Comparison of Micro-Pipette Method and Hydrometer Method in Soil Particle Size Analysis (Micro-Pipette법과 Hydrometer법에 의한 토양 입경 분석의 비교)

  • Determination of soil texture, particularly the clay fraction, is an important measurement in most researches related with soil. In this study micro-pipette method For soil particle size analysis was compared with the standard hydrometer method. Micro-pipette method can eliminate the need for bulky laboratory equipment and long settling times associated with the standard hydrometer or pipette method. In the results of this investigation, the particle size data obtained with micro-pipette method were in good agreement with those found using standard hydrometer method. And with this method one person could run analysis much larger numbers of soil sample per day than with hydrometer method in relatively small laboratory space.

Method for Rapid and Accurate Measurement of Chitosan Viscosity

  • A simple and rapid method to estimate the viscosity of chitosan using laboratory pipettes was developed. The voscosities of nine different chitosan samples, prepared ini 1 % acetic acid at a 1% concentration , were measured with a standard viscometer. Prior to measurement of flow time of 1% chitosan solution with a pipette, twelve pipettes were assorted into three groups with flow times of 4, 5 and 6 sec after measuring passage of 9 ml of 1% acetic acid througth a 10 ml pipette. With each group of pipettes. flow time of 1% chitosan solution was determined by measuring the delivery time of 5 ml of the 10ml solution through a 10 ml pipette. Results of regression analyses revealed high linear relationship(R2=0.9812, 0.9663, and 0.9754) between viscosities calculated with a viscometer and flow times measured with 4, 5 or 6 sec group pipettes. The viscosity of chitosan could be readily and accurately estimated from these linear regression equation by measuring flow times based on pipette delivery.

Measurement of Retention, Repellency and Penetration of Pesticide for Protective Clothing (방제복에 대한 농약의 체류성, 발수성 및 침투성 측정)

  • BACKGROUND: The personal protective equipments (PPEs) is the most important factor for reducing the pesticide exposure during the preparation and spraying of pesticides. This study was to investigate the retention, repellence and penetration of protective clothing of PPEs according to the ISO 22608 'Liquid penetration resistance-pipette test. Protective clothing for agriculture worker is very important for their health. However, test method for measurement of protective clothing is not presented in Korea. METHODS AND RESULTS: In order to measure the retention, repellence and penetration of protective clothing, a apparatus for pipette method in accordance with the ISO Guideline was prepared. The test was conducted at $25{\pm}5^{\circ}C$, $60{\pm}10%$ of relative humidity and pipette applied to a apparatus was validated to take exact amount(0.2 mL). The retention, repellence and penetration of five types of protective clothing and one type of shirt were analyzed by GC/MS. Pendimethalin(5% a.i, emulsion) was used as a test pesticide to measure above factors. The retention were less than 11.0% with the exclusion of two types(F4 and shirt) and the repellency was more than 67.0% with the exclusion of shirt material. The penetration was less than 5.4%, however, that of shirt was 66.7%. CONCLUSION: This results indicated that all protective clothing were suitable to use as PPEs according to the criteria specified by ISO Guide 22608. However, shirt was not suitable due to high penetration. This test method established for measurement the retention, repellence and penetration of protective clothing will help to establish the test notice of pipette method.

A Precision Test of Hydrometer Method for Determining Soil Texture (비중계법에 의한 토성분석시 정도 실험)

  • A precision test of hydrometer method, used to determine soil texture, was conducted on selected 10 soil samples, compared to pipette method. Soil texture measurements with hydrometer method were performed with monitoring the temperature of soil suspension in settling cylinder. The temperature and its fluctuation during settling time had a range of $13^{\circ}C-28^{\circ}C$ and $0.2^{\circ}C-4.4^{\circ}C$, respectively. The difference of clay content between hydrometer and pipette method were distributed from -6.4% to 4.0%. Positive end of difference in clay content was observed at soil having very low clay content, whereas negative end at soil having high organic matter content and exchangeable cations. Except both ends, difference in clay content of soils was less than 3%, and expecially closed to 0% in soils having clay content more than 25%. The difference of sand content were distributed from -1.5% to 4.2%. Similar to clay content, positive end soil was soil sample having lowest sand content.

  • Grain size distributions of sediment samples were analyzed by the different methods, and the results were compared. Reported data of the bottom sediments from the Korean seas show significant deviations among the institutes, and the inter-lab comparison and calibration procedures are considered to be necessary. Grain size data by different methods show different results. Laser diffraction method provides good precision in replicate analysis, but underestimates the amounts of finer (smaller than 2-3 ${\mu}m$) fraction. Data from particle settling method, on the other hand, represent significant errors in the coarse silt (5-6 ${\phi}$) fraction, and slightly overestimate the clay fraction. In the sieve and pipette methods, best results were taken in 0.5 ${\phi}$ interval of analysis. Especially in the coarse silt fraction, pipette analysis is suggested to be made with 0.5 ${\phi}$ interval. During the dry sieving procedure in 1 ${\phi}$ interval, serious errors occur when large amounts of sample materials were concentrated in a particular sieve.

