Effects of Hansu-Daebowon (HDW) on RANKL-induced Osteoclast Differentiation and Bone Loss in Mammal Model (한수대보원이 포유동물인 생쥐 모델에서 골 손실 및 RANKL 유도 파골세포 분화에 미치는 영향)

  • Objective: This study investigated the effects of Hansu-Daebowon (HDW) on bone resorption in vitro and bone loss in vivo. Methods: Osteoclast differentiation was measured by counting TRAP (+) MNC formed from RAW 264.7 in the presence of RANKL. Bone pit formation was determined in an artificial bone slice loaded with RANKL-stimulated osteoclasts. To elucidate the mechanisms of the inhibitory effects of HDW on bone resorption and osteoclast differentiation, osteoclastogenic genes (i.e. TRAP, MMP-9, NFATc1, c-Fos, and Cathepsin K) were measured using real time PCR. Furthermore, bone loss was observed using micro-CT in an LPS-treated mammal model. Results: HDW inhibited the bone pit formation in vitro and inhibited bone loss in vivo. Moreover, HDW decreased the number of TRAP (+) MNCs in the presence of RANKL, and HDW inhibited the expressions of cathepsin K, MMP-9, TRAP, NFATc1, and c-Fos in the osteoclasts. Conclusion: HDW exerts inhibitory effects on bone loss and bone resorption resulting from the inhibitions of osteoclast differentiation and osteoclastogenic gene expression.

A SURVEY ON THE USING STATUS AND PERCEPTION OF PIT AND FISSURE SEALANT (치면열구전색제 사용실태와 인식에 관한 조사)

  • The property of pit and fissure sealant has been improved and many studies on the bond strength, penetration, microleakage have been published. But there are few studies on the using status and perception of pit and fissure sealant within the country. Therefore, this study made a survey on it. Pedodontists and non-pedodontists were surveyed by interview. The Results were as follows; 1. On caries prevention effect, 96.7% of the pedodontists replied that sealants were effective on both permanent teeth and primary teeth. On the other hand, 13.5% of the non-pedodontists replied that sealants weren't effective on both. 2. All of the pedodontists and 27% of the non-pedodontists used rubber dams. 83.3% of the pedodontists and 40.5% of the non-pedodontists used bonding agents. 3. Non-pedodontists used enameloplasty more frequently than Pedodontists but the pattern was not significantly different. 4. The causes of sealant failures included salivary contamination, caries under sealant, low strength, low flowability, overfilling. 5. In the pedodontists, 90% replied that PRR application was desirable and PRR applications were more frequent than sealant application.

Effect of surface anodization on stability of orthodontic microimplant

  • Objective: To determine the effect of surface anodization on the interfacial strength between an orthodontic microimplant (MI) and the rabbit tibial bone, particularly in the initial phase aft er placement. Methods: A total of 36 MIs were driven into the tibias of 3 mature rabbits by using the self-drilling method and then removed aft er 6 weeks. Half the MIs were as-machined (n = 18; machined group), while the remaining had anodized surfaces (n = 18; anodized group). The peak insertion torque (PIT) and the peak removal torque (PRT) values were measured for the 2 groups of MIs. These values were then used to calculate the interfacial shear strength between the MI and cortical bone. Results: There were no statistical differences in terms of PIT between the 2 groups. However, mean PRT was significantly greater for the anodized implants ($3.79{\pm}1.39$ Ncm) than for the machined ones ($2.05{\pm}1.07$ Ncm) (p < 0.01). The interfacial strengths, converted from PRT, were calculated at 10.6 MPa and 5.74 MPa for the anodized and machined group implants, respectively. Conclusions: Anodization of orthodontic MIs may enhance their early-phase retention capability, thereby ensuring a more reliable source of absolute anchorage.

