Analysis of Cone and Seed Characteristics from Different Mating Design Strategies of Pinus densiflora for. multicaulis

  • This study aimed to enhance seed productivity and secure genetic resources for Pinus densiflora for. multicaulis. We analyzed the characteristics of cone and seed generated by control pollination between Pinus densiflora (PD) and Pinus densiflora for. multicaulis (PDM). The highest number of cone scales (63.0) was obtained from the self-pollinated (sp) PDM clone B (PDM-sp-B), whereas the lowest number of cone scales (44.7) was obtained from two combinations designated as PDM-A×PD-075 and PDM-A×PD-0111. Both female parents of the hybrids were PDM-A. The highest seed production capacity (80.8) was obtained from the open-pollinated (op) PDM clone B (PDM-op-B). The seed potentials of PDM-B×PD-0111, PDM-op-A, and PDM-sp-B were 67.4, 66.5, and 63.1, respectively. The highest number of fertile scales (41.5) was obtained from PDM-op-B, and the lowest number of fertile scales (28.8) was obtained from PDM-A×PD-075. The total number of aborted ovules and 1st aborted ovules was not statistically significant in the mating design. The cross combination of PDM-B×PD-0111 had the highest number (34.8) of filled seeds and the lowest number of 2nd aborted ovules (5.2) and empty seeds (9). PDM-op-B had the highest number of developed seeds (47.6), although the number of empty seeds was the highest (41.2). Therefore, we conclude that the mating design of PDM-B×PD-0111 is useful for future breeding programs to improve seed yield of PDM. Our results showed that there was a strong correlation between the following two parameter pairs: number of scales and number of fertile scales, and the number of fertility scales and seeds potential (r=0.89 and r=0.84, respectively; both p<0.01).

  • To develope food preservative, antimicrobial activities of Pinus densiflora (PD) ethanol extract against Listeria monocytogenes Scott A. Listeria monocytogenes Brie I and Listeria monocytogenes ATCC 19111 were investigated. The ethanol extracts of PD showed strong antimicrobial activities on Listeria monocytogenes. The crude ethanol extracts of PD were further fractionated by ether, ethyl acetate and butanol. The ether fraction from ethanol extract showed the strongest antimicrobial effects on Listeria monocytogenes in tryptic soy broth containing 40 mg/mL ether fractions compared with other fractions. The effect of ethanol extract of pinus densiflora against Listeria monocytogenes culture for growth stage in tryptic soy broth at 35$^{\circ}C$ showed the strongest antimicrobial activites for lag phase. The morphological changes of the cells were observed with transmission electron microscope (TEM) and scanning electron microscope (SEM) and the cells were injured by treatment of 40 mg/mL ethanol extract of Pinus densiflora.

  • This study was carried out to investigate the effect of ethanol extract of Pinus densiflora (PD) on the growth of lactic acid bacteria (LAB), A-1, B-9, K-7, M-7 isolated from kimchi. The growth of isolated LAB was inhibited significantly in the modified MRS broth containing 40 mg/mL PD ethanol extract. Ethyl acetate fraction showed the strongest antimicrobial activities against LAB strains compared to other fractions. The addition of PD ethanol extract to kimchi did not change the pH of kimchi greatly compare with the control during the fermentation for 25 days. Change of titratable acidity in control was more higher than in the PD ethanol extract added kimchi during fermentation. The growth of total bacteria and LAB was inhibited about 1 to 2 log cycle by the addition of PD ethanol extract during the kimchi fermentation for 25 days at 1$0^{\circ}C$. Sensory quality of PD ethanol extract added kimchi was lower than that of control.

Identification of two common types of forest cover, Pinus densiflora(Pd) and Querqus mongolica(Qm), using the 1st harmonics of a Discrete Fourier Transform