Effects of Noradrenaline on the Membrane Potential of Prostatic Neuroendocrine Cells of Rat

  • The prostate gland contains numerous neuroendocrine cells that are believed to influence the function of the prostate gland. Our recent study demonstrated the expression of both ${\alpha}1$- and ${\alpha}2$-ARs, signaling the release of stored $Ca^{2+}$ and the inhibition of N-type $Ca^{2+}$ channels, respectively, in rat prostate neuroendocrine cells (RPNECs). In this study, the effects of NA on the resting membrane potential (RMP) of RPNECs were investigated using a whole-cell patch clamp method. Fresh RPNECs were dissociated from the ventral lobe of rat prostate and identified from its characteristic shape; round or oval shape with dark cytoplasm. Under zero-current clamp conditions with KCl pipette solution, the resting membrane potential (RMP) of RPNECs was between -35 mV and -85 mV. In those RPNECs with relatively hyperpolarized RMP (<-60 mV), the application of noradrenaline (NA, $1{\mu}M$) depolarized the membrane to around -40 mV. In contrast, the RPNECs with relatively depolarized RMP (>-45 mV) showed a transient hyperpolarization and subsequent fluctuation at around -40 mV on application of NA. Under voltage clamp conditions (holding voltage, -40 mV) with CsCl pipette solution, NA evoked a slight inward current (<-20 pA). NA induced a sharp increase of cytosolic $Ca^{2+}$ concentration ($[Ca^{2+}]_c$), measured by the fura-2 fluorescence, and the voltage clamp study showed the presence of charybdotoxin-sensitive $Ca^{2+}$-activated $K^+$ currents. In summary, adrenergic stimulation induced either depolarization or hyperpolarization of RPNECs, depending on the initial level of RMP. The inward current evoked by NA and the $Ca^{2+}$-activated $K^+$ current might partly explain the depolarization and hyperpolarization, respectively.

  • Many important traits have been tagged allowing plant breeders to apply marker assisted selection (MAS) in rice. PCR itself is simple to set up, and requires little hands-on time. However, a crucial limiting step of MAS programs is the reliable and efficient extraction of DNA which can be performed on thousands of individuals. In this study, We describe a modification of the DNA extraction method, in which cetyltrimethylammonium bromide (CTAB) is used to extract DNA from leaf tissues for suitable MAS in rice. We followed the standard 2% CTAB extraction method in all the procedure. In addition we used the 1.2 ml 8-strip tube instead of 1.5 ml E-tubes to fit the 8-multichannel pipette and employ the 96 well plate to use the swing bucket centrifuge. Our modified CTAB DNA extraction method offers several advantages with respect to traditional and simple methods. 1) adult leaf samples collected in paddy field are applicable. 2) 96 leaf samples can be homogenized only one-time by using tungsten carbonate bead and 96well block. 3) semiautomatic loading method using 8-multichannel pipette from DNA extraction to electrophoresis of PCR products. 4) our system can extract about 400 leaf samples per day by only one technicion. Therefore, this method could be useful for marker assisted breeding in rice.

The Electric Control Method on the Packaging Technology for Non-Conductive Materials Using the Surface Processing Cavity Pressure Sensor (표면 가공형 캐비티 압력센서를 이용하여 비전도성 물질용 패키지 기술에 전기적 제어방식 연구)

  • In this study, a pressure sensor for each displacement was fabricated based on the silicon-based pressure sensor obtained through simulation results. Wires were bonded to the pressure sensor, and a piezoresistive pressure sensor was inserted into the printed circuit board (PCB) base by directly connecting a micro-electro-mechanical system (MEMS) sensor and a readout integrated circuit (ROIC) for signal processing. In addition, to prevent exposure, a non-conductive liquid silicone was injected into the sensor and the entire ROIC using a pipette. The packaging proceeded to block from the outside. Performing such packaging, comparing simple contact with strong contact, and confirming that the measured pulse wavelength appears accurately.

On Board Comparison of Total Hydrogen Ion Concentration (pH) and Total Alkalinity Measurements in Seawater (해수의 총수소이온농도(pH)와 총알칼리도 측정방법에 대한 선상 비교 연구)

  • On board comparison of pH and total alkalinity measurement in seawater was carried out during the JES expedition on R/V Roger Revelle in the East Sea using 550 real seawater samples from the surface to the bottom. Spectrophotometry and potentiometry without liquid junction were compared for pH measurement. The pH values of two methods are generally in a good agreement. Spectrophotometry with a pipette provides higher value compare with the potentiometry in the deep layer, where pH values are lower. However, spectrophotometry without a pipette shows same values with potentiometry within their precision range. The pipetting procedure may remove of $CO_2$ in the sample, which causes increase of pH. Potentiometric titration methods using a closed-cell and an open-cell were compared for the total alkalinity measurement. Values from open cell method are smaller by about 5~10 ${\mu}mol\;kg^{-1}$ than those from closed cell method. This may be caused by the bubble formed in the closed cell during the experiment. Although any analytical method compared in this study for the pH or TA measurement can be applied, special attentions should be paid for satisfactory results.

Studies on the Efficient Separation of Primordial Germ Cells and Introduction of Foreign DNA in the Chicken (닭에서 원시생식세포의 효율적 분리 및 외래 유전자 전이에 관한연구)

  • This study was conducted to determine the embryonic stages for the isolation of the highest number of PGCs and to improve PGCs enrichment method. The primordial germ cells(PGCs) from different sources of chick embryos were isolated. The embryonic stage having the highest number of PGCs from each sources was selected ; 1-day-old embryos for germinal crescent (stage 6-8), 2.5-day-old embryos for blood (stage 17-18) and 5.5-day-old embryos for gonad (stage 27-28). The number of PGCs from one embryonic germinal crescent, blood and gonad was about 87$\pm$1.8, 103$\pm$4.0, and 932$\pm$10.9, respectively. The viability of PGCs after Ficoll from each sources was similar, showing approximately 70%. the PGCs enrichment method was improved using Ficoll density gradient centrifugation. After this step the purity of PGCs from germinal crescent, blood, and gonad was 45$\pm$9.10%, 85$\pm$1.18%, and 86$\pm$0.19%, respectively. Also, PGCs were picked up by mouth pipette to improve the purity. This improved method for the separation of PGCs from different sources will serve as a useful too to preserve the foundation stocks of poultry and to produce germline chimeras.