A New Ceanothane-Type Triterpenoid Saponin Isolated from Gouania leptostachya DC. var. tonkinensis Pit. and Its Underlying Anti-Inflammatory Effects

  • Metabolites from medicinal plants continue to hold significant value in the exploration and advancement of novel pharmaceuticals. In the search for plants containing compounds with anti-inflammatory effects, we observed that the ethanol (EtOH) extract obtained from the aerial components of Gouania leptostachya DC. var. tonkinensis Pit. exhibited substantial suppression of nitric oxide (NO) in vitro. In a phytochemical study on an EtOH extract of G. leptostachya, 11 compounds were purified, including one unreported compound namely gouanioside A (1). Their chemical structures were unambiguously determined through the use of various spectroscopic techniques, such as 1 and 2D NMR, IR, and HR-ESI-MS, and by producing derivatives via chemical reactions. The EtOH extract, fractions, and a new compound exerted inflammatory effects by altering NO synthesis in murine RAW264.7 macrophage cells stimulated with lipopolysaccharide. The underlying inflammatory mechanism of the new compound 1 was also explored through various in vitro experiments. The results of this study indicate the potential usefulness of new compound 1 from G. leptostachya as a treatment for inflammatory diseases.

Morphological Variation of Vessel Elements in the Korean Diffuse-porous Woods (한국산(韓國産) 주요(主要) 산공재(散孔材)의 도관요소(導管要素) 형태변이(形態變異))

  • This study was carried out to investigate the variations of dimension and exterior shape of vessel elements, morphology of spiral thickening and ray-vessel pit in korean diffuse-porous woods (56 species, 18 families), The tangential pore diameter and vessel element length was increased, whereas the pore number per unit area($1mm^2$) was decreased from pith outwards. The tangential pore diameter was decreased but the length of vessel element was not changed from earlywood to latewood within an annual ring. However, the dimensional variations of vessel element was not able to be recognized among relative positions in peripheral variation. The exterior shape of vessel element could be classified into four types; Type 1 is without tail, Type 2 with ligulate tail, Type 3 with broad taper tail and Type 4 with the very short length between perforations. The distribution frequency of Type 2 and 3 was relatively high in comparision with the others. According to the prominence, distribution position and branched shape, the spiral thickenings could be divided into five types. The spiral thickenings occurred 52% in the species observed. Thus it was doubtful to consider the simple presence of spiral thickening as diagnostic index in diffuse-porous woods. The morphology of ray-vessel pit could be grouped as reticulate, scalariform, oval, linear and coalescent type. Most of species examined showed oval and linear type.

Effects of Gardeniae Jasminoides on RANKL-induced Osteoclastogenesis and Bone Resorption (치자 추출물이 RANKL 유도 파골세포 형성 및 골 흡수에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of Gardenia jasminoides extract (GJ) on osteoclast differentiation and bone resorption in vitro. Methods: To investigate the effect of GJ on osteoclast differentiation, the mouse leukemic myeloid cell line RAW 264.7 was stimulated by RANKL (receptor activator of nuclear factor kB ligand). Osteoclast differentiation was measured by counting TRAP (+) MNC in the presence of RANKL. To elucidate the mechanism of the inhibitory effect of GJ on osteoclast differentiation, gene expression of TRAP, Cathepsin K, MMP-9, NFATc1, c-Fos, MITF, DC-STAMP, CTR, OC-STAMP and Atp6v0d2 was measured using reverse transcription-PCR (RT-PCR). Bone resorption was measured using the bone pit formation assay. Results: GJ decreased the number of TRAP (+) MNCs in the presence of RANKL. GJ inhibited the expression of cathepsin K, MMP-9, TRAP, MITF, NFATc1, c-Fos, iNON, OC-STAMP, Atp6v0d2, and DC-STAMP in the osteoclast, and inhibited bone pit formation in vitro. Conclusions: The results suggest that GJ has inhibitory effects on bone resorption resulting from inhibition of osteoclast differentiation and gene expression.

Fabrication of $MgB_2$ tape with metal powder addition (금속분말이 첨가된 $MgB_2$ 선재의 제조 및 특성)

  • The $MgB_2$ tapes with several metal powder addition were fabricated by PIT method with or without heat treatment. The $J_c$ value of $5.600A/cm^2$ and $16.000A/cm^2$ at 4.2 K and 5 T were obtained for the $MgB_2$ tape and 10 vol % of Cu added $MgB_2$ tape without heat treatment respectively. The $J_c$ value of $8.000A/cm^2$ and $35,000A/cm^2$ at 4.2 K and 5 T were obtained for the $MgB_2$ tape and 10 vol. % of Al added $MgB_2$ tape with heat treatment, respectively. The $J_c-B$ curve shows enhancement in $J_c$ under magnetic field. which suggests enhancement in workability and grain connectivity with several metal powder addition.