  • The time-series normalized difference vegetation index (NDVI) product has proven to be a powerful tool to investigate the phenological information because it can monitor the change of the forests with very high time-resolution, This study described the application of the DFT analysis over the 9 year MODIS data for the identification of the two types of vegetation cover, Pinus densiflora(Pd) and Querqus mongolica(Qm) which are dominant species of evergreen and broadleaved deciduous forest, respectively, The total number of samples was 5148 reference cycles which consist of 2160 Pd and 2988 Qm. They were extracted from the pixel-based MODIS scenes over the 9 years from 2000 to 2008 of South Korea. The DFT analysis was mainly focused on the 0th and $1^{st}$ harmonic components, each of which represents the mean value and the variation amplitude of the NDVI over the years, respectively. The $0^{th}$ harmonic values of the vegetation Pd and Qm averaged over the 9 years were 0.74 and 0.65, respectively. This implies that Pd has a higher NDVI than Qm. Similarly obtained $1^{st}$ harmonic values of Pd and Qm were 0.19 and 0.27, respectively. This can be intuitively understood considering that the seasonal variation of Qm is much larger than Pd. This distinctive difference of the $1^{st}$ harmonic value has been used to identify evergreen and deciduous forests. Overall agreement between the Fourier analysis-based map and the actal vegetation map has been estimated to be as high as 75%. This study found that the DFT analysis can be a concise and repeatable method to separate and trace the changes of evergreen and deciduous forest using the annual NDVI cycles.

Antimicrobial Effects of Pinus densiflora Sieb. et Zucc. Extract on Pathogenic Microorganisms (병원성 미생물에 대한 소나무(Pinus densifiora Sieb. et Zucc.) 추출물의 항균효과)

  • To develop natural food preservative, antimicrobial activity of Pinus densiflora Sieb. et Zucc. (PD) ethanol extract against pathogens were investigated. The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtils KCTC 1021, Bacillus megaterium KCTC 3007 and Vibrio parahaemolyticus ATCC 17802 was inhibited as much as 4 to 6 log cycle in tryptic soy broth containing 40mg/㎖ of PD extract but Salmonalla typhimurium ATCC 14028 and Pseudomonas fluorescens ATCC 21541 were not inhibited. The total, gram negative and psychrotrophic bacterial count were inhibited in minced pork containing 40mg/㎖ of PD extract for 20 days at 4$\^{C}$. The sensory quality of hamburger patty such as taste, flavor and overall acceptability were not decreased by the addition of 40mg/㎖ PD(p<0.05).

Effect of Ethanol Extracts in Pinus densiflora, Lithospermum erythrorhizon on the Lipid Oxidation of Oil Emulsion (식물체(솔잎, 자초)의 에탄올 추출물이 유탁액의 지방산화에 미치는 영향)

  • This study was carried out to investigate the effects of ethanol extracts on lipid oxidation of oil emulsion. The results are as follows; The scavenging ability of plant extracts for hydroxyl radical was found, and plant extracts played an important role as a strong chelating agents to bind iron if Fe2+ ion exists in oil emulsion. Pinus densiflora(PD), Lithospermum erythrorhizon(LE) and PD+LE acted as strong chelating agents to bind iron to reduce lipid oxidation in oil emulsion. The content of Fe2+ ion in ethanol extracts from LE and PD+LE were significantly higher(p<0.05) than that of ethanol extracts from PD. The content of total iron has same tendency. The ascorbic acid content of PD(16.36ppm) was slightly higher than those of LE(13.08ppm). Electron donating ability of PD was significantly higher(p<0.05) than those of LE. However, the superoxide(SOD) like ability of LE showed a little higher than those of LE and PD+LE, which means the strong antioxidant activity of LE. The nitrite scavenging effects were dependent on pH value, however, they decreased as pH value increased. Especially, they almost didn't show the nitrite scavenging effect in pH 6.0. In conclusion, the PD and LE extracts may be used as natural antioxidant sources to reduce lipid oxidation in oil emulsion.

Activation of Intestinal Immune System by an Orally Administered Methanol Extract from Pine Needles (솔잎 메탄올추출물의 마우스 경구투여에 의한 장관면역 활성)

  • When pine (Pinus densiflora) needles were fractionated into cold water (PD-CW), hot water (PD-HW) and methanol extract (PD-M), PD-M showed potent simulating activity (1.19-fold of the saline control) for proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. MeOH extracts were prepared by homogenization, stirring or reflux to identify the method of methanol extraction, and MeOH extract by reflux method showed significantly highest intestinal immune system modulating activity (1.30-fold) in vitro. The intestinal immune system modulating effect of orally administered PD-M fractionated from pine needles also were studied in mice. Analyzing intestinal immune system modulating activity mediated Peyer's patch cells from C3H/He mice which had been fed with PD-M at different doses for 7 days, 1.0 g/kg of BW/day indicated that the bone marrow cells had proliferated (3.65-fold of 3% EtOH administered group). In addition, the amounts of IL-6 in the culture supernatant of Peyer's patch cells at 1.0 g/kg of BW/day were increased (1.13-fold) whereas the production of GM-CSF was not dose dependent. These results indicate that oral administration of PD-M enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-6 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation.

