bed soil and growth regulator which are effective for the macro propagation by cutting of Cudrania tricuspidata Bureau. The rooting of Cudraniae tricuspidata showed good result when the bed soil was coarse soil, rooted hardwood cutting 56.9%, and softwood cutting 62.3%. The rooting of Cudraniae tricuspidata showed good result when treated by growth regulator IBA 100 ppm, rooted hardwood cutting 57.9%, and softwood cutting 89.2%.

  • The soybean (Glycine max(L.) Merrill), an important food crop in the world, is popular because of its high quality protein and oil content. Soybeans as a food have long been known for their beneficial effects on health and are well-recognized globally. Isoflavones, significant soybean secondary metabolic products, may be crucial in avoiding some cancers and lowering the risk of cardiovascular disorders. This study investigates the correlation between plant growth regulator and the effect on the isoflavone levels in soybean leaves. The study was carried out in the green-house of the southern crop department in miryang. Soybeans(Seonpung) were cultivated in 1/2000 of the Wagner pot. Ethephon(500, 1000, 2000 ppm) and ABA(100, 200, 400 ppm) were used as plant growth regulators, and they were each treated on R2, R5, and R7 stage. After treatment, leaves were sampled three times at intervals of 5 days, and the content of 6 isoflavones and coumestrol was analyzed. Soybean isoflavones were analyzed using Ultra Performance Liquid Chromatography (Acquity UPLC H-Class system, Waters). The isoflavones content showed an overall highly in the R5 stage, and the level was similar to that of no treatment in the R2 and R7 stage. The difference between the growth regulators was found to be higher than that of ethephon when ABA was treated. The coumestrol content was confirmed to be high in the order of R7, R5, and R2 on the treatment time, and it was found that the content increased as the treatment time was delayed. In the treatment with the growth regulator, the coumestrol content tended to be higher when ethephon was treated than ABA.

Efficient Plant Regeneration from Shoot Tip and Young Leaf in Rhodiola sachalinensis A. Bor.

  • The shoot tip and young leaf of Rhodiola sachalinensis were cultured to invest the plant growth regulator condition for callus induction, shoot and root regeneration. When the shoot tip was sterilized in 2.0% of NaOCl for 20min., the contamination rate was the lowest. And the survival rate of the culture material was good in carbenicillin 500mg/L treatment group. Callus was obtained from shoot tip and young leaf segments. NAA 0.1-1.0mg/L and 2,4-D 0.1-0.5mg/L alone treatment were shown to have a good response on callus induction from shoot tip culture. In the case of young leaf culture, NAA and 2,4-D 0.1-0.5mg/L alone treatment were good in callus induction. In culturing shoot tip NAA 0.5mg/L and BA 0.5mg/L, NAA1.0mg/L and BA 0.lmg/L combination treatment was good in shoot regeneration. The regenerated shoots were rooted on MS medium supplemented with NAA and BA combination treatment. Especially, NAA 1.0mg/L and BA 0.1mg/L combination treatment was effective for root regeneration.

  • This study was conducted to evaluate the effect of several fungicides and growth regulators on rice seedling growth and damping-off in seedling boxes for machine transplanting. Fungicide treated plots were better seedling growth, shoot regrowth, rooting ability, change of moisture content than those of nontreated plot. Metalaxyl application of Samgangbyeo and SF8002 application of Nagdongdyeo apparently increased plant height, length of the third leaf and fourth leaf. And metaiaxyl application highly increased dry weight. Fungicide treated plots were highly effective in reducing the incidence of damping-off. Benzyladenine application of Samgangbye and $GA_3$ application of Nagdongbyeo apprently increased plant height. But ABA application highly decreased plant height. ABA application and aCE application resulted in highly increased rooting ability. Fungicide and $GA_3$ treated plots, Metalaxyl and growth regulator treated plots resulted in highly increased plant height. I soprothiolane and growth regulator treated plots resulted in decreased plant height. Dachigaren and lAA treated plot apprently increased dry weight and shoot dry weight/plant height. Fungicide and growth regulator treated plots were highly effective in reducing the incidence of damping-off.

  • A bioassay is a test system using a living organism (in whole or in part) to determine the presence or relative potency of chemical substances. The development and uses of bioassay are intimately linked to the discovery and characterization of the major classes of plant hormones. An application of this relationship is helpful for understanding the concept of plant hormones as well as the use of bioassay. And plant bioassay have been development and employed not only for the discovery and characterization of the biological activity of plant growth regulators but also have served several important secondary roles. The ideal bioassay should possess the characteristic of high specificity. great sensitivity. short response time, low cost and ease of obtaining plant material. acceptable ease of manipulation, and minimal space and equipment requirements.