Evaluation of Microleakage and Penetration Ability of Flowable Resin in Occlusal Fissure (유동성 레진의 교합면 열구내 침투도와 미세누출 평가)

  • The aim of this study was to compared the penetration scores and microleakage levels of flowable resin in comparison to resin-based pit and fissure sealant. A total 120 extracted premolars were used and classified with group I (resin-based pit and fissure sealant), group II (flowable resin), group III (bonding agent + flowable resin), IV (fissurotomy + bonding agent + flowable resin) depending on the materials and the application methods. The penetration scores of the group treated with flowable resin following fissurotomy were similar to those of the group treated with resin-based sealant, but higher compared to those of the other groups treated with flowable resin without fissurotomy or bonding agent. The group treated with flowable resin following fissurotomy also exhibited the lowest microleakage levels. The group treated with resin-based sealant and the one treated with a bonding agent prior to the application of flowable resin showed similar microleakage levels. In conclusion, flowable resin may be applied as a pit and fissure sealant, and its application along with fissurotomy could increase the penetration scores.

THE EVALUATION OF MICROLEAKAGE OF PIT AND FISSURE SEALANT BY USAGE OF SELF-ETCHING ADHESIVE SYSTEM (자가 산부식 접착제를 이용한 치면열구전색의 미세누출 평가)

  • The purpose of this study was to evaluate the effectiveness of fissurotomy and double application time of acidic primer of self-etching adhesive system of acid treatment on enamel surfaces for prevention of microleakage of pit and fissure sealants. The microleakage of pit and fissure sealants was evaluated by measuring the penetration depth of methylene blue solution. Specimens were divided by 4 groups according to the method of treatment. Group I: 35% phosphoric acid etching, 20 seconds. Group II: Priming with self-etching primer, 20 seconds. Group III: Priming with self-etching primer, 40 seconds. Group IV: Priming with self-etching primer, 20 seconds, after fissurotomy. The etched pattern produced on enamel was observed using a scanning electron microscope. Obtained data were analysed statistically using Kruscal-Wallis test followed by Mann-Whitney test for comparison of groups. The results are as follows: 1. Microleakage scores of group IV priming with self-etching primer 20 seconds after fissurotomy was the lowest. 2. Microleakage scores between group II and group III were not shown significant difference. 3. Enamel for 20 seconds etching with 35% phosphoric acid was observed the most effective etching pattern. And the etching pattern on the fissure enamel with self-etching adhesive 20 seconds after fissurotomy was more prominent than group II, III only using self-etching primer.

EVALUATION OF THREE FLOWABLE COMPOSITES AS PIT AND FISSURE SEALANTS WITHOUT USING BONDING AGENTS (접착제 미 사용시 치면열구전색제로서의 유동성 복합레진 평가)

  • The aims of this study were to determine if flowable composites can be used as pit and fissure sealants without bonding agents. Three flowable composites(Filtek Flow, Tetric Flow, Charmfil Flow) and a filled sealant (Ultraseal XT plus) were used. The pattern of resin tag formation in the four sealant materials were compared using SEM. For the microleakage assessment, 54 extracted human premolar teeth were randomly divided into 3 groups containing 18 premolars each. In each group, a conventional filled sealant and one of the three flowable composites were applied to occlusal fissures. The teeth were thermocycled(1200 cycles between $5^{\circ}{\pm}2^{\circ}C\;and\;55^{\circ}{\pm}2^{\circ}C$ with a dwell time of 30 seconds) and immersed in a 1% methylene blue solution for 48 hours. Each tooth was sectioned and examined to determine the extent of dye penetration. Three flowable composites and a filed sealant showed a similar resin tag formation pattern. The three flowable composites showed significantly more microleakage in each group than the filled sealant. The level of microleakage was similar in the three flowable composites. Flowable composites are not recommended as pit and fissure sealants because more microleakage can occur even when occlural fissures are mechanically widened.