Heavy Metal Concentration of Soils and Plants in Baekdong Serpentinite Area, Chungnam - A Case of Pinus densiflora and Pinus rigida - (충남 백동 사문암지역의 토양 및 식물체내 중금속 함량 - 소나무 및 리기다소나무를 중심으로 -)

  • Heavy metal concentrations in rocks and soils from serpentinite(SP) and in plants (Pinus densiflora: PD and Pinus rigida: PR) were examined at Baekdong mine in Hongsung, Chungnam. Parent rocks were compared with amphibole schist(AS) and gneiss(GN) and plants divided the above grounds and roots were examined, respectively. In rocks, Ni, Cr, Co, Fe concentrations in SP were higher than those in AS and GN. The concentrations of top soils had the similar differences to their rocks; especially Ni, Cr, Co, Fe concentrations were the highest in SP, Zn and Sc concentrations, however, were the highest in AS. Average Ni, Cr, Co, Au, As, Sb, W concentrations of PD were the highest in SP and especially Ni, Cr, Co concentrations were accorded with changes of rocks and top soils. Zn and Sc concentrations in AS were higher and Fe and Mo concentrations in GN were higher than those in SP. Compared with two plants in the same serpentinite sites, most elements of PR were higher than those of PD. Therefore, these suggested PR absorbed much heavy metal than PD. Most element concentrations of roots in two plants and three rocks were higher than those of the above ground. Relative ratios (average plant concentration/soil concentration) of Ni, Cr, Co, Zn, Sc, Fe in AS and GN were higher than those of SP. Especially, relative ratios of most elements except Zn in GN were the highest.

A Study on the Activities of Five Natural Plant Essential Oils on Atopic Dermatitis (자생식물 Essential Oil 5 종의 항 아토피피부염 활성 연구)

  • This study is an experiment to evaluate the anti-atopy efficacy of five kinds of natural plant essential oils; Artemisia annua L. (AA), Citrus junos Sieb. ex TANAKA (CJ), Chrysanthemum boreale Makino (CB), Pinus koraiensis (PK), and Pinus densiflora for. erecta (PD). Through Agar diffusion test, five species of native plant essential oils were treated in a total of four strains, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Candida albicans. In order to invest the anti-inflammatory effect, five kinds of natural plant essential oils were treated in HaCaT cells-induced by TNF-α and IFN-γ (TI). AA, CJ, CB, PK and PD showed antibacterial effects on Candida albicans at a concentration of 10 mg/mL. We also found that the thymus and activation-regulated chemokine (TARC) expression was suppressed in 0.1 ㎍/mL of PK, 1 ㎍/mL of AA, CB, and PK. macrophage-derived chemokine (MDC) expression was suppressed in 1 ㎍/mL of AA and PK. IL-6 expression was suppressed in 0.1, 1 ㎍/mL of AA, PK in HaCaT cells. Hence it suggests that AA, CB, and PK have the anti-inflammatory effects, and it could contribute to atopic dermatitis relief by reducing the infiltration of immune cells to inflamed area.

Correlation Between Vegetation Structure and Environmental Factors of Corylopsis coreana Uyeki Habitats (히어리 자생지 식생구조와 환경요인 간 상호관계)

  • The purpose of this study is to understand the vegetation structure of the Corylopsis coreana habitats in South Korea and their correlation with environmental factors, in order to provide basic data for preparing conservation. A total of 40 vegetation survey plots were established in around Southern region, Gangwon-do and Gyeonggi-do Province. The cluster analysis revealed four distinct clusters: Quercus mongolica-Pinus densiflora (QmPd), Castanea crenata (Cc), Quercus mongolica (Qm), and Pinus koraiensis (Pk). Among them, the QmPd cluster was selected as the representative community. Soil texture analysis that most areas consisted of loamy soil and were distributed on the northern aspects. The plantation, represented by Pk, exhibited a high content of exchangeable aluminum (5.227±0.342 mg/kg), suggesting the need for soil improvement and monitoring in these habitats. The canopy openness (forest gap) ranged from 11% to 21%, indicating a relatively closed canopy in many survey plots. Non-metric multidimensional scaling analysis indicated heterogeneous species composition between the QmPd cluster and the Pk cluster (total R2 = 0.608).
